      • Differences in the metabolic profiles and antioxidant activities of wild and cultivated black soybeans evaluated by correlation analysis

        <P>Wild soybeans are considered a potential resource for soybean domestication and an important source of genetic diversity for soybean crop improvement. Understanding metabolite-caused bioactivity differences between cultivated and wild soybeans is essential for designing a soybean with enhanced nutritional traits. In this study, the non-targeted metabolic profiling of 26 soybean varieties, 15 wild black soybeans (WBS) and 11 cultivated black soybeans (CBS), using liquid chromatography-mass spectrometry (LC-MS) in combination with multivariate analysis revealed significant differences in 25 differential metabolites. Among these, the soyasaponins Ab and Bb were found to be characteristic metabolites expressed more substantially in CBS than in WBS. Three different antioxidant assays and correlation analysis identified major and minor antioxidants that contributed to WBS having an antioxidant activity 4- to 8-fold stronger than that of CBS. Epicatechin, procyanidin B2, and cyanidin-3-O-glucoside were identified by both association analysis and the online LC-ABTS radical scavenging assay as being major antioxidants.</P>

      • A Functional SNP in the MDM2 Promoter Mediates E2F1 Affinity to Modulate Cyclin D1 Expression in Tumor Cell Proliferation

        Background: The MDM2 oncogene, a negative regulator of p53, has a functional polymorphism in the promoter region (SNP309) that is associated with multiple kinds of cancers including non-melanoma skin cancer. SNP309 has been shown to associate with accelerated tumor formation by increasing the affinity of the transcriptional activator Sp1. It remains unknown whether there are other factors involved in the regulation of MDM2 transcription through a trans-regulatory mechanism. Methods: In this study, SNP309 was verified to be associated with overexpression of MDM2 in tumor cells. Bioinformatics predicts that the T to G substitution at SNP309 generates a stronger E2F1 binding site, which was confirmed by ChIP and luciferase assays. Results: E2F1 knockdown downregulates the expression of MDM2, which confirms that E2F1 is a functional upstream regulator. Furthermore, tumor cells with the GG genotype exhibited a higher proliferation rate than TT, correlating with cyclin D1 expression. E2F1 depletion significantly inhibits the proliferation capacity and downregulates cyclin D1 expression, especially in GG genotype skin fibroblasts. Notably, E2F1 siRNA effects could be rescued by cyclin D1 overexpression. Conclusion: Taken together, a novel modulator E2F1 was identified as regulating MDM2 expression dependent on SNP309 and further mediates cyclin D1 expression and tumor cell proliferation. E2F1 might act as an important factor for SNP309 serving as a rate-limiting event in carcinogenesis.

        Polymer Nanostructured Components Machined Directly by the Atomic Force Microscopy Scratching Method

        This study aims to machine complex three dimensional nano-structures on polymer surfaces directly using the Atomic Force Microscopy (AFM)-based nano-mechanical processing method. Processing parameters including the normal load, the feed rate, the machining speed and the scanning direction are optimized by the Taguchi method. Their effects on the machined depth and the surface roughness of the machined structures are analyzed. Meanwhile, the AFM PZT tube is controlled to achieve the tip’s three-dimensional motions in space. Using the optimized processing parameters, three-dimensional nanostructures on the surface of PMMA are directly fabricated. It is a novel low-cost manufacturing method for machining complex three-dimensional nano-structures or nano parts on polymer surfaces directly.

      • Protective Effect of Nicotine on Gastrin-induced Gastric Mucosal Damage in Rats

