        MMP and TIMP production in periodontal ligament fibroblasts stimulated by Prevotella nigrescens lipopolysaccharide

        이 연구에서는 Prevotella nigrescens (P. nigrescens)의 lipopolysaccharide (LPS)로 자극한 치주인대 섬유아세포에서 matrix metalloproteinase (MMP)와 tissue inhibitor of metalloproteinase (TIMP)의 생성 양상과, LPS를 수산화칼슘으로 처리했을 때의 영향을 평가하였다. P. nigrescens에서 추출, 정제한 여러 농도의 LPS와 수산화칼슘으로 처리한 LPS로 치주인대 섬유아세포를 자극하여, Immunoprecipitation법으로 MMP-1, -2, TIMP-1의 단백질 생성 양상을, real-time polymerase chain reaction법으로 MMP-1의 mRNA 발현 양상을 분석하였다. 이 연구의 결과는 아래와 같다. 1. MMP-1은 단백질과 유전자 수준 모두 자극 시간과 비례하여 증가하여 48시간에 최대값을 보였다. 2. MMP-2단백질 생성은 1, 10 mg/ml에서 자극 시간과 비례하여 증가하였다. 3. TIMP-1 단백질 생성은 24시간까지 증가하다가 48시간에 감소하였고, 0.1과 1 ${\mg}g/ml$에서 증가하였으나 10 ${\mu}g/ml$ 에서 억제되었다. 4. P. nigrescens의 LPS를 수산화칼슘으로 처리시 MMP-1의 mRNA 발현은 현저하게 감소하였다. The purpose of this study was to monitor the secretion of matrix metalloproteinase (MMP) and tissue inhibitor of metalloproteinase (TIMP) by human periodontal ligament (PDL) fibroblasts stimulated with Prevotella nigrescens lipopolysaccharide (LPS), and to examine the effect of calcium hydroxide treatment on P. nigrescens LPS. LPS was extracted and purified from anaerobically cultured P. nigrescens. PDL fibroblasts were stimulated by the LPS (0, 0.1, 1, 10 ${\mu}g/ml$) or LPS (10 ${\mu}g/ml$) pretreated with 12.5 mg/ml of $Ca(OH)_2$ for 3 days, for various periods of time (12, 24, 48 h). Immunoprecipitation were performed for protein level analysis of MMP-1 MMP-2 and TIMP-1. Total RNA was isolated and real-time quantitative polymerase chain reaction (PCR) was performed for quantification of MMP-1 mRNA. According to this study, the results were as follows: 1. The p개duction of MMP-1 by stimulation with P. nigrescens LPS increased in time-dependent manner, and showed maximum value at 48 h in both protein and mRNA level. But there was no dose-dependent increas. 2. MMP-2 production time-dependently increased when stimulated with 1 and 10 ${\mu}g/ml$LPS, but there was no dose-dependent increase. 3. TIMP-1 p개duction increased to 24 h, but decreased at 48 h. It increased when stimulated with 0.1 and 1${\mu}g/ml$, but suppressed at 10 ${\mu}g/ml$ .4. P. nigrescens LPS pretreated with $Ca(OH)_2$ markedly downregulated MMP-1 gene expression.

        『트리스트람 섄디』에 나타난 실천적 종교관

        According to the recent studies that aim to read Laurence Sterne`s works in the context of his Anglicanism, Tristram Shandy contains a lot of examples of Christianity that reflect Sterne`s preference for practical divinity over polemic divinity. Even its sceptical narrative of experimental techniques is considered to result from the fideistic scepticism which acknowledges the limits of reason and accepts interpretative suspense with Christian humility. The central figure of Christianity in Tristram Shandy is Yorick who, like Sterne, favors the practical divinity and tries to do his duty as the parson of the parish. Yorick arranges for a poor widow to obtain a licence to practice as a midwife. In fact, the contemporary midwife, being licensed by church, was required to be faithful to God since she played an important social role as guaranteeing the legitimacy of the child, who became a new member of a family and a parish. Being trusted by his parishioners, Yorick supports their simple but religious ideas, too. When Toby asserts his idea about love which is based on Anglican Church`s Common-Prayer Book, Yorick approves the plain sense in it while he stands against Walter`s pedantic dogmatism. He supports Mrs. Shandy`s Christian belief in the importance of the procreation of children in marriage. He also praises Trim who puts fifth commandment into practice, providing regularly his parents with money. Filled with simple, practical, Christian truths expounded by these characters, Tristram Shandy itself is an essence of Sterne`s practical divinity, indeed.

        또래수용성 및 또래괴롭힘과 청소년의 자아존중감간의 관계

        Relationships between peer acceptance, peer victimization, and self-esteem in adolescents were examined in a sample of 392 eighth grade students(189 boys and 203 girls). Data were collected using questionnaires regarding peer acceptance(social preference and social status), peer victimization, and self-esteem of adolescents. High social preference was positively related to self-esteem. Social preference varied as a function of social status both in boys and in girls. Popular boys had higher self-esteem than neglected and rejected boys; popular girls had higher self-esteem than neglected girls, who had higher self-esteem than rejected girls. The more boys and girls were victimized by peers, the lower their self-esteem. The higher their social preference, the less they were victimized. Rejected boys and girls experienced peer victimization much more than either popular or neglected boys and girls.

