        미세조류 추출물 및 MAAs 함유 자외선차단제의 자외선차단지수(SPF) 및 자외선 A 차단지수(PFA) 평가

        천연물 유래 자외선차단제 개발을 위해 미세조류 추출물 및 mycosporine-likeaminoacids(MAAs)의 활용가능성을 연구하였다.Chlamycomonashedleyi추출물과 추출물 분획물인 MAAs가 각각 7% 포함된 자외선차단크림을 제조하고 인체 적용시험을 실시하였다.이를 통하여 자외선차단지수(SPF)및 자외선 A차단지수(PFA)를 평가하였다.Ch.hedleyi추출물과 MAAs함유 자외선 차단 크림의 자외선 차단지수는 각각 9.07및 9.42로 나타났으며,자외선 A차단지수는 각각 2.43과 2.41 로 측정되었다.자외선 A차단지수가 모두 2 상으로 측정되어 두가지 자외선 차단크림의 자외선 A차단등급은 [PA+]으로 분류되었다.이상의 연구결과는 미세조류 추출물 및 MAAs함유 자외선차단제가 자외선을 효과적으로 막지 못하지만,무기 자외선 차단제가 혼합되면 만족할만한 자외선 차단효과를 보일 수 있을 것으로 판단된다. For the sunscreen development from natural resources, a possible usage of microalgal extracts or mycosporine-like amino acids (MAAs) was investigated. Sunscreens containing 7% of microalgal extracts or MAAs derived from microalgae, Chlamydomonas hedleyi, were prepared and they were applied to human research. Through this clinical research, the values of Sun Protection Factors, Sun Protector Factor (SPF) and Protection Factor of UVA (PFA), of sunscreen containing microalgal extracts or MAAs were determined: SPF values of microalgal extracts and MAAs indicated 9.07 and 9.42, respectively, while PFA ones did 2.43 and 2.41. Due to more than 2 of PFA value in both sunscreens, they can be classified into [PA+]. Taken together, although sunscreen containing microalgae-derived extracts or MAAs does not effectively protect UV irradiation, its capacity can be satisfied if inorganic UV-protecting compounds are added.

        생리활성 펩타이드의 피부미용학적 특성 및 활용

        Bioactive peptides (BAP) showed excellent cosmetic activity than bio-materials such as caffeic acid (CA), gallic acid (GA), and nicotinic acid (NA). Caffeoyl tripeptide-1 (CT-1) is a BAP that is stabilized with Gly-His-Lys (GHK) tripeptide and CA by using Fmoc solid phase peptide synthesis. Digalloyl tetrapeptide-19 (DT-19) is stabilized by combining Lys-Glu-Cys-Gly with GA and nicotinoyl tripeptide-1 (NT-1) is synthesized by GHK and NA. According to experiments, CT-1 has an excellent anti-oxidant function even with a very small amount of 10 ppm CT-1. DT-19’s tyrosinase inhibition activity has the better effect of about 28.57% in 0.01% and 33.33% in 0.005% of concentration and about 7.89% in 0.001% concentration than vitamin-C. In addition, NT-1 is safer than the NA. Almost BAPs like pal-KTTKS, acetyl hexapeptide, and copper tripeptide-1 have the anti-wrinkle effect while DT-19 and NT-1 are applicable for potential BAPs focused on the whitening effect. The three kinds of BAPs like CT-1, DT-19, and NT-1 consisting of amino acids are safe to the skin, and have more excellent stability than bio-materials which are found to be unstable and cause skin irritation. Due to the high biological activity of BAP in the field of skin care, its utilization will increase constantly.

        딱지꽃 부위별 추출물의 피부세포에서의 효능 연구

        As natural plant-based industry has been expanded, the use of oriental medicinal plants as cosmeceutical material was received a lot of attentions in the cosmetic industry. Among various medicinal plants, Potentilla chinensis have been interested in its biological effects. Although several attempts were tried to study its biological effect as medicinal plant, only limited studies tried to evaluate its biological effect as cosmeceutical material. In this study, we examined the possibility of Potentilla chinensis extract as a cosmeceutical material. After extraction of Potentilla chinensis, biological evaluation including anti-inflammation, anti-wrinkle, whitening effect and moisturizing effect was performed as cosmeceutical material. As a result, the Potentilla chinensis extract showed remarkable biological activities through stimulating elastin and aquaporin-3, and inhibiting melanin synthesis, cyclooxygenase-2 expression and expression of metallopeptidase-1, respectively. These results show the potential use of Potentilla chinensis as a functional cosmetic material for anti-inflammatory, wrinkle improvement, skin hydration and whitening effects.

