        Alcaligenes faecalis에 의한 $\beta$-glucan의 생합성과 구조 분석

        본 연구에서는 Alcaligenes faecalis에 의해 생산되는 세포 외 다당류인 $\beta$-glucan의 생합성에서 탄소원, 질소원의 영향과 생합성된 curdlan의 구조를 살펴보았다. 먼저 $\beta$-glucan의 생합성을 확인하기 위해 Aniline Blue 배지에서 청색 colony들을 선별하고 Cellufluor 배지에서 형광성을 보임으로 $\beta$-1,3 glucan의 생합성을 확인하였다. Flask 배양에서는 탄소원이 glucose이고 질소원이 ($NH_4$)$_2$$SO_4$일 때 가장 많은 $\beta$-glucan을 생산하였다. 탄소원을 glucose로 질소원을 (NH$_4$)$_2$$SO_4$로 사용한 발효조 배양에서는 flask 배양에 비해 생산량이 7.2%가 증가되었다. 질소원을 제한한 발효에서는 최종 생산수율이 38.82%였고 질소원을 제한하지 않은 발효보다 3.02%가 증가 되었다. 분리 정제된 curdlan의 FT-IR의 분석에서는 지문영역인 890$cm^{-1}$ /에서 $\beta$-form이 관찰되었고 $^{13}C$-NMR 분석에서는 $C_1$-103.5 ppm, $C_2$-74.3 ppm, $C_3$-89.8 ppm, $C_4$-68.8 ppm, $^{5}C$-75.5 ppm, $C_2$-61.8 ppm에서 검출되어 homogeneous $\beta$-1,3 glucan으로 확인되었다. curdlan의 triple-helix coil 형태에서 random coil 형태로의 전이는 0.1에서 0.25 N NaOH의 농도에서 일어남이 확인되어 중성 상태의 curdlan은 triple-helix coil 형태를 가지지만 알칼리 상태에서는 그 구조가 약해져 random-coil로 전이 되어짐을 알 수 있었다. 이는 본 연구에서 분리 정제된 curdlan이 정확한 $\beta$-결합을 가진 glucan임이 확인되었으며 생합성된 curdlan의 구조와 분자적인 특성을 확인하였다. 본 연구를 통하여 $\beta$-결합을 가지는 glucan의 신속한 선별법을 구축할 수 있었고 이는 새로운 기능을 가지는 $\beta$-glucan 생합성 효소나 분해효소의 탐색에 이용될 것으로 기대된다. Biosynthesis of curdlan( ${\beta}$-1,3 glucan) was shown by fluorscence on cellufluor medium. The highest production of curdlan was produced when glucose was used as a carbon source and ($NH_4$)$_2$$SO_4$ was used as a nitrogen source. ${\beta}$ -form of curdlan was detected in the fingerprint region (890 $cm^{-1}$) by FT-IR spectrum and shown homogeneous ${\beta}$ -1,3 glucan by $^{13}C$ NMR spectrum ($C_1$-103 ppm, $C_2$-73.2 ppm, $C_3$-86.4 ppm, $C_4$-68.7 ppm, $C^{5}$-76.63 ppm, $C_{6}$-61.2 ppm). Transition of structure from triple helix coil form to random coil form was appeared at 0.1 ∼0.25 M NaOH concentration. It was shown that natural curdlan is a triple helix form in neutral but becomes weak in alkaline condition.

