      • Molecular genetic diversity and population structure of a selected core set in garlic and its relatives using novel SSR markers

        <P> <I>With 7 figures and 6 tables</I> </P><P><B>Abstract</B></P><P>Garlic is widely consumed for its culinary and medical benefits. Six hundred and thirteen accessions of garlic and its relatives with diverse origin were evaluated for genetic diversity at eight recently novel simple sequence repeat loci in this study. A total of 113 alleles were detected, the average allelic richness was 14.1 alleles per locus. Using a heuristic approach, a core set of 95 accessions was successfully developed, which showed 100% coverage of alleles with minimum redundancy. The model‐based structure analysis here revealed the presence of four subpopulations in the selected core set, which was basically consistent with clustering based on the genetic distance. The analysis of molecular variance based on this core set showed that between‐population component of genetic variance is <15.6% in contrast to 84.4% for the within population component. Overall <I>F</I><SUB>ST</SUB> value was 0.1560, indicating a moderate differentiation among the four groups. These results will provide an effective aid for future allele mining, association genetics, mapping and cloning gene(s), germplasm conservation, and improvement programs.</P>

      • Soluble c‐Met protein as a susceptible biomarker for gastric cancer risk: A nested case‐control study within the Korean Multicenter Cancer Cohort

        <P><B>Abstract</B></P><P>This study was conducted to evaluate the relevance of the soluble form of c‐Met protein, a truncated form of the c‐Met membrane receptor involved in the CagA pathway, as a potential biomarker for gastric cancer. Among 290 gastric cancer case‐control sets selected from the Korean Multicenter Cancer Cohort, the plasma concentrations of soluble c‐Met protein were measured with enzyme‐linked immunosorbent assays. Using analysis of variance and covariance models with age, sex, smoking, <I>Helicobacter pylori</I> infection, and CagA seropositivity, the mean concentrations of soluble c‐Met protein between cases and controls were compared. To evaluate the association between gastric cancer and a c‐Met protein level, odds ratios and 95% confidence intervals were estimated using conditional logistic regression models. Interactions between CagA‐related genes and the soluble c‐Met protein concentration were also investigated. The overall median plasma concentration of soluble c‐Met among cases was significantly lower than those of controls (1.390 <I>vs</I>. 1.610 ng/mL, <I>p</I> < 0.0001). Closer to the onset of gastric cancer, the soluble c‐Met protein level decreased linearly in a time‐dependent manner (<I>p</I> for trend = 0.0002). The combined effects between the CagA‐related genes and the soluble c‐Met protein concentration significantly intensified risks for gastric cancer. Restricted analyses including cases that had been diagnosed within 1 year after entering the cohort had a fair degree of ability (area under the receiver operating characteristic curve of 0.73–0.77) to discriminate gastric cancer cases from normal controls. Our findings demonstrate the potential of the soluble form of c‐Met protein as a novel biomarker for gastric cancer. The beneficial effects of a high soluble c‐Met concentration in human plasma are strongly supported.</P>

        Influence of Microstructure Evolution on Mechanical Properties, Wear Resistance and Corrosion Resistance of Ti(C,N)-Based Cermet Tools with Various WC Additions

        The efects of tungsten carbide addition on the microstructure, mechanical properties and corrosion resistance of Ti(C,N)-based cermets prepared by the sinter-HIP method are investigated through scanning electron microscope, X-ray difraction,electron back scattered difraction, mechanical test and electrochemical method. The results show that the appropriate WCaddition has an important infuence on promoting the formation of core-rim microstructure of cermet, increasing the wettability of ceramic phase and binder phase, and improving the mechanical properties. Ti(C,N)-based cermet with 5 wt% WCaddition has the highest hardness of 92.4 HRA and excellent wear resistance. However, excessive WC addition causes theappearance of slight white core phase containing plenty of W and Ti element in microstructure, which increases the fracturetoughness, and reduces the hardness and wear resistance of cermet. Additionally, the electrochemical result shows that theaddition of 5 wt% WC in cermet has the optimal corrosion resistance in 1 mol/L hydrochloric acid solution.

