        마우스대식세포주인 RAW 264.7에서 SD-01의 항염증 활성 연구

        Objective: The aim of this study was to investigate anti-inflammatory activity of SD-01 methanol extract in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Methods: Cytotoxic activity of SD-01 methanol extract on RAW 264.7 cells was measured using 5-(3-caroboxymeth-oxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. The nitric oxide (NO) production was measured by Griess reagent system. And proinflammatory cytokines and PGE2 were measured by ELISA method. The levels of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), I k-B-alpha and nuclear NF-k B p65 expression were detected by western blot. Results: Our results indicated that methanol extract of SD-01 significantly inhibited the LPS-induced NO, PGE2 production and iNOS, COX-2 expression accompanied by an attenuation of TNF-a, IL-1b, IL-6 and MCP-1 production in RAW 264.7 cells. Moreover, methanol extract of SD-01 treatment also blocked LPS-induced NF-k B activation. Conclusion: These findings indicate that methanol extract of SD-01 inhibits the production of pro-inflammatory mediators and cytokines via suppression of NF-k B activation. Take together, these results indicate that methanol extract of SD-01 has the potential for use as an agent of anti-chronic inflammatory diseases.

        사람 간암 세포주인 HepG2에 대한 茵陳蒿湯의 항암 효과

        Objective: Hepatocellular carcinoma is the most common primary malignant tumor of the liver worldwide. In-Jin-Ho-Tang (IJHT) has been used as a traditional Chinese herbal medicine since ancient time, and today it is widely applied as a medication for jaundice which is associated with inflammation in liver. In this study, I investigated whether methanol extract of IJHT induced HepG2 cancer cell death.Methods: Cytotoxic activity of IJHT on HepG2 cells was using XTT assay. Apoptosis induction by Ros A in HCT116 cells was verified by the induction of cleavage of poly ADP-ribose polymerase (PARP), and activation of caspase-3, -8 and -9. The release of cytochrome c from mitochondria to cytosol, the level of Bcl-2 and Bax and the expression of p53 and p21 were examined by western blotting analysis. Furthermore, MAPKs activation was analyzed by western blotting analysis.Results: IJHT induced apoptosis in HepG2 cells. And treatment of IJHT resulted in the release of cytochrome c into cytosol, decreased anti-apoptotic Bcl-2, and increased pro-apoptotic Bax expression. IJHT markedly inactivated extracellular signal-regulated kinase (ERK1/2), and activated p38 mitogen-activated protein (MAP) kinase. Sodium orthovanadate (SOV), a phosphatase inhibitor, to reverse IJHT-induced ERK1/2 inactivation and SB203580, a specific p38 MAP kinase inhibitor efficiently blocked apoptosis of HepG2. Thus, IJHT induces apoptosis in HepG2 cells via MAP kinase modulation.Conclusion: These results indicated that IJHT has some potential for use as an anti-cancer agent.

        血府逐瘀湯과 그 構成藥物群이 Alloxan 糖尿 白鼠의 血淸 組成 및 抗酸化 效果에 미치는 影響

        This study was reseached to investigate the effect of Hyulbuchukeotant(?????) and its component groups on diabetes, free radical and antioxidative defense system in alloxan-induced diabetic rats, The experimental group was divided into three groups: Hyulbuchukeotang (HCT), and its components groups, Sayeoksan(H-sa) and Dohong- samultant(H-do). The results were obtained as follows: In the study of effect on diabetes, the level of glucose was considerablely reduced by HCT, H-sa and H-do with high significancy. In the change of fat metabolism, all experimental groups significantly changed the level of triglyceride, total cholesterol and HDL cholesterol expecially HCT and H-do group, but H-sa group has less significancy. In the change of protein metabolism, all experimental groups changed the level of total protein and albumin with high significancy. In the change of kidney function, HCT decreased the level of creatinine and BUN with high significancy, H-sa and H-do did with some significancy too. In the study on free radical scavenging effect in vitro, it has shown that all experimental groups have significant suppressing effect on peroxidation of linoleic acid on concentration, and in the scavenging effect of DPPH radical, inhibitory effect of superoxide in xanthine-xanthine oxidase system and inhibitory effect on lipid peroxidation reaction by hydroxy radical in H₂O₂-Fe^2+ system, all experimental groups have some activity. In the study on antioxidative defense system in vivo, HCT reduced the amount of lipid peroxide in serum and liver with high significantcy, and H-sa, H-do group did too with some significancy. In hepatic catalase activity, all experimental groups had showed activity on catalase, but H-sa no significancy. In the level of hepatic hlutathione, HCT increased the amount of glutathione significantly, but other groups with no significancy. In the activity of glutathione-s-transferase, all experimental groups has decreasing effect with significancy. These results suggest that Hyulbuchukeotant(?????) has strong effect on diabetes and peroxidative damage by free radical, so it is useful to prevent diabetes.

