An asymmetric synthetic route for 1-iodofarnesane, a key intermediate for tocopherol side chain synthesis, starting from (+)-(R)-citronellal was developed. 1-lodofarnesane was prepared through eight steps in about 50% overall yield, and asymmetric transfer hydrogenation of the enal with chiral organocatalyst was conducted as a stereoinduction step. To measure the stereoinduction level and optical purity of the product, a convenient analytical method was developed in which a phenylcarbamate derivative of the C15 alcohol was found to be suitable to give proper polarity and UV-activity for chiral UV-HPLC analysis. ⓒ2014 Elsevier Ltd. Collagenous colitis is characterized clinically by chronic watery diarrhea and pathologically by increased subepithelial collagen deposition associated with an inflammatory infiltrate in the lamina propria. Its etiology is still unclear, although a variety of associated diseases such as rheumatic syndromes, scleroderma, and thyroid diseases have been reported. We report a case of collagenous colitis following the prolonged use of NSAIDs. A 72-year-old woman who has taken NSAIDs for many years due to some dermatologic problems was admitted to the hospital because of chronic watery diarrhea and colicky abdominal pain of 3 months duration. There was no abnormal physical finding except cachectic appearance due to weight loss of 10㎏ during 3 months. Stool examination for ova and parasites and fat was negative, and stool culture for bacterial pathogens was negative. In complete blood count, there were relative eosinophila and mild anemia. Total serum protein and albumin was low, and thyroid function, RA factor, FANA were all normal. Results of upper and lower gastrointestinal contrast radiographs were normal. Sigmoidoscopy revealed normal colonic mucosa but she had a thick subepithelial collagenous deposit and chronic inflammation in lamina propria on colonic biopsy. Based on the above findings, she was diagnosed as collagenous colitis. Diarrhea improved after withdrawing NSAIDs and the treatment with oral prednisolone. In the post-treatment biopsy, the thickness of the collagen hand was diminished. Collagenous colitis is now recognized as one of the common causes of chronic diarrhea of obscure origin and NSAIDs may play an etiological role in some patient with collagenous colitis.

Objective: To evaluate the efficacy of ultrasound-guided sclerotherapy with 95% ethanol in the treatment of endometrioma in terms of ovarian reserve. Methods: We retrospectively reviewed the records of 67 patients with ovarian endometrioma, 32 of whom were treated with sclorotherapy and 35 with laparoscopic cystectomy. Results: Clinical characteristics including patient age, duration of infertility, cyst location, and symptoms showed no significant difference between the two groups. Changes in AMH level were smaller in the sclerotherapy group compared to laparoscopic cystectomy group (sclorotherapy group: 1.24 ± 1.4, laparoscopic cystectomy group: 1.96 ± 1.9). However, the difference was not statistically significant. Conclusion: Ultrasound-guided sclerotherapy with 95% ethanol is a safe treatment for ovarian endometriosis, providing relief if related symptom while preserving healthy ovarian tissue and avoiding early menopause.

We recently reported that 6-aminoalkyl-2,4,5-trimethylpyridin-3-ols, novel series of 6-aminopyridin-3-ol-based antioxidants, have high antiangiogenic activities. In pursuit of wider variety in the analogues, we here report the synthesis and antiangiogenic activities of 6-amidoalkyl-2,4,5-trimethylpyridin-3-ols, which would not be considered excellent antioxidants because of the poorer electron-donating effect of the C(6)-amido group than the corresponding C(6)-amino group. The selected 6-amido compounds showed up to several fold-higher antiangiogenic activities and up to an order of magnitude better antitumor activities in the chick embryo chorioallantoic membrane (CAM) assay than SU4312, a positive control. We also found that paracetamol, as a direct phenolic analogue of our simplest 6-amidopyridin-3-ol, showed a moderate level of antiangiogenic activity. We propose this study will offer a basis for a scaffold of novel angiogenesis inhibitors that can perturb angiogenesis-related pathologies.ⓒ2014Elsevier Ltd. All rights reserved.

<P><B>Background:</B></P><P>Anoctamin (ANO)/transmembrane member 16 (TMEM16) proteins mediate diverse physiological and pathophysiological functions including cancer cell proliferation. The present study aimed to identify the role of ANOs in pancreatic cancer.</P><P><B>Methods:</B></P><P>In an initial screen of ANOs, ANO9/TMEM16J was overexpressed in pancreatic cancer cells, and its role in the pathogenesis of pancreatic cancer was evaluated using an integrated <I>in vitro</I> and <I>in vivo</I> approach. To determine clinical relevance of the experimental findings, the prognostic value of ANO9 was evaluated in patients with pancreatic cancer.</P><P><B>Results:</B></P><P>The ANO9 mRNA and protein levels were increased in pancreatic cancer-derived cells. Exogenous expression of <I>ANO9</I> in PANC-1 cells significantly increased cell proliferation in cell cultures and in mice. In contrast, knockdown of <I>ANO9</I> in AsPC-1, BxPC-3, and Capan-2 cells strongly inhibited cell proliferation. Mechanistic analysis suggested that physical association of ANO9 with epidermal growth factor receptor (EGFR) underlies ANO9-induced cell proliferation. Knockdown <I>of ANO9</I> augmented the effects of the EGFR inhibitor and the cytotoxic agent on pancreatic cancer cell proliferation. In addition, high ANO9 expression is a poor prognostic factor in patients with pancreatic cancer.</P><P><B>Conclusions:</B></P><P>The ANO9/TMEM16J appears to be a clinically useful prognostic marker for pancreatic cancer and a potential therapeutic target.</P>