        $\beta$-Glucan 투여에 의한 조피볼락(Sebastes schlegeli)의 세균성 질병에 대한 저항성 향상

        $\beta$-Glucan을 경구 혹은 침지투여하여 조피볼락(Sebastes schlegeli)의 비특이적 방어기작을 향상시켜 세균성 질병에 대한 저항성을 증가시키는 면역자극제로서의 효과를 알아보고자 하였다. $\beta$-Glucan을 사료에 섞어 경구투여하거나 혹은 사육수에 현탁시켜 침지투여한 후 Vibrio ordalii, Staphylococcus epidermidis 및 Edwardsiella tarda를 주사하여 인위감염으로 $\beta$-glucan의 효능을 시험하였다. V. ordalii를 주사한 결과, 1% $\beta$-glucan을 30일 동안 경구투여한 시험구는 25%의 생존율을 보였으나 $\beta$-glucan을 투여하지 않은 대조구는 3일 이내에 모두 폐사하였다. S. epidermidis를 주사한 결과, 20 및 30일 경구투여구는 95%의 높은 생존율을 보였다. 그러나 E. tarda의 인위감염시 전혀 방어효과가 없었다. V. ordalii 사균혼합구 혹은 단독 침지 시험구는 주사 후 10일 동안 전혀 방어효과가 관찰되지 않았다. 이러한 결과로 볼 때 $\beta$-glucan의 경구투여는 S. epidermidis와 V. ordalii에 효과적이나 E. tarda에는 방어효과가 없는 것으로 나타났다. The effect of $\beta$-glucan as an immunostimulant to increase resistance to bacterial diseases by enhancing non-specific defense mechanism in rockfish (Sebastes schlegeli) was examined by oral and bath administration. After oral or bath administration with $\beta$-glucan, the injection challenges with Vibro ordalii, Staphylococcus epidermidis and Edwardsiella tarda were performed to assess $\beta$-glucan efficacy. After injection of V. ordalii, oral administration for 30 days with 1% $\beta$-glucan showed 25% of survival rate. But all control fish died within 3 days after the injection. After injection of S, epidermidis, oral administration group for 20 and 30 days showed a remarkably increased survival rate of 95%. But oral administration of $\beta$-glucan to rockfish did not induce protection against experimental E. tarda infection. $\beta$-Glucan bath administration with or without formalin-killed V. ordalii showed that no protection was observed at 10 days after challenge. The results show that $\beta$-glucan to rockfish was effective to increase survival rate of bacterial infections of S. epidermidis and V. ordalii but not against E. tarda.

        Cloning and Molecular Characterization of ${\beta}$-1,3-Glucan Synthase from Sparassis crispa

        A ${\beta}$-glucan synthase gene was isolated from the genomic DNA of polypore mushroom Sparassis crispa, which reportedly produces unusually high amount of soluble ${\beta}$-1,3-glucan (${\beta}$-glucan). Sequencing and subsequent open reading frame analysis of the isolated gene revealed that the gene (5,502 bp) consisted of 10 exons separated by nine introns. The predicted mRNA encoded a ${\beta}$-glucan synthase protein, consisting of 1,576 amino acid residues. Comparison of the predicted protein sequence with multiple fungal ${\beta}$-glucan synthases estimated that the isolated gene contained a complete N-terminus but was lacking approximately 70 amino acid residues in the C-terminus. Fungal ${\beta}$-glucan synthases are integral membrane proteins, containing the two catalytic and two transmembrane domains. The lacking C-terminal part of S. crispa ${\beta}$-glucan synthase was estimated to include catalytically insignificant transmembrane ${\alpha}$-helices and loops. Sequence analysis of 101 fungal ${\beta}$-glucan synthases, obtained from public databases, revealed that the ${\beta}$-glucan synthases with various fungal origins were categorized into corresponding fungal groups in the classification system. Interestingly, mushrooms belonging to the class Agaricomycetes were found to contain two distinct types (Type I and II) of ${\beta}$-glucan synthases with the type-specific sequence signatures in the loop regions. S. crispa ${\beta}$-glucan synthase in this study belonged to Type II family, meaning Type I ${\beta}$-glucan synthase is expected to be discovered in S. crispa. The high productivity of soluble ${\beta}$-glucan was not explained but detailed biochemical studies on the catalytic loop domain in the S. crispa ${\beta}$-glucan synthase will provide better explanations.