        Numerical simulation of particle concentration and size on the slurry erosion on the surface of V-shaped groove microstructure

        Sand concentration and particle size are the main factors that afect the erosion of solid surfaces under the impact of solid particles in liquid–solid two-phase fow. In this study, fve sand concentrations (1 wt%, 3 wt%, 5 wt%, 10 wt%, and 20 wt%) and fve particle sizes (20 μm, 75 μm, 125 μm, 300 μm, and 425 μm) were used for V-groove Surface mud erosion is simulated. Gambit and Ansys Fluent were used to simulate the anti-erosion performance of the V-shaped groove. The fow feld near the V-shape grooved surface was analyzed for the explanation of the erosion behavior. Corresponding experiments were also carried out to verify the simulation. The results showed that the slurry erosion of the V-shape grooved surface increased with the increasing sand concentration. There was a critical particle size between 25 μm and 75 μm, above which the slurry erosion remained stable with the particle size. In addition, no great change occurred in the fow style for all cases with diferent sand concentrations and particle sizes. The erosion area did not change with the increasing sand concentration but enlarged with the increasing particle size. The erosion mechanism with diferent sand concentrations and particle sizes was also discussed.

        Tailoring the Impact Toughness of Sintered NdFeB Magnets via Surface Coating

        Metallic coating by electroplating is commonly attractive for improving the corrosion resistance of sintered NdFeB magnets. However, its tailoring of mechanical characteristics for sintered NdFeB magnets has been seldom concerned. Herein, the impact toughnesses of sintered NdFeB magnets with various metallic coatings (Ni or Ni/Sn) were comparatively investigated. The results indicate that the impact toughnesses of sintered NdFeB magnets are both improved by Ni coating and Ni/Sn bilayer coating. And Ni/Sn bilayer coating exhibits more enhancement of the impact toughness, increased by 41.6% compared with the original magnet. Moreover, the microstructural observations of the metallic coatings and the fracture were conducted, and the enhanced mechanism of impact toughness for the magnet is analyzed. These findings may provide a reference for toughening the brittle materials.

      • Genome-wide association study in a Chinese population identifies a susceptibility locus for type 2 diabetes at 7q32 near <i>PAX4</i>

        <P><B>Aims/hypothesis</B></P><P>Most genetic variants identified for type 2 diabetes have been discovered in European populations. We performed genome-wide association studies (GWAS) in a Chinese population with the aim of identifying novel variants for type 2 diabetes in Asians.</P><P><B>Methods</B></P><P>We performed a meta-analysis of three GWAS comprising 684 patients with type 2 diabetes and 955 controls of Southern Han Chinese descent. We followed up the top signals in two independent Southern Han Chinese cohorts (totalling 10,383 cases and 6,974 controls), and performed in silico replication in multiple populations.</P><P><B>Results</B></P><P>We identified <I>CDKN2A/B</I> and four novel type 2 diabetes association signals with <I>p</I> < 1 × 10<SUP>−5</SUP> from the meta-analysis. Thirteen variants within these four loci were followed up in two independent Chinese cohorts, and rs10229583 at 7q32 was found to be associated with type 2 diabetes in a combined analysis of 11,067 cases and 7,929 controls (<I>p</I><SUB>meta</SUB> = 2.6 × 10<SUP>−8</SUP>; OR [95% CI] 1.18 [1.11, 1.25]). In silico replication revealed consistent associations across multiethnic groups, including five East Asian populations (<I>p</I><SUB>meta</SUB> = 2.3 × 10<SUP>−10</SUP>) and a population of European descent (<I>p</I> = 8.6 × 10<SUP>−3</SUP>). The rs10229583 risk variant was associated with elevated fasting plasma glucose, impaired beta cell function in controls, and an earlier age at diagnosis for the cases. The novel variant lies within an islet-selective cluster of open regulatory elements. There was significant heterogeneity of effect between Han Chinese and individuals of European descent, Malaysians and Indians.</P><P><B>Conclusions/interpretation</B></P><P>Our study identifies rs10229583 near <I>PAX4</I> as a novel locus for type 2 diabetes in Chinese and other populations and provides new insights into the pathogenesis of type 2 diabetes.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (doi:10.1007/s00125-013-2874-4) contains peer-reviewed but unedited supplementary material, which is available to authorised users.</P>

        Measurement of the Secondary Neutron Emission Differential and Double-Differential Cross Sections between 20 and 30 MeV

        More and more interests have been paid to the differential and double-differential neutron emission cross sections (DX and DDX) at above 20 MeV region in recent years. At the China Institute of Atomic Energy (CIAE), a program to measure the DX and DDX between 20 and 30 MeV neutron energy region has been carried out. The d-T reaction neutron source was used to produce the 20 - 30 MeV neutrons with a tritium gas target at the HI-13 tandem accelerator at CIAE. The time-of-flight(TOF) technique was used to measure the neutron energy from about 1 MeV to 30 MeV with 3 BC501A liquid scintillators (with a size of Φ180 ×100 mm). A preliminary measurement has been carried out for ^9Be at 22 MeV. The measured data were analyzed by detailed Monte-Carlo simulation to perform the multiple scattering, flux attenuation and finite geometry corrections. The experimental details, the data analysis, the progress, the results and the future plan will be presented in this report.

