        Streptococcus mutans에 대한 홍삼박 n-hexane 추출물과 Arginine의 병용 효과

        본 연구에서는 항충치 활성을 보이는 지용성의 홍삼박 n-hexane 추출물과 수용성의 arginine의 병용이 S. mutans의 생육에 미치는 영향을 조사하였다. Checkerboard assay로 분석한 결과, 홍삼박 n-hexane 추출물과 arginine은 fractional inhibitory concentration index 0.396으로 S. mutans의 생육 저해에 시너지 효과를 나타내었다. 홍삼박 n-hexane 추출물과 arginine의 병용은 핵산 성분의 유출과 생균수의 감소를 초래하였으며, 이는 모두 홍삼박 nhexane 추출물의 농도에 비례하였다. 결론적으로 S. mutans의생육 저해에 대한 홍삼박 n-hexane 추출물과 arginine의 시너지효과는 주로 홍삼박 n-hexane 추출물이 기여하는 것으로 판단된다.

        홍삼박 n-hexane 추출물의 충치를 유발하는 Streptococcus mutans 저해 효과

        In this study, to investigate the anticariogenic effect of red ginseng, the antibacterial effect, cell adherence and biofilm formation inhibitory effect of n-hexane extract of red ginseng marc (HERGM) against Streptococcus mutans, the causative bacteria of caries, were measured. The growth of S. mutans was inhibited in proportion to the concentration of HERGM, and was hardly observed at a concentration above 125 μg/mL (MIC =125 μg/mL). It was found that HERGM acts on the cell membrane and the nucleic acid component of the cell was leaked. In addition, HERGM inhibited the adherence and biofilm formation of S. mutans by more than 90% at a concentration of 125 μg/mL. GTase activity was completely inhibited at a concentration of 50 μg/mL of HERGM. In conclusion, it was found that HERGM commonly inhibited the growth and biofilm formation of S. mutans. 본 연구에서는 홍삼의 항충치 효과를 조사하기 위하여 홍삼박n-hexane 추출물의 충치 원인균 Streptococcus mutans에 대한항균 효과, 균체 부착과 생물막 생성 저해 효과를 측정하였다. 홍삼박 n-hexane 추출물의 농도에 비례하여 S. mutans의 생육이 저해되었으며, 125 μg/mL 이상의 농도에서는 거의 관찰되지않았다(MIC=125 μg/mL). 홍삼박 n-hexane 추출물이 세포막에작용하여 세포의 핵산 성분이 유출시키는 것으로 나타났다. 그리고, 홍삼박 n-hexane 추출물은 125 μg/mL 이상의 농도에서S. mutans의 부착과 생물막 형성을 90% 이상 저해하였다. GTase 활성은 50 μg/mL 농도에서 완전히 저해되었다. 결론적으로 홍삼박 n-hexane 추출물은 S. mutans의 생육과 생물막 형성을 공통적으로 저해하는 것으로 나타났다.

        홍삼추출물이 LPS로 유도된 RAW 264.7 cell의 염증반응에 미치는 효과

        We conducted this study to investigate anti-inflammatory possibilities of applying cosmetic material about extracts from red ginseng. For this we carried out biological active evaluation about anti-inflammatory by using extracts of red ginseng. In order to evaluate the anti-inflammatory effects of the samples in macrophages (RAW 264.7 cells), MTT assay was used to evaluate the toxicity of red ginseng extracts and the inhibitory activity of nitric oxide production and the expression levels of inflammation-related proteins and genes. The inhibitory activity of nitric oxide in the LPS-induced RAW 264.7 cells was 71.2% at 25 μg/ml concentration and western blot analysis showed that the expression of iNOS and COX-2 protein decreased in a concentration-dependent manner. These results suggest that extracts from red ginseng may have value as the potential cosmetic materials. 본 연구는 홍삼추출물의 화장품소재로서의 항염증 효과의 가능성을 확인하는 것을 목적으로 한다. 본 연구에서는 홍삼 추출물을 사용하여 항염증에 대한 생물학적 활성평가를 수행하였다. 대식세포인 RAW 264.7 세포에서 시료의 항염증 효과를 평가하기 위해 MTT assay를 이용한 홍삼 추출물의 독성평가와 nitric oxide 생성 저해 활성 및 염증관련 단백질 및 유전자의 발현량을 확인하였다. LPS로 유도된 RAW 264.7 세포 내에서 시료의 nitric oxide 저해활성은 25 μg/ml에서 71.2 %의 우수한 효능을 나타내 었으며, western blot 시험결과 iNOS, COX-2 단백질의 발현은 농도 의존적으로 감소하는 것으로 확인하 였다. 이러한 실험 결과를 바탕으로 홍삼추출물이 항염 효과를 가진 화장품 소재로서의 가치를 제안할 수있다.