        대시호탕(大柴胡湯)이 alloxan으로 유발(誘發)된 고혈당(高血糖) 백서(白鼠)에 미치는 영향(影響)

        The purpose of this study was to reseach the effect of Daesihotang and its component groups on diabetes, free radical and antioxidative defense system in alloxan-induced diabetic rats. The experimental group was divided into three groups: Daesihotang (DS), Soyangyak (DS1), Yangmyungyak (DS2). The results were obtained as follows: The level of glucose, triglyceride, total cholesterol, creatinine in serum were considerablely reduced by Daesihotang. And the level of HDL cholesterol, albumin, total protein in serum were increased by Daesihotang significantly. And the level of BUN has some decreased, but with no significancy. In the study of Daesihotang on free radical scavenging effect in vitro, it has shown that Daesihotang and its components group have significant suppressing effect on peroxidation of linoleic acid on concentration. And in other experiments as scavenging effect of DPPH radical, inhibitory effect of superoxide in xanthine-xanthine oxidase system and inhibitory effect on lipid peroxidation reaction by hydroxy radical in $H_2O_2-Fe^{2+}$ system, Daesihatang have some activity, but with no significancy. In the study of Daesihotang on antioxidative defense system in vivo, the activity of GOT and GPT in serum were significantly increased; and the amounts of lipid peroxide in serum was reduced significantly but in liver no significancy. In hepatic catalase activity, Daesihotang has showed significant effect. In the level of hepatic glutathione, Daesihotang increased the amount of glutathione significantly. And in the activity of glutathione-s-transferase, Daesihotang has decreasing effect but has no significancy. These result suggest that Daesihotang has strong effect on diabetes and it is useful to prevent diabetes, but has less effect on peroxidative damage by free radical.

      • 북한의 대남전략ㆍ전술과 민족공조론 : 그 연관성에 관한 연구

        북한의 대남전략의 궁극적 목표는 '전한반도의 주체사상화와 공산사회 실현'에 있다. 1980년대 후반 이후 한반도의 통일환경은 급격한 변화를 맞이하였다. 그로인하여 북한은 생존전략 차원에서 체제유지에 주력하는 전술적 변화를 모색하게 되었다. 따라서 1998년 북한권력을 공식적으로 승계한 김정일은 심각한 체제위기의 현실을 감안하여 체제유지적이고 방어적인 입장에서 대남관계에 강경노선과 유화노선을 적절히 배합하는 실리추구의 전술적 변화를 가져왔다. 특히 2000년 남북정상회담과 6․15공동선언을 기점으로 '우리 민족끼리' 통일담론을 유포시키면서 민족공조를 강조하기 시작하였으며, 2002년 10월 2차 북핵문제로 미북관계가 긴장국면으로 접어들자 북한은 민족이라는 혈연적 동질성을 내세워 핵위기를 미국과 전조선 민족의 갈등으로 규정하고 반미․반전을 위한 민족공조를 더욱 강조하기 시작하였다. 2000년대 이후 북한이 강조하고 있는 민족공조의 기본목적은 한반도 평화통일을 위하여 민족끼리 공조한다는 것이 아니라 반미․반전을 위해 통일전선을 구축하자는 것이다. 북한은 정권 수립 이후 남한혁명전략의 수행을 위해 다양한 전술을 구사하고 있으며, 그 대표적인 전술의 하나가 통일전선전술인 것이다. 북한은 이처럼 대남전략 목표를 달성하기 위해 새로운 전술로서 민족공조를 강조하고 있는 것이다. 따라서 우리 정부는 북한의 민족공조 주장의 본질을 정확히 파악하고 대북정책을 수립하고 추구해야 할 것이다. 우리의 대북정책은 국민적 합의를 배경으로 일관성을 견지하여야 하며, 대북협상에서는 성과주의에서 탈피하여 철저한 기본원칙하에 임해야 할 것이며, 대북경제지원은 상호주의원칙하에서 이루어져야 할 것이다. The ultimate purpose of North Korean strategy is the realization of socicalization and sovereignty on Korean peninsula. They continued to build up this strategy by mixing the tough and soft pushing alternatively. Since 1980's, after 6․15 joint declaration and summit talks, Kim Jeong-Il has changed his leadership into getting practical profits on the base of mutual racial-partnership. The deep structure of this idea might be based on; 1)the realization of reunification by North Korean style, 2)to keep up their regime against economic supporting of South Korea, 3)the retreat of U.S armed forces in South Korea, 4)to draw the nuclear weapons and anti-war in issues between U.S.A and North Korea, 5)to disperse the socical conflict in South Korea. It appears that they don't want the mutual racial-partnrship but anti-U.S.A and building up their own strategic battle front. This strategy affects to South Korea people as follows: 1)the dilution of national security, 2)the deepening of conflict, 3)the issues of anti-U.S.A and the retreat of U.S armed forces on fire 4)the dilution of reunfication. To face with this strategy, we must be alert on: 1)the national consolidarity, 2)the loyalty on the base of national prosperity, 3)the economic support based on the mutual interest.

        시호사물탕 메탄올 추출물의 항산화 활성

        Sihosamultang has been used in treatment for puerperal fever, liver disease in traditional medicine. The present study was carried out to evaluate the antioxidant effects of Sihosamultang methnol extract(SME) in vitro. The inhibitory effect of SME on lipid peroxidation was examined in the linoleic acid autoxidation system. In this test, SME significantly inhibited the time course of the lipid peroxidation. And SME showed about 77% scavenging effect on DPPH radical, also exhibited about 89% inhibitory effect on lipid peroxide formation induced by hydroxyl radical derived from H₂O₂-Fe^2+ in the rat liver homogenate. And SME dose-dependently inhibited superoxide generation from xanthine-xanthine oxidase system. In addition, SME protected the hepatic cell death induced by tert-butyl hydroperoxide. These data indicated that SME might play a protective role against oxidative injury by free radicals.
