        육미지황탕이 흰쥐의 뇌손상 회복에 미치는 영향

        Objectives : This study was designed to investigate the effects of Yukmijihwang-tang on contusion of the mice induced with medicine. Methods : I observed the effects of light and electron microscopes. and examined hematological changes and VEGF-immunohistochemistry. Results : Hematology: Leukocytes were increased significantly in a control group of mice compared with the experimental group. Light microscope : A few neurons were condensed in the 7-day experimental group, but condensed remarkedly in the 3-day control group. Most glial cells were observed in the 3-day experimental group. Edema and dilatation of vessels occurred significantly in the 3-day control group, and these results occurred weakly in the 7-day experimental group. VEGF-immunohistochemistry : VEGF-immunohistochemical reactivity for the glial cells was the highest in the 3-day experimental group, and immunoreactivity for the vessels and neurons highly increased in the 7-day experimental group. Electron microscope : In the 3-day control group, protoplasmic astrocytes concerned with angiogenesis contained weakly developed rough endoplasmic reticulum. and a few of glial filaments were observed. In the 7-day experimental group, the bundles of glial filaments were found in the cytoplasmic process of astrocytes. Conclusion : medication using Yukmijihwang-tang of mice contused by medical stress is highly effective in inflamatory response, curing cell damage and angiogenesis.

        This study has been carried out to investigate Brain in Nei-jing. The results of this study were as follows. 1. Kidney energy becomes the marrow by warm caring function of Meong-mun(命門) and the marrow becomes brain by ascending and gathering between a hole Back-hoi(百會) and a hole pung-ji(風池) 2. Brain is a sea of the marrow that five vital organs are eraporated togather. 3. In Nei-jing(內經), there is a explanation the connectin between brain and mind, thanking by drawing in a concept of head. 4. Brain regulrates physical excercise. 5. There is a direct connection between brain and biosis. 6. Brain achieves function of sense, relating with the five sensory organs. 7. Brain controls a change of emotion. 8. Brain has immunological function.

      • The Presence of an Antioxidant Protein to Inhibit Enzyme Inactivation by an Ascorbate/Fe(III)/$O_2$, Mixed-function Oxidation System in Yeasts

        Saccharomyces와 Candida의 두 종류 효모 추출물 중에서 아스코르브산/철(III)/산소 혼합기능 산화계를 보다 선택적으로 억제하는 항산화 활성의 존재를 확인하였다. 두 효모의 조단백질을 DEAE관을 이용한 고속액체 크로마토그래피 방법으로 분리하였을 때 디티오트레이톨이나 아스코르브산을 전자공여체로 하는 혼합기능 산화계에 의한 효소의 불활성화를 억제하는 각각 세 종류의 항산화 활성도가 검출되었다. 이 중 두 효모 모두에서 동일하게 0.22M 염화칼륨 농도에서 용출되는 분획 중에 포함되어 있는 항산화 활성도는 아스코르브산/철(III) 산소 혼합기능 산화계를 선택적으로 억제하였다. 이는 디티오트레이톨/철(III)/산소와 아스코르브산/철(III)/산소 두 혼합기능 산화계를 같은 정도로 억제하는 카탈라제나 S. cerevisiae에서 정제 보고된 티올기 특이적 항산화 단백질로서 디티오트레이톨/철(III)/산소 혼합기능 산화계만을 억제하는 활성도와는 전혀 다른 것으로서 카탈라제 활성도를 나타내지 않았고 티올기 특이적 항산화 단백질에 대한 항체와 반응하는 단백질도 함유하지 않았다. 따라서 이들 효모 중에는 카탈라제와 티올기 특이적 항산화 단백질외에 아직 보고되지 않은 새로운 종류의 항산화 단백질이 있는 것으로 추정된다. From crude extracts of the yeast three different antioxidant activities against MFO systems containing dithiothreitol (DTT) or ascorbate as an electron donor could be separated by a HPLC-DEAE column chromatography. Antioxidant activity of the fraction eluted at 0.22 M potassium chloride from both yeast preparations selectively inhibited the ascorbate/Fe(III)/$O_2$ MFO system. It differed from catalase activity which inhibits DTT/Fe(III)/$O_2$ and ascorbate/Fe(III)/$O_2$ MFO systems with similar extent and thiol-specific antioxidant activity which was reported from S. cerevisiae and inhibits DTT/Fe(III)/$O_2$ MFO system specifically. The active fraction showed no catalase activity and did not carry immunoreactive protein which specifically reacts with antibody against thiol-specific antioxidant protein. The results suggest that there is another type of antioxidant protein besides catalase and thiol-specific antioxidant protein in yeast.