        영지버섯에서 추출한 $\beta$-glucan 이 자돈의 생산능력에 미치는 영향

        영지버섯에서 추출한 균사체 함량은 배지 L당 8.52g, 균체외 다당류는 4.49g으로 $\beta$-glucan 생산능력이 우수한 영지버섯을 선발하였으며, 대두분과 수용성전분 등을 이용한 최적의 산업용 버섯배양배지를 개발하였다. 원심분리와 냉동건조방법으로 제조한 $\beta$-glucan을 자돈사료에 첨가하여 이유자돈의 성장능력, 설사, 폐사율 및 경제성을 비교하여 항생제대체효과를 구명하기 위하여 실시하였다. 삼원교잡종(Landrace${\times}$Large White${\times}$Duroc) 이유자돈(8.6kg) 144두를 선발하여 28일간 사양시험을 실시하였다. 본 시험은 4처리 4반복의 난괴법 배치로 T1 처리구(-control)는 무항생제, T2 처리구(+control)는 항생제, T4 처리구는 무항생제에 $\beta$-glucan 0.2% 첨가, T3 처리구는 항생제와 $\beta$-glucan 0.2% 첨가하였다. 이유전기 (phase I, 0~14일)에서는 일당증체량과 사료섭취량은 처리간에 유의적인 차이가 없었으나 사료효율은 처리간에 차이가 있었다(p<0.05). 한편 이유후기(phase II, 15~28일)와 전 기간(0~28일)에서는 사료섭취량과 사료효율은 처리간에 유의적인 차이가 없었으나 일당증체량은 처리간에 유의적인 차이가 있었다(p<0.05). 전 기간의 일당증체량의 비교에서 설사지수와 분변의 수분함량은 T3와 T4 처리구가 다른 처리구보다 높았다(p<0.05). 이유자돈 분변의 수분함량은 T3와 T4 처리구가 다른 처리구보다 높았으나(p<0.05), 설사지수와 폐사율은 처리간에 유의적인 차이가 없었다. 이유자돈 시험의 경제성분석 결과 이유전기에서는 항생제와 $\beta$-glucan을 첨가함에 따라 사료비가 증가하였으나, 섭취량과 증체량을 고려한 사료비에서는 $\beta$-glucan을 첨가한 처리구(T3 및 T4)가 첨가하지 않은 처리구(T1 및 T2)보다 감소하여 $\beta$-glucan의 효과가 있었다. 그러나 이유후기의 $\beta$-glucan의 효과가 이유전기의 효과가 적 었다. 이상의 이유자돈시험을 결과를 종합해 보면 기능성 $\beta$-glucan은 이유자돈의 증체량, 영양소 소화율 및 경제성에 효과가 있었으며, 특히 항생제 대체제로서 일정 부분 개선 효과가 있는 것으로 사료된다. This experiment was selected a $\beta$-glucan producing mushroom strain and developed industrial media, and used to $\beta$-glucan as an alternative for antibiotics in weaned pigs. Yields of mycelial biomass and extracellular $\beta$-glucan from Youngji (Ganoderma lucidum) mushroom was 8.52g/L and 4.49g/L respectively. Also, we prepared optimum formula for mushroom cultivations. A total of 144 pigs ($8.6{\pm}0.9$ kg average body weight, weaned $20{\pm}3$ days of age) were allotted to 4 different treatment groups and replicated 4 times with 8 pigs per replicate in randomized complete block design. Treatments were T1) NC (negative control, basal diet), T2) PC (positive control, basal diet+0.255% antibiotics), T3) NC+0.2% $\beta$-glucan and T4) PC+0.2% $\beta$-glucan. The T2 and T4 treatments were significantly higher in feed efficiency by antibiotics group (p<0.05), however, there was no significant differences in terms of average daily gain (ADG) and average daily feed intake (ADFI) during phase I (0~14 days). In phase II (15~28 days), Pigs fed with antibiotics and $\beta$-glucan (T4) had greater ADG than other treatments (p<0.05), while no differences were observed in ADFI and feed efficiency. During the whole experiment period, the ADG of T4 treatment was higher than other treatments. Pigs fed with $\beta$-glucan (T3 and T4) had greater diarrhea score and moisture content than other treatments (p<0.05). Pigs fed with $\beta$-glucan (T3 and T4) had greater moisture content than other treatments (p<0.05). However, there was no significant differences in diarrhea score and mortality of weaned pigs. There was marginal reductions in feed cost measured feed cost per weight gain used in antibiotics and $\beta$-glucan added diet during phase I. In the second phase, the treatment supplemented with antibiotics had a significantly lower feed cost per weight gain compared to the other treatments. The results from these experiments suggests that $\beta$-glucan is likely able to improve the growth performance, and reduce feed cost although they do not have similar effects like antibiotics in weaning pigs.

