      • Molecular Cloning, Expression and Genomic Structure of Glyceraldehyde-3-Phosphate Dehydrogenase from the Entomopathogenic fungus, Paecilomyces tenuipes Jocheon-1

        Presently, We have constructed an olig-d(T) primed directional cDNA library from the silkworm Dongchunghacho, an entomopathogenic fungus, of which species is belonging to Paecilomyces tenuipes Jocheon-1. To isolate and screen genes in the fungus, 626 expressed sequence tags(ESTs) were generated by a partial sequencing from the cDNA library. Paecilomyces tenuipes Jocheon-1 cDNA encoding the glyceraldehyde-3-phosphate dehydrogenase(Pt-GAPDH) of Paecilomyces tenuipes Jocheon-1 was cloned from the above cDNA library. The complete cDNA sequence of Pt-GAPDHis comprised of 1,014bp encoding 338 amino acid residues. The deduced protein sequence of Pt-GAPDH showed higher homology with Beauberia bassiana-GAPDH(93% amino acid identity). Hydropathy analysis revealed that Pt-GAPDH protein is hydrophilic. The major three amino acids in its composition of amino acid residues were alanine(11.54%), valine(9.47%) and glycine(8.88%). The cDNA encoding Pt-GAPDH was expressed as a 37 kDa polypeptide in baculovirus-infected insect Sf9 cells. The Pt-GAPDH gene of Paecilomyces tenuipes entomopathogenic fungus consisted of three exons and two introns coding for 338 amino acid residues, and the genomic DNA length of the gene spans 1302bp. The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA.

        Fungi belonging to the Paecilomyces spp. have recently been used as food and herbal medicines in Korea and are greatly popular as commercially available powdered supplement or dried fruiting body. Despite this acceptance and its use, little is known of the genes related to its reactive agents. Presently, We have constructed an olig-d(T) primed directional cDNA library from the silkworm Dongchunghacho, an entomopathogenic fungus, of which species is belonging to Paecilomyces spp. based on the previous identification of ITS1 and ITS2 at the molecular level and collected from Jocheon Miryang, Korea. To isolate and screen genes in the fungus, 626 expressed sequence tags(ESTs) were generated by a partial sequencing from the cDNA library. cDNA encoding the glyceraldehyde-3-phosphate dehydrogenase(Pt-GAPDH) of Paecilomyces tenuipes- Jocheon was cloned from the above cDNA library. The complete cDNA sequence of Pt-GAPDH is comprised of 1,014bp encoding 338 amino acid residues.         PECVD법으로 증착된 전자소자용 thiophene 박막의 전기화학적 신뢰성에 관한 연구

        The corrosion failure of electronic devices has been a major reliability concern lately. This failure is an ongoing concern because of miniaturization of integrated circuits(IC) and the increased use of polymers in electronic packaging. In this paper plasma-polymerized thiophene films were considered as a possible candidate for an interlayer dielectric for multilever metallization of ultra large scale integrated (ULSI) semiconductor devices. The protective ability of above films as a function RF power in an 3.5 wt.% NaCl solution was examined by electrochemical methods and contact angle measurement. The protective efficiency of the film increased with increasing RF power, which induced the higher degree of cross-linking and hydrophobicity of the films.

      • 턴디슈용 SiC 코팅材의 특성에 관한 硏究

        턴디쉬 내장용으로 사용할 내화재료의 여러 가지 특성들을 시험하였다. 납석 70%, 점토 10% 그리고 탄화규소 20%을 혼합하여 18 가지의 조성을 가지는 시험편을 제조하였다. 탄화규소는 각각 순도 85%와 65%인 유럽산, 일본산, 중국산을 택하여 최종 제품에 미치는 영향을 알아보았다. 여러 가지 시험의 결과 10% 탄화규소를 포함한 시험편의 경우 점토질 내화벽돌 보다 수명이 2배 정도 향상되었으며 차지당 용손 두께도 1.5∼1.7㎜/ch였다. Various properties of refractory materials for the tundish lining were examined. The refractory samples with 18 different composition were prepared by mixing powders of 70 wt.% pyrophyllite. 10 wt.% clay and 20 wt.% Sic. SiC was selected from three sources, European. Japanese and Chinese-made, having 85% and 65% purities, to examine their effects on the final properties. As a results of several testings. the sample with 10% SiC(European1 was best suitable for tundish lining as a refractory coating materials. The wear depth was 1.2-1.5 mm per charge at the wall and 1.7 mm per charge a t the slag line, having shown that this service life was about 2 times longer than the ordinary lining made of clay firebricks(SiO₂-Al₂O₃).

