      • Enhanced strength and ductility in particulate-reinforced aluminum matrix composites fabricated by flake powder metallurgy

        Reinforcement agglomeration always leads to severe stress concentration and porosity, which is detrimental to the deformation ability and mechanical properties of particulate-reinforced metal matrix composites. In this study, uniform distribution of 32vol%B<SUB>4</SUB>C has been achieved in B<SUB>4</SUB>C/Al composite by means of flake powder metallurgy (Flake PM), in which flake Al powder is used as the starting material. The flake Al powder exhibits higher apparent volume than spherical powders of the same mass, and thus can provide more space to accommodate the B<SUB>4</SUB>C particles. Therefore, compared with conventional PM, Flake PM can lead to more uniform distribution of B<SUB>4</SUB>C particles in the composite powder as well as in the consolidated composite. Meanwhile, the flake Al powder has a nano skin of Al<SUB>2</SUB>O<SUB>3</SUB>, which could be fractured and dispersed inside the fine matrix grains during consolidation, and were found to induce a higher normalized strain hardening rate for the composite during deformation. As a result, the Flake PM 32vol%B<SUB>4</SUB>C/Al composite exhibits an ultimate tensile strength of 305MPa and a uniform elongation of 6.6%, 63% stronger and 13% more ductile than its counterpart fabricated by conventional PM.

        Sauchinone from Saururus chinensis protects vascular inflammation by heme oxygenase-1 induction in human umbilical vein endothelial cells

        Sauchinone, a diastereomeric lignan isolated from the roots of Saururus chinensis (LOUR.) BAILL. (Saururaceae), is reported to exert a variety of biological activities such as hepatoprotective, anti-inflammatory actions and inhibitory effects on bone resorption. In this study, we investigated the effect of sauchinone in suppressing cell adhesion molecules such as vascular cell adhesion molecule-1 (VCAM-1) and intracellular adhesion molecule-1 (ICAM-1) expression in high glucose stimulated human umbilical vein endothelial cells (HUVEC). Sauchinone inhibited the phosphorylation and degradation of IκB-α, as well as the nuclear translocation of nuclear factor kappa B (NF-κB) p65 caused by the stimulation of high glucose. In addition, sauchinone induced heme oxygenase (HO)-1 expression through nuclear translocation of nuclear factor E2-related factor 2 in HUVEC. The effects of sauchinone on the high glucose-induced expression of VCAM-1 and ICAM-1 and nuclear translocation of NF-κB p65 were partially reversed by transfection of the cells with HO-1 siRNA. These findings suggest that sauchinone-induced HO-1 expression plays a key role in the vascular protective effects of sauchinone in HUVEC.

        Comparison of Motility, Acrosome, Viability and ATP of Boar Sperm with or without Cold Shock Resistance in Liquid Semen at 17℃ and 4℃, and Frozen-thawed Semen

        This study was designed to analyze boar sperm to compare motility, acrosome morphology, viability and ATP by various preservation methods between Duroc boar A with cold shock resistance sperm and Duroc boar B with cold shock sensitivity sperm. Semen volume, sperm concentration, motility and normal acrosome between Duroc boar A and B did not show any differences within 2 h after collection. There were no differences in sperm motility and normal acrosome between boar A and B at 1 day of preservation at $17^{\circ}C$ and $4^{\circ}C$, respectively. However, sperm motility and normal acrosome from 2 day of preservation at $17^{\circ}C$ and $4^{\circ}C$, respectively, were higher for boar A than boar B. The frozen-thawed sperm motility and normal acrosome were higher for boar A than boar B. The sperm viability and ATP concentration according to storage period of liquid semen at $17^{\circ}C$ and $4^{\circ}C$ were higher for boar A than boar B. Also, the sperm viability and ATP concentration of frozen-thawed semen were higher for boar A than boar B. In conclusion, we found out that the original quality of boar semen with cold shock resistance sperm played an important role.