        식물세포배양기술을 이용한 순무 캘러스 추출물의 생리활성

        본 연구에서는 인천광역시 강화군의 특화작물을 이용한 기능성 소재를 개발하기 위해 순무 캘러스를 배양하고 열수, 초음파 그리고 에탄올 추출하여 생리활성을 평가하였다. 순무 캘러스의 열수, 초음파 그리고 에탄올 추출물에서 DPPH radical소거활성이 대조군에 비해 농도 의존적으로 증가하였다. 순무 캘러스 에탄올 추출물의 수렴효과는 열수 및 초음파 추출물보다 높게 나타났다. 콜라겐 생합성량을 측정한 결과 순무 캘러스 에탄올 추출물의 5 ppm과 10 ppm 농도에서 59%와 78%의 주름개선 효과를 확인하였다. 이러한 결과들은 순무 캘러스의 열수, 초음파 그리고 에탄올 추출물이 천연 항산화 소재로의 활용이 가능할 것으로 사료되며 특히, 에탄올 추출물은 수렴효과와 주름개선 효과가 뛰어나 기능성 화장품 천연소재로서의 활용이 가능할 것으로 기대된다. The purpose of this study was to investigate biological activities of Brassica rapa (Turnip) plant callus extracts of Ganghwa-gun of Incheon city using water, ultrasonic wave and ethanol extractions to develop functional materials. DPPH radical scavenging activities of the callus extracts were increased in a concentration-dependent manner, as compared with control. The astringent effects of the ethanol extracts were higher, as compared to water and ultrasonic extracts. In the collagen synthesis assay, the ethanol extract showed significant anti-wrinkle effects of 59% and 78% at a concentration of 5 ppm and 10 ppm, respectively. These results suggested that water, ultrasonic wave and ethanol extracts of turnip plant calluses are natural antioxidant sources. Especially, the ethanol extract can be regarded as a functional, natural cosmetic material with astringent and anti-wrinkle effects.

        Mycosporine-like amino acids (MAAs) 처리에 따른 배양세포 내 스크래피 프리온 단백질의 형성증가

        Prion은 양의 scrapie, 소의 bovine spongiform encephalopathy와 사람의 CJD와 같은 다양한 신경 퇴행성 질환을 유발시키는 단백질 병원체이다. 정상 prion 단백질인 PrP<sup>C</sup>가 병원성 PrP<sup>Sc</sup>로 바뀌는 과정에 대해서는 많은 연구가 진행되었고, PrP<sup>Sc</sup>로의 단백질 구조 변화가 다양한 환경적 요소에 의해서 영향 받는 것으로 추측된다. 바다조류로부터 분리된 MAAs는 다양한 스트레스 환경에서 조류를 보호해주는 것으로 알려져 있다. 이와 같은 사실에 기초하여 mycosporineglycine, porphyra-334와 shinorine 3종의 MAAs로 처리한 prion 감염 신경세포 주에서 prion 단백질 축적의 변화를 평가하였다. PK 저항성을 갖는 PrP<sup>Sc</sup>를 western blot 방법으로 확인한 결과, MAA에 의해서 PrP<sup>Sc</sup> 단백질의 증식을 관찰하였다. Prions are proteinaceous infectious particles that cause neurodegenerative diseases, such as scrapie in sheep, bovine spongiform encephalopathy in cattle and Creutzfeldt-Jakob disease (CJD) in humans. Although the detailed process, regarding the abnormal conversion of prion proteins (PrP), remains to be fully elucidated, a number of environmental factors appear to affect the formation of misfolded PrP, termed PrP<sup>Sc</sup>. Because oceanic algae contain mycosporine-like amino acids (MAAs), which exhibit cellular defensive activities under a variety of stress conditions, we investigated the level of PrP<sup>Sc</sup> in prion-infected neuroblastoma cells using mycosporine-glycine, porphyra-334 and shinorine. When judged by the level of protease-resistant PrP<sup>Sc</sup> in western blots, porphyra-334 and shinorine increased the level of PrP<sup>Sc</sup> in cells, but mycosporine-glycine did not. The current results indicate that the MAAs tested in this study enhance the formation of PrP<sup>Sc</sup>.

