        Retinoic Acid가 Bromodeoxyuridine 표지 PLC / PRF / 5 간암 세포주의 역동성에 미치는 효과

        N/A Retinoie acid (RA) has been reported to show antiproliferative and differentiation-inducing effects on human tumor cells. In this report, we examined the antiproliferative effect and the influence on the cell cycle in PLC/PRF/5 hepatocelluar carcinoma cells, secreting hepatitis B surface antigen (HBs Ag). MTT assay was conducted to prove the antiproliferative effect of RA, and the univariate and the bivariate distribution of BrdU/DNA were analyzed using FACScan, to evaluate the influence of RA on tumor cell kinetics in the labelled cells with the 5-bromodeoxyuridine (BrdU). Following 6 days of exposure, 1uM and 0 1 uM RA containing nontoxic DMSO level showed antiproliferative effect on the cells. The analysis from the tumor cell kinetics showed that actual doubling time (Td) is 69 hrs, potential doubling time (Tpot) is 42 hrs, the mean DNA synthesis time (T5) is 17.4 hrs, and the labelling index is 35.2%. The changes of the tumor cell kinetics in the cells treated with RA were investigated. Early stage of culture (day 2, 4) showed increase in S phase percentage of cells and decrease in G0/G1 phase. But ]ate stage of culture (day 6) show decrease in S phase percentage of cells to minimum and increase in G0/G1 phase at either 1 uM or 0.1 uM concentration of RA as compared with that of control. The influence on cell cyclc was rnore pronounced at 1 uM than at 0.luM concentration. These results demonstrated that RA inhibits the growth of PLC/PRF/5 cells and its antiproliferative effect is suggested to be driven from arresting cell cycle progression from G0/G1 to S phase in tumor cell kinetics.

      • 96년 거제에서 패류 중독 사고를 유발한 진주담치의 마비성 독소

        Food poisoning accident occurred on May, 1996 at Oepo, Geoje County, Kyeongnam Province, Korea, and two persons wee died within a few hours after ingestion of the soup prepared with wild mussel, Mytilus edulis, harvested on the sea rock. Paralytic shellfish poisons(PSP) were elucidated as the responsible toxins for the food poisong accident because the wild mussels caught after three days at the near place from the accident contained high toxicity of PSP ranged 650-1000 MU/g of edible meal by mouse bioassay. Gonyautoxin-1+4(42.7%) and C1 +C2(40.0%) were detected as the major toxins in the mussels by fluorometric HPLC method. Although, the poison extracted out with drip during freezing and thawing, and the toxicity gradually decreased by boiling for 20 minutes, over 30 MU/g of toxins remained in the soup and meat, which indicated that they could be able to make food poisoning.

        Retinoic Acid 가 Bromodeoyuridine 표지 Hep G2 간암 세포주의 세포 주기 역동성에 미치는 효과

        Objectives: Measuring the total cellular DNA and BrdU content simultaneously by using the monoclonal antibody to BrdU and propidium iodide (PI) has been possible. So it has been advanced that the making a correct diagnosis and the predicting a prognosis in cancer as well as the evaluation of the responses of anti-cancer therapy, We intended to exam new anticancer chemotherapeutic agent, retinoic acid (RA) which has been reported to show antiproliferative effect and also differentiation-inducing effect on tumor cells, for the treatment of the hepatocarcinoma prevailed in our country. This study was designed to probe beneficial effects of RA for preventing tumor recurrence and secondary metastasis through the application of it to recently wide spreaded chemoembolization therapy in heaptomas. Methods: A semi-logarithmic plot of the proliferation was made in Hep G2 cells, and the changes of the proliferative activities from adding RA to the medium were evaluated in according to various concentrations of it. After staining the Hep G2 cells with fluoroscence conjugated anti-BrdU and PI, the tumor cell kinetics was analyzed and the effects of RA on the changes of the kinetics were also evaluated in the univariate and bivariate distribution of flowcytometric measurement of DNA content/BrdU incorporation. Results: Hep G2 cells stained with FITC conjugated anti-BrdU and with propidium iodide (PI), were anal- yzed in the distribution of bivariate BrdU/DNA content or univariate DNA content to investigate the tumor kinetics. Data from this expreiments showed that actual doubling time (Td) is 51 hrs, potential doubling tiem (Tpot) is 29 hrs, the mean DNA synthesis time (Ts) is 9. 3 hrs, the labelling index is 27.6%. From the evaluation of the effects of RA, known to enhance the differentiation and inhibit the growth of tumor cells on Hep G2 cells, RA inhibited the proliferation of Hep G2 cells significantly and the inhibition lasted for 10 days. To understand the growth inhibition of RA on Hep G2 cells in terms of tumor cell kinetics, bivariate BrdU/DNA content distributions were analyzed. Early stage of culture (day 1) showed increase in S phase percentage of cells and decrease in Go/G1 phase. But late stage of culture (day 4) show decrease in S phase percentage of cell to minimum and increase in Go/G1 phase at either 1 μM or 0.1 μM concentration of RA as compared with those of control. The influences on cell cycle and the antiproliferative effect were more pronounced at 1 μM than at 0.1 μM concentration. Conclusion: These results demonstrated that RA inhibits the proliferation of Hep G2 cells and the anti-proliferative effect is suggested to be driven from arresting cell cycle progression from Go/G1 to S-phase in tumor cell kinetics.

        마비성패류독의 생물학적 제독 : 1. 마비성패류득 분해세균의 분리 및 세균학적 특성

        PSP 분해 활성 세균을 이용한 마비성패류독의 생물학적 제독 방법을 확립하기 위하여 진주담치 및 해수에서 마비성패류독 분해능이 있는 균주를 분리하였으며, 분리 균주의 독소 분해 활성 및 세균학적 특성 시험을 실시한 결과는 다음과 같다. 남해안 패류양식장에서 채취한 진주담치 및 해수 중에서 마비성패류독 분해능이 있는 8균주를 분리하였으며, 분리균주 중 GTX 1∼4 혼합물 전반에 대하여 폭넓은 분해 활성을 나타낸 CW-6를 시험 균주로 선발하였다. 선발 균주 CW-6는 배양 3일만에 18nmole/g의 GTX2를 완전히 분해하는 강한 독소 분해 활성을 나타내었다. 선발 균주 CW-6는 생화학적 특성 시험 결과, Enterobacter속으로 동정되어 Enterobacter sp. CW-6로 명명하였으며, 이 균주는 20∼40℃, pH 7∼9에서 왕성하게 증식하는 전형적인 중온세균의 특징과 염분농도 6%에서도 최적 증식의 46.8% 정도 증식 가능한 내염성을 나타내었다. For the establishment of biodetoxification method which can be acceptable for live bivalves, paralytic shellfish poison (PSP) detoxification bacteria were isolated from sea water and bivalves, and PSP detoxification activity and optimal growth condition of the isolated strains were investigated. From the bivalve and sea water samples, 8 strains of PSP detoxification bacteria were isolated. Of the isolated strains, CW-6 isolated from sea water shown strong PSP detoxification activity and decomposed completely 18 nmole/g of GTX2 after 3 days incubation in artificial medium. The selected stain CW-6 shown typical characteristics of the Enterobacter sp. and identified as Enterobacter sp. CW-6. Optimal growth condition of the Enterobacter sp. CW-6 were 35℃, pH 7 and NaCl 1%, respectively.
