        Selection and evaluation of reference genes for gene expression using quantitative real‐time PCR in Mythimna separata walker (Lepidoptera: Noctuidae)

        In order to precisely assess gene expression levels, the suitable internal reference genes must be served to quantify real‐time reverse transcription polymerase chain reaction (RT‐qPCR) data. For armyworm, Mythimna separata, which reference genes are suitable for assessing the level of transcriptional expression of target genes have yet to be explored. In this study, eight common reference genes, including β‐actin (β‐ACT), 18 s ribosomal (18S), 28S ribosomal (28S), glyceraldehyde‐3‐phosphate (GAPDH), elongation fator‐alpha (EF1α), TATA box binding protein (TBP), ribosomal protein L7 (RPL7), and alpha‐tubulin (α‐TUB) that in different developmental stages, tissues and insecticide treatments of M. separata were evaluated. To further explore whether these genes were suitable to serve as endogenous controls, three software‐based approaches (geNorm, BestKeeper, and NormFinder), the delta Ct method, and one web‐based comprehensive tool (RefFinder) were employed to analyze and rank the tested genes. The optimal number of reference genes was determined using the geNorm program, and the suitability of particular reference genes was empirically validated according to normalized HSP70, and MsepCYP321A10 gene expression data. We found that the most suitable reference genes for the different experimental conditions. For developmental stages, 28S/RPL7 were the optimal reference genes, both RPL7/EF1α were suitable for experiments of different tissues, whereas for insecticide treatments, 28S/α‐TUB were suitable for normalizations of expression data. In addition, 28S/α‐TUB were the suitable reference genes because they have the most stable expression among different developmental stages, tissues and insecticide treatments. Our work is the first report on reference gene selection in M. separata, and might serve as a precedent for future gene expression studies.

        Coefficient of Earth Pressure at Rest of a Saturated Artificially Mixed Soil from Oedometer Tests

        The present study focuses on the coefficient of earth pressure at rest (K0) of a saturated crushed binary soil mixture. The mixture isartificially mixed by crushed sandstone and mudstone particles according to presupposed weight ratios and particle size distributioncurves. The oedometer tests are performed to determine the coefficient K0. From test data, the values of coefficient K0, which rangefrom 0.242 to 0.381, with a mean value of 0.300, are obtained. Just as other mechanical parameters of soils, the K0 value is affectedby many factors. The effects of the properties of test specimen and material are discussed on the basis of the test data. Strong negativecorrelation between the values of K0 and initial dry bulk density of test specimen, one between the values of K0 and median particlesize diameter of test material, and one between the values of K0 and gravel content by weight of test material, respectively, areobserved from the test data. The mudstone particle content by weight of the mixture may also affect the value of K0. The effects ofthese factors on the K0 value may exhibit interlocking effect. Higher interlocking effect results in higher shear strength, and thereforeresults in lower K0 value.

        De novo assembly of the transcriptome for Greenbug (Schizaphis graminum Rondani) and analysis on insecticide resistance-related genes

        The greenbug, Schizaphis graminum Rondani is a major pest species of wheat crops. In this study, a transcriptome sequencing, and the expression of the 12 genes related to insecticide resistance were conducted in S. graminum. The sequencing and subsequent bioinformatics analysis outputed 46,593 unigenes, among which 28,289 unigenes were annotated to corresponding functions by blasting with high homologous genes in database, giving annotation rate of 60.72%. To gain insight into the mechanism of insecticide resistance, the expression of the 12 genes related to insecticide resistance for S. graminum was investigated. The expression level of aminopeptidase N (AN), cytochrome P450 (CYP), acetylcholinesterase 1 (AC), catalase (CAT), cytochrome c oxidas (CCC), GABA receptor (GABA), glutathione S-transferase (GST) were highest in the apterous nymphs among different developmental stages; The expression level of AN, CBL, CYP, CA, SD, and GST were relatively more abundant in the abdomen compared to head and throax. The results could give out the key information about the relationship between the expression of these genes in different developmental stages, tissues, treatments and metabolism of insecticides. These genes that were co-up-regulated significantly in third instar nymphs of S. graminum induced by imidacloprid, were consistent with their putative involvement in insecticide resistance. This provides most comprehensive transcriptome data for S. graminum to further study and managenment.

        Nasality Changes With Age in Normal Korean-Speaking Adults

        Objectives. This study was performed to investigate the effects of aging on nasality and the influence of age-related changes in nasal cavity volume and nasal patency on nasality. Methods. A total of 180 healthy Korean-speaking adult volunteers, who had no nasal or voice-related complaints, were enrolled in this study. Nasometry, acoustic rhinometry, and rhinomanometry were performed to obtain the nasalance score, nasal cavity volume, and nasal resistance, respectively. Changes in these parameters with age were analyzed. Results. Nasal cavity volume increased significantly, and nasal resistance decreased significantly, with age. The nasalance scores for the nasal passage and oronasal passage decreased significantly with age, while there were no age-related changes in nasalance scores for the oral passage. Conclusion. Nasalance scores for the passages containing nasal consonants decreased with age although significant increases were observed in nasal cavity volume and nasal patency with age. Therefore, the age-related decreases in nasalance scores may result from factors other than changes in the nasal cavity.

