        Effect of Sowing Dates on Agronomic Traits and Quality of Seed for Soybean [Glycine max (L.) Merr.] in Southern Area of Korea

        Owing to adverse weather conditions, there is a heightened focus on actively researching the regulation of the sowing date in field crop cultivation. Soybean, a prominent field crop with extensive acreage and production, is a photophilic and thermophilic crop characterized by short-day photoperiodism. Identifying the optimal sowing time is crucial for mitigating the effects of severe weather conditions on soybean yield. Precise control over the timing of soybean sowing is the key to minimizing yield reduction due to unfavorable weather conditions. Temperature, photoperiod, and their interplay are the most significant factors influencing soybean cultivation among various weather factors. We conducted an experiment using three Korean soybean cultivars with varied maturities (Hwangkeumol: early maturing and Daewonkong and Pungsannamulkong: late maturing) in 2013 and 2014. Our investigation covered aspects of soybean growth, development, yield components, isoflavones, and visual seed quality. Across all three varieties, isoflavone levels increased with later sowing dates, while other measured components exhibited significant variations based on the sowing date. This study also provides valuable insights for the selection of suitable cultivars that perform well in soybean cultivation at various durations of maturity

        Functional and Proteomic Analyses Reveal That ScpBXv Is Involved in Bacterial Growth, Virulence, and Biofilm Formation in Xanthomonas campestris pv. vesicatoria

        Segregation and condensation protein B (ScpB) is essential for replication and segregation in living organisms. Here, we reported the functions of ScpBXv (ScpBlike protein in Xanthomonas campestris pv. vesicatoria) using phenotypic and proteomic analyses. Growth of XcvΔscpBXv (ScpBXv knockout mutant) was reduced under both slow and fast growth conditions in rich medium, but comparable to this of the wild-type in plant-mimic conditions. Interestingly, the mutant was significantly less virulent than the wild-type in tomato, indicating that ScpBXv is involved in virulence. To investigate ScpBXv‑associated mechanisms, comparative proteomic analyses were carried out and the abundance of 187 proteins was altered. Among them, diverse transcriptional regulators involved in biofilm formation and virulence were abundant in the wild-type. We further showed that biofilm formation of XcvΔscpBXv was reduced. This study provides new insights into the functions of ScpBXv in bacterial replication and biofilm formation, which may contribute to the virulence of Xcv.

      • 인터넷 기사 분석을 통한 북한이탈주민에 대한 인식의 변화추이와 영향요인 분석

        통일은 우리민족의 숙원이다. 통일 이후 마주할 사회혼란을 줄이기 위해서 우리나라 국민이 북한주민에 대해 어떠한 인식을 가지고 있고, 이러한 인식에 영향을 미치는 요인은 무엇인지 파악하는 것이 필요할 것으로 보인다. 따라서 본 연구는 2011-2015년 기간 동안 인터넷에 업로드 된 북한이탈주민 관련 기사로부터 남한의 북한이탈주민에 대한 인식과 인식의 변화 경향을 파악하고, 이러한 인식에 영향을 미치는 요인을 분석하였다. 연구의 분석결과에 나타난 특징은 1)북한이탈주민 관련 기사에서 긍정단어보다는 부정단어의 사용이 많았으며, 2)북한이탈주민은 정치분야의 기사에서 가장 빈번하게 언급되고 있고, 3)기사에서 부정단어가 많이 나타나는 경우는 남북한 간의 정치적 문제가 발생한 시기에 집중되어 있었다는 점이다. 이 연구의 분석결과는 많은 선행연구와 마찬가지로 한국사회에서 북한이탈주민과 북한정권을 동일시하는 경향이 있고, 북한과의 정치적 관계가 한국사회의 북한이탈주민에 대한 인식에 영향을 주는 것으로 해석될 수 있다. 이는 통일 이전에 남북간의 화해와 협력관계의 설정과 유지가 통일 후 안정적인 국가의 유지에 중요한 과제가 될 수 있음을 시사하는 것으로 판단된다. The greatest problem after reunification of South and North Korea will be the integration of two societies. To minimize the future social confusion it might be important to find the factors that affect the South Koreans’ negative perception to the North. The purpose of this study is to analyze the South Koreans’ impressions to the North Korean defectors settled in South Korea and to find the influencing factors to the impressions. The frequency of positive and negative words used in the online press during 2011 to 2015 was analyzed. The online news articles are categorized to 4 groups-political, social, international and cultural issues- and analyzed separately. The main results of the study are as follows: First, in the articles on North Korean defectors, negative words were used more than positives. Second, the defectors are most frequently mentioned in the political field. Third, negative expressions were concentrated at the time of the political conflicts between the two Koreas. The results suggest that the establishment and maintenance of reconciliation and cooperation between the two Koreas during the pre-unification period can be an important key to a stable nation after unification. It was discussed that the ways to establish the systems for the peaceful coexistence of two Koreas.