        해수에 적응된 송사리 (Orizias latipes)아가미 염소세포의 아포토시스

        Ultrastructural changes of chloride cells in the gill from seawater-adapted killifish (Orizias latipes) were studied with a transmission electronmicroscope. Chloride cells contain many mitochondria and specifically developed tubular systems. Apical pits, formed by several neighboring chloride cells, were exposed to the environment. Degeneration and death of the chloride cells by apoptosis occurred more frequently than by necrosis. Apoptotic chloride cells shrank, became to apoptotic bodies, and eventually were phagocytosed and digested by the microphages around them. We conclude that apoptosis plays an important role in increased cellular turnover of chloride cells for the osmoregulation caused by changes in salinity of the environment.

        류머티스 관절염 환자의 원인지각과 건강추구행위에 관한 탐색적 연구

        This study was undertaken to find out the causal perception and health seeking behaviors of Rheumatoid arthritis patients, define and understand the typology and find the relationship between causal perception and health seeking behavioral types. The Q-population consisted of 236 statements of causal perception and 249 statements of health seeking behavior were collected. 38 Q-samples each of both causal perception and health seeking behavior were selected. The P-sample for this study were made up of 28 female Rheumatoid arthritis patients from a Rheumatoid arthritis specializing hospital who had only visited the hospital once. Each respondent were given 2 Q-sets (causal perception and health seeking behavior) of 76 statements and sorted them according to 9-point scale. The result of Q-sorting were coded and analyzed using QUANL PC program. 1) Typological Observation on Causal Perception (1) Physical Fatigue Type: Type 1 perceived that the illness occurred due to excessive work requiring physical labor or strain that had occurred from not resting after excessive physical labor, therefore, thinking the origin of the illness was from physical strain. (2) Dispensation of Nature Type : Type 2 perceived that the major cause for the illness is not only excessive physical labour but also fecundity and old age. (3) Causality to Environment Type: Type 3 perceived that Rheumatoid arthritis occurred from injury to the joints or bad and humid weather. (4) Conscience of Guilty Type : Type <1 consisted of people with guilty conscience for lack of religious commitment. They perceived that the illness was a punishment from God for not praying or because of bad luck. (5) Rationally Perceiving Type: People who belong in type 5 perceived the cause of illness in light of scientific facts such as genetics, unbalanced diet or lack of exercise. (6) Psychological Stress Type: People who belong in type 6 believed that excessive stress was the cause of the illness. 2) Typological Observation on Health Seeking Behaviors (1) Oriental Medical Treatment Type: People categorized in type 1 used Chinese medical treatment such as acupuncture. Ttum (maxacautery). Bu-Hwang (Chinese therapy using cupping glass to get rid of pus and bad blood) to ease pain in the joints and seek health or warm bath therapy such as sauna. 1967 warm-water bathing, fomentation to promote blood circulation. and therefore categorized oriental medical treatment type. (2) Information Seeking Type: People categorized in type 2 \"ere those who tried out the health seeking behaviors obtained from other people or the mass media. (3) Dietary Control Type : People categorized in type 3 tried to keep Rheumatoid arthritic symptoms and health intact by dietary control. (4) Western Medical Treatment Type: People categorized in type 4 were those who followed the treatment suggested by specialists from hospitals for the Rheumatoid arthritis symptoms and health care. 3) Causal Perception of Illness and Related Health Seeking Behavior. After investing the relationship between types of causal perception and health seeking behavior, we found out that there were various health seeking behaviors for each causal perception. However, if we look at elements weighted relatively highly in each type and sort out the causal perception into unchangeable internal elements and changeable external elements, we can find a relationship with health seeking behaviour types. Oriental medical treatment and information seeking type were common health seeking behaviors of all six causal perception types. Only difference being that internal causal perception type was related to hospital instruction following and external causal perception type was related to dietary controlling. Physical fatigue, dispensation of nature, rationally perceiving and psychological stress types, which are internal causal perceptions, follow the instructions of doctors thoroughly whereas, causality to environment and conscience of guilty types, which are external causal perceptions, take care of health by means of dietary control and both types of causal perception approach health in their own way. The most advisable health care is to carry out both hospital and dietary treatment together to achieve continuous treatment \\``ith positive causal perceptions.

