Development of a Multi-tactile Stimulation System for Cognitive Study

The multi-tactile stimulation system was developed to analyze cognitive function characteristics. The developed system consists of three units; control unit, drive unit and actuator that simultaneously and separately supports the vibration, heat, and pressure stimulation. From the results, vibration stimuli have a wide frequency range of 0-300 Hz comprising 20 levels, and stimulation intensity comprising 40 levels. The control temperature and stimulation temperature used for heat stimulation comprise four and eight levels, respectively. Pressure stimuli in the range of 0-600 mmHg can be stimulated comprising 12 levels. Therefore, the developed stimulator can be used for integrative tactile perception and cognition studies on various vibration, heat and pressure sensations.

The tactile air-pressure stimulator was developed to stimulate blow and suction type to control the stimulation strength. It is possible to present the consistent stimulation and the two types of stimulation at the same time to the same stimulation area. The developed stimulator consists of control, drive unit, and actuator. The control unit performed the role of controlling stimulus type, intensity, and time by using a microcontroller (ATMEGA 128, Atmel, USA). The blow and suction stimuli strength is controlled by a unit of 1 psi in the range of 0~14 psi. Drive unit moved an air-compressor (JS20, Joinmedical, South Korea) and a valve matrix. Compressed air generated from the air-compressor was transferred to the valve matrix, which was composed of three valves. Controlling the on/off capability for each valve enabled generation of air flows for the blow-type and suction-type stimuli. Air -pressure was detected using a pressure sensor (33A-005G-2210, Smate, Taiwan) and vacuum sensor (SPSB, TPC mechatronics, Korea) then feedback to the control unit. Actuator served the role of presenting two of pressure sensation stimuli in identical locations and areas. Two-and three-dimensional AUTOCAD was used for design and a three-dimensional printer, Mojo (Stratsys, USA), which was used for fabrication of the actuator. The total size was 20 × 20 × 20mm3 (length×width×height), and the stimulus presentation area was 1 mm2. It is expected to stimuli applying the perceptual and cognitive structures of air pressure.

<P><B>Abstract</B></P> <P>MeCP2 is a chromatin associated protein which is highly expressed in brain and relevant with Rett syndrome (RTT). There are AT-hook motifs in MeCP2 which can bind with AT-rich DNA, suggesting a role in chromatin binding. Here, we report the identification and characterization of another AT-rich DNA binding motif (residues 295 to 313) from the C-terminal transcription repression domain of MeCP2 by nuclear magnetic resonance (NMR) and isothermal calorimetry (ITC). This motif shows a micromolar affinity to AT-rich DNA, and it binds to the minor groove of DNA like AT-hook motifs. Together with the previous studies, our results provide an insight into a critical role of this motif in chromatin structure and function.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Basic cluster region (residues 295 to 313) in the MeCP2 binds to AT-rich DNA. </LI> <LI> NMR relaxation data identified that IKKRKTRE is critical for binding. </LI> <LI> Basic cluster peptide binds to the minor groove of DNA, similar to AT-hooks. </LI> </UL> </P>

<P><B>Abstract</B></P> <P> <I>Desulfurococcus amylolyticus</I> is an anaerobic and hyperthermophilic crenarchaeon that can use various carbohydrates as energy sources. We found a gene encoding a glycoside hydrolase family 57 amylolytic enzymes (DApu) in a putative carbohydrate utilization gene cluster in the genome of <I>D. amylolyticus</I>. This gene has an open reading frame of 1,878 bp and consists of 626 amino acids with a molecular mass of 71 kDa. Recombinant DApu (rDApu) completely hydrolyzed pullulan to maltotriose by attacking α-1,6-glycosidic linkages, and was able to produce glucose and maltose from soluble starch and amylopectin. Although rDApu showed no activity toward α-cyclodextrin (CD) and β-CD, maltooctaose (G8) was detected from reaction with γ-CD. The highest activity of rDApu was measured at pH 5.0 and 95 °C. The half-life of rDApu was 12.7 h at 95 °C and 27 min at 98 °C. Interestingly, rDApu was able to transfer a maltose unit to 6-<I>O</I>-α-maltosyl-β-CD via transglycosylation. Structure analysis using MALDI-TOF/TOF MS and nuclear magnetic resonance revealed that the new transglycosylated products were 6<SUP>1</SUP>, 6<SUP>4</SUP>-di-<I>O</I>-maltosyl-β-CD and 6<SUP>1</SUP>, 6<SUP>3</SUP>, 6<SUP>5</SUP>-tri-<I>O</I>-maltosyl-β-CD.</P> <P><B>Highlights</B></P> <P> <UL> <LI> GH57 amylopullulanase from <I>Desulfurococcus amylolyticus</I> (DApu) was characterized. </LI> <LI> DApu exhibits not only hydrolysis activity but also transglycosylation activity. </LI> <LI> Branched cyclodextrins were formed by transfer of a maltose unit by DApu. </LI> </UL> </P>
Ligand-Mediated Folding of the OmpA Periplasmic Domain from <i>Acinetobacter baumannii</i>

<P>The periplasmic domain of OmpA from Acinetobacter baumannii (AbOmpA-PD) binds to diaminopimelate and anchors the outer membrane to the peptidoglycan layer in the cell wall. Although the crystal structure of AbOmpA-PD with its ligands has been reported, the mechanism of ligand-mediated folding of AbOmpA remains elusive. Here, we report that in vitro refolded apo-AbOmpA-PD in the absence of ligand exists as a mixture of two partially folded forms in solution: mostly unfolded (apo-state I) and hololike (apo-state II) states. Binding of the diaminopimelate or glycine ligand induced complete folding of AbOmpA-PD. The apo-state I was highly flexible and contained some secondary structural elements, whereas the apo-state II closely resembled the holo-state in terms of both structure and backbone dynamics, except for the ligand-binding region. N-15-relaxation-dispersion analyses for apo-state II revealed substantial motion on a millisecond timescale of residues in the H3 helix near the ligand-binding site, with this motion disappearing upon ligand binding. These results provide an insight into the ligand-mediated folding mechanism of AbOmpA-PD in solution.</P>
CHARACTERIZATIONS OF SEVERAL SPLIT REGULAR FUNCTIONS ON SPLIT QUATERNION IN CLIFFORD ANALYSIS

In this paper, we investigate the regularities of the hyper-complex valued functions of the split quaternion variables. We define several differential operators for the split qunaternionic function. We research several left split regular functions for each differential operators. We also investigate split harmonic functions. And we find the corresponding Cauchy-Riemann system and the corresponding Cauchy theorem for each regular functions on the split quaternion field.
