        The effects of codon usage on the formation of secondary structures of nucleocapsid protein of peste des petits ruminants virus

        The nucleocapsid (N) protein of peste des petits ruminants virus (PPRV) with a conserved amino acid usage pattern plays an important role in viral replication. The primary objective of this study was to estimate roles of synonymous codon usages of PPRV N gene and tRNA abundances of host in the formation of secondary structure of N protein. The potential effects of synonymous codon usages of N gene and tRNA abundances of host on shaping different folding units (α-helix, β-strand and the coil) in N protein were estimated, based on the information about the modeling secondary structure of PPRV N protein. The synonymous codon usage bias was found in different folding units in PPRV N protein. To better understand the role of translation speed caused by variant tRNA abundances in shaping the specific folding unit in N protein, we modeled the changing trends of tRNA abundance at the transition boundaries from one folding unit to another folding unit (β-strand → coil, coil → β-strand, α-helix → coil, coil → α-helix). The obvious fluctuations of tRNA abundance were identified at the two transition boundaries (β-strand → coil and coil → β-strand) in PPRV N protein. Our findings suggested that viral synonymous codon usage bias and cellular tRNA abundance variation might have potential effects on the formation of secondary structure of PPRV N protein.

        LncRNA A2M-AS1 lessens the injury of cardiomyocytes caused by hypoxia and reoxygenation via regulating IL1R2

        Background: Myocardial ischemia and reperfusion injury (MI/RI) is a complex pathophysiological process, which can lead to severe myocardial injury. The long noncoding RNA alpha-2-macroglobulin antisense RNA 1 (A2M-AS1) has been revealed to be abnormally expressed in MI, However, its function in MI and the potential mechanism are still unclear. Objective: To evaluate the functional role of A2M-AS1 in hypoxia/reoxygenation (H/R)-induced neonatal cardiomyocytes and its potential molecular mechanism. Methods: Dataset GSE66360 was obtained from GEO database for analyzing the RNA expression of A2M-AS1 and interleukin 1 receptor type 2 (IL1R2). KEGG pathway enrichment analysis of the genes that co-expressed with A2M-AS1 was performed. Human neonatal cardiomyocytes were subjected to H/R to construct in vitro models. QRT-PCR and Western blot were adopted to test the levels of mRNA and protein. The viability and apoptosis of cardiomyocytes were tested by CCK-8 and flow cytometry assays, respectively. Results: The expression of A2M-AS1 was notably downregulated in H/R-treated cardiomyocytes. Overexpression of A2M-AS1 can notably enhance the cell viability of H/R-damaged cardiomyocytes, whereas knockdown of A2M-AS1 showed the opposite outcomes. Besides, a negative correlation was showed between A2M-AS1 and IL1R2 expression. In H/R-treated cardiomyocytes, overexpression of IL1R2 weakened the promoting proliferation and anti-apoptosis effects caused by overexpressing A2M-AS1, however, IL1R2-knockdown abolished the anti-proliferation and pro-apoptosis effects caused by silencing A2M-AS1. Conclusion: This study demonstrates the potential regulatory role of A2M-AS1/ IL1R2 axis in cardiomyocytes suffered from H/R, and provides insight into the protection of MI/RI.

      • Treatment of fever with traditional Chinese medicine according to Zheng on cancer patients (based on case reports)

        Fever in cancer patients is often due to the following causes: evil qi and toxity stagnancy, disorders of qi and blood, deficiencies of zang and fu organs, and the disorder of yin and yang. The treatments given to cancer patients with a fever are according to five: (a) Excessive inner heat and toxicants: remove heat and the toxicant, induce purgation. We use Cheng-Qi-Tang plus Qing-Wen-Bai-Du-Yin. (b) Tangle of damp and heat, and qi stagnancy: remove damp and heat, smooth the qi channel. We use Gan-Lu-Xiao-Du-Dan or San-Ren-Tang. (c) Obvious blood and heat stagnancy: remove heat and blood stasis. We use Xue-Fu-Zhu-Yu-Tang. (d) Deficiency of spleen qi, inner heat caused by a yin deficiency: nourish spleen qi and yin to remove the inner heat. We use Bu-Zhong-Yi-Qi-Tang or Xiao-Jian-Zhong-Tang. (e) Prominent yin deficiency and hectic fever: replenish yin and remove inner heat. We use Qing-Hao-Bie-Jia-Tang or Chai-Qian-Mei-Lian-San. The pathogenesis of fever in cancer patients is complicated. We can see both deficiency and excess in one differentiation. Therefore, we must make sure of it, then we can get the most effective treatment.

