        Posterior silk glands of Bombyx mori were cultured for the in vitro production of silk fibroin. Four-day-old fifth-instar larvae and Grace's media with or without silkworm hemolymph were used for experiments. With the addition of silkworm hemolymph, significant increase in the secretion of fibroin from the lumen to the outside of the gland was observed and total amount of fibroin increased by 14%. Most of the fibroin produced in the media was insoluble protein. Total amount of fibroin reached a plateau in 24 day culture without the addition of silkworm hemolymph, whereas it increased continuously in the medium supplemented with silkworm hemolymph.

        Hollow fiber membrane devices have been used as enzyme reactors because of their large surface area-volume ratio and the advantage of immobilized soluble enzymes in intact form. Theoretical analysis of the hollow fiber enzyme reactor with recirculated enzyme solution was performed and the model equations were numerically solved in previous paper. For a slow kinetics one might assume that the flowing substrate solution has radially uniform concentration profile. In this paper, the equations were solved analytically with the above assumption in the case of 1st and zero order reaction, and the approximate analytical solutions were compared with the exact numerical solutions. Exact numerical solutions of shell side concentration and of the outlet concentration at low Pe'clet number were in good agreement with the approximate analytical solutions. And the two solutions of outlet concentration at high Pe'clet number were agreed, when the Thiele modulus was small.

      • 발효시 세포 재순환의 새로운 응용

        Cell recycle has been used in fermentation and most of the previous works were concerned with increasing the cell concintration. Other utilities of cell recycle chemostat are considered in this article. Steady state should be changed from one to another in the experiment for the evaluation of kinetic parameters. In this case, the recylce system reduces the transient period between two steady states. The effect of cell recycle on the trasient period in single-stage continuous operation is theoretically analyzed. Dilution rate or another parameter, r, can be varied to get other steady states. The result shows that change of the dilution rate rather than r gives shorter transient period. For the same specific growth rate, combination of higer dilution rate and lower r gives shorter transient period.

        저열량영양식품 섭취에 따른 비만여성의 체중감량 효과에 관한 연구

        In the method of obesity treatment, Dietary method, Exercise method, Behavior method, and Drug method, Dietary method is the most important control method of obesity. The aim of this study is that evaluate the weight reduction effect of the specific low calorie diet products (KSD I and 2) in obese women. Twenty five women who were volunteer participated in this study. They have no heart diseases and metabolic diseases. we classified two groups : obese group and standard group. Anthropometric indices were measured in all subject for four times. Venous blood sample were taken to determine the lipid profiles, glucose level, and liver function test. In obese group, body weight were reduced 2.85±1.46kg, but showed statistically non-signicicant. Body mass index were gradually reduced, 1.09±0.93kg/m^2(p<0.05).In blood analyses, total cholesterol were reduced 10.61 ±33.4mg/dℓ ( p<0.05). KSD 2 significantly inhibited maltase, sucrase and α-glucosidase from rat intestine, and porcine pancreatic lipase, although KSD I inhibited only the sucrase.

        Uncoupling of the cell growth from the production of an inducible substance gives higher performance, and two-stage continuous operation allows one to separate the cell growth from the product induction. Residence time of each individual cell in the second tank is important since the extent of the induction is dependent on the residence time of the cell in the second tank and keeps the cell to stay long enough for the product induction. The effect of cell recycle on the residence time in two-stage continuous operation has been theoretically analyzed. The cell recycle system increases the mean intracellular concentration by the increment of the mean residence time of the cell. Higher recycle ratio and lower separation parameter give longer mean residence time. Optimum dilution rate exists for the maximum productivity.