        β-Glucan 추출에 미치는 온도, 용매 농도 및 pH의 영향

        추출조건에 따른 귀리겨 ${\beta}$-glucan의 추출조건을 확립하기 위하여 추출온도($40{\sim}60^{\circ}C$), 추출용매의 에탄올 농도(0~20%) 및 pH(5~9)를 변수로 중심합성계획법으로 설계하여 ${\beta}$-glucan 추출특성을 조사하였다. 추출시간의 증가에 따라 ${\beta}$-glucan의 추출속도는 추출초기에 급격히 증가하였으나, 이후에는 완만하였다. 총괄물질전달계수는 $3.363{\sim}8.552{\times}10^{-6}cm/min$ 범위로 낮은 추출온도에서는 추출용매의 농도가 낮을수록, 높은 추출온도에서는 추출용매의 농도가 높을수록 증가하였지만 pH에 의한 영향은 적은 것으로 나타났다. 추출용매의 pH는 중성일 때보다 산성이나 알칼리성일 때 ${\beta}$-glucan의 추출량이 증가하였다. 추출조건에 따라서 ${\beta}$-glucan 수용액의 점도는 유의적인 차이를 보였으며, 총괄물질전달계수가 높을수록 crude ${\beta}$-glucan 수용액의 점도가 높게 나타났으며, 점도, 추출율 및 총괄물질전달계수의 상관관계를 분석한 결과, 각각의 요인들이 높은 양의 상관관계(p<0.01)를 나타내었다. This study investigated the effects of temperature, solvent concentration, and pH on the ${\beta}$-glucan extraction. Oat bran ${\beta}$-glucan was extracted with different extraction conditions, using various combinations of experiment factors, such as temperature (40, 45, 50, 55, and $60^{\circ}C$), ethanol concentration (0, 5, 10, 15, and 20%), and pH (5, 6, 7, 8, and 9). Under the various extraction conditions, ${\beta}$-glucan extraction rate and overall mass transfer coefficient of oat bran ${\beta}$-glucan, and viscosity of oat bran extracts were investigated. As increasing the extraction time, the extraction rate of ${\beta}$-glucan increased. The overall mass transfer coefficient of ${\beta}$-glucan ranged from $3.36{\times}10^{-6}$ to $8.55{\times}10^{-6}cm/min$, indicating the lowest at the extraction condition of $45^{\circ}C$, 15% and pH 8, and the highest at $50^{\circ}C$, 0% and pH 7. It was significantly greater with increasing extraction temperature and decreasing ethanol concentrations of extraction solvent, except for solvent pH. There were positive correlations among the overall mass transfer coefficient, the extraction rate of ${\beta}$-glucan, and the viscosity of extract.

        Aureobasidium pullulans으로 부터 분리한 β-1,3/1,6-glucan의 면역활성의 연구

        ${\beta}$-Glucan은 효모나 버섯의 세포벽이나 곡물의 섬유에 들어있는 천연 화합물이며, 면역 시스템을 활성화 한다고 알려져 있다. Aureobasidium pullulans 수용성의 ${\beta}$-1,3/1,6-glucan을 생성한다. 본 연구에서는 A. pullulans 로부터 생성된 ${\beta}$-1,3/1,6-glucan의 면역활성에 관한 효과를 조사하기 위하여, 면역활성에 관여하는 NK 세로의 활성과 대식세포에 미치는 영향을 관찰하였다. NK 세포에 의한 Yac-1 세포의 사멸율은 ${\beta}$-1,3/1,6-glucan을 처리한 군은 처리하지 않은 대조군에 비하여 63-39% 증가하는 것으로 나타났고, 대식세포의 탐식작용은 zymosan을 처리한 군보다 15-21% 증가함을 보였다. LP-BM5 바이러스 복제 억제능을 확인하기 위하여 SC-1/LP-BM5 세포주에 ${\beta}$-1,3/1,6-glucan을 처리한 결과 $200{\mu}g/mL$ 처리에서 LP-BM5 바이러스 복제능이 최대 74% 유의적으로 감소하였다. 이 결과는 ${\beta}$-1,3/1,6-glucan이 면역활성에 관여하는 NK 세포와 대식세포를 활성화하여 면역체계가 민감하게 반응할 수 있도록 만들고, LP-BM5 바이러스 복제를 억제하여 면역력을 증진할 수 있다는 것을 보여준다. ${\beta}$-Glucan is a natural compound contained in cell walls of yeast or fungi, and cereal's fiber. It is also known to boost the immune system in human. Aureobasidium is a producer of water-soluble ${\beta}$-1,3/1,6-glucan. In this study, natural killer (NK) cell and macrophage activity were tested to investigate the effects of ${\beta}$-1,3/1,6-glucan isolated from A. pullulans on immune activity. Activation of NK cell was increased about 63-39% by the treatment of $10-200{\mu}g/mL$ ${\beta}$-1,3/1,6-glucan than control. Besides, only $10{\mu}g/mL$ of ${\beta}$-1,3/1,6-glucan was enough to boost activation of NK cell. Phagocytosis of macrophage was increased to 15~21% by the treatment of $10{\sim}200{\mu}g/mL$ of ${\beta}$-1,3/1,6-glucan than zymosan-treatment. In LP-BM5 proliferating inhibition test, relative mRNA level of LP-BM5 virus was decreased in ${\beta}$-1,3/1,6-glucan-treated cell about 36~74% than control. The decline of LP-BM5 mRNA level appeared to depend on the concentration of ${\beta}$-1,3/1,6-glucan. These results suggest that pure ${\beta}$-1,3/1,6-glucan from A. pullulans might be contributing to enhancement of immune activity through the activation of NK cell and phagocytosis of macrophage. Moreover, treatment of the ${\beta}$-1,3/1,6-glucan could increase the resistance to virus infection such as LP-BM5 through the restraining of the multiplication.

