가미소암산(加味消癌散)의 혈관신생(血管新生) 억제에 관한 항암효과 연구

Soamsan is known as an anti-cancer remedy in the traditional Korean Medicine. To enhance the synergic effects of anti-cancer activity of Soamsan, this study reconstituted the original components of Soamsan with a slight modification and produced a novel herbal remedy, namely Gamisoamsan. Extracts of Gamisoamsan inhibited the growth of cultured CT-26 cells, mouse colon adenocarcinoma, in a dose-dependent manner $(1\;to\;50{\mu}g/ml)$, and $ID_{50}$ was estimated approximately $16.7{\mu}g/ml$. Using tumor-bearing mouse model, in which was produced by subcutaneous injection of CT-26 cells ($1{\times}10^5$cells). the effects of Gamisoamsan on tumor growth and host survival were examined by evaluating tumor volume and increase in life span. When Gamisoamsan extracts in variable doses of 100, 200 and 500mg/kg body weight per day were orally administered to tumor-bearing mice, following results were obtained: Improvement in the hematological parameters following Gamisoamsan treatment such as hemoglobin contents, red blood cells and white blood cells of the tumor-bearing mice have been observed. Gamisoamsan treatment also showed a prolongation of life span and a reduction of tumor volume in the CT-26 tumor hosts. The results of the present study suggest that Gamisoamsan extracts has a potential anti-tumor activity and may be an useful remedy to prevent and/or treat cancer.

The inhibitory effect of extract of Atractylodis Rhizoma alba on melanin biogenesis was studied by using B16/F10 melanoma in culture. Atractylodis Rhizoma alba significantly inhibited tryrsinase activity, and melanin contents with or without alpha-MSH and forskolin in vitro. Melanin contents and tryosinase aotivity have decreased in a dose-dependent manner, These results show that extract of Atractyledis Rhifoma alba could be developed as skin whitening components of cosmetics.

Gamisoamsan is a prescription originated in Soamsan which is known as an anti-cancer remedy in the traditional Korean Medicine. To enhance the synergic effects of anti-cancer activity of Soamsan, this study reconstituted the original components of Soamsan with a slight modification and produced a novel herbal remedy, namely Gamisoamsan. To investigate the effects of Gamisoamsan on anti-cancer reaction, I studied the effects of Gamisoamsan on angiogenesis via chorioallantoic membrane (CAM) assay, corneal neovascularization assay and the effects on expression of growth factor which are VEGF, TGF-β, bFGF and IMUP-1. Anti-cancer effects of Garnisoamsan was also abserved through hematological parameters, tumor volume and survival rate in mice. Gamisoamsan inhibited embryonic angiogenesis of blood vessels in CAM assay and inhibited neovascularization of rat cornea. Gamisoamsan reduced cell proliferation in HT1080 cells and IC50 was 2.18 mg/ml. Gamisoamsan reduced the expression of VEGF, TGF-β, bFGF and IMUP-1 which was known as vascular growth factor and this effects of Gamisoamsan was predominant than VP-16. The treatment of Gamisoamsan decreased the CT-26 cell inoculated-tumor volume in mice colon adenocarcinoma and increased mice survival which was inoculated CT-26 cells. The results of the present study suggest that Gamisoamsan extracts has a potential anti-tumor activity and may be an useful remedy to prevent and/or treat cancer.

Modified Hwanglyunhaedoktang has been used against allergic disease for generations, and still occupies an important place in Oriental medicine. In this study, the author investigated the effect of Modified Hwanglyunhaedoktang on immunoglobulin E-mediated anaphylaxis activated by anti-dinitrophenyl immunoglobulin E antibody. Modified Hwanglyunhaedoktang potently inhibited passive cutaneous anaphylaxis when intradermally, intraperitoneally, and orallly administered. On the other hand, it showed poor inhibitory activity when intravenously administered. Moreover, Modified Hwanglyunhaedoktang inhibited anaphylactic histamine release from peritoneal mast cells activated by anti-dinitrophenyl immunoglobulin E antibody. These results indicate that Modified Hwanglyunhaedoktang inhibits the passive cutaneous anaphylaxis by inhibition of histamine release from mast cells in vivo and in vitro and also suggest that the differential activity following prescription routes may be caused by the difference of bioavailability.

Caesalpinia sappan L. has been commonly used as analgesic, a cure for contusion and sprain as well as a remedy for thrombosis in the oriental medicine. The main constituent of Caesalpinia sappan L. is brazilin having a flavonoid structure. Melanin is a main pigment found in skin, hair and eyes, and melanin synthesis is controlled by a cascade of enzymatic reactions regulated at the level of tyrosinase. The aim of this study was to examine the effect of Caesalpinia sappan L. on proliferation and melanogenesis of B16/F10 melanoma cells. After 48h treatment of cells with various concentrations of EtOAc extract, the cells exhibited a dose-dependent inhibition in their proliferation without apotosis. Therefore, the growth retardation may be due to the cell arrest, not due to cytotoxicity. And there were estimated total melanin contents as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. EtOAc extract increased the melanin contents and tyrosinase activity in low concentration (0.01-10㎍/ml), whereas it decreased those in high concentration. In conclusion, it was observed that EtOAc extract of Caesalpinia sappan L. differently regulates melanization of cells dependent of its concentrations.
