김치로부터 새로운 프로바이오틱균주인 Lactobacillus sakei proBio65를 분리하고 명명하였다. 형질전환 생쥐 (Foxp3-GFP KI 생쥐)를 이용하여 L. sakei proBio65의 면역조절 메커니즘 규명 및 면역 조절능을 확인하고 in vivo 적용 질환제어 응용 가능성을 평가하였다. 조절 T 세포의 master 전사조절인자로 알려진 Foxp3+를 선정하고, L. sakei가 Foxp3+전사조절인자를 증가시키는지 확인하기 위해 확립된 세포기반 screening system을 이용하였다. 항 염증성 사이토카인 전사조질인자의 증가에 이어 Foxp3+ 전자조절 인자발현의 상당한 증가를 확인하였다. L. sakei proBio65는 염증성면역 장애의 조절에 치료적으로 유용할 것이다. We isolated and identified a novel probiotic strain, Lactobacillus sakei proBio65 from Kimchi. To determine whether L. sakei proBio65 has an immunomodulatory effect, we investigated cells via an in vitro screening system which co-cultured freshly isolated mesenteric lymphocyte with probiotics. A significant increase of Foxp3+ transcription regulatory factor expression was observed, followed by an increase in anti-inflammatory cytokines transcription regulatory factor. L. sakei proBio65 exhibited high levels of the IL-10/IL-12 production ratio and enhanced Foxp3 expression in vitro. L. sakei proBio65 may thus be therapeutically useful for the modulation of inflammatory immune disorders.

We isolated Bacillus sp. SH-517 from decomposed chicken feathers at a local poultry plant. This strain produced a keratinase that degrades poultry feathers and therefore will be very valuable for industrial use. Most feathers were degraded by the strain within 40 h at $40^{\circ}C$ by shaking culture (180 rpm). The keratinase from the culture medium of Bacillus sp. SH-517 was purified by centrifugation, 30-80% ammonium sulfate fractionation, twin-column DEAE-cellulose ion exchange chromatography and Sephadex G-150 gel filtration, to obtain a purified keratinase at 10.82% yield and 14-fold overall purification. The purified enzyme had a specific activity of 825 U/mg. A single protein band was shown on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDSPAGE). The molecular weight of the keratinase from Bacillus sp. SH-517 was estimated as 51 kDa. The optimum pH and temperature for the enzyme reaction were 7.5 and $40^{\circ}C$, respectively. The enzyme remained stable over the pH range from 4.0 to 9.0 and at temperatures below $50^{\circ}C$. Proteins such as milk casein and chicken feathers were easily hydrolyzed by this enzyme. The enzyme activity was significantly inhibited by $Hg^{2+},\;Ag^{2+}$, ethylene diamine tetraacetic acid (EDTA) and ethylene glycol tetraacetic acid (EGTA), but slightly stimulated by $K^+$ and $Na^+$.
Serotype-Dependent Inhibition of Streptococcus pneumoniae Growth by Short-Chain Fatty Acids

Streptococcus pneumoniae (pneumococcus) is an opportunistic pathogen that can cause severe infectious diseases such as pneumonia, meningitis, and otitis media. Despite the availability of antibiotics and pneumococcal vaccines against some invasive serotypes, pneumococcal infection remains a tremendous clinical challenge due to the increasing frequency of infection by antimicrobial resistant, nonencapsulated, and/or non-vaccine serotype strains. Short-chain fatty acids (SCFAs), which are produced at various mucosal sites in the body, have potent antimicrobial activity, including inhibition of pathogen growth and/or bacterial biofilm formation. In this study, we investigated the antimicrobial activity of SCFAs (acetate, propionate, and butyrate) against various serotypes pneumococci. Propionate generally inhibited the growth of S. pneumoniae serotypes included in the pneumococcal conjugate vaccine (PCV) 13, except for serotypes 3 and 7F, though butyrate and acetate showed no or low inhibition, depending on the serotypes. Of note, butyrate showed strong inhibition against serotype 3, the most prevalent invasive strain since the introduction of the PCV. No SCFAs showed inhibitory effects against serotype 7F. Remarkably, the nonencapsulated pneumococcal strain had more sensitivity to SCFAs than encapsulated parental strains. Taken together, these results suggest that propionate showing the most potent inhibition of pneumococcal growth may be used as an alternative treatment for pneumococcal infection, and that butyrate could be used against serotype 3, which is becoming a serious threat.

Impaction of an ingesfed foreigm body in the colon is uncommon but surgical or endoscopic intervention is occasionally needed when serious complicafions such as perforation, obstruction or hemorrhage occur. Several factors may predispose a patient to ingestion and subsequent impaction of lower intestinal foreign body. It is more common in the elderly who wear dentures, in the mentally infirm, in chronic alcoholics or rapid eating. The majority of impaction occurs at narrowing and angulation such as ileocecal valve or rectosigmoid junction. The diagnosis of lower intestinal foreign body should be considered, among more common conditions, in the differential diagnosis of elderly patients who present with altered bowel habits. We report the case with a literature review that successfully endoscopically removed the fish bones impacted in the rectosigmoid colon complicated by rectal bleeding.

본 연구는 MOOC등 기존 이러닝 시스템에서 꾸준히 제기되고 있는 학습자들의 낮은 수료율 및 저조한 참여율, 협동성을 높일 수 있는 방안으로서 학습자 유형별 Push메세지 발송을 통한 협동학습 증진을 목적으로 한다. 이를 위해 소셜러닝 기반의 CLAS(Cooperative Learning Activate System)플렛폼을 제안한다. 소셜러닝은 기존 형식학습(formal learning)의 단점을 보완하고 지식정보사회의 새로운 학습모델로 개인의 능동성과 다른 학습자와의 협동성을 중요시하고 있다. 하지만 상용화된 SNS의 사용은 학습자 개개인의 성향을 파악하기 힘들며, 관리주체가 부재 한다는 문제점을 안고 있다. 이에 본 논문에서는 학습자 협동학습을 활성화 할 수 있는 소셜러닝 플랫폼을 통해 학습자들의 참여 및 협동성 향상을 높일 수 있는 소셜러닝 플랫폼을 제안하고자 한다.
