        In Vitro Evaluation for Antioxidant Activities of Culture Broth of Tremella fuciformis

        To investigate antioxidant activity of the culture broth from submerged culture of Tremella fuciformis, we preferentially analyzed the chemical composition of culture broth, which was mainly composed of carbohydrate (296.39 mg/g) and protein (9.24 mg/g), respectively. Also, contents of polyphenols, flavonoids and flavonols were 16.63 mg/g, 9.19 mg/g and 83.74 ${\mu}/g$, respectively. Next, we examined the scavenging abilities on DPPH (1,1-diphenyl-2-picrylhydrazyl) radical, ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid) radical, the reducing power, and chelating ability on ferrous ions. All antioxidant activities of the culture broth were increased in proportion to its concentrations. The $IC_{50}$ values were in order as follows ABTS radical scavenging activity < OPPH radical scavenging activity < chelating power. Accordingly, these results suggest that pharmacological function of T. fuciformis might be due to, at least partially, their protective effects against oxidation and the culture broth of T. fuciformis was free radical inhibitors or scavengers at low concentration, involving possibly in termination of free radical reaction as primary antioxidants.

        된장으로부터 혈전용해능 및 β - Glucosidase 활성을 가진 균주 분리 및 효소생산 배지의 최적화

        재래식 된장으로부터 혈전용해능과 β-gluocosidase 효소를 생산하는 미생물을 분리하였으며, 분리한 균의 형태학적, 생화학적인 분류를 수행하여 Bacillus속으로 동정하였으며, Bacillus sp. KH-15로 명명하였다. 그리고 분리한 균주의 효소생산을 위한 배지조건과 배양조건을 측정하였다. 선발균주는 2% glucose, 0.5% yeast extract, 0.1% calcium chloride의 최적 배지에서 최대의 효소생산을 보였다. 초기 pH7~8과 40℃에서는 혈전용해능이 최대를 보였으며, 초기 pH6~8과 30~40℃에서는 β-gluocosidase 효소활성의 생산이 가장 높았다. Bacterial strains showing the firinolytic and β-glucosidase activity were screened from Doenjang. The strain of KH-15 revealed a high level of fibrinolytic and β-gluocosidase activity. The isolated bacterium was identified and desingnated as Bacillus sp. KH-15. The carbon, nitrogen and salts significantly influenced the fibrinolytic enzyme and β-glucosidase production. The optimized composition of medium appeared to be 2% glucose, 0.5% yeast extract, and 0.1% calcium chloride. The optimum pH and temperature for fibrinolytic enzyme and β-glucosidase activities were pH 7~8, 40℃ and pH 6~8, 30~40℃, respectively.

        Bacillus amyloliquefaciens CHP-12에 의한 대두 발효물의 Cyclo-His-Pro (CHP) 함량 증진 및 항당뇨 효과

        To enhance cyclo-His-Pro (CHP) content, soybean hydrolysate was obtained using the strains isolated from Chungkukjang and further purified by various purification steps. First, twenty two strains were screened from Chungkukjang containing high level of CHP. Among them, the strain No. 12, which showed higher productivity of CHP from soybean ferment and have homologous sequence with 16S rDNA of Bacillus amyloliquefaciens, was named B. amyloliquefaciens CHP-12. Through various purification processes, CHP was concentrated from soybean ferment using ultrafiltration, which showed the best efficiency of CHP production, with the yield (71.3%) and CHP content (2.14 mg/g). Moreover, when glucose tolerance test was performed in Type I Sprague-Dawley rat induced by streptozotocin using the soybean ferments [0.5 g soybean ferment/kg body weight (CHP-0.5 group)] and 1.0 g soybean ferment/kg body weight (CHP-1.0 group), there were significant differences in glucose levels between diabetes-control group (265.3 mg/dL) and soybean ferment-treated groups (CHP-0.5 group: 84.3 mg/dL and CHP-1.0 group: 85.3 mg/dL) 120 min after glucose injection (2 g/kg body weight) (p < 0.05). Accordingly, it is suggested that the soybean ferment containing high level of CHP might be a candidate material as an anti-diabetic supplement for manufacturing functional healthy foods.

