        Potential Control of Foodborne Pathogenic Bacteria by Pediococcus pentosaceus and Lactobacillus graminis Isolated from Fresh Vegetables

        The consumption of fresh vegetables has been related to recurrent outbreaks of foodborne diseases (FBD) worldwide. Therefore, the development of effective alternative technologies is necessary to improve the safety of these products. This study aimed to isolate and identify epiphytic lactic acid bacteria (LAB) from fresh fruits and leafy vegetables and characterize their antagonistic capacity due to their ability to produce bacteriocins or antibacterial compounds. For this, 92 LAB isolates from fruits and leafy vegetables were screened for antagonistic activity. Two strains with the highest and broadest antagonistic activities were selected for further characterization; one from cantaloupe melon (strain CM175) and one from cilantro leaves (strain C15). The cell-free supernatants (CFS) of CM175 and C15 were found to exhibit antagonistic activity against FBD-causing pathogens. The CM175 and C15 strains were identified as Pediococcus pentosaceus and Lactobacillus graminis, respectively. Notably, the P. pentosaceus CM175 CFS stopped the growth of Salmonella Typhimurium, Salmonella Saintpaul, Staphylococcus aureus, and Listeria monocytogenes, and delayed Escherichia coli O157:H7 growth. Moreover, L. graminis C15 CFS delayed the growth of all indicator pathogens, but did not completely stop it. Organic acids and bacteriocin-like molecules were determined to be possibly exerting the observed antagonistic activity of the identified LAB strains. Thus, application of the antagonistic compounds produced by Pediococcus pentosaceus and Lactobacillus graminis could be a novel and ecological strategy in developing antimicrobial biopreservatives for the food industry and mitigating FBD by reducing the biological contamination in fruit and vegetable orchards, mainly via their potential in controlling both gram-negative and gram-positive pathogenic bacteria.

        The successful development of advanced nuclear systems for sustainable energy production and nuclear waste management depends on high quality nuclear data libraries. Recent sensitivity studies and reports [1][2][3] have identified the need for substantially improving the accuracy of neutron cross-section data for minor actinides. The n_TOF collaboration has initiated an ambitious experimental program for the measurement of neutron capture cross sections of minor actinides. Two experimental setups have been constructed for this purpose: a Total Absorption Calorimeter (TAC) [4] for measuring neutron capture cross-sections of low-mass and/or radioactive samples and a set of two low neutron sensitivity C_6D_6 detectors for the less radioactive materials.

        Impact of radiotherapy on mandibular bone: A retrospective study of digital panoramic radiographs

        Purpose: The purpose of this study was to investigate the impact of radiotherapy on mandibular bone tissue in head and neck cancer patients through an analysis of pixel intensity and fractal dimension values on digital panoramic radiographs. Materials and Methods: Thirty patients with radiographic records from before and after 3-dimensional (3D) conformational radiotherapy were selected. A single examiner carried out digital analyses of pixel intensity values and fractal dimensions, with the areas of interest unilaterally located in the right angle medullary region of the mandible below the mandibular canal and posterior to the molar region. Results: Statistically significant decreases were observed in the mean pixel intensity (P=0.0368) and fractal dimension (P=0.0495) values after radiotherapy. Conclusion: The results suggest that 3D conformational radiotherapy for head and neck cancer negatively affected the trabecular microarchitecture and mandibular bone mass.

        BCL2 antagonist of cell death kinases, phosphatases, and ovarian cancer sensitivity to cisplatin

        Objective: The BCL2 family proteins are critical mediators of cellular apoptosis and, as such, have been implicated as determinants of cancer cell chemo-sensitivity. Recently, it has been demonstrated that the phosphorylation status of the BCL2 antagonist of cell death (BAD) protein may influence ovarian cancer (OVCA) cell sensitivity to cisplatin. Here, we sought to evaluate how kinase and phosphatase components of the BAD apoptosis pathway influence OVCA chemo-sensitivity. Methods: Protein levels of cyclin-dependent kinase 1 (CDK1) and protein phosphatase 2C (PP2C) were measured by immunofluorescence in a series of 64 primary advanced-stage serous OVCA patient samples. In parallel, levels of cAMP-dependent protein kinase (PKA), AKT, and PP2C were quantified by Western blot analysis in paired mother/daughter platinum-sensitive/resistant OVCA cell lines (A2008/C13, A2780S/A2780CP, Chi/ChiR). BAD pathway kinase CDK1 was depleted using siRNA transfection, and the influence on BAD phosphorylation and cisplatin-induced apoptosis was evaluated. Results: OVCA patient samples that demonstrated complete responses to primary platinum-based therapy demonstrated 4-fold higher CDK1 (p<0.0001) and 2-fold lower PP2C (p=0.14) protein levels than samples that demonstrated incomplete responses. Protein levels of PP2C were lower in the platinum-resistant versus that shown in the platinum-sensitive OVCA cell line sub-clones. Levels of PKA were higher in all platinum-resistant than in platinum-sensitive OVCA cell line sub-clones. Selective siRNA depletion of CDK1 increased sensitivity to cisplatin-induced apoptosis (p<0.002). Conclusion: BAD pathway kinases and phosphatases, including CDK1 and PP2C, are associated with OVCA sensitivity to platinum and may represent therapeutic opportunities to enhance cytotoxic efficacy.         Calcium Digestibility and Metabolism in Pigs