      • 석회석의 표면개질에 의한 기능성 원료 개발

        본 연구는 균일침전법에 의해 calcite, aragonate, vaterite의 lime(CaC0₃) 분체를 합성하였으며, 출발원료로 CaCl₂와 urea를 이용하였다. Lime의 코팅은 lithium을 이용하여 침전제와 침전제를 사용하지 않고 합성하였으며 출발원료로 Ni(NO₃)₂ㆍ6H₂0와 침전제는 urea(or NaHCO₃, NH₄HCO₃)를 사용하였다. Ni수화물은 수용액계에서 CaC0₃의 표면에 균일하게 코팅이 되었다. Lime표면에 NiO를 코팅함으로서 lime의 성질을 개선시킬 수 있었다. 표면에 코팅된 니켈 수화물은 500℃의 하소 과정에 의해 NiO로 전이되었다. 얻어진 lime분체의 특성은 주사전자현미경(SEM), X-선 회절분석기(XRD), 시차열분석기(DTA), EDS 등을 이용하여 조사하였다. Lime(CaC0₃) powders were prepared by the homogeneous precipitation method using CaC1₂, urea as the starting materials. Uniform lime(calcite, aragonite, vatrite) powders were obtained by various conditions. The surfaces of lime were modified by coating them with an lithium precursor using two different precipitation techniques: homogeneous decomposition of precipitating agents(urea, NaHCO₃, NH₄HC03) and forced hydrolysis in pure water. Selecting the Ni(NO₃)₂ㆍ6H₂O and urea(or NaHCO₃, NH₄HC0₃) as Ni-source and precipitating agent respectively, the NiO could be uniformly coated on the CaCO₃ powder in the aqueous system. The coating NiO layer dominated the surface properties of the coated lime powders. Calcination of these nickel hydroxide crystals in air at 500℃ transformed them into NiO. Scanning electron microscopy(SEM), X-ray diffractiometry, differential thermal analysis(DTA), and EDS were used to characterize the coated powders.

      • 제주도산 山草, 海藻 및 버섯類 水溶性 抽出物의 抗癌效果

        제주지역에서 생산되는 식물 중에서 파리풀(지방명;가신새), 삼지구엽초, 미역, 톳, 표고버섯 및 만년버섯(일명;영지)에서 열수 및 냉수추출물을 얻어 마우스 암모델 세포인 sarcoma-180 세포를 ICR 마우스에 이식하여 고형암 성장저지율과 수명연장율에 대한 항암성을 검색한 결과는 다음과 같다. 산초추출물의 항암효과는 파리풀에서 53.4%, 삼지구엽초에서 58.2%의 고형암 성장저지율을 보였다. 해조류추출물의 항암효과는 톳에서 63.1%, 미역에서 38.2%의 고형암 성장저지율을 보였다. 버섯추출물의 항암효과는 만년버섯에서 62.8%, 표고버섯에서 66.4%의 고형암 성장저지율을 보였다. 산초, 해조, 버섯류 각 추출물에서 복수형암에 대한 효과는 매우 낮았다. 모든 시료는 일정량을 초과 투여한 실험군에서 고형암 성장저지효과가 낮거나 수명이 단축되었다. The hot and cole water extracts were obtained from herbs (Phryma leptostachya and Epimedium koeranum), seaweeds(Hizikia fusiforme and Undaria peterseniana), and mushrooms(Gonoderma luciolum and Lentimus deodes) among which have long been known to have antitumor effects. These extracts were tested on growth inhibition rate(I.R) of solid tumor and prolongation rate(P.R) in animal experiment using ICR mouse with a mouse tumor models sarcoma-180 cell. The results were as follows ; The antitumor effects of herb extracts were 53.4% with Phryma leptostachya and 58.2% with Epimedium koeranum in growth inhibition rate of solid tumor. The antitumor effects of seaweed extracts were 63.1% with Hizikia fusiforme and 38.2% with Undaria peterseniana in growth inhibition rate of solid tumor. The antitumor effects of mushroom extracts were 62.8% with Ganoderma luciolum and 66.4% with Lentinus edodes in growth inhibition rate of solid tumor. The effects on ascite tumor of extracts from herbs, seaseeds and mushrooms were so low that no anititumor effect could be recognized. All the samples showed low growth inhibition rate of solid tumor or short life in over dose groups.

