지반의 진행성 파괴와 기초의 크기 효과를 고려한 지지력 특성에 관한 실험적 연구

얕은 기초의 지지력을 산정하는데 있어서 기존 이론의 지지력계수(Nc,Nr,Nq)는 흙의 내부마찰 각 φ만의 함수로 표현되어져 있다. 그리고 조밀한 모래지반에서는 동시파괴(전반전단파괴)가 발생한다고 가정하고 있다. 본 논문에서는 주문지 표준사를 이용하여 중간·조밀한 모래지반을 제작하여, 재하폭 B를 변화시켜 실험하였다. 실험 결과, 조밀한 모래지반에서도 진행성파괴가 발생하며, 지지력계수 Nr의 평가에 있어서 내부마찰각 φ뿐만 아니라, 재하폭 B의 영향이 고려되어져야 함을 알 수 있었다. The bearing capacity factors(Nc,Nr,Nq) of an existing formulae are the function of only internal friction angle(φ) in calculating of ultimate bearing capacity of shallow foundations. And it is assumed that the simultaneous failure(general shear failure) occurs in the dense sand. In this study, the medium to dense sand grounds are formed by Joomunjin standard sand, and the laboratory medel tests are made with the change of load-width(B). By test it is known that the progressive failure is also occurred in the dense sand ground and so the bearing capacity factor(Nr) should be considered for not only internal friction angle(φ) but also the effect of load-width(B).

Procurement, cutting, cleansing, freezing, freeze-drying, and demineralization of the allogeneic bone must be made under the germ-free stable condition without bacterial and/or viral contamination. Even thought the bone is procured under the germ free condition, we must have confidence on disinfection of all the solutions that come in contact with tissue during the whole procedure. Lots of antibacterial agents have been introduced for chemical sterilization. Recently ethylene oxide gas sterilization or radiation sterilization is frequently selected as a secendary sterilization procedure. The biological and biochemical response of the graft material differs with the type and concentration of the sterilizing agents, and various toxic reactions have been reported due to the graft material itself and the substance released by the chemicals. The authors conducted the Millipore filter test to observe the toxic effect on L929 fibroblasts according to the effect on activity of succinate dehydrogenase, during the secondary sterilization of the demineralized allogeneic bone powder with irradiation or ethylene oxide gas. The result were as follows: 1. Around the copper disk, positive control group, 10mm diameter discoloration was observed. 2. As same as the negative control group, the disk showed no discoloration. 3. The demineralized allogeneic bone which was sterilized with ethylene oxide gas or irradiation showed no cytotoxicity. 4. From this results, it is suggested that treatment with ethylene oxide gas or irradiation should be effective to sterilize the deminineralized allogeneic bone.
고추 1986年度 수集在來種의 裁培的 特性과 1987年度 蒐集在來種의 種果特性

Ten Korean local cultivars of pepper (Capsicum annuum) collected in 1986 were planted in the Kyungpook National University Experiment Farm to investigate growth characteristics. 'Punggakcho' was late in flowering, bearing large fruits, and high yielding. 'Kalmicho' was vigorous, good in fruit setting, and slightly resistant to bacterial spot but badly infected with powdery mildew. These two cultivars were the most promising materials for pepper breeding. Local cultivars collected in Younyang in Kyungpook such as 'Chilsungcho', 'Subicho', and 'Daehwacho' were poor in growth and suffered from viral complexes. As results, size and form of the original seed fruits was not reproduced but small and malformed fruits were produced. Collection of local cultivars of pepper was continued in 1987. Fruits of 14 local cultivars were collected in Youngwol in Kangwondo, Bonghwa and Munkyung in Kyungpook, Guchang in Kyungnam, and Jejudo, and their fruit characters were recorded.

The purpose of this study is to develop models and instruments for teaching and assessing scientific experiments and practices. General objectives for teaching and evaluating scientific experiments and practices in secondary schools were analyzed and categorized into three domains:they were inquiry skill, scientific thinking skill, and scientific attitude. The sub-categories for each domain were constructed, and specific objectives for each general objective were also stated in terms of behavioral objectives according to Mager's suggestion. The tables of specification (content×behavior) were developed in order to assist secondary school teachers to teach and evaluate scientific experiments in class. In addition, we introduced some useful assessment techniques which can be adopted to evaluating learning outcomes of each specific objective. The sample test items corresponding to each objective were listed for practical usage in class.

