        영양유전체학(Nutrigenomics)의 최근 경향

        With the decoding of human genome in 2004 and the recent development in nutritional science, there has been an integration of molecular biology and nutrition. As a consequence a new word “molecular nutrition” has been formed and recently the word ‘nutrigenomics’ is coined and widely being used. The field of science that showed the most positive result from grafting the science of nutrition and nutrigenomics is obesity. In 1994, Jeffrey Friedman from Rockeffeler University announced that ob gene and obesity has a close relationship and since then there’s been a huge research done on genes related to obesity from the molecular nutrition’s point of view. Even now there are many genes presented which are supposed to be related to obesity and big efforts are put into finding what exactly those genes do. Moreover studying only in the context of genes was not enough so functional genomics, which is the study of the functions of cells and the functions and effects between genes and protein products, is being studied. This review article discusses the relationship between nutrition and genes and the general idea of nutrigenomics. The article also discusses about the current research status on these subjects.

        체중조절 기전과 항비만 기능성물질

        The obese population has been increasing worldwide and obesity has become one of the socioeconomic problems. Obesity raises more concerns as more studies regarding its direct and indirect relativity to several diseases such as type II diabetes, hypertension, etc. are published. Since leptin, an important signal in the chronic control of food intake and energy expenditure, was discovered in 1994, there has been a great accumulation of knowledge on fighting obesity by facilitating pharmacological and nutritional strategies on the molecular level of the body weight control system. In particular, evidences are accumulating that particular food components affect our physiological function and gene expressions which are associated with body weight control. In this study, we review the four mechanisms for weight control and antiobesity functional agents such as HCA, L-carnitine, CLA, chitosan, calcium supplements capsaicin contained in red pepper, and oriental herbal mixture. We also describe about the efficacy and working mechanism of these functional agents on the basis of antiobesity mechanisms.

        HHD Mice를 이용한 대장암세포유래 펩타이드 특이적 CD8<sup>+</sup> T 세포의 입양전이

        Background: 1-8D gene is a member of human 1-8 interferon inducible gene family and is shown to be overexpressed in fresh colon cancer tissues. Three peptides 1-6, 3-5 and 3-7 derived from 1-8D gene were shown to have immunogenicity against colon cancer. Methods: To study tumor immunotherapy of these peptides we established an adoptive transfer model. $D^{b-/-}{\times}{\beta}2$ microglobulin (${\beta}2m$) null mice transgenic for a chimeric HLA-A2.1/$D^b-{\beta}2m$ single chain (HHD mice) were immunized with irradiated peptide-loaded RMA-S/HHD/B7.1 transfectants. Spleens were removed after last immunization, and splenocytes were re-stimulated in vitro. Lymphocytes from vaccinated HHD mice were transferred together with IL-2 to the tumor bearing nude mice that were challenged S.C. with the HCT/HHD/B7 colon carcinoma cell line that was found to grow in these mice. Results: Peptide 3-5 was found to be highly effective in CTL activity. Adoptively transferred anti-peptide 3-5 cytolytic T lymphocytes caused significant retardation in tumor growth. Conclusion: This study shows that peptide 3-5 can be the most effective candidate for the vaccine of adoptive immunotherapy against colon cancer.

        모링가(Moringa oleifera) 잎의 영양성분, 안전성 및 생리활성 효능에 관한 연구 고찰

        Moringa oleifera leaves, seeds, pods, roots, and flowers have been widely used for their medicinal and nutritional properties. Many studies have been conducted on the chemical composition and effectiveness of M. oleifera. In fact, almost every part of M. oleifera has been found to contain essential nutrients and medicinal value. Especially, the leaves of M. oleifera are known to have various nutrients and diverse efficacy. Several studies have assessed the potential toxicity of the leaves when prepared by various methods. The results showed that the M. oleifera leaves when prepared differently were safe in locally used doses and amounts. Moreover, M. oleifera is known to contain various physiological efficacies, including antioxidant, anti-inflammatory, antidiabetic, and anticancer effects and so on. In the latest research, many attempts are being made to utilize the diverse effects of M. oleifera. This research seems to be bringing a promising view of M. oleifera as a therapeutic functional food for various diseases.