        Investigation of Weld Corrosion Eff ects on the Stress Behavior of a Welded Joint Pipe Using Numerical Simulations

        In this paper, the infl uence of weld corrosion on the mechanical behavior of a welded joint pipeline was investigated usingcorrosion and mechanical simulations simultaneously. In the corrosion simulation, the modeling results (i.e., the corrosionpotential and current density) revealed that the welded joint is preferentially corroded and the corrosion rate is higher in theoutside environment due to the severe corrosion factors. The increase in corrosion degradation according to the operationtime increases the stress concentration on the welded joint, indicating that the failure risk of the welded joint is increasedwith increasing corrosion degradation. These results can be used to evaluate the lifetime of welded joints exposed to corrosionand suggest guidelines for the maintenance of structures.

      • 백서 해마에서 카이닌산 유도 경련에 의한 JNK 경유 신호전달경로 활성화의 발달 단계에 따른 변화

        연구목적 : 어린 백서의 해마에서 전기경련충격(electroconvulsive shock, ECS) 후 활성화되는 것으로 알려져 있는 MAPK계 신호전달경로가 카이닌산(Kainic acid, KA)에 의한 경련 후 어떤 양상으로 활성화되는 지를 초기 발달 단계에 따라서 관찰하고자 하였다. 방 법 : 생후 7일, 14일, 21일 된 어린 백서와 성년 백서의 해마에서 면역블롯 방법으로 MAPK계 효소의 인산화 효소 중 하나인 SAPK/ERK kinase-1(SEK-1), MAPK계 효소인 c-Jun N terminal protein kinase(JNK), 전사인자 c-Jun 및 조기유전자 단백질 Fos의 단백질 양과 기저 인산화를 측정하였고, 백서에게 KA를 주사하여 경련을 유발한 후 위 단백질들의 인산화 변화의 양상을 같은 방법으로 관찰하였다. 결 과 : SEK-1, JNK, c-Jun 모두 연령 증가에 따른 단백질 양 자체에는 변화가 없었다. JNK와 c-Jun은 기저 인산화의 양적 변화도 없었으나 SEK-1의 경우 기저 인산화 양이 생후 7일에 가장 높고 이후 연령이 높아짐에 따라 감소하였다. JNK의 경우 KA 주사 후 인산화 변화 양상이 관찰되지 않았으나 SEK-1과 c-Jun은 생후 14일부터 연령의 증가와 함께 인산화 양도 증가하였다. Fos 역시 생후 7일부터 발현되어 연령의 증가와 함께 발현양이 증가하였다. 결 론 : 백서 해마에서의 MAPK계 신호전달계의 성숙은 연령의 증가에 따라, 또 신호전달 물질에 따라 차이를 보였다. KA 유도 경련 후 JNK는 생후 21일까지도 활성화가 되지 않는 것으로 보아, 세포주에서 알려진 SEK-1 - JNK-c-Jun - Fos로 연결되는 신호전달 경로가 어린 백서의 해마에서는 적용될 수 없음을 시사한다. Objective : We observed the developmental pattern of activation of MAPK signal transduction pathways known to be activated by eletroconvulsive shock(ECS) in young rat hippocampus after kainic acid(KA)-induced seizure. Methods : We used the method of immunoblotting for examining the basal protein amount and basal level of phosphorylation of MAPK kinase(SAPK/ERK kinase -1, SEK-1), MAPK(c-Jun N terminal protein kinase, JNK), transcription factor(c-Jun) and immediate early gene proteins(Fos) in rat hippocampus at postnatal day 7, 14 and 21, respectively. We also examined the changes of phosphorylation of those proteins after kainic acid-induced seizure in the same way. Results : The basal protein amounts of SEK-1, JNK, and c-Jun did not show age-dependent changes and basal level of phosphorylation of JNK and c-Jun remains unchanged throughout the early developmental period. The basal level of phosphorylation of SEK-1 was peaked at postnatal 7 days and then decreased with aging. After kainic acid-induced seizure, the change of phosphorylation of JNK was not observed but those of SEK-1 and c-Jun increased after postnatal day 14. The expression of Fos was observed at postnatal day 7 and also increased with aging. Conclusion : These results show that the MAPK signal transduction system in rat hippocampus matures in accordance with aging, but the process of maturation differs depending specific proteins. This study suggests the signal transduction cascade(SEK-1 - JNK - c-Jun - Fos) which is well established in cell line studies may not be applied to rat hipposcampus because we could not observe the activation of JNK after KA-induced seizure in young rat hippocampus.
