Lipid accumulation in macrophages is a prominent phenomenon observed in atherosclerosis. Previously, intimal foamy macrophages (FM) showed decreased inflammatory gene expression compared to intimal non-foamy macrophages (NFM). Since reprogramming of lipid metabolism in macrophages affects immunological functions, lipid profiling of intimal macrophages appears to be important for understanding the phenotypic changes of macrophages in atherosclerotic lesions. While lipidomic analysis has been performed in atherosclerotic aortic tissues and cultured macrophages, direct lipid profiling has not been performed in primary aortic macrophages from atherosclerotic aortas. We utilized nanoflow ultrahigh-performance liquid chromatography-tandem mass spectrometry to provide comprehensive lipid profiles of intimal non-foamy and foamy macrophages and adventitial macrophages from Ldlr−/− mouse aortas. We also analyzed the gene expression of each macrophage type related to lipid metabolism. FM showed increased levels of fatty acids, cholesterol esters, phosphatidylcholine, lysophosphatidylcholine, phosphatidylinositol, and sphingomyelin. However, phosphatidylethanolamine, phosphatidic acid, and ceramide levels were decreased in FM compared to those in NFM. Interestingly, FM showed decreased triacylglycerol (TG) levels. Expressions of lipolysis-related genes including Pnpla2 and Lpl were markedly increased but expressions of Lpin2 and Dgat1 related to TG synthesis were decreased in FM. Analysis of transcriptome and lipidome data revealed differences in the regulation of each lipid metabolic pathway in aortic macrophages. These comprehensive lipidomic data could clarify the phenotypes of macrophages in the atherosclerotic aorta.

Background: The ankle-brachial index is a useful tool to detect lower extremity peripheral artery disease, which is affected by exercise. The aging process causes degenerative vascular changes. The aim of this study was to elucidate the association between aging and changes in the ankle-brachial index after exercise. Methods: We analyzed data from ninety-nine patients who underwent ankle-brachial index tests and graded maximal aerobic exercise tests because of chest pain. The ankle-brachial index was measured before and after the exercise test. The correlation between age and changes in the ankle-brachial index after exercise was investigated using Pearson’s correlation coefficient. Results: The ankle-brachial index decreased after exercise, compared with that at rest (before vs. after: 1.11±0.06 vs. 1.05±0.08, P<0.001, 1.12±0.06, vs. 1.06±0.08, P<0.001; right and left, respectively). Upper-extremity systolic blood pressure did not change from baseline, and upper extremity diastolic blood pressure only increased slightly. However, the lower-extremity systolic and diastolic blood pressures decreased significantly. The correlation coefficients of the association between aging and changes in both the right and left ankle-brachial index indices were 0.319 and 0.255 (P=0.001 and P=0.011), respectively. Conclusion: A positive correlation was observed between aging and changes in the ankle-brachial index after exercise.
토양 수분함량 변화에 따른 용적전기전도도 변화와 포화침출액전기전도도의 상관관계

간척농지 염농도 관리를 위해서 염농도 자동계측기 설치가 확대되고 있다. 자동계측기에서는 용적전기 전도도(Bulk electrical conductivity, EC<SUB>b</SUB>)가 측정되는데, EC<SUB>b</SUB>는 토양 수분에 크게 영향을 받는다. 따라서, EC<SUB>b</SUB>를 토양 수분함량에 독립적인 포화침출액 전기전도도(Saturated-paste extract EC, EC<SUB>e</SUB>)로 환산할 필요가 있다. 본 연구에서는, 실내 토조실험을 통해 사양토와 미사질 양토를 이용하여 EC<SUB>b</SUB>와 EC<SUB>e</SUB>의 상관관계를 구명하였다. 토조에 세 가지 수준의 염농도로 조정된 토양을 충진하고 염농도 센서(Teros12)를 설치한 후 수분을 과포화 상태로 조정하여 각각의 토양에 대해 약 2주간 실험을 수행하였다. 사양토의 EC<SUB>e</SUB>는 4.5, 7.0, 10.5 dS m<SUP>-1</SUP> 였고, 미사질양토의 EC<SUB>e</SUB>는 4.3, 7.8, 11.4 dS m<SUP>-1</SUP> 였다. 시간이 경과함에 따라 토양 수분 함량은 증발에 의해 약 0.2 cm³ cm<SUP>-3</SUP>로 감소하였다. 수분 함량이 과포화에서 약 0.3 cm³ cm<SUP>-3</SUP> 수준으로 감소하는 동안 EC<SUB>b</SUB>는 염 농축에 의해 증가하였지만, 그 이후부터는 수분 함량이 감소함에 따라 이온 활동도가 감소하여 EC<SUB>b</SUB>도 감소하였다. EC<SUB>b</SUB>가 EC<SUB>e</SUB>에 가장 근접하게 측정되는 수분함량은 토양 염농도 수분별로 상이하였는데, 사양토의 경우 ECe는 4.5, 7.0, 10.5 dS m<SUP>-1</SUP>에서 각각 0.39, 0.39, 0.28 cm³ cm<SUP>-3</SUP> 였고, 미사질양토의 경우에는 ECe는 4.3, 7.8, 11.4 dS m<SUP>-1</SUP>에서 각각 0.48, 0.62, 0.50 cm³ cm<SUP>-3</SUP> 였다. 해당 수분 함량에서 EC<SUB>b</SUB>와 EC<SUB>e</SUB>는 매우 강한 상관관계를 보였는데, 회귀식은 사양토와 미사질양토가 각각 EC<SUB>b</SUB>=EC<SUB>e</SUB> 0.80 x + 1.3, EC<SUB>b</SUB>=EC<SUB>e</SUB> –0.27 x² + 4.7 x - 9.8 였다. 본 연구결과는 염농도 자동계측기에서 측정하는 EC<SUB>b</SUB>가 토양 수분함량이 높을 때는 염농도 희석, 그리고 수분함량이 낮을 때는 이온 활동도 감소에 의해 크게 변하는 것을 보여준다. 따라서, EC<SUB>b</SUB>가 EC<SUB>e</SUB>와 유사한 토양 수분 함량 조건에서만 EC<SUB>b</SUB>를 직접 활용할 수 있는 것으로 나타났다. 따라서, 향후 염농도 자동계측기의 활용도를 높이기 위해서는 토양수분함량에 따른 EC<SUB>b</SUB> 변화를 정량화하여 토양수분함량의 영향을 제거할 수 있는 방법 개발이 필요하다.
Marine Algal Flora on Goheung Coast, Korea