        Investigation of PCR-RFLPs within Major Histocompatibility Complex B-G Genes Using Two Restriction Enzymes in Eight Breeds of Chinese Indigenous Chickens

        New polymorphism of major histocompatibility complex B-G genes was investigated by amplification and digestion of a 401bp fragment including intron 1 and exon 2 using polymerase chain reaction-restriction fragment length polymorphism (PCRRFLP) technique with two restriction enzymes of Msp I and Tas I in eight breeds of Chinese indigenous chickens and one exotic breed. In the fragment region of the gene, three novel single nucleotide polymorphisms (SNPs) were detected at the two restriction sites. We found the transition of two nucleotides of A294G and T295C occurred at Tas I restriction site, and consequently led to a nonsynonymous substitution of asparagine into serine at position 54 within the deduced amino acid sequence of immunoglobulin variableregion- like domain encoded by the exon 2 of B-G gene. It was observed at rare frequency that a single mutation of A294G occurring at the site, also caused an identical substitution of amino acid, asparagine 54-to-serine, to that we described previously. And the transversion of G319C at Msp I site led to a non-synonymous substitution, glutamine 62-to-histidine. The new alleles and allele frequencies identified by the PCR-RFLP method with the two enzymes were characterized, of which the allele A and B frequencies at Msp I and Tas I loci were given disequilibrium distribution either in the eight Chinese local breeds or in the exotic breed. By comparison, allele A at Msp I locus tended to be dominant, while, the allele B at Tas I locus tended to be dominant in all of the breeds analyzed. In Tibetan chickens, the preliminary association analysis revealed that no significant difference was observed between the different genotypes identified at the Msp I and Tas I loci and the laying performance traits, respectively.

        Nasal immunization with mannan-decorated mucoadhesive HPMCP microspheres containing ApxIIA toxin induces protective immunity against challenge infection with Actinobacillus pleuropneumoiae in mice

        <P>The development of subunit mucosal vaccines requires an appropriate delivery system or an immune modulator such as an adjuvant to improve antigen immunogenicity. The nasal route for vaccine delivery by microparticles has attracted considerable interest, although challenges such as the rapid mucociliary clearance in the respiratory mucosa and the low immunogenicity of subunit vaccine still remain. Here, we aimed to develop mannan-decorated mucoadhesive thiolated hydroxypropylmethyl cellulose phthalate (HPMCP) microspheres (Man-THM) that contain ApxIIA subunit vaccine - an exotoxin fragment as a candidate for a subunit nasal vaccine against Actinobacillus pleuropneumoniae. For adjuvant activity, mucoadhesive thiolated HPMCP microspheres decorated with mannan could be targeted to the PRRs (pathogen recognition receptors) and mannose receptors (MR) of antigen presenting cells (APCs) in the respiratory immune system. The potential adjuvant ability of Man-THM for intranasal immunization was confirmed by in vitro and in vivo experiments. In a mechanistic study using APCs in vitro, it was found that Man-THM enhanced receptor-mediated endocytosis by stimulating the MR of APCs. In vivo, the nasal vaccination of ApxIIA-loaded Man-THM in mice resulted in higher levels of mucosal sIgA and serum IgG than mice in the ApxIIA and ApxIIA-loaded THM groups due to the specific recognition of the mannan in the Man-THM by the MRs of the APCs. Moreover, ApxIIA-containing Man-THM protected immunized mice when challenged with strains of A. pleuropneumoniae serotype 5. These results suggest that mucoadhesive Man-THM may be a promising candidate for a nasal vaccine delivery system to elicit systemic and mucosal immunity that can protect from pathogenic bacteria infection.         Presynaptic Regulation of Astroglial Excitatory Neurotransmitter Transporter GLT1