      • $\beta$-glucan Contents and Their Characteristics of Winter Cereals According to Particle Sizes and Milling Recoveries

        This study was conducted to investigate the $\beta$-glucan contents and their characteristics of winter cereals according to particle sizes and milling recoveries. Sieved fractions differed in their average contents of $\beta$-glucans, and the coarse fraction had higher contents of $\beta$-glucan than finely milled fractions. In all winter cereals, the $\beta$-glucan contents of raw flours were higher than those of their brans, and the highest $\beta$-glucan contents of every cereals were observed at 100 mesh > or 100-140 mesh fractions except the Chalssalbori fractions which showed the higest $\beta$-glucan contents (12.9%) at 140-200 mesh fraction. As compared with the $\beta$-glucan content of Chalbori among the various milling recoveries, the $\beta$-glucan was distributed more evenly throughout the endosperm but $\beta$-glucan content in bran of Chalbori was only 1.5%. However, $\beta$-glucan content of Chalssalbori (hull-less waxy barley) was the highest in the subaleurone region (8.2%) and declined slightly toward inner layers of grain. This results suggest that $\beta$-glucan distribution between high (Chalbori) and low $\beta$-glucan barley (Chalssalbori) may explain the difference in milling performance of barley. On the other hand, $\beta$-glucan contents of two rye varieties (Chilbohomil, Chunchoohomil) were lower than those of two waxy barley varieties, and the higest $\beta$-glucan contents were observed at the 60% milling recoveries. In all winter cereals, the L-values (lightness) of raw flours were higher than those of brans. And the L-values of barley varieties were higher than those of oat and rye varieties. As the particle sizes and milling recovery ratios were decreased, the L-value were increased. The a-values (redness) in brans of every winter cereals were higher than those of every particle size flours and every milling ratio fractions, and this tendency was observed in the b-values (yellowness) of every particle size of cereal flours. The L and b-value of barley, the b-value of oat, and L, a, b-value of rye have the significant relationship with the $\beta$-glucan contents, respectively. This results represent the fact that $\beta$-glucans affected the color of the flours and pounded grains of winter cereals.

        글루칸 투여에 의한 한국산 메기(Silurus asotus)의 Edwardsiella ictaluri와 Aeromonas hydrophila감염증에 대한 저항성의 증가