        Effects of exogenous phytase and xylanase, individually or in combination, and pelleting on nutrient digestibility, available energy content of wheat and performance of growing pigs fed wheat-based diets

        Objective: Two experiments were conducted to determine the effects of adding exogenous phytase and xylanase, individually or in combination, as well as pelleting on nutrient digestibility, available energy content of wheat and the performance of growing pigs fed wheat-based diets. Methods: In Experiment 1, forty-eight barrows with an initial body weight of $35.9{\pm}0.6kg$ were randomly assigned to a $2{\times}4$ factorial experiment with the main effects being feed form (pellet vs meal) and enzyme supplementation (none, 10,000 U/kg phytase, 4,000 U/kg xylanase or 10,000 U/kg phytase plus 4,000 U/kg xylanase). The basal diet contained 97.8% wheat. Pigs were placed in metabolic cages for a 7-d adaptation period followed by a 5-d total collection of feces and urine. Nutrient digestibility and available energy content were determined. Experiment 2 was conducted to evaluate the effects of pelleting and enzymes on performance of wheat for growing pigs. In this experiment, 180 growing pigs ($35.2{\pm}9.0kg\;BW$) were allocated to 1 of 6 treatments according to a $2{\times}3$ factorial treatment arrangement with the main effects being feed form (meal vs pellet) and enzyme supplementation (0, 2,500 or 5,000 U/kg xylanase). Results: In Experiment 1, there were no interactions between feed form and enzyme supplementation. Pelleting reduced the digestibility of acid detergent fiber (ADF) by 6.4 percentage units (p<0.01), increased the digestibility of energy by 0.6 percentage units (p<0.05), and tended to improve the digestibility of crude protein by 0.5 percentage units (p = 0.07) compared with diets in mash form. The addition of phytase improved the digestibility of phosphorus (p<0.01) and calcium (p<0.01) by 6.9 and 7.6 percentage units respectively compared with control group. Adding xylanase tended to increase the digestibility of crude protein by 1.0 percentage units (p = 0.09) and increased the digestibility of neutral detergent fiber (NDF) (p<0.01) compared with control group. Supplementation of the xylanase-phytase combination improved the digestibility of phosphorus (p<0.01) but impaired NDF digestibility (p<0.05) compared with adding xylanase alone. In Experiment 2, adding xylanase increased average daily gain (p<0.01) and linearly improved the feed:gain ratio (p<0.01) compared with control group. Conclusion: Pelleting improved energy digestibility but decreased ADF digestibility. Adding xylanase increased crude protein digestibility and pig performance. Phytase increased the apparent total tract digestibility of phosphorus and calcium. The combination of phytase-xylanase supplementation impaired the effects of xylanase on NDF digestibility.

        Genetic Differentiation of Chinese Indigenous Meat Goats Ascertained Using Microsatellite Information

        To investigate the genetic diversity of seven Chinese indigenous meat goat breeds (Tibet goat, Guizhou white goat, Shannan white goat, Yichang white goat, Matou goat, Changjiangsanjiaozhou white goat and Anhui white goat), explain their genetic relationship and assess their integrity and degree of admixture, 302 individuals from these breeds and 42 Boer goats introduced from Africa as reference samples were genotyped for 11 microsatellite markers. Results indicated that the genetic diversity of Chinese indigenous meat goats was rich. The mean heterozygosity and the mean allelic richness (AR) for the 8 goat breeds varied from 0.697 to 0.738 and 6.21 to 7.35, respectively. Structure analysis showed that Tibet goat breed was genetically distinct and was the first to separate and the other Chinese goats were then divided into two sub-clusters: Shannan white goat and Yichang white goat in one cluster; and Guizhou white goat, Matou goat, Changjiangsanjiaozhou white goat and Anhui white goat in the other cluster. This grouping pattern was further supported by clustering analysis and Principal component analysis. These results may provide a scientific basis for the characteristization, conservation and utilization of Chinese meat goats.
