Myocilin as a Potential Factor for the Fast Type Change in the Muscle Isoform Composition: A Mini Review

        Yunsuk Choi(Yunsuk Choi),Hyunseok Jee(Hyunseok Jee) 대한운동학회 2023 아시아 운동학 학술지 Vol.25 No.1

BACKGROUND Various factors-based properties of the skeletal muscle are determined by mechanical stress, chemical stimuli, neuromuscular stimuli, different protein isoforms etc. The properties of the muscle are formed in the process of the balance between anabolism and catabolism. Especially, the anabolism on the way of the cell proliferation to its differentiation is accompanied by various controlling signal pathways. OBJECTIVE The purpose of this study focused on the mTOR/S6K system, Myosin, and Myocilin among these signaling pathways, especially on previously reported studies on the contribution of Myocilin to the skeletal muscle fiber type transformation. METHODS To identify the molecular pathways related to muscle development, the KEGG Mapper was used to present the mTOR-related signaling system that has not been identified so far. RESULTS Consequently, it suggests that it affects the myogenic process, through the path of PI3K->AKT- >MEF2->MyoC->mTOR->p70S6K->S6. CONCLUSIONS It also suggests that Myocilin serves as a potential effector to control skeletal muscle cell type. Furthermore, more detailed study on the potential target-oriented (to the fast or slow fiber type change) role of Myocilin for enhancing the health state by adjusted exercise programs developed by its optimal type, intensity, frequency, and time is awaited.

      • Treatment of Multiple Dural Arteriovenous Fistulas Associated with Visual Disturbances in a Chronic Schizophrenia Patient: A Case Report

        Yunsuk Choi,Moon Jong Un,Cheolsu Jwa 대한말초신경학회 2018 The Nerve Vol.4 No.1

A 51-year-old woman presented with visual disturbances that had persisted for approximately one year. She had been taking psychiatric medication for schizophrenia. Brain magnetic resonance imaging showed engorged vessels in both hemispheres, and a delay in the mean transit time and time to peak was observed on perfusion images. The initial transfemoral cerebral angiography showed multiple dural arteriovenous fistulas (AVFs) with cortical venous reflux and venous ectasia. Both middle meningeal arteries (MMAs), superficial temporal arteries (STAs), and occipital arteries (OAs) were feeding the fistulas, and the superior sagittal sinus was the draining vein. Owing to the presence of multiple lesions and feeder arteries, a staged treatment was planned. We first performed the Onyx embolization of the left MMA, followed by the surgical ligation of the left STA, with subsequent embolization of the left OA. Finally, the venous ectasia was clipped. Importantly, after treatment, the patient’s schizophrenia-like symptoms improved. The patient could be originally misdiagnosed with schizophrenia. This case shows that staged and active treatment should be considered in case of multiple dural AVFs for improvement of symptoms.

A Type 1 Persistent Proatlantal Artery Originating from the External Carotid Artery Detected by Computed Tomographic Angiography

Yunsuk Choi,Sang Bong Chung,Myoung Soo Kim 대한뇌혈관외과학회 2018 Journal of Cerebrovascular and Endovascular Neuros Vol.20 No.4

A persistent proatlantal artery (PA) is rare. We report a type 1 persistent PA originating from the right external carotid artery (ECA). A 78-year-old woman presented with dizziness. Computed tomographic (CT) angiography showed a persistent PA originating from the right ECA. This persistent PA did not pass through the atlas transverse foramen. The extracranial segment of this artery in the atlas transverse process level had a more lateral position than a normal left vertebral artery. CT angiography well demonstrated the relationship with bony structures and the course of this persistent PA. This anomalous artery in our patient presented as an incidental finding. Surgeon should recognize a persistent PA when performing carotid endarterectomy or ligation of the ECA for avoidance of complication.

Prevalence and Anatomy of Aberrant Right Subclavian Artery Evaluated by Computed Tomographic Angiography at a Single Institution in Korea

Choi, Yunsuk,Chung, Sang Bong,Kim, Myoung Soo The Korean Neurosurgical Society 2019 Journal of Korean neurosurgical society Vol.62 No.2