        고지방식이 비만 유도 마우스에서 자색옥수수 추출물의 항당뇨 및 항염증 효과

        Metabolic syndrome, including obesity, glucose intolerance and elevated blood pressure, is related to type 2 diabetes and cardiovascular disease. Previous studies have reported the anti-oxidative, anti-inflammatory and anti-diabetic effects of purple corn extract. We investigated the efficacy of purple corn extract (PC) against high-fat diet (HFD)-induced obesity and glucose intolerance, and examined the underlying mechanisms by analyzing expression of proteins and genes involved in glucose regulation and macrophage infiltration. C57BL/6 mice were fed with normal chow diet (ND), or HFD treated with distilled water (DW, control) or PC, for 10 weeks. Although body weights were similar in the HFD-fed groups, we observed a decrease in the liver and epididymal adipose tissue (EAT) weights, and enhanced glucose tolerance test (GTT) results in the PC group, as compared with DW group. Liver showed increased Akt phosphorylation in the PC-treated mice; however, no changes were observed in the EAT, for all groups. In PC-treated mice, decreased macrophage infiltration was seen in the EAT, with a reduced expression of macrophage marker genes. Finally, proinflammatory cytokine gene expressions were decreased by PC in the EAT, and a modest trend for downregulation was observed in the liver. Hence, we conclude that PC may decrease glucose intolerance by increasing the phosphorylation of Akt and reducing the macrophage infiltration into the EAT.

        Today, scientific technology has become an essential part of human beings' everyday lives and is affecting him in every aspect. With the discovery of the three-dimensional structure of DNA by Watson and Crick in 1953, the life science started to take the central place in scientific development and initiated the emergence of a new field of science, molecular biology. The development of life science made us understand the physiological phenomena underlying some of the major human diseases such as aging, cancer, AIDS, and brain diseases. The successful cloning of sheep, Dolly, using the technique of developmental biology and genetic engineering brought about a big change in the conventional scientific concept that the nucleus of a differentiated cell cannot reverse its genetic ability like that of an undifferentiated cell which has totipotency for full development. The birth of Dolly also brought immediate response and worry that the same technique might be used to clone a human being. The people of this modern era have been watching the fast development of the life science and are amazed by its power and usefulness. However, they also fear its seemingly endless potential. Scientific development has tow faces, giving two opposite effects on our lives. It improves the quality of our lives in one hand and destructs the foundation on the other hand. why did God allow men to develop science? why did He commend us to "be fruitful and increase in number: fill the earth and subdue it"? Here we discuss the Christian perspective on the current issue of life-cloning. We also mention about the roles of religious idolatry and philosophical concepts such as naturalism and progressivism that have settled in our consciousness and have instigated human beings to manipulate life and clone it on their own. And finally, we discuss about the roles and attitudes of Christians and Christian scientists who are living in this age of science and technology.

      • 포항 부추의 생화학적 기능성 효과

        포항 지역에서 생산되는 부추를 중심으로 부추의 식품기능성 분석을 한 결과 암예방 지표 효소인 QR의 유도능을 확인하였고 항산화 효소의 유도능은 음성대조구에 비해 약 2배 이상의 활성을 보여 앞으로 더 많은 연구를 기대하게 한다. 지역에 따른 QR 유도능과 SOD 유사 활성 유도능의 차이는 각 지역의 토질과 환경에 의한 차이에 기인하는 것으로 판단된다. 이에 대한 구체적인 연구는 토양분석 등 기후 특성에 대한 연구가 이루어져야 할 것으로 사료된다. 부추의 항비만기능성 식품으로서의 가능성을 지방세포 분해능에 의해 측정한 결과 50㎍/㎕ 농도에서는 음성 대조구의 약 3배 이상의 분해능을 나타내어 항비만 기능성 식품으로서 효과가 큰 것으로 기대된다. Leek(Allium tuberosum Rotter) is a korean traditional herb of which leaves are used for daily food. In order to improve the potency of leek as a functional food, we investigated the several biochemical functions in vitro. The quinone reductase activity, and anticarcinogenic enzyme, was induced 2 times at the concentration of 1㎎/㎖ of leek as compared to that of negative control(80% methanol). The induction of SOD(Superoxide dismutase) activity was also demonstrated. But the functional activities of leek were shown to be a little difference depending on the place where the leek was produced. The difference might to be due to the different environmental factors where affects the growth of leek. In an experiment to test the lypolytic activity of leek, the glycerol release was significantly increased(about 3 times to that of negative control). This was suggested that leek can be considered as a potential food for diet. The above results provide the potential food, and more profound research is requested.
