        심근경색 모델 흰쥐에서 스트레스가 B-type Natriuretic Peptide 발현 및 심박변이율 변화에 미치는 영향

        심박변이율의 감소는 심근 경색 환자에서 예후의 악화와 관련되어 있다. 심박변이율 중 very low frequency가 심부전환자의 예후를 판단할 수 있는 독립적 인자로서 사용이 가능하다고 보고된 바 있으며, 심실의 압력 증가에 반응해서 생성되는 B-type natriuretic peptide가 심근 경색의 예후 인자로서 사용할 수 있음 역시 보고된 바 있다. 본 연구에서는 심근경색 모델을 제작하여 일정의 회복 기간을 거친 다음 다시 일정 기간의 스트레스를 겪게 한 후 심박변이율의 변화 및 심실 조직에서의 BNP 발현 정도를 비교하여 스트레스에 의한 영향을 관찰하고자 하였다. Sprague-Dawley계 수컷 흰쥐 15마리를 대상으로 하였다. 실험동물은 인위적 스트레스 및 수술을 받지 않은 대조군(CON, n=5), 심근경색 수술을 받은 후 restraint stress를 받지 않은 군(MI+No Stress, n=5), 심근경색 수술을 받은 후 1주일 동안 restraint stress를 받은 군(MI+Stress, n=5)으로 분류하였다. 심근경색 모델 제작 직후 15분 이상 심전도의 변화를 기록하였으며, 2개월 정도 후 심근경색 수술을 받은 동물을 두 군으로 나누어 그 중 한 군에는 1주일 동안 restraint stress를 가한 후 restraint stress를 가하지 않은 동물과 심박변이율을 비교 분석하였다. 심실 조직은 좌심실 전벽 부위의 위축을 관찰할 수 있었고, 대식세포에 의하여 응고, 괴사된 심근세포의 탐식과 혈관이 풍부한 육아조직 및 섬유 변화를 볼 수 있었다. 심박변이율은 심근경색 수술을 받은 직후 심박수는 유의한 증가를 보였고, standard deviation of the normal to normal intervals (SDNN), very low frequency (VLF) 및 low frequency (LF)의 유의한 감소를 보였다. 심근경색 수술을 받았던 동물에서 두 달 정도의 회복 기간을 거친 후 스트레스를 받지 않은 군은 심박수 및 기타 심박변이율 분석에서 SDNN 값을 제외하고는 정상군과 유의한 차이를 관찰할 수 없었던 반면 1주일 동안 스트레스를 받은 군에서는 심박수가 다시 유의하게 증가했을 뿐만 아니라 SDNN, VLF 및 LF 역시 정상군과 유의한 차이를 보였다. 이상의 결과를 종합하면 심근경색 동물에 대해 1주일 동안 스트레스를 가한 결과 심박변이율이 감소하고 심실에서의 BNP 발현은 더욱 증가하여 악화함을 보여주고 있다. Diminished heart rate variability (HRV) is associated with less favorable prognosis after myocardial infarction (MI). It has been reported that very low frequency (VLF) power in HRV analysis is an independent risk predictor in patients with congestive heart failure and B-type natriuretic peptide (BNP) can be used as a prognostic factor of MI. The purpose of this study was to investigate whether stress has an effect to the changes of BNP expression and/or heart rate variability in MI model in rats. Male Sprague-Dawley rats were divided into 3 groups: control group (CON), MI group (MI+No Stress), and MI followed by stress group (MI+Stress). MI+Stress group rats were raised for a two month recovery period after the operation, followed by being exposed to restraint stress for 2 hours per day for 1 week. Electrocardiogram was recorded after the operation and the last day after 1 week of stress. The frequency components of HRV were calculated in the frequency domain such as VLF, low frequency (LF), high frequency (HF) and so on. In HRV analysis, standard deviation of the normal to normal intervals (SDNN) was significantly reduced in both groups compared to the control group. VLF and LF also were significantly reduced in MI+Stress group compared to the control groups. In addition, BNP expression in western blotting was shown the strongest bands in MI+ Stress group among experimental groups. These results suggest that BNP and HRV were aggravated by stress in MI rat model. (Korean J Str Res 2010;18:275∼285)

