        산지별 고려엉겅퀴의 Pectolinarin 함량 및 항산화 활성

        This study was performed to provide basic data of Cirsium setidens Nakai by cultivars that will be applied for development of functional foods and ingredients. We assessed pectiolinarin content, total flavonoids content and antioxidant effects (DPPH radical scavenging activity and ORAC assay) of C. setidens Nakai. Our results showed that the pectolinarin and total flavonoids contents of C. setidens Nakai by cultivars ranged from 3.95±0.05 to 7.29±0.07 mg/g and from 40.42±0.91 to 76.70±2.24 mg pectolinarin equivalent (PNE)/g, respectively. Among C. setidens Nakai by cultivars, the pectolinarin content was highest in GW-D extract. Futhermore, the DPPH radical scavenging activity of C. setidens Nakai ranged from 31.25 to 81.93%, respectively. The oxygen radical absorbance capacity (ORAC) value was highest in GW-D and GW-E extracts (514.49 and 501.73 μM TE/g, respectively). These results suggest that C. setidens Nakai extract could be considered as a good sources of natural antioxidants and functional food ingredients.

        Persicaria senticosa Ameliorates Imiquimod-induced Psoriasis-like Skin Lesions in Mice via Suppression of IL-6/STAT3 Expression and Proliferation of Keratinocytes

        Psoriasis is one of the most common inflammatory skin disorders, with a global prevalence of 2% –3%. It is an autoimmune skin disorder characterized by excessive generation of plaques on the skin with typical long-lasting red, itchy, and scaly lesions. In this study, we aimed to elucidate the anti-psoriatic effect of the methanolic extract of Persicaria senticosa (PS), a bioactive edible plant extract used in traditional medicine, using a mouse model of imiquimod (IMQ)-induced psoriasis. The daily topical application of IMQ could induce human psoriasis-like lesion. The extract ameliorated IMQ-induced psoriasis. Furthermore, hematoxylin and eosin staining and the Psoriasis Area and Severity Index (PASI) scores indicated that topical application of PS led to an improvement in erythema, scaling, and thickness scores of the mouse dorsal skin and a considerable decrease in the epidermal thickness of the ear and dorsal skin in the IMQ-induced psoriatic mouse model. We also studied the effect of PS on the proliferation of keratinocytes using HaCaT cells. The extract inhibited cell proliferation and IL-6 and pSTAT3 expression induced by M5 cocktail (comprising interleukin [IL]-1α, IL-17A, IL-22, oncostatin M, and tumor necrosis factor-α) in HaCaT cells. Thus, PS might serve as a potential therapeutic agent for the treatment of psoriasis.

        6-O-Galloylsalidroside, an Active Ingredient from Acer tegmentosum, Ameliorates Alcoholic Steatosis and Liver Injury in a Mouse Model of Chronic Ethanol Consumption

        We have previously reported that Acer tegmentosum extract, which is traditionally used in Korea to reduce alcohol-related liver injury, suppresses liver inflammation caused by excessive alcohol consumption and might improve metabolism. The active ingredient, 6-O-galloylsalidroside (GAL), was isolated from A. tegmentosum, and we hypothesized that GAL could provide desirable pharmacological benefits by ameliorating physiological conditions caused by alcohol abuse. Therefore, this study focused on whether GAL could ameliorate alcoholic fat accumulation and repair liver injury in mice. During chronic alcohol consumption plus binge feeding in mice, GAL was administered orally once per day for 11 days. Intrahepatic lipid accumulation was measured in vivo using a noninvasive method, 1H magnetic resonance imaging, and confirmed by staining with hematoxylin and eosin and Oil Red O. The serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were measured using a Konelab system, and the triglyceride content was measured in liver homogenates using an enzymatic peroxide assay. The results suggested that GAL alleviated alcohol-induced steatosis,e as indicated by decreased hepatic and serum triglyceride levels in ethanol-fed mice. GAL treatment also correlated with a decrease in the Cd36 mRNA expression, thus potentially inhibiting the development of alcoholic steatosis via the hepatic de novo lipogenesis pathway. Furthermore, treatment with GAL inhibited the expression of cytochrome P450 2E1 and attenuated hepatocellular damage, as reflected by a reduction in ALT and AST levels. These findings suggest that GAL extracted from A. tegmentosum has the potential to serve as a bioactive agent for the treatment of alcoholic fatty liver and liver damage.

        DSS로 유도한 마우스의 대장염에 대한 돌배 추출물의 개선효과

        Pyrus pyrifolia (Burm.f) Nakai fruit has been used as a traditional medicine for fever, cough, asthma, and rheumatic arthritis. This study aimed to investigate the anti-inflammatory effects of P. pyrifolia Nakai fruit extract (PPE) against DSS-induced colitis in mice. PPE—water extract (PWE), ethanol extract (PEE), or fermented extract (PFE)—was administered for 30 days. After 21 days of PPE treatment, ulcerative colitis was induced by oral administration of 2.5% DSS for 5 days. The mice were divided into seven groups: one normal control (NC) group and six groups with different treatments—DSS, DSS + 100 mg/kg PWE (PWL), DSS + 400 mg/kg PWE (PWH), DSS + 100 mg/kg PEE (PEL), DSS + 400 mg/kg PEE (PEH), DSS+ PFE (1:4 dilution, PFE), respectively. Mice treated with DSS showed severe bloody stools, diarrhea, and weight loss. Colon tissue in DSS group exhibited a marked decrease in its length. However, PPE treatment improved the disease activity index score and colon length. PPE alleviated DSS-induced inflammatory colon lesions. The level of IL-1β, a proinflammatory cytokine, was significantly decreased in PFE group. Moreover, colonic myeloperoxidase activity was significantly reduced in all PPE-treated groups, except in PWL group. These results show that PPE can be used for developing effective and safe functional health products for inflammatory bowel disease.

