The interaction of antioxidant ligands, propyl gallate (PG) and methyl gallate (MG) with bovine serum albumin (BSA) was studied by means of fluorescence spectroscopy. Intrinsic fluorescence of BSA was monitored as a function of ligand concentrations, and fluorescence quenching assays were used to characterize the interaction between BSA and the ligands and to determine various quenching and binding constants. The quenching constants obtained for both PG and MG suggest a static quenching mechanism, and complex formation between BSA and PG or MG was confirmed by UV/Vis absorption spectroscopy. The site marker displacement experiments were performed to identify the binding site of BSA using warfarin and ibuprofen as site markers. The temperature dependence of the BSA-ligand binding interaction was investigated, and ?H and ?S for the binding process were determined using the van't Hoff equation. The results indicate that the binding interaction between BSA and PG are distinctively different in nature from that between BSA and MG.
Antihyperlipidemic effects of Korean ginseng in high-fat diet-fed ovariectomized rats

Because of decreased estrogen levels, postmenopausal women are at a high risk ofdeveloping dyslipidemia, obesity, and other metabolic diseases. The effects of Korean ginseng wereinvestigated to determine whether it can be used as an adjunct supplement for hyperlipidemia. Ovariectomized Sprague-Dawley rats were used to mimic postmenopausal conditions and wererandomly divided into five groups (n=6): normal control, high-fat diet, high-fat diet+statin, high-fatdiet+white ginseng, and high-fat diet + heated ginseng. In vitro results revealed that heated ginsenghad higher ginsenosides. Meanwhile, in vivo results showed that heated ginseng inhibited excessivelipogenesis and promoted lipolysis. It also had a better lipid profile than white ginseng. Finally, plasmaadipokine levels indicated a lower risk of developing obesity-induced hyperlipidemia in rats fed heatedginseng. Overall, these results suggest that heated ginseng ameliorates hyperlipidemia and that it canbe a suitable alternative to white ginseng.

BACKGROUND/OBJECTIVES Oxidative stress is a major cause of cancer. This study investigated the effects of the ethanol extracts from germinated and non-germinated Keunnunjami rice, a blackish-purple pigmented cultivar with giant embryo, on selected human cancer cell lines and on the antioxidant defense system of mice fed with a high-fat diet. MATERIALS/METHODS: High fat-fed mice were orally administered with either distilled water (HF) or extracts (0.25%, w/w) from brown (B), germinated brown (GB), Keunnunjami (K), and germinated Keunnunjami (GK) rice. RESULTS: In comparison with the brown rice extract, Keunnunjami extract showed higher anticancer effect against cervical and gastric cell lines but lower anticancer activity on liver and colon cancer cells. Mice from the HF group showed significantly higher lipid peroxidation and lower antioxidant enzyme activities than the control group. However, the oxidative stress induced by high-fat diet markedly decreased in B, GB, K, and GK groups as compared with the HF group. CONCLUSIONS: Germination may be an effective method for improving the anticancer and antioxidative properties of Keunnunjami rice and extracts from germinated Keunnunjami rice may serve as a therapeutic agent against cervical and gastric cancers and oxidative damage.

A cytopathology test for cervical cancer diagnosis has been introduced to complement the histopathology test. This study developed a new cervical cancer diagnostic method that complements the existing liquid cytology to reduce the diagnostician’s skill dependence and to improve diagnostic accuracy. With the ease of introduction of surface functional groups and their high biocompatibility and stability, gold nanomaterials have been used to diagnose numerous biomolecules. CA125 and p16 markers were chosen as protein markers uniquely present in cervical cancer. Bioconjugation technology was used to form a stable bond between the target antibody and gold nanorods. Antibody-gold nanorods were administered to cervical cancer cells as a factor for optical microscopic analysis. The degree of cancer cell labeling was determined through additional fluorescence microscopy analysis. Additionally, a liquid-based cytology test mimicking the clinical environment was prepared, and the false-positive rate, false-negative rate, and accuracy were determined when the study strategy was implemented. The strategy validated in this study (antibody-gold nanorod-based cervical cancer diagnosis) suggests a method that can reduce the dependence on experts for the cancer cytology and can increase test accuracy.