        Calcium (Ca) and phosphorus (P) are minerals that have important physiological functions in the body. For formulation of diets for pigs, it is necessary to consider an appropriate Ca:P ratio for an adequate absorption and utilization of both minerals. Although both minerals are important, much more research has been conducted on P digestibility than on Ca digestibility. Therefore, this review focuses on aspects that are important for the digestibility of Ca. Only values for apparent total tract digestibility (ATTD) of Ca have been reported in pigs, whereas values for both ATTD and standardized total tract digestibility (STTD) of P in feed ingredients have been reported. To be able to determine STTD values for Ca it is necessary to determine basal endogenous losses of Ca. Although most Ca is absorbed in the small intestine, there are indications that Ca may also be absorbed in the colon under some circumstances, but more research to verify the extent of Ca absorption in different parts of the intestinal tract is needed. Most P in plant ingredients is usually bound to phytate. Therefore, plant ingredients have low digestibility of P due to a lack of phytase secretion by pigs. During the last 2 decades, inclusion of microbial phytase in swine diets has improved P digestibility. However, it has been reported that a high inclusion of Ca reduces the efficacy of microbial phytase. It is possible that formation of insoluble calcium-phytate complexes, or Ca-P complexes, not only may affect the efficacy of phytase, but also the digestibility of P and Ca. Therefore, Ca, P, phytate, and phytase interactions are aspects that need to be considered in Ca digestibility studies.

        Laboratory and field performance of Trichopria drosophilae reared on Drosophila melanogaster and D. Suzukii

        Two colonies of the parasitoid Trichopria drosophilae (Perkins) (Hymenoptera: Diapriidae) are reproduced using a mass rearing approach in Mexico. The first colony uses the worldwide commercial host Drosophila melanogaster Meigen (Diptera: Drosophilidae) reared on an artificial diet (=T. drosophilae-Dm), while the second colony uses D. suzukii Matsumura reared on fresh banana (=T. drosophilae-Ds). Since theoretically the type of colony host affects parasitoid field performance, we conducted concatenated laboratory and field studies of both parasitoid lines. In the laboratory trials, T. drosophilae-Ds exhibited greater longevity, larger adult size, and higher fecundity than T. drosophilae-Dm. Both lines of parasitoids oviposited indistinctly in three drosophilid species commonly found in the berry zones of Colima: D. melanogaster, D. suzukii, and Zaprionus indianus Gupta. In the field trials, T. drosophilae-Ds had a larger population size and provided 2.4-fold higher pest reduction compared to T. drosophilae-Dm. Under the particular conditions of these field trials (berry fields separate from each other ≤ 3 km, and intensive agronomic management), these results demonstrate that T. drosophilae-Ds can significantly contribute more to the integrated management of D. suzukii than when reared on the commercial host D. melanogaster.

        Advanced Paternal Age and Sperm DNA Fragmentation: A Systematic Review

        Purpose: Male ageing is often associated with defective sperm DNA remodeling mechanisms that result in poorly packaged chromatin and a decreased ability to repair DNA strand breaks. However, the impact of advanced paternal age on DNA fragmentation remains inconclusive. The aim of the present systematic review was to investigate the impact of advancing paternal age (APA) on DNA fragmentation. Materials and Methods: We conducted a thorough search of listed publications in Scopus, PubMed, and EMBASE, in accordance with the PRISMA guidelines. Results: We identified 3,120 articles, of which nineteen were selected for qualitative analysis, resulting in a sample of 40,668 men. Of the 19 articles evaluating the impact of APA on DFI% (DNA fragmentation Index) included, 4 were on Normozoospermic and subfertile men, 3 on normozoospermic, Oligoasthenoteratozoospermic and Teratozoospermic, 6 on fertile and infertile men, 4 on just infertile men, and 2 evaluated a general population. Seventeen of the ninrnteen studies demonstrated APA’s effect and impact on DFI%. Conclusions: Although there was no universal definition for APA, the present review suggests that older age is associated with increased DFI. In elderly men with normal semen parameters, further studies should be performed to assess the clinical implications of DFI, as a conventional semen analysis can often fail to detect an etiology for infertility.

        Negative magnetoresistance in sputtered niobium thin films grown on silicon substrates

        Nb-films (thicknesses of 20 nm, 50 nm and 100 nm), directly grown on orientated Si substrates by DC sputtering, have shown reduced superconducting critical temperatures TC as compared to the bulk value; an effect associated with their disordered granular character (the disorder phenomenon of the Nb-films depresses the density of states at the Fermi level, consequently shifts the TC value towards low temperatures). The disorder effect was also correlated to the semiconducting-like behavior observed in the RðTÞ measurements. At the normal state, while the 100 nm Nb-film is dominated by a metallic-like behavior, the thinner Nb-films (20 nm and 50 nm thick) show significant negative magnetoresistances in a small temperature range about TC. This behavior was also explained assuming their granular characters, where the applied magnetic field first destroys the global superconducting character of the Nb-films, leaving Cooper pairs localized inside Nb-grains. A further increase of the applied field strength affects the superconductivity inside Nb-grains, enhancing the normal intergranular electric transport channel, which decreases the whole resistance of the Nb-films. The present study suggests that the microscopic disorder, at the grain surfaces/interfaces, seems to be an essential point to comprehend the negative magnetoresistance effect observed in some superconducting granular systems.