        위점막 손상에 미치는 영향에 관한 nicotine의 효과는 아직 정설이 없는 형편이다. 본 연구에서는 nicotine이 위점막 손상에 미치는 영향을 보기 위하여 nicotine (5 mg/kg, 10mg/kg)을 9일간 하루에 두번씩 위내 투여하였다. 위점막 손상은 gastrin (1.2 mg/kg)을 피하 주사함과 동시에 유문부결찰을 6시간 동안 시행하므로 야기시켰다. 그 결과 nicotine 투여군에서 현저한 위점막 손상의 감소를 보였다 (대조군의 50%). 이러한 nicotine의 위점막 보호 효과에 대한 기전을 추구하기 위하여 위관류 실험을 시행하였다. Nicotine은 기초 위산 분비에는 영향이 없었으나 gastrin으로 자극된 위산 분비를 현저히 감소시켰고, 이러한 반응은 nicotine 용량에 비례하였다. 이상의 결과로 보아 nicotine의 장기간 간헐적 투여는 gastrin 투여로 인한 위점막 손상에 보호 효과가 있으며, 이러한 효과는 gastrin으로 자극된 위산 분비를 억압하는 nicotine의 효과가 관련될 것으로 생각된다. 또한 nicotine의 위점막 손상 악화 효과를 관찰한 보고들을 고려하면 nicotine의 위점막 손상에 관한 효과는 nicotine의 투여 방법에 따라 전혀 다르게 나타날 수 있을 것으로 생각된다. Conflicting data have been reported on the effect of nicotine on gastric mucosal damage. To elucidate the effect of chronic intermittent nicotine on gastric mucosal damage, intragastric nicotine (5 mg/kg, 10 mg/kg) was administered twice per day for 9 days. Gastric mucosal damage was created by s.c. injection of a large dose (1.2 mg/kg) of pentagastrin followed by pylorus ligation for 6 hours. Nicotine treated rats showed reduced gastric mucosal damage about 50% of the control. To examine the mechanism of the protective effect of nicotine, gastric perfusion experiments were done. Basal acid secretion was not affected by intragastric or intravenous nicotine. However, pentagastrin-stimulated acid secretion markedly inhibited by a bolus injection of nicotine, and this response was dose-related. These data indicates that chronic intermittent administration of nicotine protects gastric mucosa against gastrin-induced gastric mucosal damage, and nicotine-induced inhibition of gastrin-stimulated acid secretion has an important role for the protective effect of nicotine. Considering reports concerning nicotine's aggravating effect on the gastric mucosal damage, it is suggested that the methods of administration of nicotine may be an important decisive factor of the divergent action of nicotine on the gastric mucosa.

        Study of the Band-gap Structure of a 1D-photonic Crystal by Using Different Numerical Approaches

        Comparative studies between the transfer matrices method (TMM) and plane wave method (PWM) approaches have been performed on 1D photonics crystal under different conditions to show the differences between these two kinds of calculations. TMM is suitable for the design of 1D photonic crystal device with high precision and is in good agreement with experimental results, but is not suitable for the 2D and 3D photonic structures which are limited by the complicated boundary conditions at micro interfaces. The result based on the PWM approach to deal approximately with the photonic structure in approximation has not yet been strictly verified by experiment, not even for 1D photonic crystal structures. More efforts will be required to explore its validation under all physical conditions to enhance its application.

      • Association Between Single Nucleotide Polymorphisms in miRNA196a-2 and miRNA146a and Susceptibility to Hepatocellular Carcinoma in a Chinese Population

        Hepatocellular carcinoma (HCC) is one of the most prevalent cancers in the world and deeply threatens people's health, especially in China. Techniques of early diagnosis, prevention and prediction are still being discovered, among which the approaches based on single nucleotide polymorphisms in microRNA genes (miRNA SNPs) are newly proposed and show prospective potential. In particular, the association between SNPs in miRNA196a-2 (rs11614913) and miRNA146a (rs2910164) and HCC has been investigated. However, the conclusions made were conflicting, possibly due to insufficient sample size or population stratification. Further confirmations in well-designed large samples are still required. In this study, we verified the association between these two SNPs and the susceptibility to HCC by MassARRAY assay in a 2,000 large Chinese case-control sample. Significant association between rs11614913 and HCC was confirmed. Subjects with the genotype of CT+TT or T allele in rs11614913 were more resistant to HCC (CT+TT: OR (95% CI)=0.73 (0.57-0.92), P=0.01; T allele: OR (95% CI)=0.85 (0.75-0.97), P=0.02) and HBV-related HCC (CT+TT: OR (95% CI)=0.69 (0.53-0.90), P=0.01; T allele: OR (95% CI)=0.82 (0.71-0.95), P=0.01). The affected carriers of CT or TT also tended to have lower levels of serum AFP (P=0.01). This study demonstrated a role of rs11614913 in the etiology of HCC. Further research should focus on the clinical use of this miRNA SNP, so as to facilitate conquering HCC.
