      • MCT 대체 마요네즈의 물성적 특성 및 기호도에 관한 연구

        The study was carried on the rheological properties and sensory evaluations of mayonnaise substitued by MCT. Nowdays we are consideration of the fat replacer and fat substitutes. It was known that fat and oils had become causes for cardiac disease, obesity and cancer, so the new materials were needed for fat replacer. MCT(medium chain triglyceride) is one of the fat bsed substitutes used as fat replacer. In case of making of mayonnaise substituted by MCT, springiness of mayonnaise prepared with SO substituted by over 20% MCT increased highly, cohesiveness of mayonnaise made of 10-20% MCT substitution was similar to that made without MCT. And hardness, gumminess, and chewiness of mayonnaise prepared with 10-30% MCT substitution were similar or increased more or less to that made with SO only. In the result of color measurement by colorimeter brightness of that made with SO substituted by 10-30% MCT was similar to that made with SO only, redness was almost same without differences significantly in all samples with or without MCT, but yellowness was decreased highly over 10% MCT substitution. By the results of sensory evaluations, appearance, color, and flavor of cakes prepared with CO substituted by MCT until 100% were shown to be better, and taste was better in range of 30% MCT substitution, but texture had lower acceptability than those of cake made without MCT. While in case of mayonnaise, appearance, color, flavor, texture, and taste were obtained worse score, overall acceptance was similar in the range of 10% MCT substitution but was decreased highly upon MCT substitution ratios increase over 20% substitution than that made with SO only. Respecting that the necessity of fat substitute become real situation in dietary in the future, it will be suggested that many foods containing fats and oils will substitute by MCT and another fat replacer.

      • HBV : Effects of Pre-S Mutations of Hepatitis B Virus on Hepatitis B Surface Antigen Expression

        Background: In chronic hepatitis B virus (HBV) infection, quantitative HBV surface antigen (qHBsAg) level for monitoring viral replication and treatment responses has been highlighted. However, whether mutations in pre-S region have any effect on qHBsAg levels remains uncertain. We aimed to determine patterns of HBsAg expression in naturally-occurring mutations in the pre-S region. Methods: Plasmids expressing 1-8 amino acid deletion in pre- S1 (“pre-S1 mutant”) and 3-25 amino acid deletion in pre-S2 (“pre-S2 mutant”) were constructed. At 72h post-transfection into Huh-7 cells, qHBsAg levels were analyzed in supernatant and cell lysate using electrochemiluminescence immunoassay analyzer. Results: Extracellular qHBsAg levels were on average 3.87-fold higher in cells transfected with pre-S1 mutant compared to wild-type. In contrast, those in cells transfected with pre-S2 mutant were slightly lower compared to wild-type (on average 0.92-fold lower, but without statistical significance). Conversely, compared to wild-type, intracellular qHBsAg levels were on average 0.57-fold lower in pre-S1 mutant, whereas those were on average 1.60-fold greater in pre-S2 mutant. Consistent with these results, immunofluorescence staining of cellular HBsAg showed that cells transfected with pre-S1 mutant were stained to a lesser degree and that those with pre-S2 mutant were more densely stained. Co-transfections of wild-type with either pre- S1 mutant or pre-S2 mutant were performed to mimic milieus of quasispecies at the following ratios; 0:10, 1:9, 3:7, 5:5, 7:3, 9:1, and 10:0. As ratios of either pre-S1 mutant or pre-S2 mutant:wild type increased from 0:10 to 10:0, relative extracellular qHBsAg levels increased accordingly from 1.0 to 3.85 in pre-S1 mutant co-transfection, whereas those decreased from 1.0 to 0.88 in pre-S2 mutant co-transfection. Conclusions: Pre-S deletion exhibit different phenotypes regarding HBsAg expression and these effects depend on precise locations of deletion. Thus, qHBsAg level for diagnosis and prognostification should be used more cautiously, considering the emergence of pre-S deletion mutants.

