        Adiponectin Regulates Bone Marrow Mesenchymal Stem Cell Niche Through a Unique Signal Transduction Pathway: An Approach for Treating Bone Disease in Diabetes : Adiponectin Regulates BMSC Niche

        <P>Adiponectin (APN) is an adipocyte-secreted adipokine that exerts well-characterized antidiabetic properties. Patients with type 2 diabetes (T2D) are characterized by reduced APN levels in circulation and impaired stem cell and progenitor cell mobilization from the bone marrow for tissue repair and remodeling. In this study, we found that APN regulates the mobilization and recruitment of bone marrow-derived mesenchymal stem cells (BMSCs) to participate in tissue repair and regeneration. APN facilitated BMSCs migrating from the bone marrow into the circulation to regenerate bone by regulating stromal cell-derived factor (SDF)-1 in a mouse bone defect model. More importantly, we found that systemic APN infusion ameliorated diabetic mobilopathy of BMSCs, lowered glucose concentration, and promoted bone regeneration in diet-induced obesity mice. In vitro studies allowed us to identify Smad1/5/8 as a novel signaling mediator of APN receptor (AdipoR)-1 in BMSCs and osteoblasts. APN stimulation of MC3T3-E1 osteoblastic cells led to Smad1/5/8 phosphorylation and nuclear localization and increased SDF1 mRNA expression. Although APN-mediated phosphorylation of Smad1/5/8 occurred independently from adaptor protein, phosphotyrosine interaction, pleckstrin homology domain, and leucine zipper containing 1, it correlated with the disassembly of protein kinase casein kinase 2 and AdipoR1 in immunoprecipitation experiments. Taken together, this study identified APN as a regulator of BMSCs migration in response to bone injury. Therefore, our findings suggest APN signaling could be a potential therapeutic target to improve bone regeneration and homeostasis, especially in obese and T2D patients.</P>

        Enhanced recovery after surgery strategy for cirrhosis patients undergoing hepatectomy: experience in a single research center

        Purpose: To evaluate the safety and effectiveness of an enhanced recovery after surgery (ERAS) programme after curative liver resection in cirrhotic hepatocellular carcinoma (HCC) patients. Methods: One hundred sixty-two patients were enrolled in the study; 80 patients whose data were collected prospectively were assigned to the ERAS group, and 82 patients whose data were collected retrospectively were assigned to the control group. Preoperative clinicopathologic factors, surgical factors, and postoperative outcomes of the 2 groups were compared. Logistic regression was applied to explore potential predictors of hospital stay and morbidity. Results: The postoperative hospital stay, postoperative complication rate, and recovery of liver function on postoperative day 5 seemed to be better in the ERAS group. The composition of complications was different in the 2 groups; pleural effusion and postoperative ascites were more common in the control group, and indocyanine green retention at 15 minutes, operation time, preoperative alanine aminotransferase, and number of liver segmentectomies were associated with postoperative complications rather than ERAS intervention. Conclusion: The ERAS programme is safe and effective for HCC patients with chronic liver disease undergoing hepatectomy, but it seems that surgical factors, such as operation type, have a greater impact on morbidity than other factors. Operative characteristics such as the method of blood loss control and the volume of liver resection should be augmented into ERAS protocol of hepatectomy.

        Macroscopic and Mesoscopic Characteristics of a Small-Span Metro Tunnel in the Development of a Disaster Under Load

        A similarity model with a volumetric similarity ratio of 1:100 and a granular flow model for the tunnel were designed. By comparing macroscopic and mesoscopic information (such as fracture process, strain evolution, stress transfer, crack propagation, and stress distribution) of the tunnel models under load, the failure mechanism of the metro tunnel under load was investigated. The result showed that: 1) under the loading path, the instability area of the tunnel is mainly distributed in the straight wall on both sides. When the load is 1.5 MPa, a large number of cracks on both sides of the straight wall run through, resulting in the initial failure of the rock mass; 2) the surface rock mass of arch bottom is under tensile stress and the deep rock mass is under pressure stress, therefore, the fracture does not develop continuously. The surface of straight wall produces continuous development crack under the action of tensile stress; 3) the arch bottom first responds during the stress redistribution of the small-span tunnel; the top and middle parts of the side walls of the running tunnel with greatest potential for damage respond most; 4) in the process of stress redistribution, the peak stress of the deep measuring points of the straight wall is greater than that of the free surface; 5) at the initial stage of loading, tensile cracks account for a high proportion of all cracks found. When the load is 1.5 MPa, the proportion of shear cracks increases to 28%, and when the load is 1.6 Mpa, the proportion of shear cracks increases to 31%. Finally, the tensile-shear effect triggers the failure of the tunnel.

