Streptomyces속 균주 WH 17-2에 의한 Xylanase의 생산 조건 및 그 효소학적 성질

Highly active xylanase was purified from Streptomycessp. Wh 17-2 isolated from the soil. The optimum condition for the production of xylanase was incubation at 32℃ for 4 days in medium containing 3% α -cellulose, 0.6% soy bean meal, 0.1% K2HPO4, 0.05% MgSO4 ㆍ 7H2O. After slow addition of cold acetone to the culture fluid to the concentration of 70% it was centrifuged at 3000g for 15 minutes. The pellet was harvested, and the enzymatic property was studied from the crudely purified enzyme. The optimum pH 5.0 and the optimum temperature was 50℃. The enzyme was most stable in the pH range from pH 7.0 to pH 8.0, but the enzyme activity was reduced to 50% at pH 11.0. Incubation for one hour at up to 60℃ preserve more then 80% of the enzyme activity, but incubation for 10 minutes at 80℃ reduced the enzyme activity to 20%. Addition of sodium thioglycolate, L-cystein and ascorbic acid to the reaction mixture increased the enzyme activity, but 90% of the enzyme activity was reduced by the addition of sodium dodecyl sulfate. The study of substrate specificity revealed that dividing activity of the enzyme was reduced except for xylan. As a result of the study for the morphologic, cultural characteristics and sugar fermentation, this bacteria was identified as Streptomyces plicatus or similar one.

1957 년 aperture-impedance 혈구계 산법 이 개 발 된 이래 자동혈구계산기는 검사실 내의 일반혈액검사의 업무를 혁신적으로 변화시켰다 1) 즉 단시간에 다량의 검체를 능률적으로 재현성 있는 검사를 자동혈구계산기를 이용해서 할 수 있게 되었다. 저자들은 일반혈액검사업무의 자체 정도관리 중에 서울대학교 병원에서 사용하는 혈구계산기인 Coulter 5 Plus IV (Coulter Electronics, INC, Hialeah: 이 하 Coulter) 기종의 검사값 중에서 백혈구 histogram에 이상을 보이븐 값들이 수기법으로 재검한 값과 차이가 있음을 발견하였다. 이들은 기계에서 여러 형태로 표지를 해주게 되는데 이 표지된 백혈구 histogram 에 이 상이 있는 검 사값이 International Committee for 5tandardization in Hematology (IC5H) 가 추천하는 기준검사법인 수기법과 차이가 있는지, 또 다른 원리 인 light scattering을 이 용하는 혈구계 산기 인 H -1 5ystem (T echnicon Instrument Corporation. 분자생물학적 방법인 PCR-REBA를 이용한 대중목욕탕 수질 중 수인성병원성미생물 검출

수인성 병원성 미생물에 의한 공중목욕탕의 오염은 질병발생의 원인이 된다. 본 연구에서는 공중목욕탕내에 존재하는 수인성 병원성미생물들을 확인하고자 하였다. 서울지역내의 30 곳 공중목욕탕에서 욕조수 시료를 채수하여 진행하였다. 수인성 병원성미생물의 검출은 0.45 ㎛의 여과막을 이용하여 전통적인 배양방법으로 분리 및 동정하였 다. 분자생물학적 기법을 사용하기 위해 미생물학적인 배양을 하지 않고 핵산을 추출하여 16S rRNA유전자를 표적으 로 polymerase chain reaction-reverse blot hybridization (PCR-REBA)을 실시하였다. 미생물학적 배양방법에서는 지표 세균인 Escherichia coli와 Shigella spp.가 검출되었으며, 분자생물학적 기법인 PCR-REBA을 수행한 결과 E. coli, Shigella spp., Salmonella spp., Pseudomonas spp., Mycobacterium spp. 등의 수인성 병원성미생물이 7곳에서 검출되었 다. 본 연구결과를 토대로 공중목욕탕의 욕조수내에 수인성병원성미생물에 의한 감염을 줄이기 위해 적절한 위생관 리과 E. coli를 포함한 유해미생물을 선정하여 지속적인 모니터링이 필요할 것으로 사료된다. Contamination of public bath water by waterborne pathogens can cause disease outbreaks and contribute to background rates of disease. The aim of this study is to determine the prevalence of waterborne pathogens in public baths. A total of 30 water samples were collected from 30 different public baths in seoul, Korea. Pathogens in water samples were concentrated by 0.45 ㎛ nitrocellulose membrane filter, analyzed by both cultivation and polymerase chain reaction-reverse blot hybridization (PCR-REBA) of partial 16S rRNA gene. Various microorganisms including Escherichia coli and Shigella spp. were identified by microbiological cultivation. E. coli, Shigella spp., Salmonella spp., Pseudomonas spp. and Mycobacterium spp. were identified by PCR-REBA. Our results suggest that appropriate hygiene practice and continuous monitoring is needed for reducing health risk associated with public bath houses.

The two new female cases of Angelman syndrome (AS) were described, which diagnosed on the basis of clinical features (dysmorphic facial features, severe mental retardation with absent speech, peculiar jerky movements, ataxic gait and paroxysms of inappropriate laughter) and neurophysiological findings. Failure to detect the deletion of the long arm of chromosome 15 or the absence of epileptic seizure were not considered sufficient to exclude a diagnosis of AS. Feeding problems, developmental delay and early signs of ataxia, especially tremor on handling objects and unstable posture when seated, proved effective as the clinical markers for early diagnosis of AS. Most of the authors agreed about the existence of three main EEG patterns in AS which may appear in isolation or in various combinations in the same patient. The most frequently observed pattern in children has prolonged runs of high amplitude rhythmic 2-3 Hz activity predominantly over the frontal region with superimposed interictal epileptiform discharges. High amplitude rhythmic 4–6 Hz activity, prominent in the occipital regions, with spikes, which can be facilitated by eye closure, is often seen in children under the age of 12 years. The EEG findings are characteristic of AS when seen in the appropriate clinical context and can be helpful to identify AS patients at an early age when genetic counselling may be particularly important. .
