In this study, we investigated the immunosuppressive effects of water-soluble barley β-glucan (BP)in mouse immune cells. The BP was prepared from barley (Hordeum vulgare), and it suppressed the proliferation of splenocytes stimulated with concanavalin A (Con A),interleukin-2 (IL-2), and alloantigenic splenocytes in a dose-dependent manner (0.25, 0.5, and 1mg/mL). Furthermore,the death-inducing activities of cytotoxic T lymphocytes (CTLs) and lymphokine-activated killer cells (LAKs) in alloantigen-reactive splenocytes were significantly suppressed by more than 50%. Also, the production of IL-2 in the splenocytes decreased. These results suggest that these barley-originated polysaccharides can be used as potential immunosuppressive agents.

Laminaria japonica의 polysaccharides(LP)가 마우스 비장 세포에서 현저한 면역 증강 활성을 보였다. Balb/c 마우스 비장 세포를 mitogen인 Con A, T cell growth factor인 IL-2 및 alloantigen인 anti-C57로 자극한 뒤, LP를 농도별(0.25, 0.5, 1 ㎎/㎖)로 첨가하여 마우스 비장 세포의 증식 활성을 측정한 결과, 비장 세포 증식능이 유의적으로 증가함을 확인하였다. 이러한 결과를 근거로 항원 특이적인 세포 독성 T 림프구(CTLs)의 활성과 IL-2에 의해 야기되는 비특이적인 림포카인 활성 살해 세포(LAKs)의 세포독성능을 측정하였다. 작동 세포와 표적 세포의 비율을 10:1로 혼합한 뒤, LP를 농도별로 첨가하여 세포 용해 시 발생되는 LDH를 측정한 결과, 동종 항원 반응 비장 세포의 살해 세포 작용이 유의적으로 증가하였으며, LAKs의 활성 또한 유의적으로 증가함을 확인하였다. 그리고 T 림프구의 활성에 직접적으로 관여하는 IL-2의 생산능을 측정한 결과, LP를 첨가한 경우 농도 의존적으로 IL-2의 생산량이 증가됨을 확인하였다. 이상의 결과로, 참다시마에서 획득한 LP가 마우스 비장 세포의 T 림프구 및 NK 세포의 증식에 영향을 미치는 면역 증강 활성을 가지고 있음을 입증하였다. Laminaria japonica polysaccharides (LP) were prepared from L. japonica through hot water extraction, ultrafiltration and gel chromatography. In this study, we investigated the immunomodulating activity of LP (0.25-1 ㎎/㎖) on the mitogen/alloantigen reactive proliferation and killing activity of the Balb/c mouse splenocytes. The LP directly induced the proliferation of splenocytes that was stimulated with mitogen or alloantigen in a dose-dependent manner. The killing activity of cytotoxic T lymphocytes (CTLs) and lymphokine activated killer cells (LAKs) were enhanced significantly in the LP treated cells. Also, the treatment of splenocytes with LP increased production of interleukin-2 (IL-2). These results suggest that polysaccharides from L. japonica show a substantial immunomodulating activity in mouse immune cells.
건선의 한약치료 효과에 대한 체계적 문헌 고찰과 메타 분석

Objectives : This study aimed to validate the effect of herbal medicine intervention to relieve the symptoms of psoriasis using systematic review and meta-analysis and provide the newest reason of effectiveness of Korean medicine to psoriasis. Methods : Data were collected through electronic database including Pubmed, Cochrane CENTRAL, NDSL OASIS, Koreantk. Two experts assessed risk of bias of randomized controlled trials by Cochrane group's Risk of Bias tool after searching, reviewing and selecting papers. Data were analyzed using Review Manager(RevMan) 5.3 and Comprehensive Meta Analysis 2.0. Results : Total number of selected trials was 16 randomized controlled trials. This study evaluated the risk of bias and effectiveness of herbal medicine to psoriasis. There were high frequency uncertain in selection bias, performance bias and detection bias. In this meta-analysis, Korean medicine treatment was more effective than western medicine (ES:0.507, 95%CI:0.147-0.867) and placebo (ES:0.955, 95%CI:0.598-1.312). Conclusions : Herbal medicine intervention can be an effective for treatment in psoriasis. But enhancing levels of evidence, we must try to accumulate clinical researches of herbal medicine to psoriasis in Korea.
인체 흑색종 세포에 대한 와송 추출물의 세포주기 억제를 통한 항암효과와 기전 연구

Objectives : The purpose of this study was to identify the anticancer effect of biological substances of ethylacetate(EtOAc) fraction from Orostachys japonicus(OJEF), their effect on human melanoma A375 cells and the related molecular mechanisms. Methods : The MTS assay was used to confirm the inhibition of cancer cell proliferation in A375 cells. And the MUSE™ analyzer was used to determine the ability of OJEF to induce cell cycle arrest. Western blotting was used to determine the changes in protein expression in A375 cells after treatment with OJEF. Results : OJEF showed cytotoxicity to A375 cells. And cell cycle arrest occurred in G1 phase and G2/M phase owing to inhibition of CDK1, cyclin B1, CDK4, and cyclin D, which are related to cell cycle regulation and cell division control. Conclusion : OJEF is effective in regulating cell cycle of human melanoma cells and thus can be a good theraputic agent to treat patients with melanoma.

The molecular mechanisms of the ethylacetate(EtOAc) fraction from Orostachys japonicus (OJE) (includinggallic acid, kaempferol, and quercetin) for anti-canceractivities in HepG2 cells were investigated. Apoptosis wasdetected using morphological observation of nuclear changesand investigation of phosphatidylserine exposure at thecytoplasmic membrane using FITC-Annexin V/PI staining. Activities of the apoptotic factors bcl-2, bax, cytochrome c,pro-caspase-3, 8, and 9, as well as MAPKs levels, weremeasured using western blotting. Some morphologic featuresof apoptosis were identified using confocal microscopy. OJE caused the expression of apoptotic proteins to change,as evidenced by an increased bax/bcl-2 ratio and increasedexpression of cytochrome c, and decreased expressions ofpro-caspase-3, 8, and 9. After HepG2 cells were exposed toOJE, expressions of p-JNK and p-ERK1/2 increased in adose dependent manner. OJE exhibits anti-cancer activityvia apoptosis induction through a mitochondria dependentsignaling pathway in HepG2 cells.