        $\beta$-glucan의 투여가 세균감염증에 대한 저항성에 미치는 영향을 알아보기 위하여 복강주사 또는 침지에 의해 한국산 메기(Silurus asotus)에 투여하였다. 투여 농도는 어체중 100 g 당 200, 500, $1000{\mu}g$을 1회 또는 3일 간격으로 2회 복강주사하거나, $100{\mu}g$/ml의 $\beta$-glucan 부유액에 30분 또는 1시간 침지시켰다. $\beta$-glucan 투여 3일후에 Edwardsiella ictaluri와 Aeromonas hydrophila의 생균액($2{\times}106$ CFU/0.1 ml)을 복강주사한 다음 10일간 사육하면서 생존율을 조사하였다. 복강주사와 침지에 의한 투여결과 복장투여만이 생잔율을 높일 수 있었으며, 투여 농도에 있어 어체중 100 g당 $200\sim1000{\mu}g$의 범위에 있어서는 농도간에 차이가 없었다. 또한 투여회수는 1회 투여보다는 3일간격으로 2회 투여가 효과적이었으며, E. ictaluri보다는 A. hydrophila에 대한 저항성이 높은 것으로 나타나, 한국산 메기에 $\beta$-glucan을 복강투여함으로서 세균감염증의 초기단계에 있어 생잔율을 높일 수 있었다. The effect of $\beta$-glucan administration on the resistance of Korean catfish(Silurus asotus) to experimental Edwardsiella ictaluri and Aeromonas hydrophila infection was evaluated. Fish were either intraperitoneally received $\beta$-glucan($200\sim1000{\mu}g$/100g body weight) dissolved in physiological saline once or twice at an intervals of 3 days, or placed in $\beta$-glucan bath($100{\mu}g/m\ell$) prepared with filtered tap water for 30 or 60 min. Bacterial challenge was performed by intraperitoneal injection of 0.1 ml of bacteria suspension($2{\times}10^7 CFU/m\ell$) 3 days after $\beta$-glucan administration. The $\beta$-glucan injected fish showed an significantly enhanced resistance against experimental infections. The resistance was much higher in the twice-injected fish than in the once-injected fish. But glucan bath did not affect survival rate after the challenge. The protective effect in the $\beta$-glucan injected fish was higher to A. hydrophila than to E. ictaluri. These results indicate that $\beta$-glucan injection can increase the resistance of Korean catfish against experimental E. ictaluri and A. hydrophila infection and that twice injection of $\beta$-glucan at an 3 day-intervals is more effective to the resistance.