        계지(桂枝)에서 정제한 보체계 활성화 다당의 작용양식과 당쇄구조

        계지에서 분리한 보체계 활성화 다당의 보체 활성화양식과 다당의 당쇄결합에 관하여 검토하였다. 계지 보체계 활성화 다당획분들(CC-Ⅲa, CC-Ⅲb, CC-Ⅲc)들은 정상인의 혈청중에 존재하는 주요 보체성분인 C3를 활성화하여 C3a와 C3b 보체 산물을 생성 시켰는데 이 과정은 anti-human C3를 이용한 면역전기영동을 실시하여 확인하였다. Ca^(++) 유무시의 보체계 활성을 측정한 결과 계지에서 분리된 CC-Ⅲa-3는 고전적 경로(classical pathway)와 부경로(alternative pathway)를 동시에 활성화시킴을 확인하였다. CC-Ⅲa-3을 메틸화하여 GC/MS로 분석한 결과 비환원 말단의 galactopyranose와 glucopyranose가 확인되었고 1,2-linked rhamnopyranose, l,6-linked mannopyranose, 1,3-linked glucopyranose, 1,6-linked galactopyranose 등이 확인되었다. The paper describes the activation mode and the glycosic linkage of anti-complementary polysaccharide isolated from Cinnamomum cassia. The polysaccharide fractions, CC-Ⅲa, CC-Ⅲb, and CC-Ⅲc, activated C3 component existed in normal human serum and produced C3 cleavage segments, C3a and C3b. The polysaccharide, CC-2-Ⅲa-3 activated the complement system both in the presence and absence of Ca^(++), suggesting that it involved in both classical and alternative complement path¬ways. Methylation of CC-2-Ⅲa-3 was performed with methylsulphinyl carbanion and methyl iodide in DMSO. The methylated products was hydrolyzed, then converted into the partially methylated alditol acetates. Gas chromatography-mass spectrometry(GC-MS) revealed derivatives of terminal Glcρ and Galρ, l,2-linked Rhaρ, 1,6-linked Manρ, l,3-linked Glcρ, 1,6-linked Galρ etc.

        양파껍질에서 분리한 플라보놀의 Xanthine Oxidase 저해기작

        양파껍질의 90% 메탄올 추출물은 12.8%의 추출율, 661.3㎎%의 플라보노이드 함량과 88.7%의 xanthine oxidase 저해율을 보였다. 이 추출물을 ODS column chromatography와 Sephadex LH-20 column chromatography에 의해 F₁과 F₂의 두개의 분획을 얻었으며, 자외선과 가시광선에서의 흡수패턴에 의해 3-OH가 유리상태로 존재하는 플라보놀임을 알 수 있었다. F₁과 F₂ 분획의 농도를 달리하여 저해활성을 측정한 결과, 분획의 농도가 증가할수록 xanthine oxidase의 활성이 감소되는 경향을 보였으며, F₁과 F₂의 IC_(50)값은 0.95㎍과 0.67㎍이었다. 반응계내에 단백질(혈청단백질) 농도의 양을 증가시킴에 따라 F₁과 F₂ 분획 공히 80% 이상의 저해활성을 보임에 따라 양파 껍질에서 분리한 플라보놀의 xanthine oxidase에 대한 저해활성은 효소에 대한 특이적인 것으로 추측된다. F₁과 F₂에 의한 xanthine oxidase에 대한 저해는 비경쟁적과 무경쟁적 저해 형태가 서로 혼합되어 있는 형태이다. The influence of flavonols from onion skin on xanthine oxidase was investigated, Methanol extract was showed 12.8% of yield, 661.3㎎% of flavonoids contents and 88.7% of inhibitory effect on xanthine oxidase, F₁ and F₂ fractions were obtained from the methanol extract by ODS and Sephadex LH-20 chromatography, F₁ and F₂ fractions flavonols(3-OH free) identified by UV/visible spectroscopy. Inhibitory effects of F₁ and F₂ on xanthine oxidase were increased with increasing concentration. IC_(50S) of F₁ and F₂ were 0.95㎍ and 0.67㎍, respectively. To confirm the specificity of F₁ and F₂ against xanthine oxidase, albumin was added to the reaction mixture. The inhibition of F₁and F₂ may be due to specific binding to xanthine oxidase. The modes of their inhibitions were of mixed type with respect to xanthine as a substrate.