        Radiofrequency에 노출된 Chlamydomonas sp.의 mycosporine-like amino acids 생합성 유전자 발현

        Mycosporine-like 아미노산(MAAs)은 UV 흡수물질이며, 다양한 해양생물들은 MAAs의 합성과 축적을 통하여 환경 자외선의 직‧간접적인 영향을 감소시키는 기능을 진화시켜 왔다. 이 연구에서 우리는 radiofrequency(RF) 발생장치를 제작 하였고, 이를 미세조류 배양에 적용하였다. 0.35±0.05 mHz의 RF를 Chlamydomonas sp. 배양기에 공급하였고, 정해진 시간(0, 0.5, 1 및 2 시간)에 시료를 채취하였다. MAAs 생합성 관련 유전자인 dehydroquinate synthase homolog (DHQS-like)와 nonribosomal peptide synthetase homolog (NRPS-like) 유전자를 Chlamydomonas sp.로부터 클로닝하였고, RF 노출에 대한 유전 자 발현을 qRT-PCR을 이용하여 분석하였다. 연구결과 RF에 노출된 Chlamydomonas sp.의 DHQS-like와 NRPS-like 유전자의 발현은 노출 1시간에 각각 1.46 배 및 1.19 배 증가하였다. 이러한 결과는 DHQS-like와 NRPS-like 유전자가 Chlaydomonas sp.의 MAAs 생합성을 진단할 수 있는 좋은 바이오마커 후보가 될 수 있음을 의미한다. Mycosporine-like amino acids (MAAs) are UV-absorbing substances, and diverse marine organisms have the evolved the capacity to diminished the direct and indirect damaging effects of environmental ultraviolet radiation by synthesis and accumulation of MAAs. In this study, we manufactured a radiofrequency (RF) generation device and applied to microalgal culture. 0.35±0.05 mHz of RF was supplied to culture vessel for Chlamydomonas sp. and samples were harvested at the designated time intervals (1, 0.5, 1 and 2 hr). MAAs biosynthetic genes, dehydroquinate synthase homolog (DHQS-like) and nonribosomal peptide synthetase homolog (NRPS-like), were cloned from Chlamydomonas sp. and their gene expressions under the RF exposure were analyzed using qRT-PCR. DHQS-like and NRPS-like gene expressions of Chlamydomonas sp. exposed to RF were increased 1.46 and 1.19 fold at 1 hr, respectively. These results means that DHQS-like and NRPS-like genes can be good biomarker candidates for diagnosis of MAAs biosynthesis in the Chlamydomonas sp.

        금/백금 프레임 나노입자의 합성과 응용

        Rare metals have been classified as highly valuable metals from ancient times. In modern society, rare metals are used in foods, sanitary products, cosmetics, household goods. Nanoparticles of gold and platinum have been used as therapeutic adjuvant and sensor efficiency amplification materials. Especially, it is a big goal to control the morphology in a beneficial direction to the human body by using properties that change depending on the form of the gold platinum nano particle. In this study, morphology control of gold and platinum with octahedron and frame structure was made possible to increase the volume to surface area ratio. As a result of experiments on human dermal keratinocytes, it was confirmed that high antioxidant activity is effective in anti-aging.

        광양만 해역에서의 고효율 질소-인 제거 해양박테리아 탐색 및 분리

        371 균주의 해양박테리아를 광양만에서 분리하였다. 우점종은 Pseudomonas aeruginosa, Aeromonas hydrophila, P. fluorescens, P. paucimobilis, Chryseomonas luteola, P. vescularis 등이었다. 영양염과 유기물을 제거할 수 있는 해양박테 리아를 탐색하기 위하여 암모니아성 질소(100 mg/L), 질산성 질소(100 mg/L) 및 인(10 mg/L)이 각각 포함된 10 mL의 marine broth 2216 (DIFCO)에 해양박테리아를 접종(1.0%, v/v)하고 12시간 배양하였다. 스크리닝 테스트 결과 25% 이상 COD을 제거하는 해양박테리아는 16종, 15% 이상의 암모니아 질소를 제거하는 해양박테리아는 9종, 60% 이상의 질산 질소를 제거하는 해양박테리아는 11종 그리고 90% 이상의 인을 제거하는 해양박테리아는 13종이 분리되었다. Aeromonas hydrophila, Chryseomonas indologenes, Pseudomonas diminuta, Vibrio parahaemolyticus 균주가 유기물 및 영양염류 제거 실험을 위해 선정되었다. 회분식 시험을 위해 4종의 해양박테리아를 COD<sub>Cr</sub> 250 mg/L, NH<sub>3</sub>-N 40 mg/L, NO<sub>3</sub>-N 40 mg/L, PO<sub>4</sub><sup>3-</sup>-P 10 mg/L이 각각 첨가되어 있는 변형 marine broth에 접종하고, 10시간 배양하면서 제거효율을 측정하였다. 371 strains of marine bacteria were isolated from Gwangyang bay in Korea. The dominant species were identified as Pseudomonas aeruginosa, Aeromonas hydrophila, P. fluorescens, P. paucimobilis, Chryseomonas luteola and P. vescularis. To screen marine bacteria capable of removing nutrients and organics, marine bacteria was inoculated in 10 mL of marine broth 2216 (DIFCO) with NH<sub>3</sub><sup>-</sup>N (100 mg/L), NO<sub>3</sub><sup>-</sup>N (100 mg/L), and PO<sub>4</sub><sup>-</sup>P (10 mg/L) with 1.0% (v/v), and incubated for 12 h. Results from the screening test, showed that the removal efficiencies for COD<sub>Cr</sub>, ammonia niterogen, nitrate nitrogen, and phosphate were over 25% for 16 strains, 15% for 9 strains, 50% for 63 strains, and 15% for 80 strains, respectively. Aeromonas hydrophila, Chryseomonas indologenes, Pseudomonas diminuta, Vibrio parahaemolyticus were selected for nutrients removal experiments. For the batch test, 4 species of marine bacteria were inoculated in modified marine broth containing with nutrients(COD<sub>Cr</sub> 250 mg/L, NH<sub>3</sub><sup>-</sup>N 40 mg/L, NO<sub>3</sub><sup>-</sup>N 40 mg/L, PO<sub>4</sub><sup>-</sup>P 10 mg/L, respectively), incubated for 10 hr and the removal efficiencies were measured.