        핀테크 사용에 대한 정보프라이버시 염려와 이용자 저항에 대한 연구

        최근 금융과 IT가 결합된 핀테크에 대한 관심이 증가하고 있다. 현재 핀테크 서비스는 확산 초기 단계에 있으며, 사용 확산을 위한 이슈로서 정보 보안 이슈가 대두되고 있다. 본 연구에서는 핀테크 서비스 실 사용자들이 인지하는 정보프라이버시 염려를 중심으로 사용 저항 정도를 분석하고, 이들에 영향을 주는 요인들로 사회적 영향 정도와 모바일 인터넷 활용 능력을 분석하였다. 특히, 본 연구에서는 사용자의 조절초점성향과 정보프라이버시 염려의 상호작용이 사용자 저항에 미치는 효과를 검증하였다. 이를 통해, 핀테크 서비스에 대한 저항 감소 및 사용 확산을 위한 전략적 시사점을 제공하고자 한다. Recently, there is a growing interest in Fintech which is a business based on using IT to provide financial services. At the early stage of diffusion of Fintech, one of the issues raised is the concern for information security. This study examines actual users’ information privacy concern and user resistance to Fintech services. We also investigates the effects of social influence and mobile Internet literacy on information privacy concern and user resistance. In particular, this research verifies the interaction effect between regulatory focus and information privacy concern to influence user resistance. Based on the findings, the results of this study provide strategic implications for firms which attempt to reduce user resistance to Fintech services and proliferate Fintech usage.

        상피세포 성장인자의 경피흡수 : 정상피부 , 각질제거피부 및 화상피부에 있어서

        In vivo and in vitro skin permeation of recombinant ^(125)I-EGF through normal. stripped and the first degree burn skin were studied. The in vitro skin permeation rate through the first degree burn skin (296 cpm/㎠/hr) and the stripped skin (1131 cpm/㎠/hr) were 3.5 times and 13 times higher, respectively, as compared with the one through normal skin. In vivo absorption study with the first degree burn skin, the peak concentration of EGF in the skin was achieved at 1-3 hr and decreased afterward up to 8 hr with an elimination constant of 1.31 × 10³g/㎖/hr. To investigate the higher elimination rate of EGF in burn skin, binding and metabolism studies were conducted. No significant metabolism of EGF in burn skin (100℃, 5second burning) was observed. With the presence of unlabelled-EGF, ^(125)I-EGF permeation through the burn skin showed higher permeation rate than the one without unlabelled-EGF. The result may indicate that EGF-receptor binding play a role in determining the skin permeation rate.

        Sitagliptin attenuates metformin-mediated AMPK phosphorylation through inhibition of organic cation transporters

        <OL><LI><P>To assess potential interactions between sitagliptin and metformin, we sought to characterize the <I>in vitro</I> inhibitory potency of sitagliptin on the uptake of MPP<SUP>+</SUP> and metformin, representative substrates for OCTs, and to evaluate the pharmacological pathways that may be affected by the combination of metformin and sitagliptin.</P></LI><LI><P>Among the OATs and OCTs screened, OAT3-mediated salicylate uptake and OCT1- and OCT2-mediated MPP<SUP>+</SUP> uptake were inhibited by sitagliptin. The <I>K</I><SUB>i</SUB> values of sitagliptin for OCT1- and OCT2-mediated metformin uptake were 34.9 and 40.8 μM, respectively.</P></LI><LI><P>As OCT1 is the gate protein for metformin action in the liver, we investigated whether sitagliptin-mediated OCT1 inhibition affected metformin-induced activation of AMPK signalling. Treatment with sitagliptin in MDCK-OCT1 and HepG2 cells resulted in a reduced level of phosphorylated AMPK, with <I>K</I><SUB>i</SUB> values of 38.8 and 43.3 μM, respectively.</P></LI><LI><P>These results suggest that the inhibitory potential of sitagliptin on OCT1 may attenuate the first step of metformin action, that is, the phosphorylation of AMPK. Nevertheless, the likelihood of a drug-drug interaction between sitagliptin and metformin is believed to be remote in usual clinical setting.</P></LI></OL>

      • Residential green environments and pregnancy outcomes: systemic review and meta-analysis