        난소절제 흰쥐에서 신령버섯의 β-Glucan과 난각 Ca 복합체가 골 대사에 미치는 효과

        본 연구는 칼슘 급원으로서 $\beta-glucan-egg$ shell calcium complex($\beta-glucan-Ca$ 복합체)의 유효성을 확인하여 칼슘 보충제 개발에 활용하고자 실시하였다. 신령버섯에서 추출한 $\beta-glucan$과 난각의 Ca을 결합시켜 $\beta-glucan-Ca$ 복합체로 제조하여 골다공증 실험동물에서 $\beta-glucan$ 및 $\beta-glucan-Ca$ 복합체가 골 대사에 미치는 효과를 비교하였다. 10주령의 Sprague-Dewley 암컷 흰쥐의 양쪽 난소를 절제(OVX)한 후 실험식 이를 급여하였으며 Sham-대조군(Sham-C)은 실험군과 동일한 스트레스를 주기 위해 난소를 절제하지 않고 개복수술만 실시한 후 실험하였다. 식이조성은 Sham-C, OVX-대조군(OVX-C)와 $OVX-\beta-glucan$군(OVX-G)은 $CaCO_3$로 Ca 함량이 0.5%가 되도록 조정하였으며, OYX-G군은 $\beta-glucan$을 첨가하여 제공하였다. $OVX-\beta-glucan-Ca$ 복합체군(OVX-GE)은 $\beta-glucan-Ca$ 복합체의 Ca 농도가 0.5% 함유하도록 조정하여 실험식이를 각각 조제하여 6주간 급여하였다. 체중 100 g당 건조시킨 대퇴골의 무게는 Sham-C>OVX-GE>OVX-C와 OVX-G의 순으로 나타나 $\beta-glucan-Ca$ 복합체를 제공한 OVX-GE에서 다른 OVX군에 비해 상대적으로 골 무게가 증가하는 경향을 보였다. 대퇴골의 골밀도는 각 군 간의 유의적인 차이를 보였으며 Sham-C에 비해 $\beta-glucan$을 첨가한 식이를 제공받은 군에서 골밀도가 상대적으로 높은 수준을 보였고 특히 OVX-GE에서 가장 높은 골밀도를 보였다. Ca체내 흡수율과 보유량은 각 군 간에 유의적인 차이를 보였으며 OVX-C가 Sham-C에 비해 높았으며 OVX군에서는 $\beta-glucan$를 첨가한 군이 높은 수준을 보였다. ALP의 활성은 OVX군에서는 $\beta-glucan$을 첨가한 군이 다소 낮은 경향을 보였다. 요 중의 골 흡수 지표인 DPD crosslink value는 각 군 간의 유의적인 차이를 보였고 OVX군의 DPD crosslink value가 Sham-C에 비해 현저하게 높은 수준으로 나타나 OVX군의 골 흡수가 높음을 확인할 수 있었다. OVX군에서 $\beta-glucan$을 첨가한 식이를 제공한 OVX-G와 OYX-GE의 DPD crosslink value가 OVX-C에 비해 유의적으로 낮은 수준이었으며 $\beta-glucan$이 난소 절제 흰쥐 모델에서 골 대사에 영향을 미쳐 골 흡수율을 낮추어 골 보호효과를 나타내는 것으로 보인다. 이상의 결과들로 미루어 볼 때, 비록 $\beta-glucan$을 제공한 군과 $\beta-glucan-Ca$ 복합체를 제공한 두 군 간에는 유의적인 차이를 보이지 않았으나 난소절제 흰쥐모델의 골 대사에서 $\beta-glucan-Ca$복합체 형태가 골다공증 예방을 위한 기능성 식품으로서의 가능성을 보여주었으며, 앞으로 골다공증 예방을 위한 $\beta-glucan-Ca$ 복합체 형태의 효과적인 농도는 계속적으로 연구가 진행되어야 할 것으로 사료된다. This study was designed to evaluate the effect of Agaricus blazei $\beta-glucan$ and egg shell calcium complex on bone metabolism in ovariectomized (OVX) rats. Forty Sprague-Dewley female rats, 10 weeks of age $(248{\pm}1.7g)$, were divided into 4 groups and fed on the experimental diets for 6 weeks: sham operated control treated with normal diet containing 0.5% calcium (Sham-C), OVX-control treated with normal diet containing 0.5% calcium (OVX-C), $OVX-\beta-glucan$ group treated with $\beta-glucan$ diet containing 0.5% calcium (OVX-G), and $OVX-\beta-glucan$ egg shell calcium complex treated with $OVX-\beta-glucan$ egg shell calcium complex containing 0.5% calcium (OVX-GE). Bone weight of femur was higher in the OVX-GE group than in the other OVX groups. Bone mineral density of femur was significantly different (p<0.05) among the experimental groups and showed the highest level in the OVX-GE group. Calcium absorption rate and retention were higher in the $\beta-glucan$ supplement groups than in the other groups (p<0.05). Alkaline phosphatase activities and osteocalcin levels of serum showed lower in the $\beta-glucan$ supplement groups than in the OVX-C group. Deoxypyridinoline crosslink values of urine, indicator of bone absorption, showed the lowest in the OVX-GE group. The $\beta-glucan$ supplemented groups had a lower bone resorption ratio than in the OVX-C group. We concluded that bioavailability of calcium is higher in $\beta-glucan$ supplement groups compared to those in OVX rats. From the above results, these findings suggest the possibility of using $\beta-glucan$ egg shell calcium complex as a functional food material related to bone metabolism, even though there is no significant difference between the groups of $\beta-glucan$ and $\beta-glucan-egg$ shell calcium complex supplementation.

        Varietal and Annual Variations of β-Glucan Contents in Korean Barley (Hordeum vulgare L.) and Oat (Avena sativa L.) Cultivars

        Varietal and annual variations in the contents of ${\beta}$-glucan fractions per weight grain samples were examined in sixteen covered and eighteen naked barley and five oat cultivars developed in Korea. Also, the effect of pearling on ${\beta}$-glucan content was investigated. Average contents of total, soluble and insoluble ${\beta}$-glucan fractions were 5.25, 3.72, and 1.53%, respectively, in covered barley, and 5.86, 3.51, and 2.35%, respectively, in naked barley. Soluble ${\beta}$-glucan content was higher in covered barley, though total ${\beta}$-glucan content higher in naked barley. The total and insoluble ${\beta}$-glucan contents were higher in pearled grains. Total ${\beta}$-glucan content was higher in waxy barley than in non-waxy barley. Duwonchapssalbori, a two-rowed and waxy naked barley cultivar, was highest in total, soluble and insoluble ${\beta}$-glucan contents. Highly significant positive correlations were observed between total ${\beta}$-glucan and soluble ${\beta}$-glucan contents both in covered and naked barley. There were significant annual variations in total ${\beta}$-glucan content in barley. Average contents of total, soluble and insoluble ${\beta}$-glucans of oat cultivars were 4.33, 3.44, and 0.89%, respectively. Contents of all fractions of ${\beta}$-glucans were higher in barley than in oat. These results would be useful for the breeding of high ${\beta}$-glucan variety and also for the use barley and oat as valueadded food ingredients.
