        Characterization and profiling of MicroRNAs in posterior silk gland of the silkworm (Bombyx mori)

        MicroRNAs (miRNAs) regulate expression of genes at post-transcriptional level by binding on complementary sequences of target mRNAs and play multiple roles in biological processes. To investigate the differential expression of miRNAs in posterior silk gland (PSG) of silkworm (Bombyx mori) in different periods and regulation of miRNAs on the expression of fibroin genes, Solexa sequencing technology was used to detect miRNAs in PSGs of fourth-instar day-2 larvae and fifth-instar day-3 larvae, respectively. As a result, 466 previously reported miRNAs, and 35 novel miRNAs were detected, and 499 of these detected miRNAs are predicted to target 13,383 genes by target prediction softwares. Additionally, 29 miRNAs expressed differently between the PSG of fourthinstar day-2 larvae and fifth-instar day-3 larvae were found, and the differential expression of these miRNAs may play an important role in the expression of fibroin genes.

        The molecular mechanisms and factors affecting the feeding habits of silkworm (Lepidoptera: Bombyxidae)

        Food is critical to the survival and development of insects. Bombyx mori (Lepidoptera: Bombycidae), a Lepi doptera model insect with economic significance, is a well-known oligophagous insect that mainly feeds on mulberry leaves. The feeding characteristics of this particular species provide an excellent model for studying the food selection of insect host plants. In recent years, there has been an increasing number of studies on the factors affecting the feeding of silkworms, especially with the development of molecular technology. Many mysteries have been gradually solved. This paper summarizes and discusses the factors that affect the feeding of silkworms, with a focus on the molecular mechanisms of silkworm feeding. It is hoped that this work will be helpful for further research on artificial diets for silkworms and disease control in lepidopteran insects.

      • Miraculin gene transformation in embryogenic Citrus callus and regeneration

        Miraculin can modify a sour taste into a sweet taste and is an alternative to traditional sweeteners. Mirculin gene was introduced into the Miyagawa Wase Satsuma mandarin (Citrus unshiu Marc.) callus by Agrobacterum-mediated transformation to produce citrus transgenic plant. Transgenic plant candidates were selected via hygromycin resistance. Transformation was confirmed by Polymerase Chain Reaction, Southern blotting, and Western blot analysis. Expression of this gene in transgenic citrus resulted in the accumulation of miraculin protein in the leaves. Multiple Shoots of transgenic citrus planets were micrografted onto trifoliate rootstocks in the sterile soil. Plants were established in the greenhouse 2 years after planting.

        Electronic and Optical Properties of Noble Metal Oxides M2O (M = Cu, Ag and Au): First-principles Study

        In this work, first-principles calculations for the structural, electronic, and optical properties of noble metal oxides M2O (M = Cu, Ag, Au) with the cuprite structure are performed by using a plane wave pseudopotential method in the framework of density functional theory (DFT) and the generalized gradient approximation (GGA). The structural, electronic, and optical properties are investigated and discussed. For Cu2O and Ag2O, good agreement was achieved between calculated and experimental results. Within the same framework, Au2O is predicted to be a semiconductor. In comparison with the copper and the silver oxides, the gold oxide has less ionic bonding between Au and O, and the intra-atomic hybridization is expected to be more evident as the depletion of the Au 5d shell appears to be more profound than it is for the Cu 3d and the Ag 4d shells.         Theoretical Study on the Point Defects in N-Doped Anatase TiO2

        In this work, first-principles calculations for the electronic and the optical properties of titanium dioxide (TiO2) with point defects are performed by using a plane wave pseudopotential method in the framework of the density functional theory and the generalized gradient approximation. The point defects, substitutional and interstitial nitrogen, in anatase TiO2 are investigated to understand the origin of the visible-light sensitivity of nitrogen (N)-doped TiO2. The results show that bands originating from substitutional N 2p states appear above the top of the valence band. These states tend to be delocalized and to mix with the valence band as the nitrogen concentration is increased. The optical absorption in the range between 400 and 700 nm is enhanced by the substitutional N impurities. The eect of interstitial N on the electronic and the optical properties of TiO2 depends on the interaction between the nitrogen dopants and the surrounding oxygen. The bonding between N and O introduces a series of localized occupied states under and above the upper valence band, and the electron transitions from these states improve the visible-light absorption of TiO2.

