The possible mechanism of the antiproliferative and apoptotic effects of Boswellia carterii water extract were studied in HL-60 human leukemia cells. The cytotoxicity of HL-60 cells after the treatment of Boswellia carterii water extract showed dose- and time-dependent manner. The appototic effect of 300 mcg/ml Boswellia carterii water extract was demonstrated by DNA laddering. The activity of capase 3-like protease was markedly increased in HL-60 cells treated with Boswellia carterii water extract. Furthermore, the level of Bcl-2 was time-dependently reduced, whereas Bax protein level was enhanced by Boswellia carterii water extract treatment. In conclusion, our results suggest that apoptotic effect of Boswellia carterii water extract treatment. In conclusion, our results suggest apototic effect of Boswellia carterii water extract may partly mediated through activations of capase-3-activity, and Bax expression, and inhibition of Bcl-2 expression.

To investigate whether glycyrrhizae inhibits melanogenesis, we utilized B16 melanoma cells. Glycyrrhizae alone decreased the tyrosinase promoter activity of B16 melanoma cells in dose-dependent manner. Glycyrrhizae attenuated tyrosinase promoter activity and tyrosinase activity. Glycyrrhizae also prevents phosphotransferase activity of c-Jun N-termianl kinase(JNK1) and transcriptional activation of AP-1 in α-MSH-treated B16 melanoma cells. In conclusion, the current studies for the first time demonstrated that an inhibitory effect of melanogenesis by glycyrrhizae on B16 mouse melanoma cell might be a new mechanism which down-regulates JNK activity via inhibition of AP-1 activity.

Gliotoxin은 Penicillium, Aspergillus, Gliocladium 및 Thermoascus와 같은 곰팡이들로부터 생성되는 대사산물중 하나로, 공통적으로 epipolythiodioxopiperazine (ETP)계 구조를 가지고 있다. 이 ETP계 구조에 의해 질병이나 면역조정기능과 같은 다양한 생리적 효과를 나타낸다. 심근세포인 H9c2세포에 gliotoxin을 처리한 결과 caspase-3의 활성에 의해 DNA fragmentation 을 유발시켜 세포의 손상을 유도하였다 Phorboll2-myristate 13-acetate(PMA)를 시간별로 처리한 경과, gliotoxin을 처리하기 전에 PMA를 15분전에 처리한 경우에는 caspase-3의 활성을 억제시켜 세포의 손상을 억제시켰으나, PMA를 gliotoxin과 동시에 처리하거나 후처리한 경우에서는 세포의 손상을 억제시키지 못했다. 따라서 ,PMA 천처리에 의해서 protein kinase C (PKC)가 활성화되며, 이 활성화된 PKC가 caspas-3의 활성을 차단하므로서, 아폽토시스를 억제한다는 것을 알 수 있었다.

Apoptosis is a complex process that removes aging or damaged cells from the body and occurs in a wide variety of organisms. Malignant gliomas are well known for their intrinsic heterogeneity that contributes to tumour recurrence, regardless current aggressive multimodality of therapy. New approaches of non-surgical intervention are formulated, according to specific characteristics of glioma biology including angiogenesis, invasion, DNA-repair mechanisms, cell cycle regulation and apoptosis. Specific proapoptotic approaches may overcome many of the obvious obstacles to achieve a satisfactory management of brain tumors. Various therapeutic approaches toward successful eradication of glioma by an induction of apoptosis have been developed, however these approaches may be hampered frequently by anti-apoptotic mechanism. One promising experimental strategy in treatment of glioma is to activate apoptosis pathway within tumor cells, which will employ the death ligands such as CD95(Fas/Apo1) ligand or Apo2 ligand(TRAIL), cytosolic protease, growth factor-related signal molecules, and intracelluar organelles(mitochondria and golgi complex), etc. In this review, we described the representative apoptotic pathways including Bcl-2, caspase, death receptor, and tumor suppressor gene to suggest their possibilities of adaption in comprehensive treatment to overcome chemoresistance or radioresistance of human brain tumor cells.

This study was to investigate the effect of antioxidant supplements, vitamin C and E on VO2max and anti-oxidant related gene expressions in college handball players. Fourteen subjects were divided into two groups, the antixoidant supplement group (ASG) and the placebo supplement group (PSG). The ASG group had 12 weeks of vitamin C and E supplementations with high intensity training, while the PSG group had placebo tablets with the same training intensity and duration that the ASG group did. The VO,max values after 12 weeks in the ASG and PSG groups were 64.43 ± 1.27 ml/kg/min and 62.14 ± 1.35 ml/kg/min, respectively. The ASG group compared to the PSG group had significantly higher VO₂max (p<0.01). The RT-PCR analysis data showed that the PSG group compared to the ASG group had significantly higher mRNA. expression levels of the anti-oxidant related genes such as SOD-1, GPx-4, GSSG-R, CAT and GSH-S at sixth week. Taken together, these results suggest that antixoidant supplements such as vitamin C and E may have some beneficial effects to enhance endurance capacity through the up-regulation of the antioxidant system.
