        Genome-Wide Association Analysis to Search for New Loci Associated with Lifelong Premature Ejaculation Risk in Chinese Male Han Population

        Purpose: Genetic factors play an indispensable role in the pathogenesis of lifelong premature ejaculation (LPE). The suscep-tibility genes/SNPs that have been discovered are very limited and can only explain part of the genetic effects of LPE. There-fore, discovering more genetic polymorphisms associated with the occurrence and development of LPE will help reveal the pathogenesis of LPE. Materials Materials and and Methods:Methods: We conducted a genome-wide association study of LPE in 486 Chinese male Han people (cases and controls). We used Gene Titan multi-channel instrument and Axiom Analysis Suite 6.0 software for genotyping. Imputation was performed by IMPUTE2 software and the 1000 Genomes Project (Phase3) was used as reference for haplotype. Finally, logistic regression analysis was performed on all loci that passed the quality control. The odds ratio and 95% confidence in-terval were calculated to determine the association between each SNPs and Chinese male Han population LPE risk. Results:Results: The results showed that a total of 33 genetic variants in 13 genes (LACTBL1 , SSBP3 , ACOT11 , LINC02486 , TMEM154, LINC01098, NONE, HCG27, HLA-C, TNFSF8, TNC, FAM53B, SULF2) have a suggestively significant genome-wide association with LPE risk (p<5×10-6). Conclusions:Conclusions: This study is the first to conduct a GWAS on LPE in Chinese male Han population 33 genetic polymorphisms have a suggestive genome-wide association with LPE risk. This study have provided data supplement for the genetic loci of LPE risk, and laid a scientific foundation for the pathogenesis and the targeted therapy of LPE.

        Numerical Simulation of the Twist Deformation for the Stiffened Panel with Laser Forming

        In the laser forming process of the panel with crossed reinforcing bars (PWCRB), a kind of stiffened panel, twisting of the panel occurred when it was formed under the integral continuous scanning strategy (ICSS). The twisting was restrained obviously when the middle stiffener was heated. In order to reveal the twisting mechanism, a FE model was established and corresponding simulations were conducted. Furthermore, an equivalent model of the sectional structure was established, and a model of the buckling critical load of the stiffener was established based on principle of minimum potential energy. It revealed that the buckling critical load of the middle stiffener was smaller than that of the marginal stiffener. The simulation results showed that the stiffeners suffered from pressure and bending moment during the forming process, which tended to induce the buckling of the stiffeners. Besides, the residual stress of the middle stiffener was larger than that of the marginal stiffener. The bending angle of the middle stiffener increased when it was heated. Besides, the lateral-torsional deformation of the middle stiffener decreased and the twisting of the panel was restrained. It demonstrated that the twisting phenomenon of the PWCRB was caused by the middle stiffener, which suffered from lateral-torsional buckling when the panel was formed with the ICSS.

        Genetic Relationships Among Panicle Characteristics of Rice (Oryza sativa L.) Using Unconditional and Conditional QTL Analyses

        Using mixed-model-based composite interval mapping and conditional statistical methods, we studied quantitative trait loci (QTLs) with epistatic effects and QTLs by environment interaction effects for rice seed set percent (SSP), filled grain number per plant (FGP), and panicle length (PL). A population of 241 recombinant inbred lines was used which was derived from a cross between “Zhenshan 97” and “Minghui 63.” Its linkage map included 221 molecular markers. Our QTL analysis detected 28, 25, and 32 QTLs for SSP, FGP, and PL, respectively. Each QTL explained 1.37%∼13.19% of the mean phenotypic variation. A comparison of conventional and conditional mapping provided information about the genetic control system involved in the synthetic process of SSP, FGP, and PL at the level of individual QTLs. Conditional QTLs with reduced (or increased) effects were identified for SSP, which were significantly influenced by FGP or PL. Some QTLs could express independently for the given traits, thereby providing possibilities for simultaneous improvement of SSR and PL, and SSR and FGP. Epistasis was more sensitive to environmental conditions than were additive effects.