Objective : Aberrant right subclavian artery (ARSA) is a rare anatomical variant of the origin of the right subclavian artery. ARSA is defined as the right subclavian artery originating as the final branch of the aortic arch. The purpose of this study is to determine the prevalence and the anatomy of ARSA evaluated with computed tomography (CT) angiography. Methods : CT angiography was performed in 3460 patients between March 1, 2014 and November 30, 2015 and the results were analyzed. The origin of the ARSA, course of the vessel, possible inadvertent ARSA puncture site during subclavian vein catheterization, Kommerell diverticula, and associated vascular anomalies were evaluated. We used the literature to review the clinical importance of ARSA. Results : Seventeen in 3460 patients had ARSA. All ARSAs in 17 patients originated from the posterior aspect of the aortic arch and traveled along a retroesophageal course to the right thoracic outlet. All 17 ARSAs were located in the anterior portion from first to fourth thoracic vertebral bodies and were located near the right subclavian vein at the medial third of the clavicle. Only one of 17 patients presented with dysphagia. Conclusion : It is important to be aware ARSA before surgical approaches to upper thoracic vertebrae in order to avoid complications and effect proper treatment. In patients with a known ARSA, a right transradial approach for aortography or cerebral angiography should be changed to a left radial artery or transfemoral approach.

Treatment outcome and prognostic factors of Korean patients with chronic lymphocytic leukemia: a multicenter retrospective study

        ( Yunsuk Choi ),( Jung-hee Lee ),( Chul Won Jung ),( Jae-cheol Jo ),( Jin Seok Kim ),( Inho Kim ),( Silvia Park ),( June-won Cheong ),( Sang-hyuk Park ),( Sung-yong Kim ),( Hong-ghi Lee ) 대한내과학회 2021 The Korean Journal of Internal Medicine Vol.36 No.1

Background/Aims: Compared with Western countries, chronic lymphocytic leukemia (CLL) rarely occurs in Asia and has different clinical characteristics. Thus, we aimed to evaluate the clinical characteristics, treatment outcomes, and prognostic significance of Korean patients with CLL. Methods: We retrospectively analyzed 90 patients with CLL who had received chemotherapy at 6 centers in Korea between 2000 and 2012. Results: Compared with Western patients with CLL, Korean patients with CLL express lambda (42.0%) and atypical markers such as CD22 and FMC7 (76.7% and 40.0%, respectively) more frequently. First-line chemotherapy regimens included chlorambucil (n = 43), fludarabine and cyclophosphamide (FC) (n = 20), fludarabine (n = 13), rituximab-FC (n = 4). The remaining patients were treated with other various regimens (n = 10). The 5-year overall survival (OS) and progression-free survival (PFS) rates were 79.3% and 28.1%, respectively. Multivariate analyses showed that hyperleukocytosis (≥ 100 × 103/μL), extranodal involvement, and the Binet C stage were significant negative prognostic factors for OS (hazard ratio [HR] 4.75, p = 0.039; HR 21.6, p = 0.002; and HR 4.35, p = 0.034, respectively). Cytogenetic abnormalities including complex karyotypes (≥ 3), del(11q), and del(17) had a significantly adverse impact on both OS and PFS (p < 0.001 and p = 0.010, respectively). Conclusions: Initial hyperleukocytosis, extranodal involvement, complex karyotype, del(17) and del(11q) need to be considered in the risk stratification system for CLL.

Inhibition of DNMT3B and PI3K/AKT/mTOR and ERK Pathways as a Novel Mechanism of Volasertib on Hypomethylating Agent-Resistant Cells

        Choi Eun-Ji,Koo Bon-Kwan,Hur Eun-Hye,Moon Ju Hyun,Kim Ji Yun,Park Han-Seung,Choi Yunsuk,Lee Kyoo-Hyung,Lee Jung-Hee,Choi Eun Kyung,Lee Je-Hwan 한국응용약물학회 2023 Biomolecules & Therapeutics(구 응용약물학회지) Vol.31 No.3

Resistance to hypomethylating agents (HMAs) in myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML) is a concerning problem. Polo-like kinase 1 (PLK1) is a key cell cycle modulator and is known to be associated with an activation of the PI3K pathway, which is related to the stabilization of DNA methyltransferase 1 (DNMT1), a target of HMAs. We investigated the effects of volasertib on HMA-resistant cell lines (MOLM/AZA-1 and MOLM/DEC-5) derived from MOLM-13, and bone marrow (BM) samples obtained from patients with MDS (BM blasts >5%) or AML evolved from MDS (MDS/AML). Volasertib effectively inhibited the proliferation of HMA-resistant cells with suppression of DNMTs and PI3K/AKT/mTOR and ERK pathways. Volasertib also showed significant inhibitory effects against primary BM cells from patients with MDS or MDS/AML, and the effects of volasertib inversely correlated with DNMT3B expression. The DNMT3B-overexpressed AML cells showed primary resistance to volasertib treatment. Our data suggest that volasertib has a potential role in overcoming HMA resistance in patients with MDS and MDS/ AML by suppressing the expression of DNMT3 enzymes and PI3K/AKT/mTOR and ERK pathways. We also found that DNMT3B overexpression might be associated with resistance to volasertib.