        4종 대황(大黃)류 추출물의 항혈전 효능 비교

        Objective: The purpose of this study is to compare anti-thrombotic activities of 4 Rhubarb species. Methods: Rhubarb has been used as medical and edible resources worldwide for the therapy of cancer, constipation and inflammation, etc. To compare the anti-thrombotic activities among the species within rhubarb, we investigated and compared the inhibitory properties of water extracts from R. palmatum(RPE), R. officinale(ROE), R. nobile(RNE), and R. franzenbachii (RFE) on the fibrinolysis, blood coagulation and platelet aggregation. APTT and PT, blood coagulation time, were measured by apparatus of auto blood coagulation analysis that called ACL-7000. The measurement of fibrinolysis was measured and compares with the width of clear zone from melted fibrin plate. The platelet aggregation was measured and compared with inhibition rate of aggregation. Results: We found out the best species from rhubarb showing the significant fibrinolytic, anti-coagulant and anti-platelet aggregation activity. Our results show that Rheum nobile (RNE) has the most effective anti-thrombotic activity among 4 tested rhubarb. Conclusions: This study, therefore, RNE will be able to be used the development of drug which is thrombosis treatment.

        생강추출물의 항위염.항궤양 작용

        The rhizoma of Zingiber officinalehas been used as antiemetic, expectorants, stomachache relieving drugs and digestive accelerators. From the observation of antigastritic action of the methanol extract of the rhizoma, it was fractionated with hexane, chloroform, ethyl acetate and butanol, followed by bioassay on antigastritic and antiulcerative activity. The hexane and the chloroform fraction reduced significantly HCl.ethanol induced gastric lesion at the dose of 370 and 210mg/kg, p.o., respectively. On the gastric ulceration and gastric secretion in pylorus-ligated rats, the hexane fraction decreased the volume of gastric secretion and acid output, and also increased pH at the dose of 370mg/kg, i.d.. It showed considerable curative ratio of acetic acid induced ulcer without inhibition of indomethacin induced gastric lesion. The methanol extract showed low acute toxicity with minimum lethal dose of more than 3000mg/kg, p.o. in mice. In conclusion, Zingiberis rhizoma exhibited antigastric and antiulcerative activity which might be attributable to inhibition of gastric secretion. It is revealed that the active component may be present in the hexane fraction.

        호장근 추출물의 항혈전 효능 및 혈소판 응집 억제작용

        항혈전 효능을 탐색하기 위하여 한약재의 호장근 추출물로 혈전 용해능 활성과 혈액 응고시간 지연효과 즉 PT(prothrombin time), APTT(activated partial thromboplastin time)와 혈소판 응집억제 활성 등에 대해 항혈전 효능을 평가하였다. 혈전용해도를 측정하는 fibrin plate가 용해되어 형성된 투명환의 넓이를 측정하는 실험을 진행한 결과 혈전용해도가 농도의존적으로 탁월한 효능을 나타내었다. 혈액응고 cascade에 미치는 영향을 알아보기 위해 혈액 응고시간 지연 및 단축 효과를 확인하고자 APTT와 PT에 미치는 영향을 조사한 결과 PT의 경우 대조군보다 단축됨을 보였다. APTT의 경우 농도의존적으로 우수한 지연효과를 보였다. 혈소판의 응집에 따라 형성되는 두 전극 사이에 형성된 전기적 저항의 변화로 나타나는 실험을 시행한 결과 호장근의 ADP로 유도한 결과에서 뛰어난 응집억제 활성을 보였다. 따라서 호장근을 향후에 혈전 질환의 치료제 개발에 효과적으로 이용될 수 있을 것으로 사료된다. We performed this study to develop antithrombotic agents from oriental medicine herb extracts. Polygonum cuspidatum has been traditionally used as an edible medical resources for the treatment of cancer, pyodermatitis, hepatitis, cystitis, and inflammation. However, the effects of Polygonum cuspidatum on thrombosis and platelet activation are not precisely understood. The antithrombotic and antiplatelet activities of Polygonum cuspidatum were investigated by assessing the effect of a 70% ethanol extract of Polygonum cuspidatum on blood coagulation, fibrinolysis, and platelet aggregation. Polygonum cuspidatum showed effective fibrinolytic activity at 10 mg/mL. Polygonum cuspidatum also inhibited adenosine diphosphate induced platelet aggregation. Furthermore, evaluation of anticoagulant activity showed that an extract of Polygonum cuspidatum prolonged coagulation time via activated partial thromboplastin time (APTT). Our results show that Polygonum cuspidatum can be a potential candidate for antiplatelet activity as well as a fibrinolytic agent.
