      • Isolation and Biochemical Properties of Ricin from Ricinus communis

        피마자(Ricinus communis)의 씨로부터 독성물질인 ricin을 DEAE-cellulose와 Sepharose-4B column을 이용하여 정제하였다. 분리된 ricin은 $K_{562}$ 세포의 증식을 억제시켰으며 적혈구 응집력이 있었다. 적혈구 응집력은 ricinus agglutinin이 갖는 응집력의 0.3%에 불과 했으며, 응집력의 억제는 galactose나 lactose, melibiose, 또 galactose를 포함하지 않은 ${\alpha}-L-rhamnose$나 L(+)arabinose에 의하여 일어났다. Ricin을 주입받은 ICR생쥐는 60ng에서 나흘만에 반이 치사되였다. The toxic lectin, ricin, present in the seeds of Korean castor bean, Ricinus communis, have been isolated and the biochemical properties were studied.         The toxic lectin, ricin, present in the seeds of Korean castor bean, Ricinus communis, have been isolated and the biochemical properties were studied. The isolation procedure involves ion exchange chromatography on DEAE-cellulose, affinity chromatography on Sepharose 4B column. The multiplication rate of K_(562) cell decreased and the death rate increased proportionally by the addition of the isolated ricin. It has hemagglutination activity but only 0.3% of the activity of the other lectin, ricinus agglutinin. The activity was inhibited by lactose, melibiose, galactose as well as α-L-rhamnose and L-arabinose. A dose of 60 ng of ricin caused death in 50% after 4 days in ICR mice.

        The yolk sac membrane(YSM) of the avian egg supports embryonic development by digesting yolk granules and delivering the digestion products to the embryo via the blood supply. Some of the hydrolytic enzymes of the YSM are serine esterases and proteases, possible targets of the organo-phosphorus (OP) insecticides. We sought to identify these potential targets and determine whether inhibition of them is functionally related to the teratogenic or embryotoxic action of the OP compounds in chick embryos. The ability of the OP compounds, administered either in ovo or in vitro, to pre-empt binding of [³H]DFP to the non-sedimentable proteins of the YSM cell-free extract was measured. The extract from untreated eggs had three major DFP-binding proteins with electrophoretically determined Mw values of 94, 83 and 72 KD. The physiological roles of these proteins are unknown. The 72 KD protein of the YSM, which ordinarily contains 80-90% of the incorporated [³H]DFP, was very sensitive to the actions of the OP compounds. OP binding of the 83 KD protein was most pronounced with DZN.

        To see any relationships between the American pokeweed(Phytoladda americana) and soil acidity, the survey of distribution of the plant and the measurement of soil acidity of Mt.Bonglae was conducted. Mt.Bonglae in Youngdo island is located in the very souty emd of Pusan which is the second largest city of Korea. The American pokeweed was distributed widely below a mountainside, that is near a village and a forest, while the plant was not found in a mountain ridge and rocky souty side. The average number of the plant per survey square was 4 which was fewer than half compare to Mt.Hwangryung. It because may be the wind from ocean dilute away the exhaustion gas of heavy traffic. There was no distinct relationship between the distribution of the American pokeweed and soil acidity of the Mt.Bonglae.

      • 흰쥐 십이지장의 점액질에 미치는 Ricin 독성에 대한 조직화학적 연구

        피마자 (Ricinus communis) 씨에서 추출한 ricin 75㎍/Kg을 복강내 투여했을 때 십이지장샘과 술잔세포에 미치는 독성을 조직화학적으로 조사하였다. 대조군에서 십이지장샘은 중성점액실만 함유하였으나 십이지장의 술잔세포는 중성점액실과 산성점액질의 혼합성이였다. 술잔세포의 산성점액질 성상은 대부분이 sulfomucin과 sialomucin의 혼합성이나 sulfomucin 또는 sialomucin만을 가진 술잔세포도 관찰되었다. 십이지장샘내에서 대조군과 ricin 투여군사이의 형태적 및 점액질성상 변화는 관찰 할 수 없었으나 융모와 장샘에서는 현저한 변화가 있었다. Ricin 투여후 24시간부터 술잔세포 중에는 커진 세포, 공포 변성된 세포, 세포막이 파괴된 세포 및 미성쑥세포 등이 마타났다. Ricin 투여후 술잔세포수는 융모에서 6시간군부터 감소하다 48시간군부터 다소 증가하였으며, 장샘에서 24시간군부터 감소하다가 120시간군부터 다소 증가하였다. Ricin 투여 후 산성점액질의 감소가 현저하였으며 특히 sulfomucin만을 분비하는 술잔세포가 감소한데 비해 sialomucin 만을 분비하는 세포는 다소 증가하였다. The toxicity of purified ricin from Ricinus communis to duodenal mucosubstances of rat was examined by histochemical method. Sprague-Dawley rats were injected intraperitoneally with 75㎍/Kg body weight of ricin, and were sacrified at intervals of 6, 24, 48 and 120 hours after injection. In the control groups, the duodenal gland contained only neutral mucosubstances, but a mixture of neutral and acidic ones was detected in the goblet cells of duodunum. About the properties of acidic mucosubstances of the goblet cells, the cells containing both sulfomucin and sialomucin were observed in most cells, but two types of cell, one producing sulfomucin and the other producing sialomucin, were also found. In the ricin injected group, the morphology and properties of mucosubstances in the duodenal gland remained the same as the control groups. However, the morphological changes in the villi and intestinal glands including enlarged, vacuolated, demolished and immature goblet cells were observed from 24 hours after injection. And a slight decrease fo goblet cells in total number was noted from 6 and 24 hours after injection in villi and crypts respectively, but reincreased from 48 hours after injection in villi and from 120 hours in crypts. With these changes, the amount of the acidic mucosubstances was significantly decreased in the goblet cells. Especially, the goblet cells secreting sulfomucin decreased in number, while those cells secreting sialomucin increased.

        흰쥐 간에 미치는 Ricin의 독성에 대한 조직학적 및 조직화학적 연구

        The toxicity of purified ricin from Ricinus communis to rats was examined by histological and histochemical methods. Sprague-Dawley rats were injected intraperitoneally with 75㎍/㎏ body weight of ricin and were sacrified at intervals of 6, 24, 48 and 120 hours after injection. The major morphological changes, such as cloudy swelling, hydropic degeneration, necrosis, fatty change, blood congestion, increase of Kupffer cells in number and extension of sinusoids, were obvious in the liver of experimental group. These morphological changes of hepatic cells were mainly observed in both the periportal and midlobular region of hepatic lobule. The extension of sinusoids was obvious in the centrolobular region. And glycogen distribution of hepatic cells tended to decrease in the same region showing morphlogical changes as compared with the control group.