        Deciphering the Role of Tyrosine Sulfation in Xanthomonas oryzae pv. oryzae Using Shotgun Proteomic Analysis

        A bacterial tyrosine sulfotransferase, RaxST, is re-quired for activation of rice XA21-mediated im-munity, and it catalyzes sulfation of tyrosine residues of Omp1X and RaxX in Xanthomonas oryzae pv. oryzae, a causal agent of bacterial blight in rice. Although RaxST is biochemically well-characterized, bio logical functions of tyrosine sulfation have not been fully elucidated. We compared protein expression patterns between the wildtype and a raxST knockout mutant using shotgun proteomic analysis. Forty nine proteins displayed a more than 1.5-fold difference in their expression between the wildtype and the mutant strains. Clusters of orthologous groups analysis revealed that proteins involved in cell motility were most abundant, and phenotypic observation also showed that the twitching motility of the mutant was dramatically changed. These results indicate that tyrosine sulfation by RaxST is essential for Xoo move-ment, and they provide new insights into the biological roles of RaxST in cellular processes.

        Rhanella aquatilis 유래 당단백질과 항암제 혼합물에 의한 인체 대장 암 HT29세포에 대한 항암상승효과

        In order to investigate the anticancer activity of an anti-yeast substance (AYS), a proteoglycan produced by Rhanella aquatilis AY2000, the cytotoxicity of the AYS against cancer cells was determined in vitro. The AYS was not cytotoxic to the human Jurkat T cell or the mouse sarcoma 180 cell, but was cytotoxic to the human colon cancer TH20 cell. The AYS was increasingly cytotoxic against human colon cancer cells in a dose-dependent manner at range from 62.5 to 500 μg/ml. Anticancer activity by combination of the AYS and an anticancer agent was also determined. The anticancer agent combined with the AYS was shown to possess greater synergistic anticancer activity against human colon cancer cells, as compared with the anticancer agent alone. Rhanella aquatilis AY2000 균주가 생산하는 일종의 당단백질인 항효모성 물질 (AYS)에 대한 항암활성을 조사하기 위해 in vitro에서 암세포에 대한 AYS의 세포독성을 조사하였다. 그 결과 AYS는 인체의 Jurkat T 세포와 마우스의 sarcoma 180 세포에 대해서는 세포독성을 나타내지 않았으나, 인체 대장암세포인 colon cancer TH20 세포에는 세포독성을 나타내었다. 또한 이 AYS는 62.5에서 500 μg/ml까지 농도의존적으로 인체대장암세포에 대해 세포독성을 증가시켰다. 뿐만 아니라 이 AYS와 시판 항암제를 혼합하여 처리한 결과 시판 항암제를 단독으로 처리한 것 보다 인체대장암세포에 대한 항암효과가 더욱 상승되었다.

        Effect on Immune Cells of Proteoglycan Originating from Rhanella aquatilis

        So as to evaluate its use an immune stimulator in humans, the toxicity and action against immune cells by an anti-yeast substance (AYS), a bacterial proteoglycan, were investigated. The AYS did not possess hemolytic activity with human red blood cells (hRBC). Nor did it exhibit cytotoxicity against human peripheral blood mononuclear cells (hPBMC). In addition, the AYS did not induce hPBMC proliferation, but it did agglutinate hPBMCs in vitro. Moreover, hPBMC induced inflammatory cytokines such as IL-6, IL-5, IFN-γ and TNF-α with the AYS during culture. Compared with alum, the AYS as an adjuvant has an increased antibody production rate against bovine serum albumin (BSA) in mice.

        하브루타 학습법을 활용한 온라인 토론이 간호대학생의 의사소통능력, 자기주도적 학습능력 및 학업적 자기효능감에 미치는 효과

        본 연구는 5주간의 하브루타를 활용한 온라인 토론을 적용한 후 의사소통능력, 자기주도학습능력, 학업적 자기효능감의 차이를 확인하고자 하는 것이다. 본 교육 프로그램은 간호대학생을 대상으로 온라인 플랫폼을 활용하여 질문중심, 친구가르치기, 논쟁중심 하브루타 방식으로 짝을 지어 토론하도록 하였다. 본 연구대상자들은 일 지역의 2학년 간호대학생으로 선정하였으며 2020년 10월에 사전조사하고 5주간 교육 프로그램 진행 후 2020년 12월에 사후조사 하였다. 본 연구에서 하브루타 활용 온라인 토론 전과 후의 의사소통능력, 자기주도적 학습능력은 통계적으로 유의한 차이가 없었고, 학업적 자기효능감은 유의하게 상승하였다. 비록 항목에서는 유의한 차이를 보이지 않았지만, 교육 프로그램 적용 후 사후조사에서 모든 항목이 증가하여 긍정적인 양상을 볼 수 있었다. 결론적으로 이 연구를 통하여 하브루타를 활용한 온라인 토론은 간호대학생에게 학습역량을 향상시킬 수 있는 효과적인 교육방법임을 제시하고 있다. 향후의 연구에서는 교육 프로그램의 장기적인 적용기간, 학업의 정의적인 요소, 동등성 대조군 전후 설계 연구 등을 포함한 연구들이 진행될 필요성을 가지고 있다. This study examined the effects of the Havruta-based online debate on communication competency, self-directed learning, and academic self-efficacy in nursing students. We used a one-group pretest-posttest design. Data were collected using structured online questionnaires from November 2020 to December 2020 in 81-second grade nursing college students attending a university in A city. The paired t-test analyzed a difference between pre-and post- Havruta-based online learning. In the results, there was no statistically significant difference in communication ability and self-directed learning, although it showed a post-intervention increase. However, it had a notable increase in academic self-efficacy. With this result, we hope this study can guide the development of a Havruta-based nursing education program. Also, applying various teaching methods to nursing education programs could be considered to enhance communication competency and self-directed learning for nursing students.