      • Degrees of Freedom for Complex MIMO Multiple-Way Relay Channel

        The amount of diversity or the number of degrees of freedom (DOF) can be increased by multiple antennas in wireless communication systems. In this paper, we study the DOF of multiple-input multiple-output (MIMO) multiple-way relay channel. There are two groups of source nodes and several one relay node which are equipped with multiple antennas in this kind of channel. Each source node in one group exchanges independent message with the source node in the other group via the relay node. We first show the upper bound on the total DOF for this channel. Then we extend it to the complex MIMO multiple-way relay channel and present the corresponding upped bound.

        Cinnamaldehyde Derivatives Inhibit Coxsackievirus B3-Induced Viral Myocarditis

        The chemical property of cinnamaldehyde is unstable in vivo, although early experiments have shown its obvious therapeutic effects on viral myocarditis (VMC). To overcome this problem, we used cinnamaldehyde as a leading compound to synthesize derivatives. Five derivatives of cinnamaldehyde were synthesized: 4-methylcinnamaldehyde (1), 4-chlorocinnamaldehyde (2), 4-methoxycinnamaldehyde (3), α-bromo-4-methylcinnamaldehyde (4), and α-bromo-4-chlorocinnamaldehyde (5). Neonatal rat cardiomyocytes and HeLa cells infected by coxsackievirus B3 (CVB3) were used to evaluate their antiviral and cytotoxic effects. In vivo BALB/c mice were infected with CVB3 for establishing VMC models. Among the derivatives, compound 4 and 5 inhibited the CVB3 in HeLa cells with the half-maximal inhibitory concentrations values of 11.38 ± 2.22 μM and 2.12 ± 0.37 μM, respectively. The 50% toxic concentrations of compound 4 and 5-treated cells were 39-fold and 87-fold higher than in the cinnamaldehyde group. Compound 4 and 5 effectively reduced the viral titers and cardiac pathological changes in a dose-dependent manner. In addition, compound 4 and 5 significantly inhibited the secretion, mRNA and protein expressions of inflammatory cytokines TNF-α, IL-1β and IL-6 in CVB3-infected cardiomyocytes, indicating that brominated cinnamaldehyde not only improved the anti-vital activities for VMC, but also had potent anti-inflammatory effects in cardiomyocytes induced by CVB3.

        Synthesis of coryphantha elephantidens-like SnO2 nanospheres and their gas sensing properties

        In this work, coryphantha elephantidens-like SnO2 with porous structures were prepared successfully by a simple hydrothermal route, through adjusting the temperature of hydrotherm. Its morphology was characterized by Xray diffraction (XRD), scanning electron microscopy (SEM) and Brunauer–Emmett–Teller (BET). Compared to the regular nanospheres, the coryphantha elephantidens-like SnO2 nanospheres had obviously higher gas-sensing response, owing to the special structure with large specific surface area (161.16m2 g−1). It surprised us that the coryphantha elephantidens-like SnO2 sensor could easily distinguish between ethanol and acetone, whose chemical property were similar. Moreover, it also exhibited wide measurement range, fast response speed (less than 10 s) and good repeatability at a low temperature (180 °C) to ethanol. The desirable specific surface area and pore volume were conducive to molecules adsorption and diffusion, which were believed to be the major cause of the improvement of gas sensing performance.

