        A method of X-ray source spectrum estimation from transmission measurements based on compressed sensing

        A new method of X-ray source spectrum estimation based on compressed sensing is proposed in this paper. The algorithm K-SVD is applied for sparse representation. Nonnegative constraints are added by modifying the L1 reconstruction algorithm proposed by Rosset and Zhu. The estimation method is demonstrated on simulated spectra typical of mammography and CT. X-ray spectra are simulated with the Monte Carlo code Geant4. The proposed method is successfully applied to highly ill conditioned and under determined estimation problems with a good performance of suppressing noises. Results with acceptable accuracies (MSE < 5%) can be obtained with 10% Gaussian white noises added to the simulated experimental data. The biggest difference between the proposed method and the existing methods is that multiple prior knowledge of X-ray spectra can be included in one dictionary, which is meaningful for obtaining the true X-ray spectrum from the measurements.

      • Opportunities and Challenges of HTTP Adaptive Streaming

        HTTP-based Adaptive Streaming (HAS) has emerged as the prominent technology for the delivery of audiovisual content over the Internet in recent years and has a major impact on network traffic. Although traditional stateful session-based streaming solution based on UDP was used initially for media content delivery, researchers and practitioners soon realize that HAS technology, due to get through firewalls friendly ,transfer NAT easily ,effectively utilize the existing networking infrastructure and provide uninterrupted video streaming service to users with dynamic network conditions and heterogeneous devices, has the potential to improve the Quality of Experiments compared with traditional streaming technologies. Consequently, various HAS media streaming solutions have been proposed and deployed successfully. This paper reviews the state-of-the-art of HAS technology and discoveries achieved by numerous researchers. We describe the basic taxonomy of HTTP adaptive streaming systems and summarize the major issues associated with HAS systems’ design. Then we outline the key challenges and open problems and highlight possible avenues for future directions.

      • Fenofibrate Increases Radiosensitivity in Head and Neck Squamous Cell Carcinoma via Inducing G2/M Arrest and Apoptosis

        Radiation therapy is an important treatment for head and neck squamous cell carcinoma (HNSCC). However, how to promote radiation sensitivity in HNSCC remains a challenge. This study aimed to investigate the radiosensitizing effects of fenofibrate on HNSCC and explore the underlying mechanisms. HNSCC cell lines CNE-2 and KB were subjected to ionizing radiation (IR), in the presence or absence of fenofibrate treatment. Cell growth and survival, apoptosis and cell cycle were evaluated. In addition, CNE-2 cells were xenografted into nude mice and subjected to IR and/or fenofibrate treatment. The expression of cyclinB and CDK1 was detected by Western blotting. Our results showed that fenofibrate efficiently radiosensitized HNSCC cells and xenografts in mice, and induced apoptosis and G2/M arrest via reducing the activity of the CDK1/cyclinB1 kinase complex. These data suggest that fenofibrate could be a promising radiosensitizer for HNSCC radiotherapy.

        The F-box Protein KIB1 Mediates Brassinosteroid-Induced Inactivation and Degradation of GSK3-like Kinases in <i>Arabidopsis</i>

        <P><B>Summary</B></P> <P>The glycogen synthase kinase-3 (GSK3) family kinases are central cellular regulators highly conserved in all eukaryotes. In <I>Arabidopsis</I>, the GSK3-like kinase BIN2 phosphorylates a range of proteins to control broad developmental processes, and BIN2 is degraded through unknown mechanism upon receptor kinase-mediated brassinosteroid (BR) signaling. Here we identify KIB1 as an F-box E3 ubiquitin ligase that promotes the degradation of BIN2 while blocking its substrate access. Loss-of-function mutations of KIB1 and its homologs abolished BR-induced BIN2 degradation and caused severe BR-insensitive phenotypes. KIB1 directly interacted with BIN2 in a BR-dependent manner and promoted BIN2 ubiquitination in vitro. Expression of an F-box-truncated KIB1 caused BIN2 accumulation but dephosphorylation of its substrate BZR1 and activation of BR responses because KIB1 blocked BIN2 binding to BZR1.         Estrogen weakens muscle endurance via estrogen receptor-p38 MAPK-mediated orosomucoid (ORM) suppression

        Gender differences in fatigue manifest as females being more prone to feel exhaustion and having lower muscle endurance. However, the mechanisms of these effects remain unclear. We investigated whether orosomucoid, an endogenous anti-fatigue protein that enhances muscle endurance, is involved in this regulation. Female rats exhibited lower muscle endurance, and this gender difference disappeared in orosomucoid-1-deficient mice. Female rats also exhibited weaker orosomucoid induction in serum, liver and muscle in response to fatigue compared with male rats. Ovariectomy elevated orosomucoid levels and increased swimming time, and estrogen replenishment reversed these effects. Exogenous estrogen treatment in male and female mice produced opposite effects. Estrogen decreased orosomucoid expression and its promoter activity in C2C12 muscle and Chang liver cells in vitro, and estrogen receptor or p38 mitogen-activated protein kinase blockade abolished this effect. Therefore, estrogen negatively regulates orosomucoid expression that is responsible for the weaker muscle endurance in females.