        <P><B>Summary</B></P><P>The neuron-astrocyte synaptic complex is a fundamental operational unit of the nervous system. Astroglia regulate synaptic glutamate, via neurotransmitter transport by GLT1/EAAT2. Astroglial mechanisms underlying this essential neuron-glial communication are not known. We now show that presynaptic terminals regulate astroglial synaptic functions, GLT1/EAAT2, via kappa B-motif binding phosphoprotein (KBBP), the mouse homolog of human heterogeneous nuclear ribonucleoprotein K (hnRNP K), which binds the GLT1/EAAT2 promoter. Neuron-stimulated KBBP is required for GLT1/EAAT2 transcriptional activation and is responsible for astroglial alterations in neural injury. Denervation of neuron-astrocyte signaling by corticospinal tract transection, ricin-induced motor neuron death, or neurodegeneration in amyotrophic lateral sclerosis all result in reduced astroglial KBBP expression and transcriptional dysfunction of astroglial transporter expression. Presynaptic elements dynamically coordinate normal astroglial function and also provide a fundamental signaling mechanism by which altered neuronal function and injury leads to dysregulated astroglia in CNS disease.</P>

      • A compact fourth-order finite difference scheme for the three-dimensional Cahn-Hilliard equation

        <P>This work extends the previous two-dimensional compact scheme for the Cahn-Hilliard equation (Lee et al., 2014) to three-dimensional space. The proposed scheme, derived by combining a compact formula and a linearly stabilized splitting scheme, has second-order accuracy in time and fourth-order accuracy in space. The discrete system is conservative and practically stable. We also implement the compact scheme in a three-dimensional adaptive mesh refinement framework. The resulting system of discrete equations is solved by using a multigrid. We demonstrate the performance of our proposed algorithm by several numerical experiments. (C) 2015 Elsevier B.V.         <P>Four new sesqui-lignans, (7R, 7'R, 7 '' S, 8S, 8'S, 8 '' S)-4',5 ''-dihydroxy-3,5,3',4 ''-tetramethoxy-7,9':7',9-diepoxy4,8 ''-oxy-8,8'-sesquineo-lignan-7',9 ''-diol (1), (7R, 7'R, 7 '' S, 8S, 8'S, 8 '' S)-4',3'-dihydroxy-3,5,3',5',4'pentamethoxy-7,9':7',9-diepoxy-4,8'-oxy-8,8'-sesquineo-lignan-7',9'-diol (2), (7R, 7'R, 7'S, 8S, 8'S, 8'S)-3',4'dihydroxy-3,5,4',5 ''-tetramethoxy-7,9':7',9-diepoxy-4,8'-oxy-8,8'-sesquineo-lignan-7',9'-diol (3) and acanthopanax A (7) together with three known compounds (4-6) were isolated from the EtOAc-soluble extract of Acanthopanax senticosus. Their structures were elucidated on the basis of spectroscopic and physicochemical analyses. All the isolates were evaluated for in vitro inhibitory activity against DGAT1 and DGAT2. Among them, compounds 1-6 were found to exhibit selective inhibitory activity on DGAT1 with IC50 values ranging from 61.1 1.3 to 97.7 1.1 111\4 and compound 7 showed selective inhibition of DGAT2 with IC50 value 93.2 1.2.         Raman scattering investigation of hydrogen and nitrogen ion implanted ZnO thin films

        Acceptor nitrogen and a donor–acceptor combination of hydrogen and nitrogen ions at 23 keV with fluences in the range from 1 × 1015 to 5 × 1016 ions cm-² have been implanted into ZnO films grown on Si substrate. DYNAMIC-TRIM calculations have been performed to obtain theoretical quantitative implantation profiles for various fluences. Ion beam analysis measurements confirmed the presence of implanted ions and were in agreement with the predicted concentrations. Raman spectroscopy was used to measure the characteristic vibrations of the various implanted ZnO/Si thin films. A correlation was found between the vibrational modes and the ion fluences. Silent B1 modes are observed due to disorder-activated Raman scattering in the as-grown films, indicating that structural disorder is present. The enhanced features seen in the co-implanted sample indicate that the disordered phase is increased.
