<P>A pharmacokinetic interaction between oral DA-8159 and amlodipine was evaluated in male Sprague–Dawley rats. In rats pretreated with troleandomycin (a main inhibitor of CYP3A1/2 in rats), the AUC<SUB>0−6 h</SUB> of amlodipine was significantly greater than the controls (34.5±6.01 compared with 28.0±4.70 µg min/ml), indicating that amlodipine is metabolized via CYP3A1/2 in rats. It was reported that the metabolism of DA-8159 and the formation of DA-8164 (a metabolite of DA-8159) were mainly mediated via CYP3A1/2 in rats, and amlodipine significantly inhibited the CYP3A2 in rats. Therefore, a pharmacokinetic interaction between the two drugs could be expected. However, after oral administration of DA-8159 at a dose of 30 mg/kg with or without oral amlodipine at a dose of 5 mg/kg to rats, the pharmacokinetic parameters of DA-8159 and DA-8164 were not significantly different between the two groups of rats. Similar results were also obtained from amlodipine between with and without DA-8159. The above data indicated that the pharmacokinetic interaction between oral DA-8159 and amlodipine was almost negligible in rats. Copyright © 2006 John Wiley & Sons, Ltd.</P>

High-quality MgB2 thin films have been fabricated by using the laser-assisted chemical vapor deposition (LACVD) method. For the in-situ LACVD process, we used a Mg target, Mg pieces, and a mixture of 5% B2H6 gas in H2. The Mg and the B sources were obtained from pulsed laser ablation and the inlet B2H6 gas, respectively. The MgB2 thin films were grown on Al2O3 (0001) substrates at various temperatures in the range of 160 – 400 C under pressures from 100 to 200 mTorr. As-grown samples showed superconducting transition temperatures (Tc) of 27 – 40 K. We obtained the best quality MgB2 thin film with a maximum Tc of 40 K at a substrate temperature of 400 C. The residual resistivity ratio (RRR) was observed to be about 4.4, which is similar to that of single-crystalline MgB2.

<P>We found that beta-lapachone (beta-lap), a novel bioreductive drug, caused rapid apoptosis and clonogenic cell death in A549 human lung epithelial cancer cells in vitro in a dose-dependent manner. The clonogenic cell death caused by beta-lap could be significantly inhibited by dicoumarol, an inhibitor of NAD(P)H:quinone oxido-reductase (NQO1), and also by siRNA for NQO1, demonstrating that NQO1-induced bioreduction of beta-lap is an essential step in beta-lap-induced cell death. Irradiation of A549 cells with 4 Gy caused a long-lasting upregulation of NQO1, thereby increasing NQO1-mediated beta-lap-induced cell deaths. Although the direct cause of beta-lap-induced apoptosis is not yet clear, beta-lap treatment reduced the expression of p53 and NF-kappaB, whereas it increased cytochrome C release, caspase-3 activity, and gammaH2AX foci formation. Importantly, beta-lap treatment immediately after irradiation enhanced radiation-induced cell death, indicating that beta-lap sensitizes cancer cells to radiation, in addition to directly killing some of the cells. The growth of A549 tumors induced in immunocompromised mice could be markedly suppressed by local radiation therapy when followed by beta-lap treatment. This is the first study to demonstrate that combined radiotherapy and beta-lap treatment can have a significant effect on human tumor xenografts.</P>
Molecular Mechanisms of Microglial Deactivation by $TGF-{\beta}-inducible$ Protein ${\beta}ig-h3$

[ ${\beta}ig-h3$ ] is a secretory protein that is induced by $TGF-{\beta}$ and implicated in various disease conditions including fibrosis. We have previously reported that ${\beta}ig-h3$ expression is implicated in astrocyte response to brain injury. In this study, we further investigated potential roles of ${\beta}ig-h3$ protein in the injured central nervous system (CNS). We specifically assessed whether the treatment of microglial cells with ${\beta}ig-h3$ can regulate microglial activity. Microglial cells are the prime effector cells in CNS immune and inflammatory responses. When activated, they produce a number of inflammatory mediators, which can promote neuronal injury. We prepared conditioned medium from the stable CHO cell line transfected with human ${\beta}ig-h3$ cDNA. We then examined the effects of the conditioned medium on the LPS- or $IFN-{\gamma}-mediated$ induction of proinflammatory molecules in microglial cells. Preincubation with the conditioned medium significantly attenuated LPS-mediated upregulation of $TNF-{\alpha},\;IL-1{\beta}$, iNOS and COX-2 mRNA expression in BV2 murine microglial cells. It also reduced $IFN-{\gamma}-mediated$ upregulation of $TNF-{\alpha}$ and COX-2 mRNA expression but not iNOS mRNA expression. Assays of nitric oxide release correlated with the mRNA data, which showed selective inhibition of LPS-mediated nitric oxide production. Although the regulatory mechanisms need to be further investigated, these results suggest that astrocyte-derived ${\beta}ig-h3$ may contribute to protection of the CNS from immune-mediated damage via controlling microglial inflammatory responses.