      • 도시 하수오니에 의한 고분자 물질의 생분해도 측정

        Factors influencing the biodegradability of polymer materials by municipal sewage sludge were studied using starch, polyethylene glycol(PEG) and polyvinyl alcohol(PVA). The degree of biodegradability was determined by measuring evolved carbon dioxide as a function of time, when the polymer material is exposed to activated sewage sludge. The biodegradability of starch reached around 70% and 40% after 30day incubation in summer and autumn, respectively. This is caused by the activity of microorganism and incubation temperature. The biodegradability of PEG 8000 and PVA was 1/7 and 1/14 of starch, respectively. Biodegradation of PEG 8000 after 30 day incubation in summer was slightly higher than that in spring while lag phase was significantly longer in spring. This shows that the activity of microorganism as well as incubation temperature is important for biodegradation.

      • 재순환 실관 효소 반응기의 이론적 해석(Ⅰ)

        Hollow fiber enzyme reactor has evolved as a spin-off of the rapidly developing membrane technologies. Hollow fiber module is now favorably considered as enzyme reactor, artificial kidney, ultrafiltration device, reverse osmosis device, and gas separator due to its high area-to-volume compared with other membrane modules such as flat or spiral modules. However, when an enzyme is immobilized in a sponge annulus, an ultrafiltration causes washout of the enzyme from the sponge annulus to result in a nonuniform distribution along the fiber. Thus, utilization of the retained enzyme is not adequate. However, this problem can be solved by recirculating the enzyme solution in the shell side, and also mass transfer resistance can be reduced by introducing the ultrafiltration. In this paper, the continuous system with recirculated enzyme solution was modeled. The numerical results were in good agreements with those of experiment. When Thiele modulus of the porous sponge region is less than 1, reaction occurs also in the shell side.

      • 人蔘 Total Saponin, Protopanaxadiol 및 Protopanaxatriol의 消炎作用에 關한 硏究

        Abstract-In an attempt to elucidate the anti-inflammatory action of ginseng total saponin, protopanaxadiol and protopanaxatriol, the anti-inflammatory activity of three compounds was investigated under various acute and chronic inflammatory models. The blood vessel permeability was slightly inhibited by total saponin and protopanaxatriol treatments. Granuloma formation induced by 2% carrageenan was inhibited by total saponin and protopanaxatriol. The cotton-pellet granuloma formation was significantly inhibited by intraperitoneal injection of total saponin. Total saponin and protopanaxadiol inhibited leukocyte emigration and protein exudation in CMC-induced pouch but protopanaxatriol increased leukocyte emigration. The swelling of rat hind paw induced by 1% carrageenan was significantly inhibited by total saponin, protopanaxadiol and protopanaxatriol both single and 2 weeks treatments. Total saponin, protopanaxadiol and protopanaxatriol decreased the anti-inflammatory activity in adrenalectomized rat. Our results suggest that total saponin, protopanaxadiol and protopanaxatriol have potent anti-inflammatory activity, this may be mediated in part through stimulation of adrenal glands.

      • 烏梅(Mume Fructus)水性엑스의 家兎 腸運動에 미치는 影響

        Effect of .Mume Fructus on intestinal function was investigated in rabbit, making use of its water extract. Mume Fructus water extract(MWE), of small doses(0.1, 0.3㎎/ml) produced contractile action but did not effect the spontaneous movement strips of high dose(1.0㎎/ml) elicited relaxative action and the inhibition of spontaneous movement in ileum strips. In lieum strips, MWE did not influence the contractile action but augmented the spontaneous movement to acetylcholine, MWE elicited the inhibition of contractile action and the agumentation of spontaneous movement to pilocarpine, MWE inhibited the both actions to histamine, did not affect to serotonin and blocked to dopamine. MWE exhibited the relaxation and the inhibition of spontaneous movement in longitadinal muscle of colon. In longitudinal muscle of colon, MWE inhibited both actions to acetylcholine, did not influence the contractile actions but augmented the spontaneous movement to histamine and serotonin. MWE elicited the increase of contractile actions and the decrease of spontaneous movement in circular muscle of colon. In circular muscle of colon, MWE augmented both actions to acetylcholine, inhibited both actions to serotonin, and then did not affect the contractile action but augmented the spontaneous movement to histamine.