        Objective: In modern society people live in large cities is important to consider the social needs of site selection and design for green spaces related with planning for environmental medicine and public health. Although the majority of published studies investigated health effect related to the cumulative greenery and air pollutions, but recently the public health community started to take a closer look at the associations between residential greenery and fetal development and pregnancy outcomes. In utero exposures to prenatal environment with air pollution exposure and greenery is an important period of susceptibility to health effects including risk of low birth weight, preterm birth, and preeclampsia. However, the modern urban-ecological systems associated with prenatal environment is limited. This study was aimed the answer the question whether living environment including green spaces and air pollutions during pregnancy are associated with adverse pregnancy outcome and what the direction of that effect is, and to propose some new methodological approaches. Methods: MEDLINE, EMBASE, Cochrane Library, K-eArticles and CINAHL databases were searched for relavant publications in English from inception up to May 2019, based on the following free-text term key- words: population (pregnancy or pregnant women), intervention (green space, parks, forest and greenness), and outcome (pregnancy outcome and birth outcome). Data were analyzed as risk ratios (RR) or odds ratios (OR), and effects were adjusted for clustering. Meta-analyses were performed using random-effects and evidence quality was assessed. For the quality assessment, Risk of Bias Assessment tool for Non-randomized Studies for observational studies were used. Results: Out of 89 records, ten studies were identified and included in the analyses. We report quality effects meta-analyses based on correlation and standardized regression coefficients as estimates of effect size. Living greenness environment within 100-m, 250m, 500m buffer was weakly and positively associated with birth weight even though air quality and civilization considered. Heterogeneity was high (I 2 = 88%) and under the quality effects model the meta-analysis produced significant pooled regression slope of 0.0016(95%CI: 0.0007, 0.0025). Conclusion: In this study even if urbanization intensifies, greenness had positive effect on pregnancy outcomes, and this study endorses the importance of green space management and planning for environmental medicine and public health future research.

      • A novel anti-melanogenic agent, KDZ-001, inhibits tyrosinase enzymatic activity

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>The demand for anti-melanogenic agents is increasing due to the unwanted side effects of current treatments. To find an effective anti-melanogenic agent, we used zebrafish as a whole animal model for phenotype-based drug and cosmetic discovery screening.</P> <P><B>Objectives</B></P> <P>The aim of this study was to identify and explore a small molecule that could be used for skin-whitening cosmetics.</P> <P><B>Methods</B></P> <P>Using zebrafish embryos, we examined the effects of 1000 compounds on zebrafish development and pigmentation. Pigmentation production was assessed by tyrosinase (TYR) enzymatic activity and melanin contents. Pigmentation marker expression in the human melanoma cell line HMV-II was analyzed by western blot. We also tested reconstituted human skin tissue and analyzed KDZ-001 with computational molecular modeling.</P> <P><B>Results</B></P> <P>We identified three compounds that affected the pigmentation of developing melanophores in zebrafish. Among them, we identified KDZ-001, a novel anti-melanogenic agent, which strongly inhibits melanin synthesis in the developing melanophores of zebrafish, HMV-II cells, and reconstituted human skin with no toxicity. We found that KDZ-001 directly inhibits TYR enzymatic activity. Notably, computational molecular modeling of KDZ-001 suggested that its interaction with copper ions in the active site of TYR is essential for melanin synthesis, further demonstrating that KDZ-001 mainly acts as a TYR inhibitor to synthesize melanin.</P> <P><B>Conclusion</B></P> <P>KDZ-001 inhibits melanin synthesis and has a potential for use in skin-whitening cosmetics.</P> <P><B>Highlights</B></P> <P> <UL> <LI> In this study, we show that KDZ-001, a novel anti-melanogenic agent, strongly inhibits melanin synthesis in the developing melanophores of zebrafish, HMV-II cells, and reconstituted human skin with no toxicity. We also show that KDZ-001 directly inhibits TYR enzymatic activity. Thus, KDZ-001 is a potential candidate for a skin-whitening agent in the cosmetic industry. </LI> </UL> </P>

        입원 신생아에 있어 환경 온·습도에 따른 체온변이

        A cross sectional explorative study was conducted to invest the relationship between the temperature and humidity of the environment that newborns were l℃ated at and the central and peripheral body temperatures of newborns in hospitalized newborns using a primary dataset on 199 high-risk infants. Data was collected on the temperature (℃) and humidity (%) of the environments (incubator, radiant warmer and room), the temperatures (℃) from the central part (right upper quadrant of abdominal surface) and the peripheral part (dorsal surface of hand) of newborns. As well the weight and length was deducted from main dataset with general characteristics of newborns. The result showed that all of newborn revealed the peripheral hypothermia regardless of 88% of them in normothermia, 9% in hypothermia and 3% in hyperthermia. The low humidity of the environment, rather than the low environmental temperature of either incubator or radiant warmer, was mainly responsible for the massive peripheral hypothermia. Proper environmental humidity should be provided to maintain the normothermia of entire body system in high-risk newborns at NICU.