        Changes of Collagen in Sea Cucumber (Stichopus japonicas) During Cooking

        Changes of the collagen in sea cucumber (Stichopus japonicas) during cooking were investigated. Crude collagen fibers (CCF) is more sensitive to heat than pepsin-solubilized collagen (PSC), absorbance at 226-232nm increased from 60 to 100^oC. PSC nearly completely degraded after cooking for 8-10 h, 4-6 h, 1-1.5 h, 40-50 min, and 10-20 min at 60, 70, 80, 90, and 100^oC,respectively. Collagen fiber shrinkage, disappearance of periodic cross striation, complete denaturation, and dispersion of denatured fibers at 40, 60, 80 and 100^oC,respectively, were demonstrated by transmission electron microscope (TEM). Above results might be instructional for sea cucumber processing and collagen usage.

        Solution-processed WOx hole injection layer for efficient fluorescent blue organic light-emitting diode

        Solution-processed tungsten oxide (s-WOx) interfacial layer for efficient hole injection in fluorescent blue organic light-emitting diode (OLED) is demonstrated. The OLED using 2-methyl-9,10-bis(naphthalen-2- yl)anthracene (MADN) as emitter shows luminous efficiency of 3.3 cd/A, power efficiency of 2.5 lm/W and external quantum efficiency of 4.6% with Commission Internationale d'Eclairage (CIE) color coordinates of (0.154, 0.102). Using MADN doped 1-4-di-[4-(N,N-diphenyl)amino]styryl-benzene as emitter, luminous efficiency of 10.8 cd/A, power efficiency of 6.4 lm/W and external quantum efficiency of 7.2% with CIE color coordinates of (0.167, 0.283) are achieved. Atomic force microscopy and X-ray photoelectron spectroscopy show that s-WOx features superior film morphology and non-stoichiometry with slight oxygen deficiency. Current-voltage characteristics and impedance spectroscopy analysis indicate that s-WOx behaves slightly enhanced hole injection and accordingly contributes to improved device performance in comparison with conventional vacuum thermal evaporation WOx. Our results pave an alternative way for broadeningWOx application with solution process and advancing fluorescent blue OLEDs.

        Family History, Tobacco Smoking, and Risk of Ischemic Stroke

        Background and Purpose Both genetic factors and smoking are associated with ischemic stroke (IS) risk. However, little is known about the potential interaction of these factors. We aimed to assess whether smoking and a positive family history interact to increase the risk of IS. Methods The nationwide prospective study recruited 210,000 men and 300,000 women in 2004 to 2008 at ages 30 to 79 years. During 9.7 years of follow-up, we documented 16,923 and 20,656 incident IS cases in men and women without major chronic diseases at baseline, respectively. Multivariable Cox regression models were used to examine associations between family history and IS. Likelihood ratio tests were used to test the smoking-family history interactions on IS. Results About 67.8% (n=135,168) of men ever smoked regularly compared with 2.7% (n=7,775) of women. Among men, a significant interaction between family history and smoking on IS was observed (P for interaction=0.03), with more pronounced association between family history and IS among ever-regular smokers (hazard ratio [HR], 1.21; 95% confidence interval [CI], 1.16 to 1.27) than among never-smokers (HR, 1.11; 95% CI, 1.01 to 1.23). The association between family history and IS among ex-smokers after more than 10 years of cessation (HR, 1.01; 95% CI, 0.85 to 1.20) appeared similar to that among never-smokers. Among women, a similar but not significant interaction between family history and smoking on IS was observed. Ever-regular smokers who had a family history of stroke had the highest risk of IS. Conclusions Among Chinese men, the association of family history with IS was accentuated by smoking, and such accentuation tended to be lowered by cessation.