      • Safety Assessment of Ovarian Cryopreservation and Transplantation in Nude Mice Bearing Human Epithelial Ovarian Cancer

        Objective: Nude mice with orthotopic transplantation of human ovarian epithelial cancer were used to investigate screening criteria for paraneoplastic normal ovarian tissue and the security of the freezing and thawing for ovarian tissue transplantation. Methods: Expression of CK-7, CA125, P53, survivin, MMP-2/TIMP-2 in paraneoplastic normal ovarian tissues were detected by RT-PCR as well as immunohistochemistry. The tissues of the groups with all negative indicators of RT-PCR, all negative indicators of immunohistochemistry, negative expression of CK-7, CA125 and survivin, positive expression of CK-7, CA125 and survivin, cancer tissues and normal ovarian tissues of nude mice were used for freezing and thawing transplantation, to analyze overt and occult carcinogenesis rates after transplantation. Results: When all indicators or the main indicators, CK-7, CA125 and survivin, were negative, tumorigenesis did not occur after transplantation. In addition the occult carcinogenesis rate was lower than in the group with positive expression of CK-7, CA125 and survivin (P<0.01). After subcutaneous and orthotopic transplantation of ovarian tissues, rates did not change (P>0.05). There was no statistical significance among rates after transplantation of ovarian tissues which were obtained under different severity conditions (P>0.05). Conclusion: Negative expression of CK-7, CA125 and survivin can be treated as screening criteria for security of ovarian tissues for transplantation. Immunohistochemical methods can be used as the primary detection approach. Both subcutaneous and orthotopic transplantation are safe. The initial severity does not affect the carcinogenesis rate after tissue transplantation. Freezing and thawing ovarian tissue transplantation in nude mice with human epithelial ovarian carcinoma is feasible and safe.

      • Test study of precast SRC column under combined compression and shear loading

        A new type of precast steel reinforced concrete (PSRC) column was put forward in this paper. In order to study the static performance of PSRC column and hollow precast steel reinforced concrete (HPSRC) column subjected to combined compression and shear loading, a parametric test was carried out and effects of axial compression ratio, concrete strength and shear ratio on the mechanical behavior of composite PSRC column and HPSRC column were explored. In addition, the cracks development, load-span displacement relationship, strain distribution and shear bearing strength of column specimens were emphatically focused. Test results implied that shear failure of all specimens occurred during the test, and higher strength of castin-place concrete, smaller shear ratio and larger axial compression ratio could lead to greater shear resistance, but when the axial compression ratio was larger than 0.36, the shear capacity began to decrease gradually. Furthermore, truss-arch model for determining the shear strength of PSRC column and HPSRC column was proposed and the calculated results obtained from proposed method were verified to be valid.

        Effects of operational parameters on performance of the air-powered needle-free injector

        The needle-free injection is a new technique for treating subcutaneous drug delivery through penetrating the skin with a high-speed liquid drug jet. The present study aims to reveal fluid dynamics characteristics of the needle-free injection. The operational parameters such as the driving pressure, the nozzle diameter and the liquid volume were investigated for an air-powered needle-free injection system using experimental techniques. The results show that the peak stagnation pressure determines the penetration performance of the jet and is significantly affected by the driving pressure, nozzle diameter and liquid volume. Furthermore, the entire injection process of fluid in the gel can be defined as two phases of penetration and diffusion. Thus, the dispersion characteristics of fluid in different phases could be controlled by an effective combination of the nozzle diameter and liquid volume. Overall, the obtained conclusions provide a sound support for understanding the operation of the needle-free injector.

        Identification and Functional Analysis of Acyl-Acyl Carrier Protein Δ9 Desaturase from Nannochloropsis oceanica

        The oleaginous marine microalga Nannochloropsis oceanica strain IMET1 has attracted increasing attention as a promising photosynthetic cell factory due to its unique excellent capacity to accumulate large amounts of triacylglycerols and eicosapentaenoic acid. To complete the genomic annotation for genes in the fatty acid biosynthesis pathway of N. oceanica, we conducted the present study to identify a novel candidate gene encoding the archetypical chloroplast stromal acyl-acyl carrier protein Δ9 desaturase. The full-length cDNA was generated using rapid-amplification of cDNA ends, and the structure of the coding region interrupted by four introns was determined. The RT-qPCR results demonstrated the upregulated transcriptional abundance of this gene under nitrogen starvation condition. Fluorescence localization studies using EGFP-fused protein revealed that the translated protein was localized in chloroplast stroma. The catalytic activity of the translated protein was characterized by inducible expression in Escherichia coli and a mutant yeast strain BY4389, indicating its potential desaturated capacity for palmitoyl-ACP (C16:0-ACP) and stearoyl-ACP (C18:0-ACP). Further functional complementation assay using BY4839 on plate demonstrated that the expressed enzyme restored the biosynthesis of oleic acid. These results support the desaturated activity of the expressed protein in chloroplast stroma to fulfill the biosynthesis and accumulation of monounsaturated fatty acids in N. oceanica strain IMET1.

        <P>Multifunctional catalysts are of great interest in catalysis because their multiple types of catalytic or functional groups can cooperatively promote catalytic transformations better than their constituents do individually. Herein we report a new synthetic route involving the surface functionalization of nanoporous silica with a rationally designed and synthesized dihydrosilane (3-aminopropylmethylsilane) that leads to the introduction of catalytically active grafted organoamine as well as single metal atoms and ultrasmall Pd or Ag-doped Pd nanoparticles via on-site reduction of metal ions. The resulting nanomaterials serve as highly effective bifunctional dehydrogenative catalysts for generation of H-2 from formic acid.</P>