To understand the marine algal flora on Goheung coast, Korea, marine algae at 8 points were collected from November 2008 to February 2009 and from April to June 2009. Thirty-seven species (2 species of angiosperms, 5 chlorophytes, 12 phaeophytes, and 18 rhodophytes) occurred from fall to winter and 52 species (2 species of angiosperms, 9 chlorophytes, 18 phaeophytes, and 23 rhodophytes) occurred from spring to summer. Commonly occurring species were Ulva pertusa, Sargassum thunbergii, Hizikia fuziformis, and Gelidium amansii, and dominant species at most points were Ulva pertusa, Sargassum thunbergii, and Gelidium amansii. The average of the ratio of total rhodophytes and chlorophytes to phaeophytes ((R+C)/P) was 1.61 in fall to winter and 1.69 in spring to summer, and the average Laminariales/Fucales/Dictyotales (LFD) ratio was 1.14 in fall to winter and 1.18 in spring to summer. These results show that the marine algal flora of Goheung could be considered as temperate. The LFD ratio was fit for showing a feature of algal flora of Goheung. Species diversity index was high at Points 4~6 while low at Points 1 and 8. Detrended correspondence analysis (DCA) showed that the similarity of occurring species at Points 3 and 4 was higher than the other points from fall to winter, whereas the occurred species at Points 1~4 were similar from spring to summer. The average values of ecological evaluation index (EEI) of the investigation points were 6.8 from fall to winter and 6.3 from spring to summer, which means that the ecological environment of the investigation points were middle class and the EEI values of outer sea points were higher than the inner bay points.

For achieving a highly selective polishing rate between a tungsten (W) film and a silicon-dioxide (SiO2) film in W chemical-mechanical planarization (CMP), we designed W-film CMP slurry by mixing a small-molecule having two carboxylic functional groups (i.e., malic acid) as a selectivity agent. The selectivity was principally controlled by the malic acid concentration: it rapidly increased with increasing malic acid concentration in the W-film CMP slurry, and a W-film polishing rate:SiO2-film polishing rate of >100:1 was achieved. We found that the selectivity was mainly determined by the chemical properties, such as the corrosion and the chemical reaction between the films and malic acid rather than by the mechanical property, i.e., the electrostatic force between the ZrO2 abrasive and the films.

Heterologous expression of a putative K+/H+ antiporter of Streptomyces coelicolor A3(2) (designated as sha4) in E. coli and Streptomyces hygroscopicus JCM4427 showed enhanced tolerance to K+ stress, acidic-pH shock, and/or geldanamycin production under K+ stress. In a series of K+ extrusion experiments with sha4-carrying E. coli deficient in the K+/H+ antiporter, a restoration of impaired K+ extrusion activity was observed. Based on this, it was concluded that sha4 was a true K+/H+ antiporter. In different sets of experiments, the sha4-carrying E. coli showed significantly improved tolerances to K+ stresses and acidic-pH shock, whereas sha4-carrying S. hygroscopicus showed an improvement in K+ stress tolerance only. The sha4-carrying S. hygroscopicus showed much higher geldanamycin productivity than the control under K+ stress condition. In another set of experiments with a production medium, the secretion of geldanamycin was also significantly enhanced by the expression of sha4.
