        Expression of Human Stem Cell Factor with Recombinant Baculovirus in BmN Cell Line and Silkworm

        A recombinant transfer vector pBacSCF was constructed by inserting huamn stem cell factor (hSCF) cDNA into plasmid pBacPAK8. BmN cells were co-transfected with modified Bombyx mori, nuclear polyhedrosis virus (BmBacPAK) DNA and the recmbinant transfer vector to construct a recombinant baculovirus containing hSCE gene. DNA dot blotting and RNA dot blotting demonstrated that the hSCE gene was contained in the recombinant virus and transcribed. The recombinant baculovirus was infectious to BmN cells and to silkworm. SDS-PAGE analysis showed a specific band of expressed product in the extract of infected cells and in the heamolymph of infected larvae. Bioactivity of the recombinant hSCE was determined with W-1 cell line and MTT colorimetric method in synergy with interlukin-3 (IL-3). These results revealed that the hSCF gene was over-expressed in cultured cells and lavae of silkworm.

        東北少數民族地區農戶土地流轉意愿影響因素硏究: 以延邊地區爲例

        본 연구에서는 연변지역 농가들에 대한 조사 결과를 바탕으로, 이분형 로지스틱 모형을 적용하여 농가들의 농지 사용권 유동의사 및 그에 미치는 영향을 분석하였다. <농업, 농촌, 농민>문제, 즉 “3농” 문제는 중국사회발전의 근본적인 문제로 간주되고 있는데 이러한 “3농” 문제의 근원은 토지문제라고 할 수 있다. 따라서 토지(농지)문제를 해결하는 것이 중국 농촌의 안정적인 발전에 기초적인 작용을 할 수 있다. 연변조선족자치주는 중국 동북지역인 길림성 동부에 위치하고 있는 중국내 최대 조선족집거지이다. 최근 들어 경제와 사회발전과 더불어 출국인원과 중국 발달도시로의 이주로 인해 대량의 농지가 방치되게 되어 농지사용권 유동에 조건을 마련하였으며, 가정농장과 전업합작사의 발전과 더불어 농지유동규모가 점차 증가하게 되었다. 이러한 배경 하에 농가들의 농지유동의사가 어떻게 표현되고 있고, 동시에 어떠한 요인들이 농지유동의사에 영향을 미쳤는지에 대한 연구가 필요한 실정이다. 따라서 소수민족지역인 연변지역을 중심으로 농가들의 농지유동의사를 규명하고, 농지 유동의사에 미치는 요인들을 규명하려 한다. 연구에 사용된 자료는 다음과 같은 방법으로 수집하였다. 2018년 8원 연변대학교 경제관리학원 15명의 학생들이 연변지역 훈춘, 도문, 용정 등 지역에서 농가를 대상으로 1대 1 설문조사를 진행하였다. 총 9개 촌을 대상으로 151부의 설문지를 회수하였다. 분석방법은 로지스틱 회귀모형을 이용하여 성별, 민족, 학력, 직업 등 농가일반사항과 로동력 수, 농기계 수, 정책에 대한 이해 정도 등 농가 경영변수들과 농지사용권 유동의사간의 영향관계를 규명하였다. 분석결과 성별, 민족, 연령, 직업, 노동력 수, 농기계 수, 정책에 대한 이해정도 등 변수들이 영향을 미치는 것으로 분석되었다. 따라서 다음과 같은 정책적 시사점을 제시하였다. 첫째, 정부 정책적 홍보작용과 인도작용을 강화하여 농지사용권 유동정책을 알기 쉽게 이해시켜 농지 유동이 원활이 진행되게 함으로써 농가들의 적정규모에서의 영농홛동을 지지하여야 한다. 둘째, 연변지역의 지역적 우위를 발위하여 농업의 규모화 경영을 도모하여야 한다. 셋째, 농촌 토지임대시장을 육성하여 농지유동의사가 있는 농가들에게 정보를 제공해야 한다. 넷째, 농촌의 사회보장체제를 완벽히 하여 농가들이 농지유동에 대한 의구심을 해소시켜야 한다. This study conducts a field survey of farmers in the Yanbian area and analyzes both their willingness to adopt land circulation and the factors that influence this willingness through a logistic model. The results reveal that gender, nationality, age, the number of the labor force, the amount of agricultural machinery owned, and an understanding of land policy are the main influential factors in farmers’ willingness to participate in land circulation. Finally, several suggestions for promoting land circulation in the Yanbian area are provided.