        간헐적 교정력 적용 후 백서 치주인대에서 UNC-50 유전자의 발현

        기계적 응력은 정상적인 발달과정 동안 조직의 항상성에 있어 중요한 역할을 한다. 기계적 응력은 치아 이동과 저작과 같은 상황을 포함한다. 치아 이동과 저작 중에 치주인대 섬유모세포는 기계적 자극을 감지하고 주위의 세포밖 물질과 생체분자 대사의 변동에 의해 반응을 보인다. 그러나 아직까지 기계적 응력 하에서 치주인대 세포에서 발현된 유전자에 관한 연구는 미비한 실정이다. 최근 기계적 응력이 초파리에 존재하는 UNC-50 유전자에 영향을 줄 수 있다고 보고되었다. 또한 UNC-50은 치은 섬유모세포와 비교해 치주인대 섬유모세포에서만 발현된다고 보고되었다. 본 연구에서는 간헐적 교정력 적용 시에 백서의 치아에서 일어나는 치근 및 치주 조직의 조직학적 변화와, UNC-50의 발현 양상을 면역조직학적 염색으로 조사하여 치주인대에서 기계적 응력과 UNC-50의 관련성을 알아보고자 하였다. Sprague-Dawely계 수컷 백서 12마리를 4마리씩 세 군으로 나누어 상악 우측 구치부에 NiTi closed coil spring을 사용하여 40 g 정도의 견인력이 발생하도록 하여 하루에 1시간씩 간헐적인 교정력을 적용한 후 1, 3, 5일 후 치주인대의 조직학적 변화를 관찰하여 다음과 같은 결과를 얻었다. 조직학적 소견에서 상악 제1대구치 근심구개치근의 치근부 1/3에서 압박측은 안장측보다 더 좁은 치주인대공간을 보였고 교정력을 적용시킨 후 3일 후부터 인장측에서 백악모세포 활성으로 인한 백악질 침착이 관찰되었다. UNC-50은 인장측의 분화 중인 백악모세포에서 강한 발현을 보였다. Osteocalcin은 인장측에서 압박측에 비해 신생 백악질에 존재하는 분화 중인 백악모세포를 따라 강한 발현을 보였다. 이상의 연구결과는 UNC-50이 간헐적 교정력 즉 기계적 응력의 변화에 따른 백악모세포의 분화과정에 중요한 역할을 함을 나타낸다. 그러나 이를 명확히 하기 위해서는 교정력 적용 후 UNC-50의 세포내 신호전달과정에 대한 보완연구가 필요할 것이다. Objective: Periodontal ligament fibroblasts have an ectomesenchymal origin and are thought to play a crucial role for not only homeostasis of periodontal tissues but also bone remodeling, wound healing and regeneration of tissues. Recently, it has been reported that UNC-50 is not expressed in gingival fibroblasts but in PDL fibroblasts. The purpose of this study was to examine the expression of UNC-50 and osteocalcin in the periodontium after application of intermittent force. Methods: Twelve rats had 40 grams of mesially-directed force applied at the upper molar for 1 hour/day. Four rats were sacrificed at 1, 3 and 5 days. Immunohistochemical localization of UNC-50 and osteocalcin antibody was carried out. The results showed apposition of new cellular cementum and a slight increase in periodontal space at the tension side. Results: Strong UNC-50 expression was observed in the differentiating cementoblasts close to PDL fibroblasts in the tension side whereas it was barely expressed at the compression side. Expression was strong at day 3, and decreased at day 5. Osteocalcin immunoreactivity expression was strong in differentiating cementoblasts at the tension side. Conclusion: It can be suggested that UNC-50 is related to the differentiation of cementoblasts, and may be responsible for the molecular event in PDL cells under mechanical stress.

        The South Korean Minjung Art in 20th century

        In 1980`s, Korean painters began to represent these terrible scenes in their arts. The movement is called ``Minjung Art``. This research explores the characteristics and meanings of the Minjung Art, primarily considering the relationship between art and Korean political situation in the 80`s. Minjung Artists thought that Korea has become a new colony of American economy, culture, art and politics. That is why Minjung Art was strongly motivated by the anti-colonial, and anti-American ideas. Minjung Art is a Socialist Art based on the Marx-Lenin theories. An influential literary and art critic of the twentieth century was a Post-Marxist who pointed out the capability of Realist Art to confront an objective reality that exists in the world. Art was a work. Bataille believed that the making of tools and weapons was the departure point for that early faculty of ration, which humanized the animals we once had been. It is obviously work that made him a human being, the reasonable animal we are. Bataille thought that if it is true that work is our origin, if it is true that work is the key to humanity, human beings would eventually distance themselves completely from animalist through work. Minjung Art did not aim at the revolution of social reality but to be an ideological art seeking for the reforms of the society that grants freedom for the people. The import of “culture” for understanding human activity and the history of its many uses provide the initial basis of Cultural Studies, an interdisciplinary field formed over forty years ago, primarily in the US and UK. Since then, interest in the field has grown exponentially. Incorporating a diverse range of new theoretical inputs, methodological innovations and objects of inquiry, Cultural issues takes up a number of issues related to contemporary culture while being aware of their specific historical formations.

        A Convergent Study on Interrelation between Color Perception and Brain Wave

        Recently in Korea, brain wave at the country"s exhibition industry has been known more and a range of support is needed. This study examined relation between color cognition process and brain wave. Color cognition can be done at background of the times, scientific development, cultural variable, change of political situation, economy and education of color Color perception process consists of natural and ordinary coloring of digital color making use of digital media of animal, plant, landscape and daily lives. The author conducted test by Art Wellness program of "S" college. First, background and scope of studies on relation between color cognition and brain wave. Second, the author described characteristics of brain wave and color to indicate cases of the art and AI fusion color cognition. Third, the study explained relation between color cognition and brain wave to indicate implications. The finding was: Color emotion with sociality was universal than accidential and personal taste. However, color sense of outcome of language, thought, experience and tradition may have different emotion. Wellness program of "S" College examines variables of sound, language and vocabulary of composite association. New art and design shall be used to develop color cognition and to make use of relation between human life and education. Segmented research is needed to build database for social and scientific application.

