      • 냉적응 생쥐의대식세포 활성화에 미치는 PKC 억제제와 Ca^(2+) Antagonist의 영향

        Human thymus 조직으로부터 cDNA library를 만들었다. 무작위로 선별한 cDNA clone중에서 468개 염기로 된 cDNA clone (PTH209)을 검색한 결과 124개의 아미노산을 code하는 EAP(Epstein-Barr asseciated protein) 유전자임을 확인하였다. Thymus의 EAP와 Epstein-Barr 바이러스 positive 및 negative cell line의 RNP에서 분리한 EAP^(13)를 비교한 결과, coding region은 동일하였으나 3'UT region은 달랐다. 더구나 EBERs(Epstein-Barr encoded RNAs) 와 La 자가항원이 결합된 RNP의 EAP와 마찬가지로, thymus의 EAP는 sea urchin의 developmental stage-specific 단백질로 알려진 217^(2) 과 매우 유사한 아미노산 서열을 보였다. 이런 결과로 미루어 thymus의 EAP는 thymic development에 중요한 역할을 하는 것으로 사료된다. To study the adaptation of immune systems in the animals kept in cold temperature, the recovery rate of peritoneal exudated mouse macrophage was measured in mouse bred at room temperature(RT) or at cold(S±2 C). NBT-reduction activities and phagocytic activities were measured too, to the macrophages recovered. The results are as follows; More than 40% of cells were recovered in the cold treated groups compaired to the RT bred groups. The NBT-reduction activities were totally blocked at 10mM EGTA, and were below 10% of the control in H7 100pM and TMB-8 0.1mM. The NBT-reduction activity of cold treated groups were higher than RT group at lower concentration than H7100pM and TMB-8 0.1mM . The H7 resulted no changes in phagocytic activities both cold and RT groups at 0.1-100pM. The TMB-8 resulted in significant decrease in phagocytic activities at 0.1 mM and 1mM. The conclusions are 1) the cold treated animals showed an increase in the NBT-reduction activities and in the macrophage recovery rates, 2) H7 was unable to block phagocytosis of yeasts up to 10011M concentration, 3) lectin-like yeast cell surface molecules have different signaling mechanism form IgG molecules.

      • cAMP와 polyamine 生合成抑制가 大食細胞의 腫瘍 致死活性에 미치는 영향

        This study was to investgate the effects of intracellular cAMP and polyamine biosynthesis inhibition on microbicidal activity, tumoricidal activity and tumor necrosis (actor production against L929 fibroblast cell line of LPS-activated rat per itoneal macrophages in vitro. The macropohages were treated with cAMP analogues(DbcAMP, prostaglandin E2), cAMP inhibitors(alloxan, indomethacin) and two ODC inhibitors(DFMO, MAP). cAMP analogues inhibited L929 tumor cell cytotoxity, NBT reduction and TNF production. When treated with DFMO, tumoricidal activity was significantly decreased. But, MAP had no effect on macrophage tumoricial activity, regardless of concentration. Exogeneous putrescine and interferon-γ, a macrophage activating factor, reverse the effect of DFMO.

      • 細胞老衰에 미치는 人蔘의 影響

        To study the effects of ginseng on the cell senescence, the experimental groups of young (about 80 days old) and older (about 210 days old) mice were injected subcutaneously with 0.3mg of alcohol extract of ginseng, which was diluted with Tyrode solution, per 20g of body weight daily. After 2 groups of mice were treated for 7 days, the alkaline phosphatase activity in the jejunum, kidney and liver homogenate and serum were determined with sodium β-glycerophosphoshate as substrate. Also the effects of water extract of ginseng on the division of Saccharomyces cervisiae and on the inhibition of the division at low temperature were studied. 1. Visceral and serum alkaline phosphatase activity of older mice were decreased compared with that of young mice. 2. The enzyme activity of young or older mice was increased by ginseng treatment. 3. After administration of ginseng extract diluted with Tyrode solution, the decreased enzyme activity in the older mice was recovered considerably, and increased to the activity of young blank normal mice. 4. Different dose of water extract of ginseng to the yeast medium promoted considerably the division of yeast at 18℃ or 25℃, however it could be confirmed the optimal dose of ginseng extract on the division. 5. The optimal dose of ginseng extract over-recovered the inhibition of the division at low temperature (18℃) than the division of control group at 25℃. In accordance with the results it could be concluded ginseng extract can regenerate the cell degeneration or delay the cell senescence.

      • TPO 활성에 대한 체중 변화의 영향과 X-선에 의한 이원적 영향

        The effect based upon changes of body weight and effect of X-ray irradiation on tryptophan pyrrolase (TPO) activity in liver homogenate of albino rats was studied. 1. The average TPO activity of control group of 33 rats weighing $95{\sim}172g$\;was\;2.372{\pm}0.165{\mu}$ moles kynurenine/g protein/hr. 2. Correlation between body weight of 33 rats and its TPO activity was not showed practically(r=0.011). 3. TPO activity of whole-body X-ray irradiated rats (700r) was increased about double at 3hours and increased for a period of 4 days after irradiation, but after 6 days it was decreased gradually. This result is that whole-body X-ray irradiation showed dual effect on TPO activity in rat liver.

      • 人蔘 成分이 Pepsinogen의 分泌에 미치는 影響

        Abstract The effects of ginseng components on pepsinogen secretion in isolated rabbit gastric glands were studied. In the release groups the pepsinogen secretion was increased at 10-1% and 100% concentration of total saponin, and 10-1% concentration of diol- and trios-saponin significantly by 240%, but did not show any significant change of pepsinogen secretion at the 10-2%, 103% of total saponin, diol- and trios-saponin. In the synthesis groups the pepsinogen secretion was inscreased by 20% at 10-1% and 100% concentration of three ginseng components and showed same level of control at lower concentration but these all values were not significant.

        Petroleum ether fraction and ethyl acetate fraction of ginseng has known as inhibitory components to cancer cell. In other to study the effects of these components on the normal cell, the proliferation rate of yeast cell treated by these components were measured and some results were obtained. On the glucose afar or glucose broth medium added by 5, 10, 20, or 40㎍ of petroleum ether fraction, all proliferation rates of experimental yeast cell groups were not observed by significant change statistically. On the glucose agar medium added by 5 and 10㎍ of ethyl acetate fraction the proliferation rates of feast cell were slightly increased, but on the afar medium added by 20 or 40㎍ arid on the broth medium added by 5, 10, 20 of 40㎍ of ethyl acetate fraction the proliferation rates were not changed significantly.

        The effects of ginseng on the penetration of glucose into Saccharomyces cerevisiae were studied by determining the amount of glucose in the supernatant of the glucose broth medium in which some dose of water extracts of ginseng or saponin were added and the yeast was cultured for 1 hour. 1. In the 1 hour culture with 0.04%, 0.08%, 0.16%, 0.32% and 0.64% dry ginseng glucose broth medium, the penetration rate of glucose into yeast was increased 4.76%, 7.96%, 10.09%, 9.08% and 7.48% respectively. 2. In the 1 hour culture with $10^{-6}%,\;10^{-5}%,\;10^{-4}%,\;10^{-3}%,\;and\; 10^{-2}%$ of saponin glucose broth medium, the penetration rate of glucose into yeast was increased 3.05%, 6.57%, 5.595, 4.44%. and 2.87% respectively. 3. Since the highest increasing percentage of penetration of ginseng and saponin were 10.19% and 5.89% respectively, the increasing rate of the ginseng was about twice that of the saponin. Therefore it could be concluded that the saponin contained in ginseng and other unkown components of ginseng affect the cell permeability.