        이질성 시각에서의 축산물 수출을 위한 사회적 선호 비교 분석

        쇠고기 등급제는 부정육 유통근절, 축산기술 일반화 등 한국 축산업 역사에서 가장 성공적인 축산정책으로 불리우며 축산업 생산기술의 발전에 중요한 역할을 하고 있다. 또한 시장 개방이 확대되면서 쇠고기 등급제가 한국 육우산업의 경쟁력을 높이는 데 결정적 역할을 하였지만 과도하게 마블링 수준에만 의거하여 평가하는 단점이 있다. 이러한 실정에서 소비자들은 등급제가 전달하려는 객관적 정보 외, 다른 정보들도 추가하여 받아들일 수 있는 여지가 충분히 존재할 가능성이 있다. 따라서 소비자들이 쇠고기 등급제를 인지함에 있어 품질 척도 이외, 어떠한 인식을 형성하고 있는지 에 대한 이질성 시각에서의 확인이 필요하다. 본 연구에서는 <2018 식품소비행태조사>데이터를 이용하여 쇠고기 등급 선호와 축산물 구매 특징, 인구 통계 특징 간의 영향 관계를 기술하고, 한우 쇠고기 생산 유통 체계 하에서 소비자의 소비 패턴과 인식 특징 등의 문제점들을 분석하였다. 분석결과 축산물 구입시 고려기준에서 가격보다는 안정성, 신선도와 영양을 고려하는 소비자일수록 높은 등급의 쇠고기를 선호하는 것으로 나타났다. 즉 소비자는 높은 등급 쇠고기가 낮은 등급 쇠고기보다 안전하고 신선하며 영양이 있다고 판단하는 것으로 나타났다. 또한 이변량 프로빗 모델을 이용해 변수 간의 내생성을 측정하고 쇠고기 등급제도의 정보비대칭성을 검증하였다. Purpose: The main purpose of this study is to clarify the relationship among beef grading preference and livestock products purchase characteristics and demographic characteristics, and to analyze the existing problems in the Korean beef production and circulation system. Research design, data, methodology: The current Korean beef production and circulation system were investigated and consumer consumption patterns and cognitive characteristics were analyzed. Data were drawn from the “Consumer Food Consumption Behavior Survey 2018” published by the Korea Rural Economic Research Institute. The Bivariate-Probit model was used to detect the endogeneity between variables, and the asymmetry of the information. Results: Using bivariate probit models, the study found that consumers in general have a misperception that high-grade beef is safer, fresher, and more nutritional than mid or low-grade beef. Conclusion: The current grading system cannot be a correct guide for consumers’ beef purchase behavior. It is proposed that when formulating the beef grading system, the relevant stakeholders involved should carefully convey the basic information about the beef grading system.

      • Research on Multipath Virtual Network Mapping Based on Spatial and Temporal Correlation

        Network virtualization is an important technology that allows multiple heterogeneous networks running simultaneously share substrate network, it can effectively solve the problem of network ossification. Earlier research has made many heuristic algorithms, but it does not consider the time factor in the embedding process, existing algorithm often make decision according to the present available substrate network resource. This paper, we first explore the time and space of the two dimensional network model, form the perspective of the two-dimensional space-time, we proposed two-dimensional discrete weighted method and multi commodity flow virtual network embedding with time-varying model. We also proposed a virtual network algorithm based on space–time. The simulation result show that the proposed algorithm can effectively utilize the network resources and make substrate network resources maintain the load balance for a long term.

      • Virtual Network Mapping Algorithm Based on Load Balancing

        Recent studies for network virtualization have shown a promising way to overcome the Internet ossification. The one of the key issues in network virtualization is a virtual network mapping problem, i.e., mapping a virtual network to the physical network. The situations of dynamic arrivals of virtual network request and the limited life cycle of the virtual networks pose significant challenges to the virtual network mapping problem. A balance between the resource allocation of the physical network and the number of mapped virtual networks. In this paper, we have considered the time characteristics that virtual network requests when mapping algorithms so as to achieve the objective that the node load and link load can simultaneously reach a balance. Giving full consideration to mutual restraints of time and resources, we propose a two-dimensional discrete weighted model based on time and resources, and establish a mathematical programming model of minimizing the degree of two-dimensional load balancing. Moreover, we devise a VN embedding algorithms LB-VNE. Simulation experiments show that the proposed algorithms can increase the acceptance ratio and the revenue by the substrate network in the long term.

