        Cytotoxic Activity and Quantitative Structure Activity Relationships of Arylpropyl Sulfonamides

        B13 is a ceramide analogue and apoptosis inducer with potent cytotoxic activity. A series of arylpropyl sulfonamide analogues of B13 were evaluated for their cytotoxicity using MTT assays in prostate cancer PC-3 and leukemia HL-60 cell lines. Some compounds (4, 9, 13, 14, 15, and 20) showed stronger activities than B13 in both tumor cell lines, and compound (15) gave the most potent activity with IC50 values of 29.2 and 20.7 μM, for PC-3and HL-60 cells, respectively. Three-dimensional quantitative structure-activity relationship (3D-QSAR) analysis was performed to build highly reliable and predictive CoMSIA models with cross-validated q2 values of 0.816 and 0.702, respectively. Our results suggest that long alkyl chains and a 1R, 2R configuration of the propyl group are important for the cytotoxic activities of arylpropyl sulfonamides. Moreover, the introduction of small hydrophobic groups in the phenyl ring and sulfonamide group could increase biological activity.

        Efficacy and Safety of Sunitinib on Metastatic Renal Cell Carcinoma: A Single-Institution Experience

        Purpose: We assessed the efficacy and safety of the tyrosine kinase inhibitor sunitinib in Korean patients with metastatic renal cell carcinoma (mRCC). Materials and Methods: Between September 2007 and December 2009, all twenty-one patients who had mRCC with a clear-cell component were retrospectively reviewed. Sunitinib was administered orally at a dose of 50 mg daily until disease progression or intolerance to treatment occurred. The primary end point of this study was the objective tumor response assessed by Response Evaluation Criteria in Solid Tumors (RECIST), and the secondary end points were progression-free survival (PFS) and overall survival (OS) rates as well as assessment of adverse effects. Results: After a median of 17.4 months (range, 5.7-33.1 months) of treatment, 11 patients (52.4%) had an objective response with a complete response in 1 patient (4.8%), and a partial response in 10 patients (47.6%) as the best tumor response. The median PFS was 13.4 months (95% confidence interval [CI], range, 12.3-14.5 months), and the median OS was 28.1 months (95% CI, 21.8-34.4 months). All patients experienced adverse events of some sort, but the studied treatment protocol was well tolerated and most patients experienced reversible grade 1 or 2 toxicities. Conclusions: Sunitinib was efficacious in the treatment of metastatic clear-cell RCC, and was well tolerated in Korean patients. Although sunitinib treatment-related adverse events such as hand-foot syndrome and facial/generalized edema were observed with a higher incidence than in Western trials, they were mainly mild to moderate, and readily managed.

      • 니켈 촉매를 사용하여 바이오매스 촉매 열분해 및 촉매 가스화의 분해 비교 분석

        신재생에너지원 중 하나인 바이오매스는 화석연료를 대체할 수 있는 에너지원으로 많은 주목을 받고 있으며 활발한 연구가 진행 중에 있다. 열분해 및 가스화는 바이오매스를 고부가가치의 물질인 바이오오일이나 합성가스로 전환하는 기술로 열화학적 분해공정을 통해 고부가가치 물질로 전환되며 생성되는 물질의 개질 및 수율을 높이기 위하여 촉매가 사용되기도 한다. 촉매 중에서 니켈 촉매는 다른 촉매와 비교적 저렴하기 때문에 상용화에 유리하여 많은 연구에서 사용되고 있지만 실제 바이오매스에 적용한 연구는 드물다. 따라서 본 연구에서는 니켈 촉매를 바이오매스의 열분해와 가스화에 적용하였으며 분해능을 비교 분석하였다. <sup>**</sup> 본 연구는 2016년도 정부(미래창조과학부)의 재원으로 한국연구재단(NRF)의 지원을 받아 수행된 기초연구사업임(No. 2016R1D1A1B03935954).

      • 혁신 자본의 등장과 한국 스타트업 생태계에 대한 연구

        과거의 스타트업 자본은 엔젤투자와 벤처캐피털(VC)로 비교적 단순하게 양분됐다. 스타트업은 이들 자본을 바탕으로 제품 및 서비스 설계에서 마케팅, 해외진출에 이르는 창업의 전 과정을 개별 기업 단위로 추진했다. 그러나 ‘혁신 자본’과 ‘혁신 플랫폼’의 등장과 진화는 스타트업 생태계 확장과 질적 성장에 눈에 띄는 변화를 불러 일으켰다. 혁신 자본이란 단순히 스타트업 성장에 필요한 자금을 수혈하는 것에 그치지 않고 아이디어 플랫폼, 개발 플랫폼, 제조 플랫폼 등 다양한 혁신 플랫폼의 역할을 함께 수행한다. 이는 소규모 자본으로도 성공적인 창업을 가능케 만들었다. 혁신 자본이 과거 스타트업이 홀로 짊어졌던 창업의 리스크를 분산시키고 성공확률을 높이는 역할을 하고 있는 것이다. 본 연구는 혁신 자본 및 혁신 플랫폼의 등장과 진화가 스타트업 생태계 성장에 미치는 영향을 분석한다. 이를 위해 국내외 스타트업 투자자와 관련 전문가를 대상으로 혁신자본 및 혁신 플랫폼의 역할과 진화 양상에 대한 인터뷰 조사를 실시해 질적 분석을 진행한다. 또, 폐쇄적인 스타트업 생태계에서 개방 · 혁신을 기치로 한 스타트업 생태계로의 진화과정에서 얻게 되는 효율을 비용 · 편익 모델을 구성해 살펴본다.