        Fatigue Evaluation of Cable-stayed Bridge Steel Deck Based on Predicted Traffic Flow Growth

        Statistical analysis of the measured traffic flow over the Nanjing No. 3 Yangtze River Bridge from 2006 to 2010 was conducted. The predictions of traffic flow growth model were established. Random traffic flow was simulated by Monte-Carlo method to study dynamic impact effects under vehicle load. Considering the dynamic impact effects and wheel mark transverse distribution, the nominal stress, hot spot stress, and notch stress in the steel bridge deck details was calculated. The results of three evaluation method were compared. Suggestions of analysis models and fatigue evaluation were proposed. The study showed that the maximum magnification coefficient of the dynamic impact effects was close to the suggestion of BS5400. The simplified model could be used to analysis local stress ignoring the superimposed effects of longitudinal multi-vehicles. The evaluated service life under hot spot stress was comparatively longer. In terms of bridge safety and convenience, notch stress for the roof weld and U rib-to-diaphragm weld and hot spot stress for diaphragm arc gap were more suitable for fatigue evaluation. And the roof weld was most likely to crack.

        Effective leaching and extraction of valuable metals from electrode material of spent lithium-ion batteries using mixed organic acids leachant

        The present work focuses on simultaneous recycling of Li and Co from crushed products of mixedelectrode materials using mixed organic acids, in which benzenesulfonic acid and formic acid werecooperatively used as the leaching reagents. Results show that the optimal leaching efficiency of 97% Coand 99% Li were obtained under the conditions of 1.3 mol/L benzenesulfonic acid, 1.5 mol/L formic acid, asolid to liquid (S/L) ratio of 30 g/L, and 40 min reaction time at 50 C. Meanwhile, the leaching of Li and Cofits well to logarithmic rate model with apparent activation energy of 32.7 and 47.0 kJ/mol in this givenleaching system, respectively. Besides, cobalt was directly recovered from the leach liquor as pure cobaltbenzene sulfonic with the recovery efficiency of 99%, and lithium can be entirely precipitated by addingphosphoric acid. Further, the reaction mechanism involves the leaching-hydrating-complexing model ofLiCoO2 particles was proposed based on the dissolution behavior of metals and then verified bymorphological and phase characterization (i.e. FT-IR, XRD and SEM-EDS) of the recycling product. Thewhole process is found to be effective and sustainable for recovery of Li, Co and graphite from mixedindustrial crushing product of spent LIBs.

        Analysis of methylation-sensitive amplified polymorphism in different cotton accessions under salt stress based on capillary electrophoresis

        A methylation-sensitive amplification polymorphism method based on capillary electrophoresis was used to analyze DNA methylation levels in three cotton accessions, two salt-tolerant accessions CCRI 35 and Zhong 07 and one salt-sensitive accession CCRI 12. Many categories of DNA methylation happened in the three cotton accessions under salt treatment, including hypermethylation, hypomethylation, and other patterns. Hypermethylation happened at a significantly higher rate than that of hypomethylation in salt-tolerant accessions CCRI 35 and Zhong 07. On the contrary, in salt-sensitive accession CCRI 12, hypomethylation happened at a significantly higher rate than that of hypermethylation. In general, the global DNA methylation level significantly increased under salt stress in both salt-tolerant accessions CCRI 35 and Zhong 07, whereas there was no significant difference in the salt-sensitive accessions CCRI 12. Our results suggested that salt-tolerant cotton might have a mechanism of increasing the methylation level when responding to salt stress; the increase of the global level of DNA methylation and also different methylation patterns might play important roles in tolerance to salt stress in cotton. Some interesting genes were found through cloning and analysis of differently methylated DNA sequences, which might contribute to salt tolerance in cotton.

      • Association Between MDM2 SNP309 T>G and Risk of Gastric Cancer: A Meta-analysis

        Background: As a negative regulator of P53, MDM2 plays an important role in carcinogenesis; a polymorphism in its promoter region. SNP309 T>G, is known to increase the expression of MDM2, thus being considered related to higher susceptibility to neoplasia. However, no agreement has been achieved regarding its effects on gastric cancer. Methods: The present systematic meta-analysis was performed based on comprehensive literature search from Pubmed, Web of science and CBM databases. Results: It was suggested from 6 independent studies that the GG genotype is associated with a significantly increased risk of gastric cancer (Recessive: OR = 1.43, 95% CI = 1.08-1.91, P = 0.013), and subgroup analysis also confirmed the relationship (English publications-recessive model: OR = 1.45, 95% CI = 1.10-1.91, P = 0.009; Studies in China-recessive model: OR = 1.58, 95% CI = 1.08-2.30, P = 0.017). No publication bias was detected. Conclusion: The meta-analysis indicated a significant inverse association between GG genotype carriage and elevated risk of gastric cancer. However, more studies and detailed information are needed to fully address the topic.