        Molecular cloning and characterization of biphenyl hydrolase-like (BPHL) protein gene from silkworm, Bombyx mori

        The cDNA of a biphenyl hydrolase-like (BPHL) protein from Bombyx mori was cloned via rapid amplification of cDNA ends and submitted to GenBank under accession number JN020647. The full-length BmBPHL cDNA was 1161 bp, with four exons and three introns. It consisted of a 208 bp 5′-terminal untranslated region (UTR) and a 191 bp 3′-UTR with three polyadenylation signal sequences AATAAA and a poly(A) tail. The BmBPHL cDNA encodes a 253–amino acid polypeptide with a theoretical isoelectric point of 8.67 and a predicted molecular weight of 28.9 kDa. The deduced amino acid sequence of BmBPHL contained an abhydrolase_6 domain and the Gly-X-Ser-X-Gly motif that is characteristic of serine hydrolases. Sequence comparison showed that BmBPHL is 51% identical to Tribolium castaneum BPHL and 50% identical to Nasonia vitripennis BPHL. Phylogenetic analysis revealed that BmBPHL is grouped with insect BPHL proteins, separating from vertebrate BPHLs. The BmBPHL mRNA transcripts were mainly detected in hemolymph and fat body using fluorescent quantitative real-time PCR. In addition, infectionwith B. mori cytoplasmic polyhedrosis virus (BmCPV) upregulated the relative BmBPHL expression in the hemolymph and midgut. Therefore, BmBPHL may have an important function in the response of silkworms to BmCPV infection.

        Differential and spatial regulation of the prophenoloxidase (proPO) and proPO-activating enzyme in cuticularmelanization and innate immunity in Bombyx mori pupae

        Insect cuticular melanization is regulated by the prophenoloxidase (proPO)-activating system, which is also involved in the innate immune reaction. Here, we demonstrate how the differentiation of the proPO-activating system is regulated toward a cuticular melanization or innate immunity function in silkworm (Bombyx mori) pupae. Our results indicate that the differential and spatial regulation of key components, such as the proPOactivating enzyme and proPOs, primes the proPO-activating system for either cuticular melanization or innate immunity. This dual strategy for cuticular melanization in development and innate immunity upon infection demonstrates a two-pronged defense mechanism that is mediated by the priming of the proPO system.

        Molecular Cloning of a Profilin cDNA from Bombyx mori

        The actin-binding protein profilin cDNA was firstly isolated from the lepidopteran insect, silkworm Bombyx mori. The B. mori profilin cDNA contains an open reading frame of 378 bp encoding 126 amino acid residues and possesses three cysteine residues. The deduced amino acid sequence of the B. mori profilin cDNA showed 80% identity to Apis mellifera profilin and 72% to Drosophila melanogaster profilin. Northern blot analysis showed that B. mori profilin is highly expressed in epidermis and less strongly in silk gland. In addition, Northern blot analysis revealed the presence of B. mori profilin transcripts in all tissues examined, suggesting that B. mori profilin gene is expressed in most, if not all, body tissues.

        Emulsifying properties of different soy hull pectin polysaccharides and application in mayonnaise

        Soy hull pectic polysaccharide (SHPP) is a kindof biological macromolecule prepared from soy hull, whichhas certain thickening and gel properties. In present study,SHPP was extracted with citric acid and sodium citratefrom soybean hulls under the assistance of microwave,respectively. SHPPs were then compared with commercialpectin polysaccharide to test their emulsification ability. The emulsion prepared from SHPP extracted with sodiumcitrate has the best emulsifying effect, small particle sizeand uniform distribution. The rheological properties andparticle size distribution of mayonnaise did not changesignificantly after the addition of different SHPPs andcommercial pectin polysaccharides. However, microscopicobservations revealed that the droplets were more uniformlydistributed in the mayonnaise after the addition ofSHPP extracted with sodium citrate under the assistance ofmicrowave and commercial pectin APC141. SHPPextracted with sodium citrate may play an important role inmaintaining emulsion stability in the future.

        Characterization and profiling of MicroRNAs in posterior silk gland of the silkworm (Bombyx mori)

        MicroRNAs (miRNAs) regulate expression of genes at post-transcriptional level by binding on complementary sequences of target mRNAs and play multiple roles in biological processes. To investigate the differential expression of miRNAs in posterior silk gland (PSG) of silkworm (Bombyx mori) in different periods and regulation of miRNAs on the expression of fibroin genes, Solexa sequencing technology was used to detect miRNAs in PSGs of fourth-instar day-2 larvae and fifth-instar day-3 larvae, respectively. As a result, 466 previously reported miRNAs, and 35 novel miRNAs were detected, and 499 of these detected miRNAs are predicted to target 13,383 genes by target prediction softwares. Additionally, 29 miRNAs expressed differently between the PSG of fourthinstar day-2 larvae and fifth-instar day-3 larvae were found, and the differential expression of these miRNAs may play an important role in the expression of fibroin genes.
