Engineered bone tissue is thought to be the ideal alternative for bone grafts in the treatment of related bone diseases. BMP9 has been demonstrated as one of the most osteogenic factors, and enhancement of BMP9-induced osteogenesis will greatly accelerate the development of bone tissue engineering. Here, we investigated the effect of insulin-like growth factor 1 (IGF1) on BMP9-induced osteogenic differentiation, and unveiled a possible molecular mechanism underling this process. We found that IGF1 and BMP9 are both detectable in mesenchymal stem cells (MSCs). Exogenous expression of IGF1 potentiates BMP9-induced alkaline phosphatase (ALP), matrix mineralization, and ectopic bone formation. Similarly, IGF1 enhances BMP9-induced endochondral ossification. Mechanistically, we found that IGF1 increases BMP9-induced activation of BMP/Smad signaling in MSCs. Our findings demonstrate that IGF1 can enhance BMP9-induced osteogenic differentiation in MSCs, and that this effect may be mediated by the enhancement of the BMP/Smad signaling transduction triggered by BMP9. We have found that the previously uncharacterized bone morphogenetic protein-9 (BMP9) is one of the most osteogenic factors. However, it is unclear if BMP9 cross-talks with TGFβ1 during osteogenic differentiation. Using the recombinant BMP9 adenovirus, we find that low concentration of rhTGFβ1 synergistically induces alkaline phosphatase activity in BMP9-transduced C3H10T1/2 cells and produces more pronounced matrix mineralization. However, higher concentrations of TGFβ1 inhibit BMP9-induced osteogenic activity. Real-time PCR and Western blotting indicate that BMP9 in combination with low dose of TGFβ1 potentiates the expression of later osteogenic markers osteopontin, osteocalcin and collagen type 1 (COL1a2), while higher concentrations of TGFβ1 decrease the expression of osteopontin and osteocalcin but not COL1a2. Cell cycle analysis reveals that TGFβ1 inhibits C3H10T1/2 proliferation in BMP9-induced osteogenesis and restricts the cells in G0/G1 phase. Our findings strongly suggest that TGFβ1 may exert a biphasic effect on BMP9-induced osteogenic differentiation of mesenchymal stem cells. [BMB Reports 2012; 45(9): 509-514]
Study on the Fracture Toughness of Polypropylene-Basalt Fiber-Reinforced Concrete
Ninghui Liang,Lianxi Ren,Shuo Tian,Xinrong Liu,Zuliang Zhong,Zhiyun Deng,Ru Yan 한국콘크리트학회 2021 International Journal of Concrete Structures and M Vol.15 No.5
To study the hybrid effects of polypropylene fiber and basalt fiber on the fracture toughness of concrete, 13 groups of notched concrete beam specimens with different fiber contents and mass ratios were prepared for the three-point bending test. Based on acoustic emission monitoring data, the initiation cracking load and instability load of each group of specimens were obtained, and the fracture toughness parameters were calculated according to the double- K fracture criterion. The test results show that the basalt fiber-reinforced concrete has a greater increase in initial fracture toughness, and the toughness of coarse polypropylene fiber-reinforced concrete is more unstable. Moreover, after the coarse polypropylene fiber content reaches 6 kg/m³ and the basalt fiber content reaches 3 kg/m³, increasing the content will not significantly improve the fracture toughness of the concrete. The polypropylene-basalt fiber will produce positive and negative effects when mixed, and the mass ratio of 2:1 was optimal. Finally, the fitting analysis revealed that the fracture process of polypropylene-basalt fiber-reinforced concrete (PBFRC) can be objectively described by the bilinear softening constitutive curve improved by Xu and Reinhardt.

The capacity and equivalent switching frequency of parallel interleaved inverters can be increased, but there are problems with neutral point potential balance and parallel bridge circulating current. This paper regards the parallel three-level inverter as a five-level inverter and five-level space vector integrated modulation is applied. On this basis, a neutral-point potential control strategy based on the mid-point charge to calculate the adjustment factor k is proposed. By further meticulously partitioning specific sector regions, the selection method of redundant vectors in the switching sequence is optimized without increasing the switching times, and the midpoint voltage ripple is reduced. Then, the redundant state allocation in the process of splitting the five-level into three-level is used as a new degree of freedom to suppress the circulating current of the parallel bridge, therefore realizing the control of voltage and current sharing. Finally, simulation and experiments verify the correctness and effectiveness of the method.
Analytical and Numerical Studies on Steel Columns with Novel Connections in Modular Construction
En-Feng Deng,Jia-Bao Yan,Yang Ding,Liang Zong,Zhong-Xian Li,Xiao-Meng Dai 한국강구조학회 2017 International Journal of Steel Structures Vol.17 No.4
modular construction is a new type of promising structures. This type of structure is an excellent alternative to conventional on-site buildings with advantages of shortened construction time, improved construction quality and reduced environmental pollution. A new concept of modular construction with novel connections has been proposed. This paper mainly focuses on the behavior and design of the innovative steel column working together with tenons at both ends. The fourthdifferential equation was firstly adopted to develop a theoretical model to determine the buckling length of the column. Then, a 3-D nonlinear finite element (FE) model was developed to simulate the ultimate strength behavior of the novel column under pure compression. Parametric studies reveal that the tenons play a significant role in affecting the ultimate load and the corresponding end shortening, among which the length of the tenons is the most important factor. The design approach and recommendations are also discussed through assessing the applicability of the current design code.
Novel HMW glutenin genes from Aegilops tauschii and their unique structures
Wen-Jie Chen,Zhong-Wei Yuan,Lian-Quan Zhang,Xing Fan,Ze-Hong Yan,Ji-Rui Wang,You-Liang Zheng,Huai-Gang Zhang,Deng-Cai Liu 한국유전학회 2012 Genes & Genomics Vol.34 No.3
A pair of novel high-molecular-weight glutenin subunits (HMW-GS) 1Dx5.3t and 1Dy12.1**t were revealed and characterized from Ae. tauschii accession PI554324. SDS-PAGE band of 1Dx5.3t was between those of 1Bx6 and 1Bx7, while 1Dy12.1**t with slightly faster migration rate than that of 1Dy12. The lengths of 1Dx5.3t and 1Dy12.1**t were 2115 bp and 1986 bp, encoding 703 and 660 amino acid residues, respectively. Their authenticity was confirmed by successful expression of the coding regions in Escherichia coli. 1Dx5.3t is the shortest of the known Dx-type alleles. 1Dy12.1**t is also a special subunit since it has an additional cysteine in the front of the central repetitive domain. This cysteine that is not existed in previously reported Dy-type genes may be useful for improving bread wheat quality. Median-joining Network analysis indicated that 1Dy12.1**t may be a key site in the genealogy of the Glu-Dy.