        Induction of Apoptosis in Cervical Carcinoma HeLa Cells by Petasites japonicus Ethanol Extracts

        Petasites japonicus is a traditional medicine in Korea for treating carcinoma, bronchitis, and asthma. However, the molecular mechanisms of anti-cancer activity are not well understood. The purpose of this study was to confirm the anti-cancer activity of P. japonicus and its mechanism. Apoptosis was examined using morphological observation of nuclear changes and flow cytometry. Activation of apoptosis-related markers was detected. P. japonicus extract significantly inhibited HeLa cell growth in vitro in a dose-dependent manner. Induction of apoptosis by P. japonicus was mediated by up-regulation of Bax and down-regulation of Bcl-2. P. japonicus also activated caspase-9, capspase-3, and PARP, indicating induction of the mitochondrial intrinsic apoptosis pathway. We also confirmed that administration of P. japonicus significantly reduced tumor weight in the xenografted mice. Thus, P. japonicus was effective for inhibiting tumor growth in vitro and in vivo by inducing apoptosis, suggesting its usefulness as a potential anti-cancer agent.

      • Effect of CH<sub>4</sub> gas environment on aromatic content of bio-oils in catalytic pyrolysis of biomass

        Bio-oils is mainly composed of oxygenated compounds. To overcome these issues, bio-oil has to be upgraded by hydrodeoxygenation of bio-oil, but this process not yet been commercialized. This paper is to utilize CH<sub>4</sub> as a source of the hydrogen for the conversion of oxygenated compounds present in the bio-oil to aromatic rich fuel. It is expecting that the bio-oil produced in the pyrolysis of yellow poplar in presence CH<sub>4</sub> might selectively produces alkyl phenols. ** This work was also supported by the C1 Gas Refinery Program through the National Research Foundation of Korea(NRF) funded by the Ministry of Science and ICT(2015M3D3A1A01064899).

        Emotion Regulation Strategies as Moderators between Counterfactual Thinking Concerning Intimate Partner Violence and Trauma-related Emotions: An Ecological Momentary Assessment Study

        According to the cognitive model of posttraumatic stress disorder (PTSD), negative appraisals and negative emotions are key factors in PTSD symptoms. Moreover, emotion regulation strategies (ERS) may affect the severity of PTSD symptom differently. This study investigated the reported types and frequency of intimate partner violence (IPV) survivors, and the effects of counterfactual thinking (CFT) and ERS on trauma-related emotions in daily life via ecological momentary assessment (EMA). Data from 59 women who experienced IPV within the past year were analyzed. The results demonstrated that cognitive reappraisal may modulate the relationship between upward CFT and trauma-related emotions (B= -0.012, p= .005), but the effect of emotion suppression was not statistically significant (B= -0.006, p= .365). Especially, upward CFT may demonstrate a greater impact on trauma-related emotions in individuals who use a lower degree of cognitive reappraisal in daily life than in participants employing a higher degree. Conversely, although downward CFT also increased trauma-related emotions, neither type of ERS moderated this relationship (cognitive reappraisal: B= -0.069, p= .129; emotion suppression: B= -0.004, p= .947). These findings extend prior research on the effectiveness of cognitive reappraisal by reinforcing its ecological validity and emphasize the need for further investigations.

        Fermentation of Chestnut (Catanea crenata Sieb) Inner Shell Enhances Anti-Obesity Effects in 3T3-L1 and C3H10T1/2 Adipocytes

        Chestnut inner shell (CIS) is rich in phenols and flavonoids such as gallic acid and ellagic acid, which are known to exhibit effective antioxidant and anti-obesity properties. Fermentation using lactic acid bacteria can enhance the physiological activity by increasing the contents of such functional ingredients. In this study, we evaluated the anti-obesity effects of a CIS extract subjected to a fermentation process (fermented CIS [FCIS]). Treatment with CIS and FCIS extracts (125, 250, and 500 μg/mL) increased cell viability and did not induce apoptosis, indicating no toxicity. The extract suppressed the gene expression of adipogenic factors, peroxisome proliferation-activated receptor gamma, CCAAT/enhancer binding protein (C/EBP) alpha, and C/EBP beta (by 7.75% and 67.59%, 21.41% and 66.27% in 500 μg/mL, respectively), and consequently suppressed the expression of downstream lipogenic factors such as fatty acid synthase, stearoyl CoA desaturase-1, citrate synthase, and ATP citrate lyase. The expression of factors involved in fat catabolism and β-oxidation increased in a dose-dependent manner, thereby preventing fat accumulation. This observation was consistent with the significant decrease in the staining intensity for lipid droplets, which indicated that lipid accumulation was decreased by 15.46% and 29.44% in 3T3L-1 and 27.01% and 46.68% in C3H10T1/2. Together, these results demonstrate the higher anti-obesity effects of FCIS extract than that of CIS extract, indicating the potential applicability of FCIS as an effective natural raw material to curb obesity.
