      • 補虛湯이 免疫機能에 미치는 影響

        For the purpose of investigate the immunoadjuvant effects of BoheuTang, BoheuTang was pretreated to Balb/c mice for 10 days with intraperitoneally. After 10 days, haptenic antigen (VCR-BSA, vincristine-bovine serum albumin) complexed with Freund's complete adjuvant was immunized. The immunoadjuvant were followed after 7 days of first immunization. The immunoadjuvant experimentswere as follows : 1. The antibody production in Balb/c mouse immunized with VCR-BSA was remarkably increased in the pretreated group of BoheuTang compared with saline treated(control) group. 2. The macrophage numbers in Balb/c mouse immunized with VCR-BSA was considerably increased about 2.9 times in the pretreated groups of BoheuTang compared with control group. 3. The lysozyme activity of macrophage in Balb/c mouse that was immunized with VCR-BSA was increased gradually in the pretreated groups of BoheuTang compared with control group. 4. In the BoheuTang pretreated group, B cell(spleen cell) proliferation was increased with a dose dependent manner. At 20μl/well of BoheuTang, B cell proliferation activity was most highest. 5. Onthe other hand, in the T cell(thymocyte) proliferation BoheuTang pretreated group had less considerable activity than in the case of B cell. These results may suggest that BoheuTang will have immunoadjuvant effects on the cells which concerned with immuno mechanism.

        This paper proposed a extracting method of the region for image using segmentation and edge information. First proposed algorithm extract information using canny edge detector and the image was divided by watershed segmentation. And it extract the image with edge information by merging region. Finally we compare the proposed method with levelset method. In the result proposed method not only extract the image with accurate region but also reduce operation time.

        Purpose : To evaluate the impact of added progestogen on biochemical markers of the bone turnover undergoing hormone replacement therapy in the postmenopausal women. Materials and Methods : There are 71 postmentopausal women who are divided into three groups (Group I, Group II, Group III). Group I(continuous combined)is given conjugated equine estrogen (CEE, 0.625mg, day 1-30) plus medroxyprogesterone acetate (MPA, 2.5mg, day 1-30), Group II(unopposed estrogen) is given conjugated equine estrogen (CEE, 0.625mg, day 1-30) plus medroxyprogesterone acetate (MPA, 5mg, day 1-12). And there were measured serum osteocalcin, serum total alkaline phosphatase and urinary deoxypyridinoline excretion before starting hormone replacement therapy and 12 months later. Results : The serum osteocalcin, serum total alkaline phosphatase and urinary deoxypyridinoline excretion significantly decreased after hormone replacement therapy (Group, I, II, III). And there was no significance in decreases in biochemical markers of the bone turnover among three groups. Conclusions : It was concluded that the impact of added progestogen on the biochemical markers of the bone turnover undergoing hormone replaced therapy in postmenopausal women was not significant than nothing added group. Thus we concluded that progestogen was not impact on decreasing on the biochemical markers of the bone turnover in postmenopausal women undergoing hormone replacement therapy.

        Thermal bum of pharynx and larynx can cause dysphagia and dyspnea which can be life threatening. Pain of larynx due to thermal bum does not subside sufficiently to drug therapy, Physiologic therapy. Here a case of acute epiglottitis with severe pain, which was successfully treated with superior laryngeal nerve block, is reported.

        Background : In recent years gonorrhoea has been panendemic and remains one of the most common Sexually Transmitted Diseases in the world, especially in developing countries. Objective & Method : For the detection of a more effective therapeutic regimen and assessing the prevalence of Penicillinase Producing Neisseria Gonorrhoeae(PPNG), Ive have been trying to study the patients who have visited the VD Clinic of Choong-Ku Public Health Center in Seoul since 1980 by means of the chromogenic cephalosporin method. Results : In 1994, 168 strains of N. gonorrhoeae were isolated, among which 109 (64,9% ) were PPNG. Conclustion : Our results suggests that the prevalence of PPNG in Seoul is still increasing.

      • Inhibitory effect of Angelica gigas on cold-induced RhoA activation in vascular cells

