      • Reference to Korean Government Procurement System in China

        Korea is one of the pioneer Asian Countries which have adopted Government Procurement Policy. Accordingly, the management system and operation mechanism of Government Procurement in Korea are efficient and the procurement by law has been functioned. In addition, the Korea has completed network system and adopted modern management instrument, which has contributed significantly to improve the transparency, standardization, competitiveness and efficiency of government procurement. At present, China is at the primary stage in establishment of government procurement policy, and numerous defects exist. Therefore, it is helpful for China to use government procurement policy of Korea for reference.

        Dynamic analysis of high-speed railway train-bridge system after barge collision

        In this paper, a framework is proposed for dynamic analysis of train-bridge systems with a damaged pier after barge collision. In simulating the barge-pier collision, the concrete pier is considered to be nonlinear-inelastic, and the barge-bow is modeled as elastic-plastic. The changes of dynamic properties and deformation of the damaged pier, and the additional unevenness of the track induced by the change of deck profile, are analyzed. The dynamic analysis model for train-bridge coupling system with a damaged pier is established. Based on the framework, an illustrative case study is carried out with a $5{\times}32m$ simply-supported PC box-girder bridge and the ICE3 high-speed train, to investigate the dynamic response of the bridge with a damaged pier after barge collision and its influence on the running safety of high-speed train. The results show that after collision by the barge, the vibration properties of the pier and the deck profile of bridge are changed, forming an additional unevenness of the track, by which the dynamic responses of the bridge and the car-body accelerations of the train are increased, and the running safety of high-speed train is affected.

      • Differential Roles of Glycogen Synthase Kinase 3 Subtypes Alpha and Beta in Cortical Development

        <P>Glycogen synthase kinases 3 (GSK3) α and β are expressed in the nervous system, and disruption of GSK3 signaling has been implicated in a wide range of neurodevelopmental and psychiatric disorders. Although several studies have established a role of GSK3 signaling in the nervous system, much less is known about isoform-specific functions. Here, we have examined the role of GSK3α and GSK3β in the developing neocortex by performing <I>in utero</I> electroporation with specific small interfering RNAs targeting each isoform. We found that depletion of either GSK3α or GSK3β commonly promoted the proliferation of neural progenitor cells in the ventricular zone, but at later stages, knocking down of each isoform resulted in distinct outcomes. In particular, the transformation of radial progenitors to intermediate progenitor cells was promoted in GSK3α-depleted cells, but markedly prevented in GSK3β-depleted cells. Moreover, knocking down of GSK3β but not GSK3α prevented the generation of upper-layer Cux1<SUP>+</SUP> neurons. Consistent with the distinct outcomes, protein levels of c-Myc and β-catenin, well-known substrates of GSK3, were differentially affected by depletion of GSK3α and GSK3β. Together, these results suggest that GSK3α and GSK3β might play distinct roles in the genesis and differentiation of neuronal lineage cells during neocortex development by differential regulation of downstream signaling pathways.</P>

      • CREATING A "THIRD SPACE" : MAGICAL REALISM IN TONIMORRISON'S EARLY NOVELS

        Toni Morrison’s portrayal for African American’s living conditions and her elaborate arrangements are frequently discussed in African American literature in a way that reconstruct postcolonialist concerns from the persp ective of history in the United States. The postcolonial theorist Homi Bha bha points out a “third space” through which the regeneration of meaning needs cultural positionality and cultural reference to be mobilized in the passageway. This paper, from a new perspective of a “third space,” explor es Morrison’s magical realism as a possibility to investigate the traumatic real of the institutionalized slavery and ideologized racism in a white dom inated society. In Morrison’s early novels, those issues are tactically discu ssed through her magical realism which creates a “third Space.” This pape r, through examining magical realism from the perspective of a “third Spa ce,” attempts to unravel Morrison’s hybridity and complexity, and explore how she visualizes the shadow and speaks the unspeakable. Morrison’s na rratives seem to be magical while they are actually real. In her early novel s, she repeatedly utilizes the magical myth, folklore, and fragmentation to generate the authentic real related to the slavery history as well as the cur rent white dominated society.

