        Prevention of Natural Flowering in Pineapple (Ananas comosus) by Shading and Urea Application

        The year-round production of pineapple (Ananas comosus var. comosus) is often interrupted by natural flowering during winter in Taiwan. A stable and promising technique for preventing natural flowering is required. In this study, we have tested the effect of shading and urea application on the flowering of pineapple. Shading 90% of sun light(s) before mid-November delayed the natural flowering of pineapple and the delay was affected by the plant age. For pineapples planted less than 11 month before mid-November, applying 1% urea plus shading treatment (SN) completely inhibited natural flowering. All the flowering-inhibited plants could be forced to flower by applying calcium carbide. Long-term shading might result in a decreased number of fruitlets within a fruit. However, 6 weeks of recovery before forcing flowering was sufficient for plants to produce fruits with quality equal to that of the control plants. In winter, pineapple plants that were prone to flowering tended to accumulate more leaf starch and increase their C/N ratio. The SN treatment increased the leaf nitrogen concentration and thus effectively maintained a low leaf C/N ratio. Furthermore, flowering-inhibited plants exhibited a constantly low level of leaf starch during the winter and their flowering forced by calcium carbide did not accompany with leaf starch accumulation.

      • Depression and Predictors in Taiwanese Survivors with Oral Cancer

        Background: Depression is a comorbid disabling problem and potentially affects patient likelihood of survival. The aims of this study were to recognize the characteristics of depression and investigate associated predictor factors in patients with oral cancers. Methods: A cross-sectional and correlational design was used to collect data for this study conducted in northern Taiwan. A set of questionnaires was used to measure depression, symptom distress, performance status, social support, and demographic and disease-related information. Logistic regression was conducted to determine important factors predicting depression. Results: A total of 132 oral cancer patients participated in this study. Of these, 18.2% were identified as depression cases. The patient average performance status score was 90 or higher. Patients reported mild-to-moderate levels of symptom distress. The majority of social support was from families. Religious belief, alcohol use, symptom distress, and social support from family were found to be important factors predicting depression. Patients with religious belief with alcohol use reported greater symptom distress, and those with lower levels of social support from families were significantly more likely to develop depression. Conclusions: Clinicians should assess patient emotional status and manage symptoms in a timely manner to enhance coping abilities. Supportive care provides assurance during the acute survivor phase.

        Safety and Efficacy of Adalimumab for Patients With Moderate to Severe Crohn`s Disease: The Taiwan Society of Inflammatory Bowel Disease (TSIBD) Study

        Background/Aims: Only moderate to severe Crohn`s Disease (CD) patients without a satisfactory conventional therapy effect are eligible to get reimbursement from the National Health Insurance of Taiwan for using adalimumab. These are more stringent criteria than in many Western countries and Japan and Korea. We aim to explore the efficacy of using adalimumab in CD patients under such stringent criteria. Methods: A retrospective analysis was conducted in nine medical centers in Taiwan and we collected the results of CD patients receiving adalimumab from Sep 2009 to Mar 2014. The clinical characteristics, response measured by CDAI (Crohn`s Disease Activity Index), adverse events and survival status were recorded and analyzed. CR-70, CR-100, and CR-150 were defined as attaining a CDAI decrease of 70, 100 or 150 points compared with baseline. Results: A total of 103 CD patient records were used in this study. Sixty percent of these patients received combination therapy of adalimumab together with immunomodulators. CR-70 was 68.7%, 74.5% and 88.4% after week 4, 8 and 12 of treatment, respectively. The steroid-free rate, complications and survival were 47.6%, 9.7% and 99% of patients, respectively. In considering the mucosal healing, only 25% patients achieve mucosal healing after treatment for 6 to12 months. Surgery was still needed in 16.5% of patients. Combination treatment of adalimumab with immunomodulators further decreased the level of CDAI at week 8 when compared with the monotherapy. Conclusions: Even under the stringent criteria for using adalimumab, the response rate was comparable to those without stringent criteria. (Intest Res 2014;12:287-292)

        The Effects of Tai Chi Exercise on Elders with Osteoarthritis: A Longitudinal Study

