        Dynamic Mechanical Properties of Red Sandrock-Polypropylene Fiber Reinforced Concrete Composite under Impact Load

        To investigate the dynamic mechanical properties and fracturing behavior of the red sandrock-polypropylene fiber reinforced concrete (R-PPFRC) composite, R-PPFRC composite specimens with different Polypropylene fiber (PPF) dosage were fabricated. Conducted quasi-static compression experiments and dynamic experiments by using Split Hopkinson pressure bar (SHPB) apparatus and testing machine. The fracture characteristics and process of composite specimens were analyzed with the assistance of ultra-high-velocity camera photography. It can be concluded that the dynamic increase factor and the dynamic compressive strength of composite specimens R-PPFRC increase with the increase of load level, indicating a remarkably strain rate strengthening effect. The optimal PPF content is determined finally. The dynamic impact toughness, ultimate toughness, and dissipation energy of R-PPFRC are significantly affected by the load level and the PPF content. The fracturing process of the composite specimen is divided into four phases, crack initiation, crack expansion, crack cluster and ultimate failure. After going through these phases, the ultimate failure pattern is mainly shear and split mixed failure.

        SPINK1 promotes cell growth and metastasis of lung adenocarcinoma and acts as a novel prognostic biomarker

        Serine protease inhibitor Kazal type 1 (SPINK1) plays a role in protecting the pancreas against premature activation of trypsinogen and is involved in cancer progression. SPINK1 promoted LAC cells growth, migration, and invasion. Mechanistically, we found that SPINK1 promoted LAC cells migration and invasion via up-regulating matrix metalloproteinase 12 (MMP12). We observed that SPINK1 expression was only up-regulated in lung adenocarcinoma (LAC) tissues, and was an independent prognostic factor for poor survival. Our results indicate that SPINK1 might be a potential biomarker for LAC that promotes progression by MMP12. [BMB Reports 2018; 51(12): 648-653]

        Research on effects of key influencing factors upon fuel injection characteristics of the combination electronic unit pump for diesel engines

        A numerical model of the combination electronic unit pump (CEUP) fuel injection system was developed in AMESim environment. The effects of five key influencing factors, including cam profile velocity, plunger diameter, length of high pressure fuel pipe, inner diameterof high pressure fuel pipe and nozzle flow rate on injection characteristic parameters, were analyzed by using the developed numericalmodel. On the basis, a correlation analysis between the influencing factors and injection characteristics was performed by usingthe design of experiments (DoE) method, and the influences of these factors were quantized accordingly. Relevant results show that boththe single influencing factor and the interaction among these factors correlates with the injection characteristics, and the correlation representsa complex law with the cam rotational speed. The effect of plunger diameter on the injection pressure, cycle fuel injection quantityand injection duration is the most obvious, especially at a cam rotational speed of 500 r/min and the correlation coefficient is up to 0.82. The length of high-pressure pipe (HP pipe) has the most obvious influence on the coefficient of fuel feeding at cam rotational speed of500 r/min and 800 r/min, and the correlation coefficient is negative. Overall, the independent influence of the factors is more significantthan the combined influence of various factors. The CEUP fuel injection system is a complicated multi-input multi-output (MIMO)nonlinear system in fact.

        Effects of echinomycin on endothelin-2 expression and ovulation in immature rats primed with gonadotropins

        Echinomycin is a small-molecule inhibitor of hypoxia- inducible factor-1 DNA-binding activity, which plays a crucial role in ovarian ovulation in mammalians. The present study was designed to test the hypothesis that hypoxia-inducible factor (HIF)-1α-mediated endothelin (ET)-2 expressions contributed to ovarian ovulation in response to human chorionic gonadotropin (hCG) during gonadotropin-induced superuvulation. By real-time RT-PCR analysis, ET-2 mRNA level was found to significantly decrease in the ovaries after chinomycin treatment, while HIF-1α mRNA and protein expression was not obviously changed. Further analysis also showed that these changes of ET-2 mRNA were consistent with HIF-1 activity in the ovaires, which is similar with HIF-1α and ET-2 expression in the granulosa cells with gonadotropin and echinomycin treatments. The results of HIF-1α and ET-2 expression in the granulosa cells transfected with cis-element oligodeoxynucleotide (dsODN) under gonadotropin treatment further indicated HIF-1α directly mediated the transcriptional activation of ET-2 during gonadotropin- induced superuvulation. Taken together, these results demonstrated that HIF-1α-mediated ET-2 transcriptional activation is one of the important mechanisms regulating gonadotropin-induced mammalian ovulatory precess in vivo.         A Dynamic Placement Mechanism of Service Function Chaining Based on Software-defined Networking

        To cope with the explosive growth of Internet services, Service Function Chaining (SFC) based on Software-defined Networking (SDN) is an emerging and promising technology that has been suggested to meet this challenge. Determining the placement of Virtual Network Functions (VNFs) and routing paths that optimize the network utilization and resource consumption is a challenging problem, particularly without violating service level agreements (SLAs). This problem is called the optimal SFC placement problem and an Integer Linear Programming (ILP) formulation is provided. A greedy heuristic solution is also provided based on an improved two-step mapping algorithm. The obtained experimental results show that the proposed algorithm can automatically place VNFs at the optimal locations and find the optimal routing paths for each online request. This algorithm can increase the average request acceptance rate by about 17.6% and provide more than 20-fold reduction of the computational complexity compared to the Greedy algorithm. The feasibility of this approach is demonstrated via NetFPGA-10G prototype implementation.

        Imbalance of Innate and Adaptive Immunity in Esophageal Achalasia

        Background/AimsPrevious studies reveal that immune-mediated neuroinflammation plays a key role in the etiology of esophageal achalasia. However, the understanding of leucocyte phenotype and proportion is limited. This study aim to evaluate the phenotypes of leukocytes and peripheral blood mononuclear cells transcriptomes in esophageal achalasia. MethodsWe performed high-dimensional flow cytometry to identified subsets of peripheral leukocytes, and further validated in lower esophageal sphincter histologically. RNA sequencing was applied to investigate the transcriptional changes in peripheral blood mononuclear cells of patients with achalasia. Cell-type Identification by Estimating Relative Subsets of RNA Transcripts (CIBERSORT) was used for estimating the immune cell types. A differential gene expression analysis was performed and the differential expressed genes were subjected to gene ontology, Kyoto Encyclopedia of Genes and Genomes network, protein-protein interaction network construction. ResultsAn imbalance between innate and adaptive immune cells occurred in achalasia. Specifically, neutrophils and CD8+ T cells increased both in peripheral blood and lower esophageal sphincter in achalasia. Eosinophils decreased in peripheral blood but massively infiltrated in lower esophageal sphincter. CIBERSORT analysis of peripheral blood mononuclear cells RNA sequencing displayed an increased prevalence of CD8+ T cells. 170 dysregulated genes were identified in achalasia, which were enriched in immune cells migration, immune response, etc. Proton pump inhibitor analysis revealed the intersections and gained 7 hub genes in achalasia, which were IL-6, Toll-like receptor 2, IL-1β, tumor necrosis factor, complement C3, and complement C1q A chain. ConclusionPatients with achalasia exhibited an imbalance of systematic innate and adaptive immunity, which may play an important role in the development of achalasia.