Heme oxygenase-1 (HO-1) degrades heme to carbon dioxide, biliverdin, and Fe2+, which play important roles in various biochemical processes. In this study, we examined the protective function of HO-1 against oxidative stress in SH-SY5Y cells and in a Parkinson`s disease mouse model. Western blot and fluorescence microscopy analysis demonstrated that PEP-1-HO-1, fused with a PEP-1 peptide can cross the cellular membranes of human neuroblastoma SH-SY5Y cells. In addition, the transduced PEP-1-HO-1 inhibited generation of reactive oxygen species (ROS) and cell death caused by 1-methyl-4-phenylpyridinium ion (MPP+). In contrast, HO-1, which has no ability to transduce into SH-SY5Y cells, failed to reduce MPP+-induced cellular toxicity and ROS production. Furthermore, intraperitoneal injected PEP-1-HO-1 crossed the blood-brain barrier in mouse brains. In a PD mouse model, PEP-1-HO-1 significantly protected against 1-methyl-4-phenyl- 1,2,3,6-tetrahydropyridine (MPTP)-induced toxicity and dopaminergic neuronal death. Therefore, PEP-1-HO-1 could be a useful agent in treating oxidative stress induced ailments including PD.

Background: Panax ginseng (i.e., ginseng) root is extensively used in traditional oriental medicine. It is a modern pharmaceutical reagent for preventing various human diseases such as cancer. Ginsenosidesdthe major active components of ginsengdexhibit immunomodulatory effects. However, the mechanism and function underlying such effects are not fully elucidated, especially in human monocytes and dendritic cells (DCs). Methods: We investigated the immunomodulatory effect of ginsenosides from Panax ginseng root on CD14⁺ monocytes purified from human adult peripheral blood mononuclear cells (PBMCs) and on their differentiation into DCs that affect CD4⁺ T cell activity. Results: After treatment with ginsenoside fractions, monocyte levels of tumor necrosis factor (TNF)-a, interleukin (IL)-6, and IL-10 increased through phosphorylation of extracellular signal-regulated kinase (ERK)1/2 and c-Jun N-terminal kinase (JNK), but not p38 mitogen-activated protein kinase (MAPK). After treatment with ginsenoside fractions, TNF-α production and phosphorylation of ERK1/2 and JNK decreased in lipopolysaccharide (LPS)-sensitized monocytes.We confirmed that DCs derived from CD14⁺ monocytes in the presence of ginsenoside fractions (Gin-DCs) contained decreased levels of the costimulatory molecules CD80 and CD86. The expression of these costimulatory molecules decreased in LPS-treated DCs exposed to ginsenoside fractions, compared to their expression in LPS-treated DCs in the absence of ginsenoside fractions. Furthermore, LPS-treated Gin-DCs could not induce proliferation and interferon gamma (IFN-γ) production by CD4⁺ T cells with the coculture of Gin-DCs with CD4⁺ T cells. Conclusion: These results suggest that ginsenoside fractions from the ginseng root suppress cytokine production and maturation of LPS-treated DCs and downregulate CD4⁺ T cells.

In order to produce monoclonal antibody to bovine IgG_1, attempts were made to isolate and purify the bovine IgG_1 from colostrum, and the prepared IgG_1 heavy chain was used as an antigen for the immunization of BALB/c mice. Spleen cells from BALB/c mice immunized with bovine IgG_1 heavy chain were successfully fused with SP2/O mouse myeloma cells. The antibody produced by the hybrid cells was specified by enzyme-linked immunosorbent assay. Of 48 initial hybridoma clones, 21 produced antibodies to bovine IgG_1. Through cloning by limiting dilutions, 2(G1D2 and G1H12) of these hybrid clones retained specificity and were grown in mass culture. The monoclonal(anti-bovine IgG_1) antibody G1D2 and G1H12 were reacted to bovine IgG_1 specifically in their dilutions of more 10^2- and 10-folds, respectively.