        Taxonomy of fungal complex causing red-skin root of Panax ginseng in China

        Background: Red-skin root of Asian ginseng (Panax ginseng) significantly reduces the quality and limits theproduction of ginseng in China. The disease has long been thought to be a noninfectious physiologicaldisease, except one report that proved itwas an infectious disease. However, the causal agents have not beensuccessfully determined. In the present study, we were to reveal the pathogens that cause red-skin disease. Methods: Ginseng roots with red-skin root symptoms were collected from commercial fields in NortheastChina. Fungi were isolated from the lesion and identified based on morphological characters alongwith multilocus sequence analyses on internal transcription spacer, b-tubulin (tub2), histone H3 (his3),and translation elongation factor 1a (tef-1a). Pathogens were confirmed by inoculating the isolates inginseng roots. Results: A total of 230 isolates were obtained from 209 disease samples. These isolates were classifiedinto 12 species, including Dactylonectria sp., D. hordeicola, Fusarium acuminatum, F. avenaceum, F. solani,F. torulosum, Ilyonectria mors-panacis, I. robusta, Rhexocercosporidium panacis, and three novel speciesI. changbaiensis, I. communis, and I. qitaiheensis. Among them, I. communis, I. robusta, and F. solani had thehighest isolation frequencies, being 36.1%, 20.9%, and 23.9%, respectively. All these species isolated werepathogenic to ginseng roots and caused red-skin root disease under appropriate condition. Conclusion: Fungal complex is the causal agent of red-skin root in P. ginseng.

        The effects of non-native listeners` L1 background and L2 proficiency on the perception of foreign accent for pitch-manipulated native and Chinese accented English speech

        This study reports a perception experiment of foreign accent where both native and non-native Chinese and Korean listeners participated in judging the foreign accent of native and Chinese accented English speech, but unlike many previous studies, pitch was manipulated to increase and decrease at the peak in statements and at the valley in yes-no questions to find out if non-native listeners` sensitivity to pitch conforms to native listeners. Moreover, the current study attempts to find the effects of non-native listeners` L1 background and L2 proficiency in the extent to which the agreement occurs between native and non-native listeners` ratings. Results showed that the prosodic system pertaining to pitch in their L1 influenced L2 listeners` responses to the synthesized changes of pitch in L2 speech. Non-native Chinese and Korean listeners` perception of foreign accent was also different between statement and yes-no question stimuli as pitch serves as a distinctive function in two syntactic structures of their L1. This all provides the evidence for the effect of non-native listeners` L1 background. Non-native listeners` L2 proficiency also turned out to contribute to the foreign accent judgment only for Chinese high talkers` yes-no question stimuli, which serves as the evidence for the effect of L2 proficiency. However, it held true only when the listeners did not share their L1 with the talkers. (University of Seoul)

      • HLA-A, HLA-B, HLA-DRB1 Polymorphisms and Risk of Cervical Squamous Epithelial Cell Carcinoma: A Population Study in China

        Cervical cancer is the second most common cancer in women. HLA class I and II alleles polymorphisms have been shown to be associated with cervical cancer risk, but results have varied among different populations. In this study, the HLA-A, -B, and -DRB1 alleles among 100 southern Chinese women with cervical squamous cell carcinoma (SCC) were compared to 254 controls. Our results showed that $B^*51$:01:02 allele frequency was significantly higher in patients with SCC than in healthy controls ($P=3.17{\times}10^{-5}$, $P_c$=0.005, OR=26.7). Statistical analysis also revealed a significantly decreased frequency of $B^*51$:01:02 ($P=7.01{\times}10^{-4}$, $P_c$=0.03, OR=0.12) in patients with SCC when compared with healthy controls. These results indicate that HLA-$B^*51$:01:02 may confer susceptibility to SCC and HLA-$B^*51$:01:02 may contribute to resistance to the development of SCC in Chinese women. None of the HLA-A-B or HLA-A-B-DRB1 haplotypes were significantly different in cases and controls after multiple testing corrections, indicating the individual allele associations to be independent of the identified haplotypes. These results support the hypothesis that some HLA-B alleles could be involved with susceptibility for developing SCC.