        A Slot Allocated Blocking Anti-Collision Algorithm for RFID Tag Identification

        In many Radio Frequency Identification (RFID) applications, the reader recognizes the tags within its scope repeatedly. For these applications, some algorithms such as the adaptive query splitting algorithm (AQS) and the novel semi-blocking AQS (SBA) were proposed. In these algorithms, a staying tag retransmits its ID to the reader to be identified, even though the ID of the tag is stored in the reader`s memory. When the length of tag ID is long, the reader consumes a long time to identify the staying tags. To overcome this deficiency, we propose a slot allocated blocking anti-collision algorithm (SABA). In SABA, the reader assigns a unique slot to each tag in its range by using a slot allocation mechanism. Based on the allocated slot, each staying tag only replies a short data to the reader in the identification process. As a result, the amount of data transmitted by the staying tags is reduced greatly and the identification rate of the reader is improved effectively. The identification rate and the data amount transmitted by tags of SABA are analyzed theoretically and verified by various simulations. The simulation and analysis results show that the performance of SABA is superior to the existing algorithms significantly.

        Self-Rated Health Status and Risk of Incident Stroke in 0.5 Million Chinese Adults: The China Kadoorie Biobank Study

        Background and purpose Self-rated health (SRH) is a consistent and strong predictor of all-cause and cardiovascular mortality in various populations. However, the associations between SRH measures and risk of first-ever or recurrent stroke were rarely explored. We thus aim to prospectively investigate the associations between SRH measures and risk of total and subtypes of stroke in Chinese population. Methods A total of 494,113 participants from the China Kadoorie Biobank without prior heart diseases or cancer (486,541 without stroke and 7,572 with stroke) were followed from baseline (2004 to 2008) until December 31, 2013. General and age-comparative SRH were obtained from baseline questionnaires. First-ever stroke or recurrent events were ascertained through linkage to disease registry system and health insurance data. Results We identified 27,662 first-ever stroke and 2,909 recurrent events during an average of 7.0 years of follow-up. Compared with excellent general SRH, the hazard ratios (HRs) and 95% confidence intervals (CIs) for first-ever stroke associated with good, fair, and poor general SRH were 1.04 (1.00 to 1.08), 1.19 (1.15 to 1.23), and 1.49 (1.42 to 1.56) in the multivariate model, respectively. Compared with better age-comparative SRH, the HRs (95% CIs) of same and worse age-comparative SRH were 1.13 (1.10 to 1.17) and 1.51 (1.45 to 1.58), respectively. The relations of SRH measures with ischemic stroke, hemorrhagic stroke, and recurrent stroke were similar to that with total first-ever stroke. However, the magnitude of associations was much stronger for fatal stroke than for non-fatal stroke. Conclusions This large-scale prospective cohort suggests that self-perceived health status is associated with incident stroke, regardless of stroke subtype.

        Family History and Stroke Risk in China: Evidence from a Large Cohort Study

        Background and Purpose Large cohort studies on relationship between family history of stroke(FHS) and stroke risk are lacking in Asians. We aimed to systematically evaluate the association ofFHS with stroke risk in a cohort study of 0.5 million Chinese adults. Methods Information about FHS was self-reported. The median follow-up time was 7.16 years andthe end-point of follow-up was incident stroke, which was entered directly into the China KadoorieBiobank system. Multivariate analyses were performed with Cox proportional hazards model, andinteraction analyses were carried using likelihood-ratio tests. Results Compared with participants without FHS, the hazard ratio (HR) (95% confidence interval,CI) of stroke for participants with FHS was 1.50 (1.46-1.55). The HRs increased with the number offirst degree relatives with stroke (HRs=1.41, 1.98 and 2.47 for 1, 2 and ≥3 relatives, respectively,Ptrend <0.001). The HRs were 1.57 (95% CI: 1.50-1.66) and 1.49 (95% CI: 1.45-1.54) for siblinghistory and parental history, respectively. Similar associations with offspring stroke risk wereobserved between paternal history (HR=1.48, 95% CI: 1.43-1.54) and maternal history (HR=1.49,95% CI: 1.43-1.55). Moreover, significant interactions were detected between FHS and health-riskbehaviors (tobacco smoking and alcohol drinking). Conclusions FHS is an independent risk factor for stroke in Chinese. The more first degree relativesare affected by stroke, the higher are individuals’ risk of suffering from stroke. The management ofthe health-risk behaviors for reducing stroke should be highlighted, especially for the individualswith FHS.