        Purpose Tai Chi exercise has been proven to be beneficial among elders with osteoarthritis (OA). The long-term effects of this exercise remain unclear. The purpose of this study was to evaluate the effects of the Tai Chi exercise for Arthritis (TCEA) program on the physical status and quality of life of OA elders. Methods This was a time series study with one group design. Subjects diagnosed with OA of the lower extremities, aged 60 years or over, were recruited from an outpatient clinic at a community teaching hospital. Thirteen participants joined a TCEA exercise class three times per week for 2 years. Physical status including body mass index (BMI), lean body mass, hand grasp strength, flexibility, and equilibrium were measured four times—at baseline, 3 months, 1 year, and 2 years of the TCEA exercise class. Quality of life was also measured at these time points. Results Lean body mass was significantly decreased within the study period (p< .05). Participants experienced significant improvements in physical functioning, role limitations, and social functioning on the dimensions of quality of life (SF-36) (p < .05). Conclusions Elderly people with OA should be encouraged to exercise using Tai Chi for maintaining physical function and improving quality of life.       • CFD simulation of compressible two-phase sloshing flow in a LNG tank

        Impact pressure due to sloshing is of great concern for the ship owners, designers and builders of the LNG carriers regarding the safety of LNG containment system and hull structure. Sloshing of LNG in partially filled tank has been an active area of research with numerous experimental and numerical investigations over the past decade. In order to accurately predict the sloshing impact load, a new numerical method was developed for accurate resolution of violent sloshing flow inside a three-dimensional LNG tank including wave breaking, jet formation, gas entrapping and liquid-gas interaction. The sloshing flow inside a membrane-type LNG tank is simulated numerically using the Finite-Analytic Navier-Stokes (FANS) method. The governing equations for two-phase air and water flows are formulated in curvilinear coordinate system and discretized using the finite-analytic method on a non-staggered grid. Simulations were performed for LNG tank in transverse and longitudinal motions including horizontal, vertical, and rotational motions. The predicted impact pressures were compared with the corresponding experimental data. The validation results clearly illustrate the capability of the present two-phase FANS method for accurate prediction of impact pressure in sloshing LNG tank including violent free surface motion, three-dimensional instability and air trapping effects.

        Platelet-Derived Growth Factor Receptor-α Subunit Targeting Suppresses Metastasis in Advanced Thyroid Cancer In Vitro and In Vivo

        Thyroid cancer is the most common endocrine malignancy. Patients with well-differentiated thyroid cancers, such as papillary and follicular cancers, have a favorable prognosis. However, poorly differentiated thyroid cancers, such as medullary, squamous and anaplastic advanced thyroid cancers, are very aggressive and insensitive to radioiodine treatment. Thus, novel therapies that attenuate metastasis are urgently needed. We found that both PDGFC and PDGFRA are predominantly expressed in thyroid cancers and that the survival rate is significantly lower in patients with high PDGFRA expression. This finding indicates the important role of PDGF/PDGFR signaling in thyroid cancer development. Next, we established a SW579 squamous thyroid cancer cell line with 95.6% PDGFRA gene insertion and deletions (indels) through CRISPR/Cas9. Protein and invasion analysis showed a dramatic loss in EMT marker expression and metastatic ability. Furthermore, xenograft tumors derived from PDGFRA geneedited SW579 cells exhibited a minor decrease in tumor growth. However, distant lung metastasis was completely abolished upon PDGFRA gene editing, implying that PDGFRA could be an effective target to inhibit distant metastasis in advanced thyroid cancers. To translate this finding to the clinic, we used the most relevant multikinase inhibitor, imatinib, to inhibit PDGFRA signaling. The results showed that imatinib significantly suppressed cell growth, induced cell cycle arrest and cell death in SW579 cells. Our developed noninvasive apoptosis detection sensor (NIADS) indicated that imatinib induced cell apoptosis through caspase-3 activation. In conclusion, we believe that developing a specific and selective targeted therapy for PDGFRA would effectively suppress PDGFRA-mediated cancer aggressiveness in advanced thyroid cancers.