      • A review of vanadium electrolytes for vanadium redox flow batteries

        Choi, Chanyong,Kim, Soohyun,Kim, Riyul,Choi, Yunsuk,Kim, Soowhan,Jung, Ho-young,Yang, Jung Hoon,Kim, Hee-Tak Elsevier 2017 RENEWABLE & SUSTAINABLE ENERGY REVIEWS Vol.69 No.-

Abstract

There is increasing interest in vanadium redox flow batteries (VRFBs) for large scale-energy storage systems. Vanadium electrolytes which function as both the electrolyte and active material are highly important in terms of cost and performance. Although vanadium electrolyte technologies have notably evolved during the last few decades, they should be improved further towards higher vanadium solubility, stability and electrochemical performance for the design of energy-dense, reliable and cost-effective VRFBs. This timely review summarizes the vanadium electrolyte technologies including their synthesis, electrochemical performances, thermal stabilities, and spectroscopic characterizations and highlights the current issues in VRFB electrolyte development. The challenges that must be confronted to further develop vanadium electrolytes may stimulate more researchers to push them forward.

      • Prognostic implications of CD14 positivity in acute myeloid leukemia arising from myelodysplastic syndrome.

        Choi, Yunsuk,Lee, Je-Hwan,Kim, Sung-Doo,Kim, Dae-Young,Lee, Jung-Hee,Seol, Miee,Kang, Young-Ah,Jeon, Mijin,Jung, Ah Rang,Lee, Kyoo-Hyung Elsevier Science Publishers 2013 International journal of hematology Vol.97 No.2

Secondary acute myeloid leukemia (s-AML) arising from myelodysplastic syndrome (MDS) shows different clinical features from de novo AML. We assessed the prognostic significance of immunophenotypic markers in patients with s-AML arising from MDS. Sixty-five adults diagnosed with AML arising from MDS between 1996 and 2010 were retrospectively analyzed. Immunophenotyping was performed for markers including CD3, CD7, CD10, CD13, CD14, CD19, CD33, CD34, CD41, CD45, CD56, CD65, CD117, HLA-DR, and TdT. Of these immunophenotypic markers, only CD14 positivity was significantly associated with lower complete remission rate (P = 0.034) and significantly shorter overall survival (OS, P < 0.001) and event-free survival (EFS, P < 0.001) on univariate analysis. On multivariate analysis, these differences remained significant in terms of OS [hazard ratio (HR) 4.49; P < 0.001] and EFS (HR 4.06; P < 0.001). Other significant prognostic variables included age 60 years [shorter OS (P = 0.003) and EFS (P = 0.020)], higher WBC count (>60,000/μL) [shorter OS (P < 0.001) and EFS (P = 0.001)], and poor cytogenetic risk group [shorter OS (P = 0.005)]. CD14 expression on leukemic blasts is an independent prognostic factor for survival outcomes in patients with AML arising from MDS.

Expression and prognostic significance of microR-NAs in Korean patients with myelodysplastic syndrome

        ( Yunsuk Choi ),( Eun-hye Hur ),( Ju Hyun Moon ),( Bon-kwan Goo ),( Dae Ro Choi ),( Je-hwan Lee ) 대한내과학회 2019 The Korean Journal of Internal Medicine Vol.34 No.2

Background/Aims: Various alterations of microRNA (miRNA) expression have been reported in myelodysplastic syndrome (MDS). We aimed to investigate the unique patterns and prognostic significance of miRNA expression in Korean patients with MDS. Methods: Bone marrow mononuclear cells were collected from eight healthy controls and 26 patients with MDS, and miRNAs were isolated and assessed via quantitative real-time polymerase chain reaction for selected miRNAs, including miR-21, miR-124a, miR-126, miR-146b-5p, miR-155, miR-182, miR-200c, miR-342- 5p, miR-708, and Let-7a. Results: MiR-124a, miR-155, miR-182, miR-200c, miR-342-5p, and Let-7a were significantly underexpressed in patients with MDS, compared to healthy controls. MiR-21, miR-126, 146b-5p, and miR-155 transcript levels were significantly lower in international prognostic scoring system lower (low and intermediate-1) risk MDS than in higher (intermediate-2 and high) risk MDS. Higher expression levels of miR-126 and miR-155 correlated with significantly shorter overall survival and leukemia-free survival. Higher miR-124a expression also tended to be related to shorter survivals. Conclusions: Although our study was limited by the relatively small number of patients included, we identified several miRNAs associated with pathogenesis, leukemic transformation, and prognosis in MDS.
