Phellinus baumii 자실체 열수 추출물의 Sarcoma-180에 대한 항암 및 면역효과

국내에서 재배되고 있는 약용버섯인 상황(P. baumii) 자실체의 열수 추출물로 부터 건강 기능성 식품 및 의약품개발의 기초자료를 제공하고자 항암실험 및 면역효과에 대한 실험을 수행하였다. 본 연구에서 사용한 PMO-P4의 경우 rDNA의 ITS부위의 염기서열 분석을 통한 계통분석 결과 P. baumii로 판명 되었으며, PMO-P4 자실체로부터 열수 추출한 성분(HWE-P4)을 160mg/kg/day의 농도로 하여 Sarcoma-180 암세포를 유발한 마우스에 경구투여한 결과 A군의 경우, 종양억제효과는 35.3%로 대조군보다 유의성 있게 감소하였으며(p<0.05), 생명연장 효과는 156%증가한 것으로 나타났다. 시료의 경구투여 방법에 있어서도 시료를 먼저 2주간 투여한 후 암세포를 접종한 A군의 경우가 시료 투여와 암세포 접종을 동시에 실시한 B군의 경우 보다 종양억제효과 및 생명연장효과가 높았음을 알 수 있었다. A군의 경우 대조군에 비하여 CD4/CD8의 비율이 71.4%증가 하였으며, CD25(IL-2receptor chain)분자의 발현을 5배정도 증가시키는 것으로 관찰되었다. 이러한 결과로 보아 P. baumii로 밝혀진 PMO-P4 자실체로부터 열수 추출한 성분의 항암효과는 암세포를 직접 공격하여 항암효과를 나타내기 보다는 T cell등의 면역세포를 활성화시킴으로써 암세포를 억제 혹은 사멸시키는 것으로 생각된다. A hot water extract(HWE-P4) was separated from the fruit bodies of PMO-P4, and its antitumor and immunomodulatory activities against sarcoma-180 in ICR mice were investigated. The internal transcribed spacer(ITS) regions from PMO-P4 were amplified using polymerase chain reaction(PCR) and sequenced. The results revealed that PMO-P4 was belong to the Phellinus baumii. When oral administration at the dose of 160mg/kg/day in the mice until the end of the experiment with 2 week's pre-feeding of the HWE-P4, the survival rate of the mice was 152% for 50days after the inoculation of sarcoma-180 and the suppression rate of the tumor growth was 35.3%(p<0.05) for 28 days after inoculation of sarcoma-180. The HWE-P4 increased 71.4% of the CD4/CD8 ratio and 5-fold of the expression of CD25(IL-2 receptor chain) compared with the control. From these results, the antitumor activity of HWE-P4 is exerted through its immunomodulating activity on the host's immune system.

The mushroom pickles were prepared as a processed mushroom product. The changes in pH, total acidity, brix, color and texture of the mushroom pickles (Lentinus edodes, Pleurotus ostreatus, Agaricus bisporus and Pleurotus eryngii) were measured during storage at 4℃. In addition, sensory evaluations on the mushroom pickles were performed. While the pH of Lentinus edodes and Pleurotus ostreatus pickles declined slowly, the pH of Agaricus bisporus and Pleurotus eryngii pickles declined rapidly during storage, The total acidity of Lentinus edodes pickles rose slowly, while the acidity of Pleurotus ostreatus, Agaricus bisporus and Pleurotus eryngii pickles rose rapidly, As for the color of mushroom pickles, "L", "a" and " b" values changed slightly. The changes in texture were determined by a rheometer, Hardness, cohesiveness, springiness, gumminess and brittleness of Lentinus edodes pickles were lower than those of the other mushroom pickles. In the sensory evaluation of mushroom pickles, Lentinus edodes showed higher acceptability ratings than the other mushrooms in taste, color, clearness, texture and overall acceptability. This study showed that among the mushrooms tested, Lentinus edodes was the most acceptable mushroom pickle for storage.

Background/Aims: We aimed to elucidate the prevalence and risk factors of HGV infection in patients with chronic renal failure undergoing hemodialysis, and to define the potential etiologic-role of HGV in the development of chronic liver disease. Methods: One-hundred and seventy-eigh patients with chronic renal failure undergoing hemodialysis were included. After extraction of RNA from serum, reverse transcription polymerase chain reaction was performed using the primers from the NS5a region. The PCR products were analyzed by liquid hybridization. Results: The prevalence was 9.0% (16/178). The prevalence rates were not statistically different according to the presence (8.5%) or abscence (10.8%) of past history of blood transfusion, patient's age or maintenance duration of hemodialysis (p$gt;0.05). Anti-HBc-positive patients (12.4%, 16/129) showed significantly higher prevalence rate (p=0.01) of HGV infection than anti-HBc-negative patients (0%, 0/49). Twelve of the 14 HGV positive patients without co-infection showed persistently normal ALT level during a year of follow-up. Conclusions: The prevalence of HGV in patients with chronic renal failure undergoing hemodialysis is significantly higher than that of volunteer blood donors. HGV may share the route of transmission with adulthood HBV infection. The etiologic-role of HGV in the development of chronic liver disease may be considerably poor. Objectives : In this study, we attempted to evaluate the effects of Careswell on human mast cell-mediated allergy inflammation in vitro and pruritogen-induced scratching behavior in vivo. Method : The Careswell was extract by distilled water. The anti-itching effects of Careswell were investigated on the compound 48/80 (50 μg/kg) or histamine (100 μg/kg) induced scratching behavior male ICR mice for 30 min by an observer blind. Terfenadine (10 mg/kg) was used as a positive control drug. The cell toxicity of Careswell was determined by 3-(4,5-dimethylthiazole-2-yl)-2, 5-diphenyl-tetrazolium bromide (MTT) assay. The regulatory effect of Careswell on interleukin (IL)-6 and tumor necrosis factor (TNF) -α levels was determined by enzyme-linked immunosorbent assay (ELISA) in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI) stimulated human mast cells (HMC-1). Also, we evaluated the effect of Careswell on PMACI induced the activation of Nuclear factor-kappa B (NF-κB) into nucleus by Western blot analysis. Result : The results revealed that the oral administration of Careswell (200 mg/kg, p.o.) attenuated the compound 48/80 or histamine-induced scratching behavior in mice. We showed that Careswell significantly reduced the PMACI-induced the production of IL-6 (0.5-1 mg/ml) and TNF-α (0.1-1 mg/ml). Additionally, Careswell significantly inhibited the activation of NF-κB in PMACI-stimulated HMC-1. Conclusion : Collectively, the findings of this study provide us with a novel insight into the pharmacological actions of Careswell as a potential molecule for use in the treatment of allergic inflammation diseases.

The ligninolytic basidiomycete, Pleurotus ostreatus K-2946, was produced a manganese peroxidase (MnP) activity when grown in liquid culture with glucose-yeast-peptone (C-Y-P) medium. However, lignin peroxidase (LiP) was not detected in this culture medium. The purification progress of MnP was purified that included chromatography on Sepharose CL-6B, Superdex 75 prep grade and Mono-Q. MnP purified by column chromatography, was 36400 dalton and a pI of 3.95. The optimal pH and temperature of the purified MnP activity were 5.0 and 55℃. The characteristics of MnP produced was quite similar to those of MnP 3 isoenzyme produced by other strains of P. ostreatus.