        <P>The herbal extract Angelica gigas (AG) has been applied as a vasodilating agent for patients suffering from vascular diseases for many years; however, the underlying mechanism has not been fully elucidated. The present study hypothesized that the anti-vasoconstrictive effect of AG may be effective in the treatment of abnormal cold-mediated vasospasms that occur in Raynaud's phenomenon (RP). The effect of AG on the activity of ras homolog gene family member A (RhoA) was investigated in cold-exposed vascular cells. Vascular cells were pretreated to AG, followed by a warm (37 degrees C) or cold (25 degrees C) incubation for 30 min and investigated with western blotting, ELISA and confocal microscopy. Cold treatment induced the activation of RhoA in pericytes and vascular endothelial cells, however this was reduced by treatment with AG. Furthermore, AG treatment reduced the endothelin-1 (ET-1)-mediated RhoA activation in pericytes; however, cold-induced ET-1 production by vascular endothelial cells was not affected by treatment with AG. In addition, AG treatment suppressed the formation of stress fibers and focal adhesion complexes, and the cold-induced phosphorylation of focal adhesion kinase, proto-oncogene tyrosine-protein kinase Src and extracellular signal-related kinase. Therefore, AG treatment demonstrated an ability to reduce cold-induced RhoA activation in pericytes and vascular endothelial cells, and attenuated ET-1-mediated RhoA activation in pericytes. In conclusion, the present study indicated that AG may be useful for the treatment of RP.</P>

        DEU-7 Derived from Ulmus macrocarpa Improved Immune Functions in Cyclophosphamide-treated Mice

        고량강(Alpinia officinarum), 산초(Zanthoxylum schinifolium), 황금(Scutellaria baicalensis Georgi), 왕느릅나무(Ulmus macrocarpa Hance), 네 종류의 식물성 한방 약재의 면역증강효능을 cyclophosphamide를 처리한 동물모델을 이용하여 조사하였다. 실험은 cyclophosphamide로 면역억제를 유도한 후 네 종류의 한방 약재를 식이하여 cyclophosphamide에 의해 억제된 면역인자의 회복 여부를 조사하였다. 네 종류의 한방 약재를 동일한 방법으로 열수 추출한 후 동일한 농도로 마우스에 처리하였다. 이 중 왕느릅나무 열수 추출물(DEU-7)이 cyclophosphamide의 영향으로 면역 억제된 마우스에서 면역인자 혈중 농도를 정상치 준하는 또는 가깝게 유지하는 것으로 나타내었다. DEU-7에 의해 비장의 무게와 비장세포수는 증가하였으나 간과 흉선과 같은 다른 장기에는 통계적으로 유의한 변화가 없었다. Ex vivo 조건에서 DEU-7은 비장세포의 사멸을 지연시키는 것으로 나타났다. 중요한 면역인자인 IL-2와 IL-4 cytokine은 면역억제로 농도가 감소되었으나 DEU-7이 약 2배와 3배 향상시키는 것으로 나타났으며, 이는 정상치의 약 1/2 수준이었다. IgM과 IgG의 농도는 cyclophosphamide 처리로 정상치의 약 1/2로 떨어졌으나, DEU-7에 의해서 정상치와 동일한 농도로 증가하였다. IL-2와 IL-4 결과에서 DEU-7은 T 림프구에 영향을 줄 수 있다고 생각되며, 또한 IgM과 IgG의 결과로서 B 림프구에 영향을 줄 수 있다고 생각된다. 선천성 면역에 중요한 면역인자인 TNF-α, IL-12과 IFN-γ 역시 cyclophosphamide에 의해 농도가 감소되었으나 DEU-7에 의해 정상치에 가깝게 회복되었다. 따라서, DEU-7은 면역 억제 또는 감소된 상태를 정상 상태로 회복 또는 유지하는 기능이 있는 것으로 생각된다. The present study investigated the immunomodulatory properties of four different medicinal plants in a cyclophosphamide-treated Balb/c mouse model. One of the four plants, Ulmus macrocarpa, showed partial resistance against immune suppression induced by cyclophosphamide. The bark of U. macrocarpa, commonly known as the Chinese elm, has been used as a pharmaceutical material in Korean traditional medicine to treat bacterial inflammation and induce wound healing. In this study, water extract of U. macrocarpa, named DEU-7, was used for its immunomodulating functional activity. DEU-7 increased the weight of the spleen and the number of splenocytes but did not significantly affect the liver, kidney, and thymus in vivo. A splenocyte viability assay confirmed that DEU-7 influenced ex vivo splenocyte survival. DEU-7 also increased the levels of cytokines, such as IL-2 and IL-4, and immunoglobulins, such as IgM, IgG, and IgA. These results indicated that DEU-7 is involved in the activation of T and B lymphocytes. In addition, DEU-7 was able to maintain the production of cytokines, such as TNF-α, IL-12, and IFN-γ, in the condition of cyclophosphamide-induced immune suppression, suggesting that DEU-7 activated innate immune cells, even under immune suppression. We concluded that DEU-7 aids immunological homeostasis, thereby preventing immune suppression, and aids both innate and adaptive immune response by maintaining the levels of various cytokines and immunoglobulins. Consequently, it is worth investigating the potential of DEU-7 as a supplemental source for immune-enhancing agents.