Background: Frontotemporal dementia (FTD) usually presents with behavioral and psycho-logical symptoms of dementia (BPSD) rather than cognitive deficits. Thus, understanding of BPSD associated with FTD is important not only from the management perspective but also from the diagnostic viewpoint. However, few studies have investigated BPSD in patients with FTD in Korea. The aim of the current study is to describe in detail BPSD associated with FTD. Methods: Firstly, a BPSD list was designed based on the symptoms reported in previous studies of FTD, items from the Neuropsychiatric Inventory (NPI), and the review of medical records of FTD patients from our clinic including FTD patients not selected in this study. Then an interviewer asked caregivers whether the BPSDs listed had been present from onset of disease to the time of interview. Subjects who participated in the interview consisted of 23 patients (9 men and 14 women) with a mean age of 65.6 (range, 41-75) years who met the FTD criteria proposed by the Lund and Manchester group. The BPSD list was completed by face-to-face interviews or by telephone interviews with the caregivers. Results: BPSDs as classified by subscales of the NPI in the order of frequency were aberrant motor behavior (95.6%), apathy (95.6%), disinhibition (91.3%), appetite/eating change (91.3%), agitation/aggression (87%), euphoria (65.2%), anxiety (60.9%), irritability/lability (52.1%), delusions (43.5%), night-time behavior (39.1%), depression/dysphoria (34.8%), hallucinations (0%). Other symptoms not listed in the NPI were stereo-typical behaviors as stereotypy of ordering, hoarding, reading signboards, wandering a fixed route, washing, and counting, utilization behaviors, and sundowning. Conclusion: Our detailed description of BPSD associated with Korean FTD patients would help understand symptoms of FTD and develop a Korean version of BPSD questionnaire for FTD in the future studies.
Time - resolved photoluminescence spectroscopy of InGaN multiple quantum wells

We have fabricated by metal organic chemical vapor deposition (MOCVD) In_(0.13)Ga_(0.87)N/GaN multiple quantum well (MQW) with thickness as thin as 10 Å and barriers also of th same width on (0001) sapphire substrate. We have investigated this thin MQW by steady-state and time-resolved photoluminescence (PL) in picosecond time scale in a wide temperature range from 10 to 290 K. In the PL at 10 K, we observed a broad peak at 3.134 eV which was attributed to the quantum well emission of InGaN. The full width at half maximum (FWHM) of this peak was 129 meV at 10 K and its broadening at low temperatures was considered to be due to compositional fluctuations and interfacial disorder in the alloy. The narrow width of the quantum well was mainly responsible for the broadening of the emission linewidth. We also observed an intense and sharp peak at 3.471 eV of GaN barrier. From the temperature dependent PL measurements, the activation energy of the InGaN quantum well emision peak was estimated to be 69 meV. The lifetime of the quantum well emission was found to be 720 ㎰ at 10 K, which was explained in terms of the exciton localization arising from potential fluctuations.

<P><B>Abstract</B></P> <P>To identify pathogenic variants in 107 Korean patients with sporadic frontotemporal dementia (FTD), 46 genes related to FTD, amyotrophic lateral sclerosis, and other dementias were screened by next-generation sequencing. Hexanucleotide repeats in <I>C9orf72</I> gene were also tested by repeat-primed polymerase chain reaction. Next-generation sequencing revealed one known pathogenic variant (c.708+1G>A) in the <I>GRN</I> gene in a patient with behavioral variant FTD (bvFTD). In addition, a novel in-frame deletion (c.2675_2683del) in the <I>CSF1R</I> gene was identified in a patient with bvFTD who had severe bifrontal atrophy with frontal subcortical white matter changes. Novel compound heterozygous variants in the <I>AARS2</I> gene, c.1040+1G>A and c.636G>A (p.Met212Ile), were found in a patient with bvFTD. Forty-six variants of uncertain significance were detected in other patients. None of the patients had expanded hexanucleotide repeats in <I>C9orf72</I>. These results show that pathogenic variants of known FTD genes are rare in Korean FTD patients but the <I>CSF1R</I> and <I>AARS2</I> genes should be screened for a genetic diagnosis of FTD or other dementias.</P>

Transfusion related acute lung injury (TRALI) is a serious, potentially life-threatening complication of transfusion therapy that is sometimes under diagnosed and under reported. Patients with TRALI present with dyspnea/respiratory distress and fever. The symptoms, signs and chest radiological findings in TRALI are similar to transfusion associated circulatory overload, which makes it is difficult to distinguish it from circulatory overload. Although the mortality rate in cases of TRALI is relatively low, TRALI is the third most common cause of fatal transfusion reactions next to ABO blood type incompatibility and hepatitis. Mild-to-moderate cases of TRALI may be misdiagnosed as volume overload. Recently, we encountered two cases where the patients suffered from dyspnea and fever after a transfusion. and review of the relevant literature. (Tuberc Respir Dis 2006; 61: 473-478)