        홍삼 추출물의 제조에서 단백질 분해효소의 활용

        In this study, proteolytic enzymatic treatment conditions for Korean red ginseng were examined to increase the extraction yield. Commercially available proteases were screened to obtain high protein and carbohydrate yield. The optimal dosage and reaction time for Alcalase, the chosen protease, were found to be 2.0% (w/w) and 1.5 h, respectively. Treatment with optimal conditions of Alcalase increased solid yield, total phenolic content and gensenosides content by 57.6, 81.8, and 33.8%, respectively, over levels in non-treated Korean red ginseng. Antioxidative activities evaluated by free radical scavenging activity, cation radical scavenging activity and reducing power were exactly similar between Alcalase-treated and non-treated extracts. 본 연구에서는 홍삼 추출물 제조과정에서 추출 수율을 향상시킬 수 있는 효소를 선별하고 최적의 반응조건을 조사하였다. 상업용 단백질 분해효소 중 Alcalase가 단백질과 탄수화물 수율향상에 효과적이었으며, 효소 사용량은 홍삼 중량의 2%, 반응시간은 1.5시간이 적당하였다. 최적의 반응조건으로 홍삼을Alcalase로 처리한 결과 대조구보다 고형분 수율은 45.1%에서 71.1%로 50% 이상, 총페놀 함량은 0.44%에서 0.80%로 80% 이상 증가하였으며, 항산화 활성은 대조군과 매우 유사하였다. 또한 진세노사이드 함량은 대조구의 1.48 mg/g에서 1.98 mg/g으로 30% 이상 증가하였다.

        축산물 및 가공 : 물 및 에탄올 추출을 통해 제조된 홍삼 추출물의 첨가가 저장 중 가열 돈육패티의 툼질특성에 미치는 영향

        The objective of this study was to investigate the effect of red ginseng extracted with water extract(WE) and 50% ethanol extract(EE) from white ginseng on cooked meat patties during storage. Different concentrations of extracts were examined (0.25%, 0.5%, and 1.%, respectively, dry base w/w). A significantly higher water holding capacity(WHC) was observed in samples supplemented with ≥0.5% WE(p<0.01); however, EE had no significant effect on the WHC of meat patties. Samples supplemented with ≥0.5% WE or EE showed a significantly higher redness(a*-value) compared to the control(p<0.01). The total plate counts(TPC) and 2-thiobarbituric acid-reactive substances (TBARS) of all treated samples were lower than those of the control. However, there were no significant differences in volatile basic nitrogen(VBN) values and sensory evaluation scores between the samples. These results suggest that red ginseng extract improves color and inhibits lipid oxidation and bacterial population at doses >0.25%, prolonging the shelf-life of meat products and acting as a natural colorant.

        Streptococcus mutans의 생물막 생성에 대한 L-arginine과 홍삼박 n-hexane 추출물의 병용 효과

        In this study, the combined effects of lipid-soluble nhexane extract of red ginseng marc (HERGM) and water-soluble L-arginine, which has been reported to have anticaries activities, on biofilm formation and glucosyltransferase (Gtase) activity of S. mutans were investigated by checkerboard assay method. The combination of HERGM and L-arginine showed additive effects on both inhibition of biofilm formation and GTase activity of S. mutans with a fractional inhibitory concentration index of 0.75, respectively. When HERGM and L-arginine were treated together, the inhibition of biofilm formation was mainly attributed to Larginine, and the inhibition of GTase activity was attributed to HERGM. These results indicate that L-arginine and HERGM complementarily inhibits the biofilm formation of S. mutans.

        추출 방법에 따른 홍삼의 항산화 및 항염 효과

        Red ginseng (RG) is known for its preventive effects on various disease, including inflammatory diseases and cancer. However, few studies have compared the effects of different RG extraction methods. In this study, the effect of RG extracts prepared by agitation or ultrasound on antioxidant and anti-inflammatory activities was investigated. Total phenol and flavonoid content and DPPH radical scavenging activity were considerably increased by the RG extracts in dose-dependent manners, but there was no significant difference between the extraction methods. Raw 264.7 macrophages treated with up to 400 μg/mL of RG extracts proliferated significantly but decreased to less than 50% at 800 μg/mL of either extract. The expression of the inflammatory marker COX-2 protein was decreased in a dose-dependent manner, especially with ultrasonicated RG extract, indicating increased anti-inflammatory activity of RG. In conclusion, our data demonstrated that RG extract prepared by either agitation or ultrasound extraction methods had antioxidant effects and promoted immune cell proliferation. However, RG extract extracted by ultrasound exerted a greater anti-inflammatory effect on immune cells, suggesting that the extraction method should be selected based on the intended application.

        Origanum Oil과 홍삼 및 녹차추출물이 구강내 미생물과 휘발성황화합물에 미치는 영향

        Objectives. The purpose of this study was to estimate the effects of origanum oil, red ginseng extract, and green tea extract on oral microorganisms and volatile sulfur compounds. Methods. Minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) were determined to estimate the growth inhibition of oral microorganisms including the 8 bacteria Streptococcus, Lactobacillus, Fusobaterium, Porphyromonas, Actinobacillus, and Prevotella. The reduction of the volatile sulfur compound (VSC) CH3SH was estimated by gas chromatography with flame photometric detector after mixing with experimental substances for 4 minutes within a 10 ml vial. Results. Among three experimental substances (origanum oil, red ginseng extract, and green tea extract), origanum oil had the lowest MIC and MBC. Origanum oil was more effective on gram (+) Streptococcus. However, the MIC and MBC of origanum oil were lower than chlorehexdine. The reduction of CH3SH on origanum oil was statistically significant (p<0.05) compared to other experimental and control substances. Conclusions. Among the three experimental substances, origanum oil showed the best reduction of oral malodor due to lower MIC and MBC, and higher reduction of CH3SH.