        Background: Cardiac dysfunction associated with brain hemorrhage was not well studied. We evaluated the incidence and characteristics of acute cardiac dysfunction related with acute brain hemorrhage. Methods: Between january and September in 2013, consecutive patient who were diagnosed with acute spontaneous or traumatic brain hemorrhage and admitted to surgical ICU were prospectively enrolled, ECG, cardiac enzyme, and echocardiography was considered as acute cardiac dysfunction related with brain hemorrhage when all the following criteria were satisfi ed. (1) Accompanied ECG change and abnormal cardiac enzyme level (2) No previous history of cardiac disease (3) Regional wall motion abnormality extending beyond a single coronary arterial distribution Otherwise, LV dysfunction was considered as cardiac dysfunction not related with brain hemorrhage. Clinical characteristics, laboratoy fi ndings, and in-hospital outcome were retrospectively reviewed. Results: Total of 107 patients (age : 59 ± 16 years, 64 men) were collected, LV systolic dysfunction on echocardiography was observed in 18 patients. Among them 11 (10%) patients were classifi ed as having acute cardiac dysfunction related with brain hemorrhage. while 5 patients in 11 patients with acute cardiac dysfunction showed typical apical balloning, 6 patients showed inverted takotsubo pattern. Other abnormalities were observed in ECG, cardiac enzyme level and echocardiography were shown in the table. In - hospital motality was observed in 19 (18%) patients. 6 patients in 11 patients with acute cardiac dysfuction had in-hospital motality (p= 0. 004). Conclusions: Acute cardiac dysfunction associated with acute brain hemorrhage was observed in 10% of patients and half of them showed inverted takotsubo pattern.

      • Effects of microalgal polyunsaturated fatty acid oil on body weight and lipid accumulation in the liver of C57BL/6 mice fed a high fat diet

        <P><B>Abstract</B></P><P>Dietary polyunsaturated fatty acids (PUFAs), which are abundant in marine fish oils, have recently received global attention for their prominent anti-obesogenic effects. Among PUFAs, eicosapentaenoic acid (EPA; 20:5n-3) and docosahexaenoic acid (DHA; 22:6n-3), which are n-3 long-chain PUFAs widely referred to as omega-3 oils, were reported to prevent the development of obesity in rodents and humans. In the present study, we evaluated the anti-obesity effects of microalgal oil on high-fat induced obese C57BL/6 mice, compared with commercial omega-3 fish oil and vegetable corn oil. Microalgal oil is an inherent mixture of several PUFAs, including EPA, DHA and other fatty acids produced from a marine microalgal strain of <I>Thraustochytriidae sp</I>. derived mutant. It was found to contain more PUFAs (>80%) and more omega-3 oils than commercial omega-3 fish oil (PUFAs >31%) and corn oil (PUFAs 59%). All three types of oils induced weight loss in high-fat-induced obese mice, with the loss induced by microalgal oil being most significant at 9 weeks (10% reduction). However, the oils tested did not improve blood lipid levels, although microalgal oil showed an apparent inhibitory effect on lipid accumulation in the liver. These findings may be attributed to the higher PUFA content, including omega-3 oils of microalgal oil than other oils. Collectively, these findings suggest that microalgal oil, derived from <I>Thraustochytriidae sp</I>. derived mutant, is a prominent candidate for replacement of omega-3 fish oils based on its apparent anti-obesity effect <I>in vivo</I>.</P>

        Obesity is one of the most serious diseases in modern society as energy intake and energy expenditure make imbalance. As treating the obesity becomes global issue, a number of treatments have been proposed and exercise is getting attention as a one way. It is well known that exercise has numerous benefits that affect whole body metabolism. Especially it can induce ‘browning’ of white adipocyte in adipose tissue. Cidea (Cell Death Inducing DFFA Like Effector A) is one of the disease associated genes including metabolic disorder and adipogenesis. It has been widely used for brown adipogenesis marker. We developed Cidea-dual reporter mouse (Cidea-P2A-Luc2-T2A-tdTomato, Luciferase/tdTomato) for applying in vivo tracing and measuring brown adipogenesis. Cidea-dual reporter mouse performed voluntary wheel running for 8 weeks. CL was injected intraperitoneally (1 mg/kg of body weight/day) for 3 days to Cidea-reporter mouse for inducing brown adipogenesis. Brown adipogenesis in Cidea-dual reporter mouse was measured not only by optical imaging system but also by histology. As a result of exercise, browning occurred depots in white adipose tissue become luminescent. We confirmed that exercise and CL can induce brown adipogenesis with in vivo reporter system. Cidea-dual reporter mouse will be widely used for detecting brown adipogenesis.