        Long non-coding RNA RP11-6O2.4 indicates poor prognosis and suppresses cell cycle progression through the p38-MAPK signaling pathway in gastric cancer

        Backgrounds: The role of long non-coding RNAs (lncRNA) in gastric cancer (GC) has been highlighted in studies conducted over the past decade. However, the potential clinical value and the mechanisms of action of RP11-6O2.4 in GC have not been thoroughly elucidated to date. The specific aim of the present study was to assess RP11-6O2.4 and to explore its role in human GC. Methods: Quantitative real-time polymerase chain reaction (qPCR) was performed to analyze the expression levels of RP11-6O2.4 in GC tissues, paired adjacent noncancerous tissues (ANTs) and GC cell lines. In addition, the correlation between RP11-6O2.4 expression and the clinical characteristics and prognosis of patients with GC was statistically analyzed. The effects of RP11- 6O2.4 on the GC cell cycle transformation through the p38-MAPK signaling pathway were explored by flow cytometry, qPCR and Western blot analysis after treatment with SB203580, a p38MAPK specific inhibitor, in vitro. Results: The expression levels of RP11-6O2.4 in GC tissues were significantly lower than the paired ANTs (P<0.05). In addition, RP11-6O2.4 expression was significantly lower in cases with older age, longer maximum tumor diameter, higher ASA grade and deeper invasive depth (P<0.05). RP11-6O2.4 expression was significantly higher in cases with well/middle differentiation than poor/no differentiation; higher in cases without lymph node metastasis than in lymph node metastasis; and higher in cases in stage Ⅰ/Ⅱ than in stage Ⅲ/Ⅳ. An in vitro assay showed that RP11-6O2.4 induced G0/ G1 phase cell cycle arrest, likely by regulating the p38- MAPK signaling pathway. Conclusion: The above mentioned data suggested that RP11-6O2.4 was a tumor-suppressor gene in GC. RP11- 6O2.4 might play an important role in the cell cycle transformation by regulating the p38-MAPK signaling pathway, thereby representing a specific biomarker and a potential molecular target for the treatment of GC.

        Predictive Significance of Claudin-3 for Epithelial Barrier Dysfunction in Chronic Rhinosinusitis With Nasal Polyps

        Purpose: The abnormal expression of tight junction (TJ) plays a vital role in the pathogenesis of chronic rhinosinusitis with nasal polyps (CRSwNP). However, there is no appropriate tool to distinguish and diagnose epithelial barrier defects in clinical practice. This study aimed to evaluate the predictive value of claudin-3 for epithelial barrier dysfunction in CRSwNP. Methods: In this study, TJ protein levels were evaluated by real-time quantitative polymerase chain reaction, immunofluorescent, and immunohistochemistry staining in control subjects and CRSwNP patients. The receiver operating characteristic (ROC) curve was created to assess the predictive value of TJ breakdown in clinical outcomes. In vitro, human nasal epithelial cells were cultured at the air-liquid interface to analyze the transepithelial electrical resistance (TER) level. Results: The expression levels of occludin, tricellulin, claudin-3, and claudin-10 were decreased (all P < 0.05), and those of claudin-1 was increased (P < 0.05) in CRSwNP patients as compared to healthy subjects. Additionally, claudin-3 and occludin levels were negatively correlated with the computed tomography score in CRSwNP (all P < 0.05), and the ROC curve indicated that the claudin-3 level had the most predictive accuracy in evaluating epithelial barrier disruption (area under the curve = 0.791, P < 0.001). Finally, the time-series analysis showed the highest correlation coefficient between TER and claudin-3 (cross-correlation function = 0.75). Conclusion: In this study, we suggest that claudin-3 could be a valuable biomarker for predicting nasal epithelial barrier defects and disease severity in CRSwNP.