      • Somatic embryogenesis and plant regeneration in Korea Native Citrus Species

        Somatic embryogenic calli were obtained from different native citrus species in Jeju island, South Korea. Undeveloped ovules were cultured on 5 different media, respectively; MSI (MS, modified, with the addition of malt extract 500 mg・L-1 and sucrose 50 g・L-1 and agar 2 g・L-1), MSII (MS, modified, with the addition of malt extract 500 mg・L-1 , kinetin 1 mg・L-1 and sucrose 50 g・L-1 and agar 2 g・L-1), MSIII (MS, modified, with the addition of malt extract 500 mg・L-1, 6-benzyladenine, 3 mg・L-1 and sucrose 50 g・L-1 and agar 2 g・L-1), EME-S (MT, modified, with the addition of malt extract 500 mg・L-1 and sucrose 50 g・L-1 and agar 2 g・L-1), 1/2 EME-S (half concentration of MT marcronutrients, half concentratrion of BH3 marcronutrients, malt extract 500 mg・L-1, glutamine 1.55 g・L-1 and sucrose 50 g・L-1 and agar 2 g・ L-1). Embryogenic calli were induced in the surface of undeveloped ovules in different manners, depending on citrus species and culture conditions. Somatic embryos developed into plantlets with a high frequency. Citrus embryogenic calli can be applied widely to somatic hybridization, genetic transformation, and in vitro germplasm conservation

        Research on the Sustainable Development of Chinese Regional Food Culture Integrating Service Design Thinking

        This study begins with the phenomenon that cultural conflict may occur between people with different social backgrounds, lifestyles and values in the context of social and economic globalization. Cultural conflict is one of the preconditions to promote cultural identity, and cultural identity is also the basis for the sustainable development of regional culture. With the rapid development of society and the substantial improvement of material conditions, food and clothing is no longer a social problem in China. People begin to pay attention to their own internal needs and have more diversified demands on the connotative value and presentation of food. As a symbol of regional culture, food can reflect the internal culture of the region and the social and cultural relationship between the region and other regions. It is an important carrier of the sustainable development of regional culture. In the face of the increasing demand at all levels, tangible single products can no longer meet the needs of users, thus promoting the market economy from product oriented to service-oriented competition. The purpose of this study is to deconstruct and reconstruct a variety of possibilities between regional food, regional culture and people, give play to the role of service design thinking in this process, and explore the value and significance of service design thinking in the process of sustainable development of regional food culture. Therefore, this paper attempts to use service design thinking to participate in the research process of the sustainable development of Chinese regional culture, and its methods and tools to participate in the process of regional culture characteristic food is not only the food remanufacturing and so on. The key is to study and analyze the relationship between the cultural attributes and lifestyle of regional food, create design value by means of symbols, vision, products, space and service process, and enhance the cultural significance and commercial attributes of food with the intervention of multi stakeholders. The research results and contents are as follows: firstly, as the determination and expression of regional food cultural connotation information, service design is not only the presentation of food material attribute information, but also the fundamental driving force to promote the development of intangible values (information, memory, interaction, etc.) such as its cultural attribute. Secondly, the food system involved in service design pays attention to the experience perception of users, promotes collaborative innovation activities of regional food stakeholders, maximizes the value of regional cultural information obtained by stakeholders in the service system, cooperatively creates possible solutions and improves the service quality of the food system. Thirdly, service design improves the user experience of regional food culture, explores the interactive relationship between people and time, region, culture, food and other contacts, and uses symbols and other means of information expression to play the role of people in promoting and inheriting the sustainable development of culture in the regional food system. Based on the research results, this paper looks forward to the ability and creativity of service design to solve the problem of sustainable development of food culture with regional characteristics, promote the advanced innovation of food culture with regional characteristics, and transmit and develop more diversified cultural values.