        Influence of compressor’s tilt angle on the performance of miniature refrigeration system

        Vapor compression refrigeration is considered as ideal miniature cooling system for miniature electronic systems and portable cooling systems. Many applications of the miniature refrigeration system are working under the situations (such as aviation field, individual combat system and automobile air conditioning, etc.) that exist bump, tilt, and even flip, which will severely degrade the performance of the miniature refrigeration system. In order to study the influence of compressor’s tilt angle on refrigeration system, a test bench was built and the performance experiments were made. The results show that the compressor’s tilt angle has a great impact on the refrigeration system performance. When the compressor worked at the maximum tilt angle of 60°, COP-c and COP-h of the system change periodically. When the compressor works in full load, the maximum tilt angle for compressor is 20°. When the compressor tilt angle is 15°, there is an optimal amount of refrigerant charge, and corresponding COP-c and COP-h reach the maximum of 2.9 and 3.9, respectively.

        A framework for carrying out train safety evaluation and vibration analysis of a trussed-arch bridge subjected to vessel collision

        Safety is the prime concern for a high-speed railway bridge, especially when it is subjected to a collision. In this paper, an analysis framework for the dynamic responses of train-bridge systems under collision load is established. A multi-body dynamics model is employed to represent the moving vehicle, the modal decomposition method is adopted to describe the bridge structure, and the time history of a collision load is used as the external load on the train-bridge system. A (180+216+180) m continuous steel trussedarch bridge is considered as an illustrative case study. With the vessel collision acting on the pier, the displacements and accelerations at the pier-top and the mid-span of the bridge are calculated when a CRH2 high-speed train running through the bridge, and the influence of bridge vibration on the running safety indices of the train, including derailment factors, offload factors and lateral wheel/rail forces, are analyzed. The results demonstrate that under the vessel collision load, the dynamic responses of the bridge are greatly enlarged, threatening the running safety of high-speed train on the bridge, which is affected by both the collision intensity and the train speed.

        CDH17 nanobodies facilitate rapid imaging of gastric cancer and efficient delivery of immunotoxin

        Background: It is highly desirable to develop new therapeutic strategies for gastric cancer given the low survival rate despite improvement in the past decades. Cadherin 17 (CDH17) is a membrane protein highly expressed in cancers of digestive system. Nanobody represents a novel antibody format for cancer targeted imaging and drug delivery. Nanobody targeting CHD17 as an imaging probe and a delivery vehicle of toxin remains to be explored for its theragnostic potential in gastric cancer. Methods: Naïve nanobody phage library was screened against CDH17 Domain 1-3 and identified nanobodies were extensively characterized with various assays. Nanobodies labeled with imaging probe were tested in vitro and in vivo for gastric cancer detection. A CDH17 Nanobody fused with toxin PE38 was evaluated for gastric cancer inhibition in vitro and in vivo. Results: Two nanobodies (A1 and E8) against human CDH17 with high affinity and high specificity were successfully obtained. These nanobodies could specifically bind to CDH17 protein and CDH17-positive gastric cancer cells. E8 nanobody as a lead was extensively determined for tumor imaging and drug delivery. It could efficiently co-localize with CDH17-positive gastric cancer cells in zebrafish embryos and rapidly visualize the tumor mass in mice within 3 h when conjugated with imaging dyes. E8 nanobody fused with toxin PE38 showed excellent anti-tumor effect and remarkably improved the mice survival in cell-derived (CDX) and patient-derived xenograft (PDX) models. The immunotoxin also enhanced the anti-tumor effect of clinical drug 5-Fluorouracil. Conclusions: The study presents a novel imaging and drug delivery strategy by targeting CDH17. CDH17 nanobodybased immunotoxin is potentially a promising therapeutic modality for clinical translation against gastric cancer.

        The effects of codon usage on the formation of secondary structures of nucleocapsid protein of peste des petits ruminants virus

        The nucleocapsid (N) protein of peste des petits ruminants virus (PPRV) with a conserved amino acid usage pattern plays an important role in viral replication. The primary objective of this study was to estimate roles of synonymous codon usages of PPRV N gene and tRNA abundances of host in the formation of secondary structure of N protein. The potential effects of synonymous codon usages of N gene and tRNA abundances of host on shaping different folding units (α-helix, β-strand and the coil) in N protein were estimated, based on the information about the modeling secondary structure of PPRV N protein. The synonymous codon usage bias was found in different folding units in PPRV N protein. To better understand the role of translation speed caused by variant tRNA abundances in shaping the specific folding unit in N protein, we modeled the changing trends of tRNA abundance at the transition boundaries from one folding unit to another folding unit (β-strand → coil, coil → β-strand, α-helix → coil, coil → α-helix). The obvious fluctuations of tRNA abundance were identified at the two transition boundaries (β-strand → coil and coil → β-strand) in PPRV N protein. Our findings suggested that viral synonymous codon usage bias and cellular tRNA abundance variation might have potential effects on the formation of secondary structure of PPRV N protein.
