N/A Purpose: Cross-modality coregistration of positron emission tomography (PET) and magnetic resonance imaging (MR) could enhance the clinical information. In this study we propose a refined technique to improve the robustness of registration, and to implement more realistic visualization of the coregistered images. Materials and Methods: Using the sinogram of PET emission scan, we extracted the robust head boundary and used boundary-enhanced PET to coregister PET with MR. The pixels having 10% of maximum pixel value were considered as the boundary of sinogram. Boundary pixel values were exchanged with maximum value of sinogram. One hundred eighty boundary points were extracted at intervals of about 2 degree using simple threshold method from each slice of MR images. Best affined transformation between the two point sets was performed using least square fitting which should minimize the sum of Euclidean distance between the point sets. We reduced calculation time using pre-defined distance map. Finally we developed an automatic coregistration program using this boundary detection and surface matching technique. We designed a new weighted normalization technique to display the coregistered PET and MR images simultaneously. Results: Using our newly developed method, robust extraction of head boundary was possible and spatial regishation was successfully performed. Mean displacement error was less than 2.0mm. In visualization of coregistered images using weighted normalization method, structures shown in MR image could be realistically represented. Conclusion: Our refined technique could practically enhance the performance of automated three dimensional coregistration.

N/A Purpose: This study was performed to search the relatively specific brain regions related to the semantic processing of Korean and English words on the one hand and the regions common to both on the other. Materials and Methods: Regional cerebral blood flow associated with different semantic tasks was examined using [150]H2O positron emission tomography in 13 healthy volunteers. The tasks consisted of semantic tasks for Korean words, semantic tasks for English words and control tasks using simple pictures. The regions specific and common to each language were identified by the relevant subtraction analysis using statistical parametric mapping. Results: Common to the semantic processing of both words, the activation site was observed in the fusiform gyrus, particularly the left side. In addition, activation of the left inferior temporal gyrus was found only in the semantic processing of English words. The regions specific to Korean words were observed in multiple areas, including the right primary auditory cortex; whereas the regions specific to English words were limited to the right posterior visual area. Conclusion- Internal phonological process is engaged in performing the visual semantic task for Korean words of the high proficiency, whereas visual scanning plays an important role in performing the task for English words of the low proficiency. 11C 표지 자동합성장치에서 루프법을 이용한 [11C](R)-PK11195의 간편한 합성법

목적: (R)-1-(2-Chlorophenyl)-N-methyl-N-1-(1-methyl-propyl)-3-isoquinoline carboxamide ((R)-PK11195)는 말초형 벤조디아제핀 수용체 (PBR)의 친화성이 높으며, 활성화된 소교세포(microglia)의 PBR에 선택적으로 결합하는 것으로 알려져 있다. 본 연구에서는 말초형 벤조디아제핀 수용체 (PBR)의 PET용 리간드인 [11C](R)-PK11195를, 절차가 빠르고 간단한 [11C]CH3I자동합성장치 및 루프법을 도입해서 합성하였다. 대상 및 방법: 사이클로트론에서 14N(p,α)11C핵반응에 의하여 생산된 [11C]CO2를 0.2 M LiAlH4/THF (0.2 mL)로 환원한 다음 HI (1 mL)과 반응하여 [11C]CH3I를 생산하였다. 반응용매인 NaH DMF complex (0.1 mL)와 녹인 전구물질 (R)-N-desmethyl-PK11195 (1 mg)을 녹인 DMSO (0.1mL)를 섞은 혼합액을 HPLC의 루프에 미리 주입하고 [11C]CH3I를 상온에서 5분 동안 질소가스로 불어준 뒤에 semi-preparative HPLC로 [11C](R)-PK11195를 분리하였다. 결과: 전구물질과 [11C]CH3I의 [11C]메틸화 반응에서의 표지효율은 71.8±8.5% 이었다. 분리 후 얻은 [11C](R)-PK11195의 비방사능은 11.8±6.4 GBq/μmol 이었으며, 방사화학적 순도는 99.2% 이상이었다. C-11 표지 후 얻어진 (R)-PK11195의 물질의 질량 분석은 m/z 353.1 (M+1)으로 물질구조를 확인 할 수 이었다. 결론: 뇌염증에 의한 활성화 소교세포의 영상화를 위한 PET용 방사성의약품인 [11C](R)-PK11195를 반복적으로 생산해야 하는 임상 적용을 위해, 합성절차가 빠르고 간단한 [11C]CH3I 자동화 합성장치를 1차 루프 방법을 이용하여 11C을 표지 할 수 있었다. 이 연구를 통해서 [11C](R)-PK11195 표지 할 때 반응 단계를 줄이고 반응을 실온에서 할 수 있어서 표지과정을 보다 단순화할 수 있었으며, NaH의 DMF 현탁액을 사용함으로써 보다 안전하게 생산할 수 있게 되었다. Purpose: ((R)-1-(2-chlorophenyl)-N-1-[11C]methyl-N-(1-propyl)-3-isoquinoline carboxamide ((R)-PK11195) is a specific ligand for the peripheral type benzodiazepine receptor and a marker of activated microglia, used to measure inflammation in neurologic disorders. We report here that a direct and simple radiosynthesis of [11C](R)-PK11195 in mild condition using NaH suspension in DMF and one-step loop method. Materials and Methods: (R)-N-Desmethyl-PK11195 (1 mg) in DMSO (0.1 mL) and NaH suspension in DMF (0.1 mL) were injected into a semi-prep HPLC loop. [11C]methyl iodide was passed through HPLC loop at room temperature. Purification was performed using semi-preparative HPLC. Aliquots eluted at 11.3 min were collected and analyzed by analytical HPLC and mass spectrometer. Results: The labeling efficiency of [11C](R)-PK11195 was 71.8±8.5%. The specific activity was 11.8±6.4 GBq/μmol and radiochemical purity was higher than 99.2%. The mass spectrum of the product eluted at 11.3 min showed m/z peaks at 353.1 (M+1), indicating the mass and structure of (R)-PK11195. Conclusion: By the one-step loop method with the [11C]CH3I automated synthesis module, [11C](R)-PK11195 could be easily prepared in high radiochemical yield using NaH suspension in DMF. Purpose: To localize and compare the neural basis of verbal and visual human working memory, we erforrned functional activation study using H2O PET. Materials and MethoDs: Repeated H213O PET scans with one control and three different activation tasks were performed on six right-handed narmal volunteers. Each activation task was composed of 13 matching trials. On each trial, four targets, a fixation dot and a probe were presented sequentially and subjects task was to press a response button to indicate whether or nat the probe was one of the previous targets. Short meaningful Korean words, simple drawings and monochromic pictures of human faces were used as matching objects for verbal or visual memory. All the images were spatially normalized and the differences between control and activation states were statistically analyzed using SPM96. Results: Statistical analysis of verbal memory activation with short words showed activation in the left Broca's area, premotor cortex, cerebellum and right cingulate gyrus. In verbal memory with simple drawings, activation was shown in the larger regions including where activated with short words and left superior temporal cortex, basal ganglia, thalamus, prefrontal cortex, anterior portion of right superior temporal gyrus and right infero-lateral frontal cortex. On the other hand, the visual memory task activated predominantly right-sided structures, especially inferior frontal cortex, supplementary motor cortex and superior parietal cortex. Conclusion: The results are :consistent with the hypothesis of the laterality and dissociation of the verbal and visual workmg memory from the invasive electrophysiological studies and emphasize the pivotal role of frontal cortex and cingulate gyrus in working memory system.
