        Rapid screening and identification of metabolites of quercitrin produced by the human intestinal bacteria using ultra performance liquid chromatography/quadrupole-time-of-flight mass spectrometry

        Ultra performance liquid chromatography/quadrupole-time-of-flight mass spectrometry (UPLC/Q-TOF MS)technique combined with MetabolynxTM software was usedfor analysis of the metabolites of quercitrin by the isolatedhuman intestinal bacteria from the human feces. Four metabolitesof quercitrin were detected and tentatively identifiedbased on the characteristics of their protonated ions. Themetabolites were metabolized by four main metabolic pathwaysincluding hydroxylation, demethylation, deglycosylationand ring-cleavage. Quercitrin was metabolized to the hydroxyquercitrinand desmethylquercitrin by themajority of theisolated intestinal bacteria such as Bacteroides sp. 54, and wasdegraded to the deglycosylated product quercetin by rhamnosidaseand further ring-cleavage metabolite 3,4-dihydroxybenzoicacid by the minority of the isolated bacteria such asBacteroides sp. 45. The metabolic pathways and most of themetabolites of quercitrin were reported for the first time.

      • Finite Element Analysis on Mechanical Behaviour of Yungui Railway Tunnel in Shallow and Unsymmetrical Strata

        Triaxial compression tests had been performed to determine the properties of the surrounding rock. The displacement and stress of surrounding rock had been analyzed according to Silin 2 at Yungui railway under three kinds of conditions: the initial state, excavation without supporting , and supporting after excavation. Plane strain FEM was established to analyze the stability of the surrounding rock. Contrasted the monitor measuring data, the results showed that the most unfavorable position in the tunnel was found at the ridge side of the arch, and the implementation of support afforded to ensure the stability of surrounding rock, and the design of the early support approach had the ability to meet the safety requirements. The results had certain significance for understanding the mechanical behavior of shallow tunnel in unsymmetrical strata. tunnel, mechanical behaviour, numerical simulation, deformation, stability.

      • Fluid Solid Coupling Analysis of Large Underground Oil Storage Caverns in Containment of Groundwater

        Triaxial compression tests had been performed to determine the properties of the rock mass around unlining underground crude oil a storage cavern which was the first one in China. The execution situation of the tunnel project and the seepage law of groundwater were taken into account. The stress and the seepage field around the tunnel in different working states were simulated by applying Comsol around a underground crude oil storage caverns. According to the test results, it was found that the excavation process may arise the local damage. The extension of the loose zone induced by excavation ranged from 0 to 15.6 m, depending on the buried depth of the caverns. According to numerical simulation results, the crown settlement and stress concentration was depended on the buried depth and the water pressure distribution after excavation of the main cavity. This research results can provide the reference for analysis on the stability of the underground cavities under low stress level and on the water sealed underground petroleum storage rock caverns.

        Application of Jasmonic Acid Followed by Salicylic Acid Inhibits Cucumber mosaic virus Replication

        Systemic acquired resistance is a form of inducible resistance that is triggered in systemic healthy tissues of local-infected plants. Several candidate signaling molecules emerged in the past two years, including the methylated derivatives of well-known defense hormones salicylic acid (SA) and jasmonic acid (JA). In our present study, the symptom on Cucumber mosaic virus (CMV) infected Arabidopsis leaves in 0.1 mM SA or 0.06 mM JA pre-treated plants was lighter (less reactive oxygen species accumulation and less oxidative damages) than that of the control group. JA followed by SA (JA${\rightarrow}$SA) had the highest inhibitory efficiency to CMV replication, higher than JA and SA simultaneous co-pretreatment (JA+SA), and higher than a JA or a SA single pretreatment. The crosstalk between the two hormones was further investigated at the transcriptional levels of pathogenesis-related genes. The time-course measurement showed JA might play a more important role in the interaction between JA and SA.

        Effects of irradiation treatment on protein structure and digestion characteristics of seed-watermelon (Citrullus lanatus var.) kernel protein

        This study examined the effects of differentdoses of irradiation treatments on protein structure anddigestion characteristic of seed-watermelon seed kernelprotein. The results showed that, the molecular structure ofseed-watermelon kernel protein was unfolded after theirradiation treatment, the content of b-sheet structure in thesecondary structure was decreased, while the content ofrandom coil structure increased. The average particle sizeof the protein increased, and the hydrophobic group buriedin the b-sheet structure was exposed hence the surfacehydrophobicity increased. Besides, the surface morphologyof seed-watermelon protein changed from smooth and flatto coarse and concave, the specific surface area in contactwith the aqueous medium increased and its solubilityincreased, the distribution of peptides in the digestabecame wider, and the small molecular weight peptidesgradually increased.