        발효녹용의 균주 선별 및 발효녹용의 생리활성

        본 연구에서는 녹용의 효율을 높이고 아울러 새로운 약리효과를 기대하기 위해 녹용을 분해하는 미생물을 탐색하였으며, 선정된 미생물 이용하여 발효시킨 녹용에 대하여 생리활성을 측정하였다. Bacillus sp., Lactobacillus sp. 및 버섯균사체중 녹용발효에 적합 균주로 B. subtilis KH-15, SCB-3과 버섯 균사체인 Cordyceps militaris, Phellinus linteus, Inonotus obliquus 26136, Inonotus obliquus 26147을 이용하여 발효녹용 제조 시 발효물의 고형분 함량은 B. subtilis KH-15와 SCB-3가 가장 높았으며, total sugar, uronic acid, S-GAG 함량은 C. militaris에 의한 발효물이 각각 1619.3 ㎍/mL, 302.0 ㎍/mL, 119.9 ㎍/mL로 가장 높은 함량을 보였으며, sialic acid 함량도 21.6 ㎍/mL로 B. subtilis KH-15의 24.8 ㎍/mL보다 다소 낮은 함량을 보였다. 면역 증강을 예측할 수 있는 항보체 활성은 B. subtilis 균주로 발효한 경우는 B. subtilis KH-15 균주의 ITCH50값이 42.3%로 SCB-3 29.9%보다 높은 값을 보였으며, 버섯균사체를 이용한 발효 녹용 중에는 C. militaris가 50.1%로 가장 높은 항보체 활성을 보였다. 항산화력을 알 수 있는 ABTS 라디칼 소거능은 B. subtilis KH-15 발효물이 4.97 ㎎/mL로 가장 높은 소거능을 보였으며, DPPH 라디칼 소거능은 Inonotus obliquus 26147가 16.98 ㎎/mL의 IC50값으로 가장 높은 라디칼 소거능을 보였다. 이상의 결과에 의하면 녹용 추출물에 비하여 녹용 발효물은 uronic acid, S-GAG, sialic acid 등 생리활성물질을 많이 함유하였으며, 항보체 활성과 라디칼 소거능이 증진되는 것을 확인하였다. 따라서 녹용자체가 지니는 조혈작용 등 생리활성 외에 항보체 활성처럼 새로운 활성이 발현될 수 있을 뿐만 아니라 라디칼 소거능처럼 기존의 활성을 증진시키는 효과를 가지고 있음을 확인하였다. This study was conducted to isolate strain for the preparation of fermented antler (Cervus cornu parvum) and evaluate its physiological activities. The growth degrees of twenty-one samples from Bacillus sp., Lactobacillius sp. and mushroom strain on antler extract agar were evaluated in this study, and Bacillus subtilis KH-15, SCB-3, Cordyceps militaris, Phellinus linteus, Inonotus obliquus 26136, and Inonotus obliquus 26147 were selected. The fermented antler extract by C. militaris had relatively higher contents of total sugar (1619.3 ㎍/mL), uronic acid (302.0 ㎍/mL), sulfated-glycosaminoglycan (S-GAGs) (119.9 ㎍/mL) and sialic acid (21.6 ㎍/mL) than any other extracts. The anti-complementary activities of all fermented antler extracts were higher than non-fermented antler extract, and among these samples, fermented antler extract by C. militaris showed the highest anti-complementary activity (inhibition of 50% total complement hemolysis, ITCH50; 50.1% at 1,000 ㎍/mL). The ability of fermented antler extract by B. subtilis KH-15 to scavenge 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical (IC50; 4.97 ㎎/mL) was significantly the highest (p<0.05), whereas the extract from I. obliquus exerted significantly (p<0.05) high 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (IC50; 16.98 ㎎/mL) among all samples. The results of this study suggest that physiological effects including immuno-modulating and antioxidant activities of the antler may be increased through fermentation process.
