      • 慶南 漆西面 自然늪의 水質과 周邊 土壤의 微生物學的 및 理化學的 性質에 對하여

        For obtaining the information of environmental conditions of a natural swamp at Chilseo-Myeon Kyungnam, where was peoposed as a site for garbage dump from Pusan and Kyungnam area, physicochemical and microbiological properties of the and soil of that site were investigated. The results are as follows : 1. Water quality of the site 2 where had been dumped with garbage was somewhat polluted such that BOD, COD and SS value were 14.6, 41.5 and 57.0 mg/1 respectively. 2. The microbial population density of the site 2 was relatively high, it showed that much orgnic waste was elucidated from garbage. Especially, the density of coloform that much organic waste was elucidated from garbage. Especially, the density of coliform bacteria was higher than other water environments. 3. The microbial population density of the soil of Chilseo-Myon area was similar to that other soil environments. From the above results, when the natural swamp is dumped with garbage, it will influence on the water environments of the Nakdong River.

        효모의 접합과정에 관여하는 유전자의 연구

        In order to elucidate and characterize the signal transduction pathway(s) whereby yeast cells respond to mating pheromone, we have isolated mutants which are able to conjugate in the absence of the alpha-factor receptor. Sixty-one suppressors of a ste2-deletion mutation which also confer a ts conditional "start" arrest phenotypw have been subjected to genetic analysis. The mutants could be assigned to three complementation groups designated CDC70, CDC72 and CDC73, which are unlinked to each other as well as to the previously identified start genes. Quantitation of mating ability of the cdc70, cdc72 and cdc73 mutations in a ste2-deletion background gives levels ranging from 0.1% to 0.3% of wild type, depending on the allele and the gene. The results indicate that the signals from mating pheromone might be mediated by the CDC70, CDC72 and CDC73 products. products.

      • 선택표지를 사용하지 않는 용이한 유전자 cloning방법

        제한효소로 자른 DNA 조각을 vector의 선택표지 내가 아닌 다른 자리에 삽입하고자 할 때, 삽입된 vector를 쉽게 얻을 수 있는 방법을 기술하였다. 기술된 방법으로 효모의 세포주기 조절유전자 CDC73을 함유하고 있는 HindⅢ 조각을 vector YCp50와 pHSS8의 HindⅢ자리에 삽입시켜 만족할 만한 결과를 얻었다. The cloning procedure in which DNA fragment inserted into the outer region of seletive marker in vectors was explained in this article. A DNA fragment containing the gene CDC73 which had been cut by the restriction endonuclease HindⅢ was cloned into the vectors YCp50 and pHSS8 by the explained procedure.

        한강에서 분리한 pwychrophilic yeasts에 관하여

        Psychrophilic yeast strains, HJ3011, HJ3023, and HJ3021, which were isolated from the Han River during winter season of December 1980 to February 1981 were identified as Candida sp., Candida nivalis, and Rhodotorula rubra respectively. The effect of temperature on growth was carried out in this study. Growth rate constants of strains HJ3011 and HJ3023 were the highest value at $15^{\circ}C\;and\;16{\sim}20^{\circ}C$ respectively, and both growth rates diminished gradually above $20^{\circ}C$ and were decreased to zero at $25^{\circ}C$. According to the results obtained in this experiment, above two strains of yeast were confirmed as psychrophile. On the other hand since the strain of HJ3021 exhibited the highest growth rate constant at $15^{\circ}C$ and could slightly grow above $25^{\circ}C$, this strain was defined as psychrotroph.

        효모의 mating과정의 기작을 연구하기 위하여, α pheromone의 수용체를 coding하는 유전자인 STE2가 결여된 균주로 하여금 mating을 할 수 있게 하는 61개의 돌연변이를 획득하였다. 이들은 상보성시험과 연관시험 결과 3개 군의 유전자에 속했는데, 3개 군을 CDC70, CDC72 및 CDC73 이라고 명명하였다. 얻어진 돌연변이는 제한 온도에서는 세포분열이 start 단계에서 정지되는 온도 감수성이며 STE2 유전자 결여를 억제하는 능력을 보여 주었는데, 이는 유전자 CDC70, CDC72, CDC73 의 생성물이 수용체로부터 전달되는 signal의 중간 전달 물질이라는 사실을 가리킨다.       • 酵母의 形性質細胞 形性과 2μm-DNA, plasmid의 分離에 關하여

        The roles and functions of 2㎛ DNA plasmid which is an important element for the research on the molecular genetics of yeasts are still unknown. When the protoplast or spheroplast is produced from yeast, its intracelluar DNA can be easily isolated. The conditions for the production of protoplast from yeast were determined in this study. Aming the factors that affect of the production of protoplast are kinds and concentrations of osmotic stabilizers, growth phase of cells, and enzymes for lysis of cell wall. Six hundred mM of KCl or 1 M of Sorbitol was most efficient for pottoplast formation as osmotic stabilizer, The cells of mid-late exponential phase were well formed to protplasts, DNA plasmid was determined with agarose gel electrophoreis.

      • 活性汚泥에 存在하는 微生物 個體群의 效果的인 測定 方法에 關하여

        Bacterial population densities in activated sludge were enumerated by using of Vortexmixer, Omnimixer, and Homogenizer and assessment was made of the relative effectiveness of bacterial clumps in activated sludge mixed liquor. The results were as follows: 1. Maximum count of bacterial population density in activated sludge was shown when Omni-mixer was operated at dial scale 3 for 30 minutes. 2. When using Homogenizer, maximum count was shown at dial scale 60 for 20 minutes. 3. Both Omini-mixer and Homogenizer were shown effectiveness of disperision when activated sludge was dispersed at lower dial scale for longer time, and higher dial scale for shorter time. 4. Vortex-mixer showed similar effect as Omni-mixer and Homogenizer when it was operated at its maximum dial scale. 活性汚泥內에 存在하는 微生物의 個體群을 效果的으로 測定하기 위해서 Vortex-mixer를 Control로 하고 Omni-mixer, Homogenizer의 相對효율을 비교 하였다. 그 결과는 다음과 같았다. 1. Onmi-mixer를 利用하여 活性汚泥內에 存在하는 細菌群을 測定한 結果 dial scale 3에서 30分間을 깼을 때 가장 효율 이 좋았다. 2. Homogenizer를 利用하였을 경우는 dial scale 60에서 20分間을 깼을 때 효율이 가장 높았다. 3. Omni-mixer, Homogenizer 모두 낮은 dial scale에서는 生活汚泥를 깨는 시간이 짧아야 효율이 좋았다. 4. Vortex-mixer의 경우 dial scale를 최대로 하여 생활오니를 깼을 때 Omni-mixer와 Homogenizer와 같은 효율을 나타냈 다.

        The focus of this paper is on the design and development of a generation of thermal energy harvesting systems that could make electrical energy from the low temperature wasted heat environment. Conventional thermoelectric devices to harvest low-temperature heat are limited due to low ZT for small temperature difference. In contrast, the proposed approach of pairing magneto-thermoelectric generator technology to triboelectric effect will provide high potential for efficient power harvesting from low-temperature waste heat generated in such as human skin. Experiments on fabricated prototype devices are conducted to measure the fundamental power properties with temperature differences as small as 5 degree Celsius. The results successfully demonstrate feasibility of efficient low-temperature energy harvesting.