        Cocoa Flavanols and Procyanidins Can Modulate the Lipopolysaccharide Activation of Polymorphonuclear Cells In Vitro

        Flavanols and procyanidins isolated from cocoa have been reported to possess multiple activities potentially relevant to oxidant defenses, vascular function, and immune function. In a combination of in vivo and in vitro studies, we and others have observed that cocoa can be an anti-inflammatory modulator and that compounds in cocoa are capable of modulating eicosanoid production, platelet aggregation, and the pool size of nitric oxide. The present study extends these findings by examining the in vitro effects of cocoa procyanidins on polymorphonuclear cells (PMNs). PMNs, part of the innate arm of the immune system, represent 50-60% of the total peripheral white blood cells and are the first cells to be recruited to the sites of inflammation or injury secondary to bacterial infections. Herein, we demonstrate that certain flavanols and procyanidins isolated from cocoa can moderate a subset of signaling pathways derived from lipopolysaccharide (LPS) stimulation of PMNs, mainly, PMN oxidative bursts and activation markers, and they can influence select apoptosis mechanisms. We hypothesize that flavanols and procyanidins can decrease the impact of LPS on the N-formyl-Met-Leu-Phe-primed PMN ability to generate reactive oxygen species by partially interfering in activation of the mitogen-activated protein kinase pathway.

        냉간신선된 고탄소강선의 굽힘피로특성

        This paper describes the bending fatigue properties of the high-carbon steel wire containing around 0.6 wt.% C, which is used as a stranded wire for ACSR(Aluminum Stranded Conductors Steel Reinforced) in transmission power lines. ACSR which consists of galvanized steel strand and aluminum one always experiences serious vibration by wind. Therefore, it is very important to clarify the fatigue property of core wire for ACSR to determine its lifetime and stability of power delivery. Since steel wires are produced by wire drawing process, they possess a very long fiber lamellar microstructure which has a very short inter-layer distance between ferrite and cementite layer. The long fiber lamellar microstructure is expected to have a different fatigue behavior compared to conventional plate and bulk-type materials. In order to characterize the fatigue property of cold-drawn high-carbon steel wires, bending fatigue tests were carried out on specimens drawn to various strains. The fatigue strength was about 38 kgf/㎟ at ε= 1.19, which was higher than the required strength of 32 kgf/㎟. This is probably due to the very fine elongated pearlitic microstructure formed by the drawing process, which suppresses formation and growth of microcracks. The several unique characteristics of drawn wires are also discussed.

        북한의 항일서사 연구시론

        북한문학에 있어서 항일투쟁문학은 김일성의 정통성과 관계되는 부분으로서 중요한 위치를 차지한다. 특히 주체시대에 항일투쟁문학은 문학의 주요 흐름으로 자리 잡았다. 사실 북한의 항일투쟁문학은 해방공간에서부터 시작되어 주체시대에 들어서서 고조를 이루었다. 즉 해방공간에 있어서 "김일성 장군의 노래"는 그 서곡을 이루었고 “백두산”은 ‘영웅서사시’를 형성했다. 주체시대에 있어서는 “불멸의 총서” 해방 전 편 ‘항일투쟁편’을 창작하여 김일성의 항일서사를 전면적이고 체계적으로 진행했다. 본고는 해방공간과 주체시대에 있어서 김일성항일서사를 고찰했다. 즉 해방공간에 있어서 김일성을 건국의 주인공으로서 ‘영웅서사시’와 주체시대에 있어서 유일사상체계를 수립에 있어서 김일성 신격화와 우상화 작업으로서의 ‘영웅서사시’를 고찰했다. 결론적으로 보면 북한에서의 항일서사는 김일성을 둘러싸고 그의 정치적 자본을 얻고 확립하는 문학의 시녀적 성격이 다분하다. 이런 ‘영웅서사시’적 작업을 통하여 김일성은 ‘절세의 애국자’, ‘만고의 빨찌산’, ‘김일성장군’ 및 천출위인, 백전백승의 강철의 영장의 형상이 수립되고 현실적으로 "위대한 수령"으로 군림하고 인민들은 "수령복"을 만끽하는 정치적 역학관계를 형성한다. The anti-Japanese War literature of North Korean literature is consistent with the orthodoxy of Kim Il-sung. Especially in the “Juche Age”, the anti-Japanese War literature was in a dominant position in the whole North Korean literature. The literature began in the liberation war period and reached its peak in the “Juche Age”. Concretely speaking, “Song to General Kim Il-sung” in the liberation war period opened the prologue, “Beakdu Mountain” promoted “heroic narrative poem”, and Immortal Series’ prequel—“anti-Japanese War piece” in “Juche Age” started to elaborate Kim Il-sung’s anti-Japanese struggle history comprehensively and systematically. Given all this, this paper respectively investigates Kim Il-sung’s anti-Japanese struggle history in the liberation war period and “Juche Age”. It refers to the “heroic narrative poem” of the Founding Father Kim Il-sung in the liberation war period and the deific and idolized “heroic narrative poem” of Kim Il-sung in the only established ideological system of “Juche Age”. According to the investigation, the narrative of North Korean anti-Japanese War took Kim Il-sung as the main line and tried to establish his political capital. By means of the “heroic narrative poem”, Kim Il-sung is shaped as “a loyal patriot”, “an immortal guerrilla”, “General Kim Il-sung”, “the great man from above”, “an ever-victorious iron soldier”, and “a great leader”. At the same time, the “heroic narrative poem” delineates the political dynamics diagram that people enjoy “the leader’s blessing”.