        Identification of Non-Muscle Nebulin Isoform in Human Brain Library

        Nebulin is a (Mr 600~900 kDa) large actin-binding protein specific to skeletal muscle and thought to act as a molecular template that regulates the length of thin filaments. Cardiac muscles of higher vertebrates have been shown earlier to lack nebulin. Recently, full-length nebulin mRNA transcripts have been detected in heart muscle, but at lower levels than in skeletal muscle. Nebulin expression also was detected in the kidney, eye, and otic canal, suggesting that nebulin isoforms may also be expressed in these organs. We have searched for nebulin isoforms in brain of human using PCR and Northern blot. Here, we provide evidence that nebulin mRNA transcripts are expressed in brain. Seven nebulin isoforms (B, C, D, E, F, G and H form) are obtained in human skeletal muscle and four isoforms (B, C, G and H form) in human brain cDNA library. We cloned the 1.3 kb of nebulin fragment from human adult bran library by PCR. The identity of the PCR product was confirmed by sequence analysis. The partial brain nebulin sequence was 99% identical to the skeletal muscle cDNA as determined by Blast alignment. It contains two simple-repeats HR1, HR2 and linker-repeals exon 135~143 except exon 140. It was different from skeletal muscle B form, which contain HR1 and HR8. These data suggest that nebulin isoform diversity occurs even more extensively than previously known, likely contributing to the distinct thin filament architecture of different striated muscles.

        Effect of hypoxia on angiogenesis-related proteins in human dental pulp cells

        Dental pulp is a highly vascularized tissue with high regenerative potential. Revascularization of severed vasculature in the tooth is required for pulp healing during avulsed tooth treatment. In this study, the relative expression of angiogenesis-related proteins was determined in human dental pulp cells using a human angiogenesis proteome profiler array. The proteome profiler array detected differentially expressed angiogenesis-related factors under conditions of hypoxia, which enhances the angiogenic potential of dental pulp cells. We confirmed that hypoxia regulates the mRNA expression of angiogenesis-related factors, including CXCL16 in dental pulp cells. Furthermore, conditioned media of hypoxic pulp cells induced tube-like structures of vascular endothelial cells, which were reduced by the neutralization of CXCL16 function. In conclusion, our data show that angiogenesisrelated factors are differentially expressed by hypoxia in dental pulp cells and suggest that CXCL16 may involve in the revascularization of hypoxic dental pulp.

        A Study on convergent and qualitative value of Haeju Baekja patterns

        The pottery is thought not to be art but to be aesthetic experience factor at daily life traditionally to express cognition and custom made experience by form and colors. The aesthetic experience puts integration and collection into practice, and outcome expresses convergent and significant value by patterns, colors and forms of Haeju Baekja. Haeju Baekja made economic, cultural, knowledge, social and educational values under various kinds of background of the times. The values made variety of convergent culture that was produced when living life with people. Haeju baekja"s patterns include factors of the times, for instance, politics, economy, industry, culture & art, regional characteristics that are applied to the lives of costume, food, construction and furniture. The desire is needed at basic life rather than dignity to have simple and daring poet sentence of not only ware but also patterns depending upon consumption inclination of public people. Haeju baekja patterns have convergence and quality values without symmetry and connection of material, size and structure to have usefulness and convenience of life of northern environment that has excluded delicateness and decoration like Cheongja and Baekja, and to have harmonious patterns. Dragon pattern with symbol of king has humorous dragon without horn to express free and quality thought in convergence way.

        Involvement of Heme Oxygenase-1 in Orexin-A-induced Angiogenesis in Vascular Endothelial Cells

        The cytoprotective enzyme heme oxygenase-1 (HO-1) influences endothelial cell survival, proliferation, inflammatory response, and angiogenesis in response to various angiogenic stimuli. In this study, we investigate the involvement of HO-1 in the angiogenic activity of orexin-A. We showed that orexin-A stimulates expression and activity of HO-1 in human umbilical vein endothelial cells (HUVECs). Furthermore, we showed that inhibition of HO-1 by tin (Sn) protoporphryin-IX (SnPP) reduced orexin-A-induced angiogenesis <i>in vivo</i> and <i>ex vivo</i>. Orexin-A-stimulated endothelial tube formation and chemotactic activity were also blocked in SnPP-treated vascular endothelial cells. Orexin-A treatment increased the expression of nuclear factor erythroid-derived 2 related factor 2 (Nrf2), and antioxidant response element (ARE) luciferase activity, leading to induction of HO-1. Collectively, these findings indicate that HO-1 plays a role as an important mediator of orexin-A-induced angiogenesis, and provide new possibilities for therapeutic approaches in pathophysiological conditions associated with angiogenesis.