        삭시톡신 분석을 위한 항체의 제조 및 항-삭시톡신 항혈청의 민감도 분석

        해양미세조류 유래 독성물질에 대한 이해와 활용에 있어서 가장 중요하지만 간과되고 있는 부분은 독성물질 을 검출할 수 있는 빠르고, 쉽고 경제적인 검출기술을 개발하는 것이다. 이 논문에서 우리는 삭시톡신(STX)에 대한 항체를 생산하였다. 헤모시아닌(mariculture keyhole limpet hemocyanin, mcKLH)과 오브알부민(ovalbumin, OVA)을 운 반단백질로 사용하였다. 면역반응을 위해서 mcKLH-STX 결합체를 BALB/c 쥐에 복강주사하였다. 채혈 후 항-STX 항 혈청을 분리하였다. 항혈청의 역가분석을 위하여 유리 STX와 OVA-STX로 코팅된 microtiter plate를 이용하여 간접 ELISA 실시하였다. 발색반응을 위한 이차항체로는 goat anti-mouse IgG-phosphatase conjugate가 사용되었다. 항-STX 항혈청은 OVA-STX와 유리 STX에 특이적으로 반응하였다. 항-STX 항혈청의 민감도는 매우 높았으며, STX를 위한 검출한계는 약 64.9 ng/kg이었다. The most essential but missing components to understand and use toxic substances from marine microalgae are developing the fast, easy and economical determining technology for detecting it. In this paper we produced the antibodies against saxitoxin (STX). Mariculture keyhole limpet hemocyanin (mcKLH) and ovalbumin (OVA) were used as carrier proteins. mcKLH-STX conjugates were injected into the peritonial cavity of BALB/c mouse for immunization. After bleeding from mouse, anti-STX antiserum was isolated. Indirect enzyme-linked immunosorbent assays (ELISA) was performed to determine antiserum titer using the microtiter plate coated with free STX and OVA-STX. A goat anti-mouse IgG-phosphatase conjugate was used as secondary antibody to enable chromogenic reaction. Reactions of anti-STX antiserum were very specific on the OVA-STX and free STX. Sensitivity of anti-STX antiserum on STX was very high and STX detection limit was to be 64.9 ng/kg for indirect ELISA.

        고삼뿌리 추출물 내 Kurarinone, Oxymatrine, Matrine 분석 방법 검증

        The root of the Sophora flavescens with excellent pharmacological efficacy has been used as a herbal medicine. The test was carried out to verify the analysis of kurarinone, oxymatrine and matrine in the root extract of the Sophora flavescens using HPLC. As a result of confirming the MS/MS by fractionation, it was confirmed that the corresponding peaks were kurarinone, oxymatrine, and matrine, respectively, as the MS/MS spectrum was consistent. As a result of confirming the accuracy test and reproducibility, and the precision within and between batches, it was possible to confirm high accuracy and precision. As a result of confirming the stability of the pre-treated sample, all three materials were stable for 48 hours. In conclusion, it was evaluated and established that the method validation for kurarinone, oxymatrine and matrine in the range with the specificity, linearity, accuracy, precision, and stability.