      • Differences in the mandibular premolar positions in Angle Class I subjects with different vertical facial types

        Objective: To compare the positions of the mandibular premolars in Angle Class I subjects according to vertical facial type. The results will provide a theoretical basis for predicting effective tooth movement in orthodontic treatment. Methods: Cephalometric parameters were determined using cone-beam computed tomography in 120 Angle Class I subjects. Subjects were categorized as short, normal, and long face types according to the Frankfort mandibular angle. Parameters indicating the position of the mandibular right premolars and the mandible were also measured. Results: The angle between the mandibular first premolar axis and buccal cortex, the distance between the root apex and buccal cortex, angle of vestibularization, arc of vestibularization, and root apex maximum movable distance were significantly greater in the short face type than in the long and norm face types. The angle between the mandibular second premolar axis and buccal cortex, the distance from root apex to buccal cortex, and the arc of vestibularization were significantly greater in the short face type than in the normal face type. Conclusions: There are significant differences in the mandibular premolar positions in Class I subjects according to vertical facial type.

        Clinicopathological aspects and prognostic value with respect to age: An analysis of 3,362 consecutive gastric cancer patients

        <B>Background and Objectives</B><P>Several studies have reported controversial results about clinicopathological features and prognoses in gastric cancer patients with respect to age, partly due to variable definitions of young age and inhomogeneity of the patient population. The aim of study was to analyze clinicopathological features and prognostic value of all stages of gastric cancer in a large consecutive series.</P><B>Methods</B><P>Between 2000 and 2005, 3,362 patients with all stages of gastric cancer were enrolled in database. Patients were divided into three groups: group 1 (≤45), group 2 (46–70), and group 3 (≥71).</P><B>Results</B><P>Upper location and linitis plastica were more frequent in group 1. Young patients had a higher proportion of poorly differentiated and signet ring cell type with elevated CA19-9 level. Depressed type was more frequent in early gastric cancer (EGC) while Bormann type IV and Lauren diffuse type were more common in AGC in group 1. In curatively resected patients, a 5-year survival rate was significantly higher in group 1 than older groups. Stage, vein invasion, curative resection, and CA19-9 level were significant prognostic factors in all gastric cancer.</P><B>Conclusions</B><P>Clinicopathological features associated with young gastric cancer include upper location, linitis plastica, histopathologically diffuse type, and unresectability.         Effects of pigment and citrinin biosynthesis on the metabolism and morphology of Monascus purpureus in submerged fermentation

        The effects of the secondary metabolite biosynthesison the metabolism and morphology of the Monascuspurpureus were investigated in this study. Hypha andseptum length became longer after deletion of genes pigRand pksCT in M. purpureus LQ-6 by Agrobacteriumtumefaciens-mediated transformation technology, highlybranched hyphae, much smaller and freely dispersedmycelial pellets were observed in M. purpureus. Comparedwith that in the wild-type, the level of intracellular NADHand NADPH was almost constant in M. purpureus DpigRat 4 days, but the NADH and NADPH levels decreased by1.58-fold and 3.71-fold in M. purpureus DpksCT. Thepresent study can not only provide a kind of strategy toimprove the Monascus pigments production, but also providetheoretical support for the further study of relationshipbetween the secondary metabolites, metabolism and morphologicalchange.

      • Streptomyces xinghaiensis sp. nov., isolated from marine sediment.

        <P>A novel actinomycete, strain S187(T), was isolated from a marine sediment sample collected from Xinghai Bay, Dalian, China. Growth occurred on ISP medium 2 containing 0-9 % NaCl and at pH 6.0-9.0 and 10-45 degrees C. The cell wall of strain S187(T) contained the isomer ll-diaminopimelic acid as the diagnostic diamino acid. The predominant menaquinones were MK-9(H(6)) (40.8 %), MK-9(H(8)) (38.2 %) and MK-9(H(2)) (8.8 %). The major fatty acids were iso-C(16 : 0) (29.6 %), anteiso-C(15 : 0) (14.0 %) and anteiso-C(17 : 0) (11.6 %). Cells contained phosphatidylethanolamine, phosphatidylinositol, phosphatidylglycerol, phosphatidylinositol mannosides and one unknown phospholipid. The G+C content of the genomic DNA was 72.01 mol%. The 16S rRNA gene sequence of the isolate had similarities of 98.1 and 97.5 % with those of Streptomyces flavofuscus NRRL B-8036(T) (=DSM 41426(T)) and Streptomyces albiaxialis DSM 41799(T), respectively, showing that the novel strain should be assigned to the genus Streptomyces. DNA-DNA hybridizations with the two above-mentioned Streptomyces species showed 31.4 and 46.9 % relatedness, respectively. Moreover, the three strains differed in some physiological and biochemical properties. Thus, on the basis of phenotypic and genotypic analyses, it is proposed that strain S187(T) represents a novel species of the genus Streptomyces, for which the name Streptomyces xinghaiensis sp. nov. is proposed; the type strain is S187(T) (=NRRL B-24674(T)=CCTCC AA 208049(T)=KCTC 19546(T)).</P>