        고려엉겅퀴 주정추출물의 안정성 조사

        This study was assessed stability of ethanolic extract from Cirsium setidens Nakai that will be applied for development of functional foods and ingredients. We evaluated pectolinarin content, total phenol content and antioxidant activity (DPPH radical scavenging activity and FRAP assay) of ethanolic extract from C. setidens Nakai against various temperature(4, 25 and 50℃) and pH(4.0, 7.0 and 10.0). Our results show that the pectolinarin and total phenol contents in ethanolic extract from Cirsium setidens Nakai slightly reduced during the storage periods. Moreover, the DPPH radical scavenging activity at 4℃ was higher than those of other temperature. Pectolinarin content, total phenol content and DPPH radical scavenging activity of ethanolic extract from Cirsium setidens Nakai at acidic (pH 4.0) and neutral (pH 7.0) pH ranges were higher than at alkaline pH ranges. These results indicate that the optimum storage condition of C. setidens Nakai ethanol extract are temperature 4℃ and pH 4.0-7.0 ranges.

        Standardized Cirsium setidens Nakai Ethanolic Extract Suppresses Adipogenesis and Regulates Lipid Metabolisms in 3T3-L1 Adipocytes and C57BL/6J Mice Fed High-Fat Diets

        Cirsium setidens Nakai, a wild perennial herb, grows mainly in Gangwon province, Korea, and has been reported to contain bioactive ingredients with various medicinal activities, including the treatment of edema, bleeding, and hemoptysis. However, the potential antiobesity effects of C. setidens Nakai have not been fully investigated. This study evaluated the antiobesity effect of standardized C. setidens Nakai ethanolic extract (CNE) in 3T3-L1 adipocytes and in obese C57BL/6J mice fed a high-fat diet. CNE suppressed the expression of lipogenic genes and increased the expression of lipolytic genes. The antiadipogenic and antilipogenic effects of CNE appear to be mediated by the inhibition of peroxisome proliferator-activated receptor γ (PPARγ) and CCAAT/enhancer-binding protein (C/EBP) expressions. Moreover, CNE stimulated fatty acid oxidation in an AMPK-dependent manner. CNE-treated groups of C57BL/6J mice showed reduced body weights and adipose tissue weight and improved serum lipid profiles through the downregulation of PPARγ, C/EBPα, fatty acid binding protein 4 (FABP4), sterol regulatory element binding protein-1c (SREBP-1c), and fatty acid synthase (FAS) and the upregulation of adiponectin and carnitine palmitoyltransferase-1 (CPT-1) in obese C57BL/6J mice fed a high-fat diet. These results suggest that CNE may have an antiobesity effect on adipogenesis and lipid metabolism in vitro and in vivo and present the possibility of developing a treatment for obesity with nontoxic natural resources.

      • 유용미생물발효를 적용한 고려엉겅퀴의 면역증강효능연구

        Background : Cirsium setidens is a perennial wild herb that belongs to the Asteraceae family. It grows in the mountainous region of Gangwon-do in Korea and is also called gondre. The bioconversion technology applied in this study has the same meaning as biosynthesis, biocatalyst, etc., and refers to a technique for producing desired products from precursors using enzymatic functions of microorganisms. Therefore, useful microorganisms having immunological activity were selected and Cirsium setidens fermentation products were prepared by applying bioconversion technology. And fermented product extracts were prepared to consider as a good sources of natural immune enhancement and functional food ingredients. Methods and Results : Lactobacillus fermentum, Lactobacillus plantarum, Saccharomyces cerevisiae, Weissella cibaria, and Lactobacillus plantarum were used as isolates from traditional foods. The fermentation product was set to a condition that the inherent physical properties did not change and did not generate a unique odor during fermentation. Cirsium setidens was fermented at 37℃ for 24 hours. And the fermented material was sterilized at 9 0℃ for 1 hour and then dried at 70℃ and pulverized. The contents of pectolinarin and pectolinarigenin, which are non - glycosides, were analyzed before and after fermentation using HPLC. Also NO production was measured in RAW264.7 cells after extract treatment at various concentrations using Griess reagent kit . The content of pectolinarin was increased in fermented Crisium setidens before fermentation, but the content of pectolinarigenin was increased after fermentation. In addition, the water extract of the fermented material accelerated the NO production compared to the pre - fermented material. Conclusion : As a result, relatively high immunostimulating effect were observed in dried Crisium setidens after fermentation, and it was confirmed that it could be a ingredient material for health functional food.

        DSS로 유도된 궤양성 대장염 모델에서의 돌단풍 추출물의 장 건강 개선효과

        Inflammatory bowel disease (IBD) is a chronic and relapsing inflammatory disease of the intestine characterized by recurrent chronic inflammation and mucosal damage in the gastrointestinal tract. Recent studies have demonstrated that Aceriphyllum rossii extract (ARE) contains abundant natural flavonoids and enhances the anticancer, antioxidant, and anti-inflammatory effects in various disease models. However, whether ARE has a protective effect against dextran sodium sulfate (DSS)-induced colitis remains unclear. In the present study, we investigated the effects of ARE treatment in a mouse model of colitis. Oral administration of DSS induced shortening of the colon and increased the disease activity index and histological damage. However, ARE treatment significantly attenuated IBD symptoms, such as weight loss, bloody stool, stool consistency, and histological damage in DSS-induced colitis. In addition, ARE treatment decreased colonic inflammatory cytokine (IL-6 and IL-22) levels. Collectively, these results suggest that ARE treatment can help improve colonic function and prevent IBD.