        말기암환자에서 가정호스피스완화 돌봄 경험

        목적: 본 연구의 목적은 말기암환자로서 가정호스피스 돌봄을 받고 있는 대상자의 삶의 경험에서 얻어지는 현상을 이해하고 경험의 본질을 이해하고자 함이다. 방법: 가정호스피스 돌봄을 받고 있는 말기암환자 10명으로부터 심층면담을 통해 자료를 수집하였고 Colaizzi의 현상학적 방법을 사용하여 분석하였다. 결과: 의미 있는 진술에서 구성된 의미가 도출되고, 구성된 의미에서 주제 및 주제모음을 확인하여 범주화 한 결과, '삶의 질 저하', '남은 삶의 수용', '신앙으로 준비된 죽음', '고마움', '부정적 대처'의 범주가 도출되었다. 결론: 가정호스피스 돌봄을 받고 있는 말기암환자들의 삶의 경험을 이해하고, 호스피스 간호를 통해 대상자가 부정적인 삶의 경험을 극복하고, 긍정적인 삶의 경험을 할 수 있도록 체계화된 호스피스 간호의 제공이 필요하다. Purpose: The purpose of this study was to understand terminal cancer patient's experiences of home-based hospice care. Methods: The data were collected from July 2011 through September 2010. Data were collected from 10 terminal cancer patients who received home-based hospice care services and by using in-depth interview. The data were analyzed using Colaizzi's phenomenological method. Results: Data were classified by 25 themes comprising 14 theme clusters and five categories. The five categories were 'life quality deterioration', 'appreciation', 'acceptance of the rest of their lives', 'Prepared for death with religion', 'negative coping'. Conclusion: Systematic hospice care should be provided to understand life experience of patients with terminal cancer who receive home-based hospice care, to help them overcome negative experiences and grow a positive perspective.

        Penicillium verruculosum 으로부터 Celobiohydrolase 의 정제 및 특성에 관한 연구

        Three cellobiohydrolase I, II and III, from culture solution of Penicillum verruculosum grown in liquid media containing cellobiose octaacetate or KC-flock, as the sole carbon source has been extensively purified and characterized. Molecular weights of the purified enzymes, I, II and III, were estimated as 60,000, 66,000 and 76,000 on 10% polyacrylamide gel containing sodium dodecyl sulfate and have 8.6, 4.2 and 8.5% of carbohydrate contents, respectively. The three enzymes showed hydrolytic activities towards crystalline celluloses such as Avicel, cotton and filter paper and soluble cellulose such as carboxymethylcellulose. Specific activities of cellobiohydrolase, I, II and III against Avicel as a substrate were 0.07, 0.10 and 0.07 unit per mg. of protein, respectively. The optimum pHs and temperatures of three enzymes were 5.0 and 50℃. All enzyme produced extensively cellobiose from Avicel, and the enzymatic hydrolysises of p-Nitrophenyl-β-D-cellobioside were inhibited by cellobiose, but not by D-glucose.
